1High-quality Arabidopsis thaliana Genome Assembly with Nanopore and HiFi Long Reads显示文摘Arabidopsis thaliana is an important and long-established model species for plant molecular biology,genetics,epigenetics,and genomics.However,the latest version of reference genome still contains a significant number of missing segments.Here,we reported a high-quality and almost complete Col-0 genome assembly with two gaps(named Col-XJTU)by combining the Oxford Nanopore Technologies ultra-long reads,Pacific Biosciences high-fidelity long reads,and Hi-C data.The total genome assembly size is 133,725,193 bp,introducing 14.6 Mb of novel sequences compared to the TAIR10.1 reference genome.All five chromosomes of the Col-XJTU assembly are highly accurate with consensus quality(QV)scores>60(ranging from 62 to 68),which are higher than those of the TAIR10.1 reference(ranging from 45 to 52).We completely resolved chromosome(Chr)3 and Chr5 in a telomere-to-telomere manner.Chr4 was completely resolved except the nucleolar organizing regions,which comprise long repetitive DNA fragments.The Chrl centromere(CEN1),reportedly around 9 Mb in length,is particularly challenging to assemble due to the presence of tens of thousands of CEN180 satellite repeats.Using the cutting-edge sequencing data and novel computational approaches,we assembled a 3.8-Mb-long CEN1 and a 3.5-Mb-long CEN2.We also investigated the structure and epigenetics of centromeres.Four clusters of CEN180 monomers were detected,and the centromere-specific histone H3-like protein(CENH3)exhibited a strong preference for CEN180 Cluster 3.Moreover,we observed hypomethylation patterns in CENH3-enriched regions.We believe that this high-quality genome assembly,Col-XJTU,would serve as a valuable reference to better understand the global pattern of centromeric polymorphisms,as well as the genetic and epigenetic features in plants.Bo Wang Xiaofei Yang Yanyan Jia Yu Xu Peng Jia Ningxin Dang Songbo Wang Tun Xu Xixi Zhao Shenghan Gao Quanbin Dong Kai Ye 2022Genomics, Proteomics & Bioinformatics2022,20,1:6

3Suppression of Vortex-Induced Vibration by Fairings on Marine Risers显示文摘Vortex-induced vibration is quite common during the operation of offshore risers or umbilical cables,commonly leading to serious damage to risers and reduced service life.Vortex-induced vibration of the offshore risers could be effectively suppressed by fairing devices.In this paper,a newly developed vortex-induced vibration fairing and large eddy simulation model of the FLUENT software were used for numerical analysis,experimental research and stimulating vortex-induced vibration at 0.1–2 ms^-1.The data of the numerical model with fairing was compared and analyzed to study the vortex shedding frequency at different Reynolds numbers and changes in drag and lift coefficients.The displacement state of 12 in risers with and without fairing was experimentally tested using a five degree-of-freedom balance.The vortex-induced vibration effect of the fairing was tested at different velocities.The result shows the drag reduction effect of the fairing is more obvious when the flow velocity is 0.4–1.2 ms^-1 and the maximum drag reduction reaches 55.6%when the flow velocity is 0.6 ms^-1.Additionally,the drag reduction effect was obvious when the flow velocity was greater than 1.3 ms^-1 and less than 0.3.The result indicates that the developed 12 in fairing,with good potential in engineering applications,has good vortex-induced vibration-suppression effects.KANG Yongtian XIAO Wensheng WANG Quanbin ZHANG Dagang ZHAO Jun 2020Journal of Ocean University of China2020,19,2:4

8Preliminary design of an SCO_(2) conversion system applied to the sodium cooled fast reactor显示文摘The supercritical carbon dioxide(SCO_(2))Brayton cycle has become an ideal power conversion system for sodium-cooled fast reactors(SFR)due to its high efficiency,compactness,and avoidance of sodiumwater reaction.In this paper,the 1200 MWe large pool SFR(CFR1200)is used as the heat source of the system,and the sodium circuit temperature and the heat load are the operating boundaries of the cycle system.The performance of different SCO_(2) Brayton cycle systems and changes in key equipment performance are compared.The study indicates that the inter-stage cooling and recompression cycle has the best match with the heat source characteristics of the SFR,and the cycle efficiency is the highest(40.7%).Then,based on the developed system transient analysis program(FR-Sdaso),a pool-type SFR power plant system analysis model based on the inter-stage cooling and recompression cycle is established.In addition,the matching between the inter-stage cooling recompression cycle and the SFR during the load cycle of the power plant is studied.The analysis shows that when the nuclear island adopts the flow-advanced operation strategy and the carbon dioxide flowrate in the SCO_(2) power conversion system is adjusted with the goal of maintaining the sodium-carbon dioxide heat exchanger sodium side outlet temperature unchanged,the inter-stage cooling recompression cycle can match the operation of the SFR very well.Hongyi YANG Xiaoyan YANG Jun YANG Quanbin ZHAO Xiaokun WANG Daotong CHONG Chanjuan TANG Chengyuan JIANG 2021Frontiers in Energy2021,15,4:1
