1Rapid, automated, and reliable antimicrobial susceptibility test from positive blood culture by CAST‐R显示文摘Antimicrobial susceptibility tests(ASTs)are pivotal in combating multidrug resistant pathogens,yet they can be time‐consuming,labor‐intensive,and unstable.Using the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman microspectroscopy.Featuring a liquid robot for sample pretreatment and a machine learning‐based control scheme for data acquisition and quality control,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman spectra.The Expedited Minimal Inhibitory Concentration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii isolates.Further tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vancomycin,and levofloxacin reveal 93%categorical agreement with BMD‐based results.The automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials.Pengfei Zhu Lihui Ren Ying Zhu Jing Dai Huijie Liu Yuli Mao Yuandong Li Yuehui He Xiaoshan Zheng Rongze Chen Xiaoting Fu Lili Zhang Lijun Sun Yuanqi Zhu Yuetong Ji Bo Ma Yingchun Xu Jian Xu Qiwen Yang 2022mLife2022,1,3:3
2Artificial intelligence-assisted automatic and index-based microbial single-cell sorting system for One-Cell-One-Tube显示文摘Identification,sorting,and sequencing of individual cells directly from in situ samples have great potential for in-depth analysis of the structure and function of microbiomes.In this work,based on an artificial intelligence(AI)-assisted object detection model for cell phenotype screening and a cross-interface contact method for single-cell exporting,we developed an automatic and index-based system called EasySort AUTO,where individual microbial cells are sorted and then packaged in a microdroplet and automatically exported in a precisely indexed,“One-Cell-One-Tube”manner.The target cell is automatically identified based on an AI-assisted object detection model and then mobilized via an optical tweezer for sorting.Then,a crossinterface contact microfluidic printing method that we developed enables the automated transfer of cells from the chip to the tube,which leads to coupling with subsequent single-cell culture or sequencing.The efficiency of the system for single-cell printing is>93%.The throughput of the system for single-cell printing is~120 cells/h.Moreover,>80%of single cells of both yeast and Escherichia coli are culturable,suggesting the superior preservation of cell viability during sorting.Finally,AI-assisted object detection supports automated sorting of target cells with high accuracy from mixed yeast samples,which was validated by downstream single-cell proliferation assays.The automation,index maintenance,and vitality preservation of EasySort AUTO suggest its excellent application potential for single-cell sorting.Zhidian Diao Lingyan Kan Yilong Zhao Jingyun Song Chen Wang Yang Liu Fengli Zhang Teng Xu Rongze Chen Xixian Wang Xiaoyan Jing Jian Xu Yuandong Li Bo Ma 2022mLife2022,1,4:2

11Characteristics of shale gas reservoirs in Wufeng Formation and Longmaxi Formation in Well WX2,northeast Chongqing显示文摘To further understand shale reservoir characteristics of Wufeng Formation and Longmaxi Formation in the Wuxi area,northeast Chongqing,based on drilling data of Well WX2,and taking the graptolite biostratigraphy as the standard marker of stratigraphic division and comparison,the geochemistry,petrology,reservoir space and properties of organic-rich black shale were well investigated,and its gasbearing capacity and controlling factors were also analyzed.The result shows that in the Wufeng Formation and Longmaxi Formation of Well WX2,the organic-rich shale is 89.8 m thick and is characterized by good kerogen type,high organic abundance,moderate maturity and favorable hydrocarbongeneration condition,and the graptolite sequence is developed completely and continuously;the organic abundance is influenced by depositional rate,and the slow depositional rate is favorable for accumulation of organic matter in the black graptolite shale;from top to bottom,content of siliceous minerals increases and content of clay minerals decrease,therefore the brittleness increases;the organicrich siliceous shale and clay siliceous shale are favorable lithofacies for development of shale reservoirs;the nanopore is dominated by the parallel-plate pore with four open sides and has good connectivity;the pore size distribution curve has the multimodal characteristic,and the pore diameter mainly is in the range of 0.42e0.62 nm and the range of 3e5 nm;organic pores