1脂肪干细胞分离、纯化和保存:研究进展与未来方向显示文摘背景:2001年,Zuk等从吸脂术抽出的脂肪中发现了脂肪干细胞,揭开了干细胞研究的新篇章。近年来的研究证实,干细胞广泛存在于体内各组织,但脂肪干细胞以其来源广泛、获取简单等优点,一直以来是整形修复科、组织工程和再生医学等相关学科的研究重点。目的:从脂肪组织的取材、脂肪干细胞的分离、纯化、传代培养和冻存等方面进行综述,讨论上述过程中影响脂肪干细胞产量、增殖和分化能力的主要因素,同时提出目前的问题和未来的研究方向。方法:于2015年9月10日在PubM ed中以(adipose stem cells[Title])OR(adipose-derived stem cells[Title])OR(adipose-derived mesenchymal stem cells[Title])作为检索式进行检索,在SinoM ed中以('脂肪干细胞'[中文标题:智能])or('脂肪间充质干细胞'[中文标题:智能])作为检索式进行检索。通过标题及摘要判断其主要内容,最终选取81篇代表性文献。文章还介绍了美国德克萨斯大学MD安德森肿瘤中心整形科和TRAMCEL实验室(Tissue Regeneration and Molecular Cell Engineering Lab)的相关经验。结果与结论:脂肪组织分布广泛,给脂肪干细胞提供了充足的来源。利用吸脂术获得脂肪组织,患者痛苦小,供区损伤低。胶原酶消化法操作简单,产量高,仍然是基础实验中脂肪干细胞的主要分离方法,但是临床应用中一般不分离脂肪干细胞,或者利用临床级的无异种胶原酶甚至无酶的分离方法。脂肪干细胞的产量、表型、增殖和分化能力等生物学特性会受到取材和分离过程中多种因素的影响,因此目前需要制定标准化的脂肪干细胞分离准则。陈犹白 陈聪慧 Qixu Zhang 韩岩 2016中国组织工程研究2016,20,10:34
2脂肪干细胞成骨分化的研究进展显示文摘创伤或肿瘤导致的骨缺损是骨科、整形修复科、口腔颌面外科等科室的常见疾病,现有的治疗手段均有一定局限性。脂肪干细胞(ASC)是来自脂肪组织的多能干细胞,基于ASC的干细胞疗法和骨组织工程,为骨缺损的修复和再生提供了新的思路。ASC成骨分化是多种基因、蛋白和信号通路相互作用的复杂过程,其中Runx2和Osterix、Wnt、骨形成蛋白、Notch、成纤维细胞生长因子、环磷腺苷/蛋白激酶A、Hedgehog、丝裂原活化蛋白激酶等均扮演了重要角色。体外化学诱导ASC的成骨分化已经非常成熟,利用细胞共培养和各种支架也可诱导ASC的成骨分化。验证ASC分化成骨的方法包括茜素红染色和相关基因与蛋白的检测。影响ASC成骨分化的因素包括供体种属、年龄、脂肪获取部位等供体因素,培养液糖浓度、ASC的代数、冻存等实验因素,血管内皮细胞生长因子、生长分化因子、胰岛素样生长因子、转化生长因子β、尼尔样1型分子、LIM矿化蛋白1、低氧诱导因子1、肿瘤坏死因子α、干扰素γ、IL-6等生长因子,糖皮质激素、雌激素、胰岛素、褪黑素、瘦素等激素,一氧化氮、组蛋白H1、白藜芦醇、曲古抑菌素A、小檗碱、硼替佐米、阿司匹林、辛伐他汀等化学因子及药物,电磁场和超声波等物理因素,拉伸力和剪切力等生物力学因素,钙、锌、锂、锶、硒等金属或非金属离子,微小RNA以及富血小板血清和富血小板纤维蛋白、降钙素基因相关肽、中药和咖啡因等其他因素。本文总结了ASC成骨分化的过程,分析了相关基因和信号通路,回顾了诱导和验证方法,讨论了主要影响因素及其机制,并展望了未来研究方向。陈犹白 陈聪慧 Qixu Zhang 韩岩 2016中华损伤与修复杂志(电子版)2016,11,2:19
3脂肪干细胞表型和标记物的研究进展显示文摘由于脂肪供体、提取方法、分离、培养、体内外微环境、鉴定时的细胞代数和检测方法等诸多因素的差异,迄今为止尚未发现脂肪干细胞(adipose-derived stem cell,ASCs)的特异性标记物。本文总结了所有ASCs的相关标记物,发现阳性标记物集中在基质细胞、间充质干细胞和成纤维细胞标记物上,而阴性标记物主要是造血系细胞标记物,ASCs的争议标记物主要以骨髓间充质干细胞(bone marrow stem cells,BMSCs)、血管内皮细胞和周细胞标记物为主,这也体现了关于ASCs体内来源的争议。体内、刚分离的和体外培养初期的ASCs表达CD34,但经长时间体外培养后,其表达逐渐降低最后完全消失。这证实了ASCs体内外的表型差异,因此不能通过检测体外长期培养后的ASCs来确定体内的ASCs表型。Stro-1等干性标记物在ASCs的表达对鉴别ASCs有重要意义,但能否作为ASCs的常规标记物用于鉴定ASCs,尚需要进一步的研究。基质血管成分(Stromal vascular fraction,SVF)和ASCs不同亚群的表型差异提示了ASCs的体内起源,即BMSCs、周细胞或成纤维细胞,但尚无定论。ASCs表达CD36、CD49d,不表达CD48f、CD104,BMSCs则相反,这是鉴别ASCs和BMSCs的表型依据。本文还介绍了美国德克萨斯大学MD安德森肿瘤中心整形科组织再生与分子细胞工程实验室(Tissue Regeneration and Molecular Cell Engineering Lab,TRAMCEL)在ASCs表型方面的研究经验,探讨了ASCs表型研究目前存在的问题和未来的发展方向。陈犹白 陈聪慧 Qixu Zhang 韩岩 2016中国美容医学2016,25,3:18

8A static technique for detecting input validation vulnerabilities in Android apps显示文摘Input validation vulnerabilities are common in Android apps, especially in inter-component communications. Malicious attacks can exploit this kind of vulnerability to bypass Android security mechanism and compromise the integrity, confidentiality and availability of Android devices. However, so far there is not a sound approach at the source code level for app developers aiming to detect input validation vulnerabilities in Android apps. In this paper, we propose a novel approach for detecting input validation flaws in Android apps and we implement a prototype named Easy IVD, which provides practical static analysis of Java source code.Easy IVD leverages backward program slicing to extract transaction and constraint slices from Java source code.Then Easy IVD validates these slices with predefined security rules to detect vulnerabilities in a known pattern.To detect vulnerabilities in an unknown pattern, Easy IVD extracts implicit security specifications as frequent patterns from the duplicated slices and verifies them. Then Easy IVD semi-automatically confirms the suspicious rule violations and reports the confirmed ones as vulnerabilities. We evaluate Easy IVD on four versions of original Android apps spanning from version 2.2 to 5.0. It detects 58 vulnerabilities including confused deputy attacks and denial of service attacks. 