and interlayer pores of clay minerals make the greatest contribution to the total pore volume,while pores of brittle minerals have the least contribution;from top to bottom,organic pores gradually increase while interlayer pores of clay minerals gradually decrease;the on-site core gas content exceeds 8 m3/t,and the gas-bearing capacity is jointly controlled by hydrocarbon generation,reservoir and preservation conditions;and the WF2-LM6 biozone of the Katian to the early Aeronian is the high-quality shale reservoir,where the LM1 biozone of the Hirnantian was the best“sweet spot”which is the target of horizontal well drilling.Jin Wu Feng Liang Wen Lin Hongyan Wang Wenhua Bai Chao Ma Shasha Sun Qun Zhao Xiaojiang Song Rongze Yu 2017Petroleum Research2017,2,4:1

13肥胖妇女减肥时运动训练强度对脂肪组织激素敏感脂酶基因表达的影响(英文)显示文摘激素敏感脂酶(HSL)是一种调节脂肪组织分解的酶,能在运动引发的脂肪组织代谢中的慢性变化发挥重要作用。本研究的目的是确定肥胖妇女减肥时,有氧运动强度是否对腹部HSL基因表达有影响。方法:30位女性(BMI=33.0±0.7 kg/m^2,年龄58±1岁)分3组各完成为期20周的干预实验:热量限制(n=8):热量限制加中等强度运动(心率储备45%-50%,每周3天,n=9);热量限制加高强度运动(心率储备70%-75%,每周3天,n=13)。各组规定的能量赤字相同:营养不良(仅热量限制状态时每周2800千卡),或者营养不良(每周2400千卡)加运动(每周400千卡)。测量身体组成和最大需氧量,在实验前之后分别采集腹部皮下脂肪组织样本,用实时RT-PCR测量脂肪组织HSL基因表达。结果:3种干预方式在减少体重、脂肪量、脂肪百分比以及腰部脂肪方面均效果显著(p<0.01)。3种方式均未改变绝对最大耗氧量,但都增加了相对最大耗氧量(p<0 05到p<0.01)。同干预实验前相比,只有热量限制加高强度运动使脂肪组织HSL基因表达显著提高(p<0.01),这组的HSL基因表达与另外两组有显著差异(只有热量限制p<0.05,热量限制加中等强度运动p<0.01)。整体来看,脂肪组织HSL基因表达的变化与绝对最大耗氧量(r=0.55,p<0.01)和相对最大耗氧量(r=0.32,p=0.09)成正相关。结论:这些结果支持肥胖妇女减肥过程中有氧运动强度对其脂肪组织代谢中HSL通路有潜在影响。Tongjian You Xuewen Wang Rongze Yang Mary F.Lyles Dawei Gong Barbara J.Nicklas 2012Journal of Sport and Health Science2012,1,3:1
14Bonding and Fracture Characteristics at α_2/γ Interface in TiAl Alloy with B Addition显示文摘The bonding characteristics of (0001)α2||(111)γ interface in two-phase TiAl alloy have beeninvestigated with the recursion method. The results of bond order integral and interaction energybetween atoms are presented. The effects of B on atoms bonding both in constituent phase andat the α2/γ interface have been studied. The correlation between the mechanical propertiesof the alloy and the bonding at the interface has been discussed. The results suggest that Bsegregation to the interface benefits the ductility. 15Quantitative characterization of horizontal well production performance with multiple indicators: a case study on the Weiyuan shale gas field in the Sichuan Basin, China显示文摘To quantitatively characterize the horizontal shale gas well productivity and identify the dominant productivity factors in the Weiyuan Shale Gas Field,Sichuan Basin,a practical productivity method involving multiple indicators was proposed to analyze the production performance of 150 horizontal wells.The normalized test production,flowback ratio,first-year initial production and estimated/expected ultimate recovery(EUR)were introduced to estimate the well productivity in different production stages.The correlation between these four indicators was determined to reveal their effects on production performance forecasts.In addition,the dominant productivity factors in the present stage were identified to provide guidance for production performance enhancement.Research indicates that favorable linear relations exist between the normalized test production,first-year initial production and EUR.The normalized test production is regarded as an important indicator to preliminarily characterize the well productivity in the initial stage.The first-year initial production is the most accurate productivity evaluation indicator after a year.The flowback ratio is a supplementary indicator that qualitatively represents the well productivity and fracturing performance.The well productivity is greatly dependent on the lateral target interval,drilling length of Longmaxi1_(1)^(1)(LM1_(1)^(1))and wellbore integrity.The first-year recovery degree of EUR is 24%–58%with a P50 value of 35%.Rongze YU Wei GUO Lin DING Meizhu WANG Feng CHENG Xiaowei ZHANG Shangwen ZHOU Leifu ZHANG 2021Frontiers of Earth Science2021,15,2:0