9Cloud Virtual Machine Lifecycle Security Framework Based on Trusted Computing显示文摘As a foundation component of cloud computing platforms, Virtual Machines (VMs) are confronted with numerous security threats. However, existing solutions tend to focus on solving threats in a specific state of the VM. In this paper, we propose a novel VM lifecycle security protection framework based on trusted computing to solve the security threats to VMs throughout their entire lifecycle. Specifically, a concept of the VM lifecycle is presented divided up by the different active conditions of the VM. Then, a trusted computing based security protecti on framework is developed, which can exte nd the trusted relati on ship from trusted platform module to the VM and protect the security and reliability of the VM throughout its lifecycle. The theoretical analysis shows that our proposed framework can provide comprehensive safety to VM in all of its states. 10The Gaojiacun Mafic-Ultramafic Complex(Sichuan,SW China):Neoproterozoic Magmatic Activity at the Western Margin of the Yangtze Craton显示文摘The Neoproterozoic Gaojiacun intrusive complex(GIC)is composed of one big intrusion(ca.9 × 7.5 km)and numerous satellite intrusions,emplaced in the low-grade schists of the Neoproterozoic Yanbian Group.The main intrusion is concentrically zoned,with peridotite bodies + leucogabbroids in its core(inner zone)and the sequence:olivine hornblende gabbronorite-porphyric pyroxene hornblende gabbronorite(transitional zone)-pyroxene hornblende gabbronorite + hornblende gabbro + diorite(outer zone),towards the margins.The satellite intrusions are composed either of a single rock type(peridotite,gabbro,diorite or granite)or of different rock types(peridotite + diorite + granite).Upper crustal contamination is evidenced by the occurrence of numerous metamorphic enclaves,migmatites and mixed rocks in the outer zone of the main intrusion and in the satellite intrusions.Hornblende(often oikocrystic)is present in all mafic and ultramafic rocks.Cu-Ni(-PGE)deposits occur in several satellite intrusions,hosted in peridotite.The geochemical and mineralogical features of GIC support the presence of a subduction zone at the western edge of the Yangtze Craton.A feeder-related genesis of the satellite intrusions is inferred.Marian MUNTEANU YAO Yong Allan H. 12CaMKIIα-driven,phosphatase-checked postsynaptic plasticity via phase separation显示文摘Ca^(2+)/calmodulin-dependent kinase Ila(CaMKIla)is essential for synaptic plasticity and learning by decoding synaptic Ca^(2+)oscillations.Despite decades of extensive research,new mechanisms underlying CaMKIIa's function in synapses are still being discovered.Here,we discover that Shank3 is a specific binding partner for autoinhibited CaMKIla.We demonstrate that Shank3 and GluN2B,via combined actions of Ca^(2+)and phosphatases,reciprocally bind to CaMKIla.Under basal condition,CaMKIla is recruited to the Shank3 subcompartment of postsynaptic density(PSD)via phase separation.Rise of Ca^(2+)concentration induces GluN2Bmediated recruitment of active CaMKIla and formation of the CaMKIIa/GluN2B/PSD-95 condensates,which are autonomously dispersed upon Ca^(2+)removal.Protein phosphatases control the Ca^(2+)-dependent shuttling of CaMKIla between the two PSD subcompartme nts and PSD condensate formation.Activation of CaMKIla further enlarges the PSD assembly and in duces structural LTP.Thus,Ca^(2+)-induced and phosphatase-checked shuttling of CaMKIla between distinct PSD nano-domains can regulate phase separation-mediated PSD assembly and synaptic plasticity.Qixu Caic Menglong Zeng Xiandeng Wu Haowei Wu Yumeng Zhan Ruijun Tian Mingjie Zhang 2021Cell Research2021,31,1:3
133D-printed models improve surgical planning for correction of severe postburn ankle contracture with an external fixator显示文摘Gradual distraction with an external fixator is a widely used treatment for severe postburn ankle contracture(SPAC).However,application of external fixators is complex,and conventional two-dimensional(2D)imaging-based surgical planning is not particularly helpful due to a lack of spatial geometry.The purpose of this study was to evaluate the surgical planning process for this procedure with patient-specific three-dimension-printed models(3DPMs).In this study,patients coming from two centers were divided into two cohorts(3DPM group vs.control group)depending on whether a 3DPM was used for preoperative surgical planning.Operation duration,improvement in metatarsal-tibial angle(MTA),range of motion(ROM),the American Orthopedic Foot and Ankle Society(AOFAS)scores,complications,and patient-reported satisfaction were compared between two groups.The 3DPM group had significantly shorter operation duration than the control group((2.0±0.3)h vs.(3.2±0.3)h,P<0.01).MTA,ROM,and AOFAS scores between the two groups showed no significant differences pre-operation,after the removal of the external fixator,or at follow-up.Plantigrade feet were achieved and gait was substantially improved in all patients at the final follow-up.Pin-tract infections occurred in two patients(one in each group)during distraction and were treated with wound care and oral antibiotics.Patients in the 3DPM group reported higher satisfaction than those in the control group,owing to better patient-surgeon communication.Surgical planning using patient-specific 3DPMs significantly reduced operation duration and increased patient satisfaction,while providing similar improvements in ankle movement and function compared to traditional surgical planning for the correction of SPAC with external fixators.Youbai CHEN Zehao NIU Weiqian JIANG Ran TAO Yonghong LEI Lingli GUO Kexue ZHANG Wensen XIA Baoqiang SONG Luyu HUANG Qixu ZHANG Yan HAN 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,10:3
14Tracking Your Browser with High-Performance Browser Fingerprint Recognition Model显示文摘As the cyber security has attracted great attention in recent years,and with all kinds of tools’(such as Network Agent,VPN and so on)help,traditional methods of tracking users like log analysis and cookie have been not that effective.Especially for some privacy sensitive users who changed their browser configuration frequently to hide themselves.The Browser Fingerprinting technology proposed by Electronic Frontier Foundation(EFF)gives a new approach of tracking users,and then our team designed an enhanced fingerprint