17Bonding Characteristics of the Intermetallic Compound Al_3Ti+Cr显示文摘Based on our structural model and a tight-binding Hamiltonian the eIectronic structure has been calculated by use of a recursion method (RM) for two models of the tetragonal DO22 and the cubic Ll2 crystal structures in Al3Ti+Cr for contrast, from which bonding characteristics have been discussed. The results for the interaction energy (IE) show that Cr prefers to replacing Al site. Cr partly replacing Al can change the IE between the atoms and make the constriction of lattice constant c and the elongation of lattice constant a for the DO22 and thereby may induce the change of crystal structure from DO22 to L12. 18Culture-free identification of fast-growing cyanobacteria cells by Raman-activated gravity-driven encapsulation and sequencing显示文摘By directly converting solar energy and carbon dioxide into biobased products,cyanobacteria are promising chassis for photosynthetic biosynthesis.To make cyanobacterial photosynthetic biosynthesis technology economically feasible on industrial scales,exploring and engineering cyanobacterial chassis and cell factories with fast growth rates and carbon fixation activities facing environmental stresses are of great significance.To simplify and accelerate the screening for fast-growing cyanobacteria strains,a method called Individual Cyanobacteria Vitality Tests and Screening(iCyanVS)was established.We show that the 13C incorporation ratio of carotenoids can be used to measure differences in cell growth and carbon fixation rates in individual cyanobacterial cells of distinct genotypes that differ in growth rates in bulk cultivations,thus greatly accelerating the process screening for fastest-growing cells.The feasibility of this approach is further demonstrated by phenotypically and then genotypically identifying individual cyanobacterial cells with higher salt tolerance from an artificial mutant library via Raman-activated gravity-driven encapsulation and sequencing.Therefore,this method should find broad applications in growth rate or carbon intake rate based screening of cyanobacteria and other photosynthetic cell factories.Jinyu Cui Rongze Chen Huili Sun Yingyi Xue Zhidian Diao Jingyun Song Xiaohang Wang Jia Zhang Chen Wang Bo Ma Jian Xu Guodong Luan Xuefeng Lu 2023Synthetic and Systems Biotechnology2023,8,4:0
19Electronic Structure Effect on Model Cluster for L1_2 Structure of Al_3Ti Intermetallic Compound with an Addition of Alloying Elements Fe, Ni and Cu显示文摘By use of self-consistent field Xα scattered-wave (SCF-Xα-SW) method, the electronic structure was calculated for four models of Ti4Al14X (X=Al, Fe, Ni and Cu) clusters. The Ti4Al14X cluster was developed based on L12 Al3Ti-base intermetallic compound. The results are presented using the density of states (DOS) and one-electron properties, such as relative binding tendency between the atom and the model cluster, and hybrid bonding tendency between the alloying element and the host atoms. By comparing the four models of Ti4Al14X cluster, the effect of the Fe, Ni or Cu atom on the physical properties of Al3Ti-based L12 intermetallic compounds is analyzed. The results indicate that the addition of the Fe, Ni or Cu atom intensifies the relative binding tendency between Ti atom and Ti4Al14X cluster. It was found that the Fermi level (EF) lies in a maximum in the DOS for Ti4Al14Al cluster; on the contrary, the EF comes near a minimum tn the DOS for Ti4Al14X (X=Fe, Ni and Cu) cluster. Thus the L12 crystal structure for binary Al3Ti alloy is unstable, and the addition of the Fe, Ni or Cu atom to Al3Ti is benefical to stabilize L12 crystal structure. 20Microfluidics for nano-drug delivery systems: From fundamentals to industrialization显示文摘In recent years,owing to the miniaturization of the fluidic environment,microfluidic technology offers unique opportunities for the implementation of nano drug delivery systems(NDDSs)production processes.Compared with traditional methods,microfluidics improves the controllability and uniformity of NDDSs.The fast mixing and laminar flow properties achieved in the microchannels can tune the physicochemical properties of NDDSs,including particle size,distribution and morphology,resulting in narrow particle size distribution and high drug-loading capacity.The success of lipid nanoparticles encapsulated mRNA vaccines against coronavirus disease 2019 by microfluidics also confirmed its feasibility for scaling up the preparation of NDDSs via parallelization or numbering-up.In this review,we provide a comprehensive summary of microfluidics-based NDDSs,including the fundamentals of microfluidics,microfluidic synthesis of NDDSs,and their industrialization.The challenges of microfluidicsbased NDDSs in the current status and the prospects for future development are also discussed.We believe that this review will provide good guidance for microfluidics-based NDDSs.Huan Zhang Jie Yang Rongze Sun Songren Han Zhaogang Yang Lesheng Teng 2023Acta Pharmaceutica Sinica B2023,13,8:0