dealing solution based on browser fingerprinting technology.Our enhanced solution plays well in recognizing the similar fingerprints,but it is not that efficient.Nowadays we improve the algorithm and propose a high-performance,efficient Browser Fingerprint Recognition Model.Our new model reforms the fingerprint items set by EFF and propose a Fingerprint Tracking Algorithm(FTA)to deal with collected data.It can associate users with some browser configuration changes in different periods of time quickly and precisely.Through testing with the experimental website built on the public network,we prove the high-performance and efficiency of our algorithm with a 20%time-consuming decrease than ever.Wei Jiang Xiaoxi Wang Xinfang Song Qixu Liu Xiaofeng Liu 2020China Communications2020,17,3:2
15Reduction behavior of cerium(Ⅲ) ions in NaCl-2CsCl melt显示文摘The cathodic process of cerium(Ⅲ) ions in NaCl-2CsCl melt was studied by cyclic voltammetry and square wave voltammetry with tungsten and gold electrodes at 873 K. The two electroanalytical methods yield similar results. The cathodic process of cerium(Ⅲ) ions consists of two reversible steps: Ce3+ + e-= Ce2+ and Ce2+ + 2e-= Ce. The half wave potentials of Ce3+/Ce2+ and Ce2+/Ce were determined as -2.525 V vs. Cl2/Cl- and -2.975 V vs. Cl2/Cl-, respectively. 16Water utilization of typical plant communities in desert steppe,China显示文摘Water is a limiting factor in the restoration and construction of desert steppe.Exploring plant water sources is necessary to understand soil-plant interactions and species coexistence;however,water sources of major plant communities within the desert steppe of Ningxia Hui Autonomous Region,China remain poorly understood.In this study,we analyzed the water uptake of plants in four typical communities:Agropyron mongolicum Keng.;Sophora alopecuroids Linn.;Stipa breviflora Griseb.,and Achnatherum splendens(Trin.)Nevski communities.Stable isotopesδD andδ^(18)O in the xylem of plant and soil water at different soil depths were analyzed.An IsoSource model was used to determine the soil depths from which plants obtained water.Results showed that A.mongolicum community obtained water predominantly from 0–20 and 40–80 cm depth,S.alopecuroids community from 0–20 cm depth,S.breviflora community from 0–40 cm depth,and A.splendens community from 0–20 and 80–140 cm depths.S.alopecuroides had a wider range of soil depths for water extraction,i.e.,utilizing different water sources depending on habitat,and the plasticity of its water uptake pattern determined its role in different communities.Water source of plants relayed heavily on the distribution of their roots.Competition for soil water exists between different plant life forms in the sierozem habitat(A.mongolicum,S.alopecuroids,and S.breviflora communities),and in the sandy soil habitat(A.splendens community).The use of soil water by A.splendens community is more spatially differentiated,and shrubs and herbs can coexist stably.Under the pattern of extended drought period in the future,sierozem habitat may be more favorable for the formation of a dominant monoculture community type of perennial fibrous plants.In aeolian sandy soil habitat,A.splendens had a strong competitive advantage,and the growth of shallow-rooted plants was easily suppressed.CHEN Juan WANG Xing SONG Naiping WANG Qixue WU Xudong 2022Journal of Arid Land2022,14,9:1
