| 1 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 2 | Active greeting technique: a mother-and-child catheter based technique to facilitate retrograde wire externalization in recanalization of coronary chronic total occlusion显示文摘Although retrograde approach has greatly improved the success rate of percutaneous coronary intervention(PCI) for coronary chronic total occlusion(CTO), retrograde wire externalization still remains challenging and time-consuming in some cases. Cases utilizing ‘‘Active Greeting Technique(AGT)', a mother-and-child catheter based technique to facilitate retrograde wire externalization, were extracted from Chronic Total Occlusion Club, China(CTOCC) database. AGT was performed by deep intubation a mother-and-child catheter(GuidezillaTMextension, 4 or 5 Fr inner catheter, and etc.) in combination with either reverse controlled antegrade or retrograde subintimal tracking(CART) technique or retrograde wire crossing technique. A total of 111 patients with 112 CTO lesions treated with this technique were retrospectively analyzed. Reverse CART technique and retrograde wire crossing technique were performed in 90.2% and 9.8% of all procedures. The utilization of GuidezillaTMextension, 4 Fr, and 5 Fr inner catheter accounted for 94.6%, 3.6%, and 1.8%, respectively. Externalization of retrograde wire was successful in all cases. No procedural complications were adjudicated to AGT. Complications independent of AGT included two target vessel perforations and two collateral perforations. No in-hospital major adverse cardiac events were found. | 3 | Endocrine Disruption Activity of 30-day Dietary Exposure to Decabromodiphenyl Ethane in Balb/C Mouse显示文摘Objective This study aimed to evaluate the hepatotoxicity,metabolic disturbance activity and endocrine disrupting activity of mice treated by Decabromodiphenyl ethane(DBDPE).Methods In this study,Balb/C mice were treated orally by gavage with various doses of DBDPE.After 30 days of treatment,mice were sacrificed; blood,livers and thyroid glands were obtained,and hepatic microsomes were isolated.Biochemical parameters including 8 clinical chemistry parameters,blood glucose and hormone levels including insulin and thyroid hormone were assayed.The effects of DBDPE on hepatic cytochrome P450(CYP) levels and activities and uridinediphosphate-glucuronosyltransferase(UDPGT) activities were investigated.Liver and thyroid glands were observed.Results There were no obvious signs of toxicity and no significant treatment effect on body weight,or liver-to-body weight ratios between treatment groups.The levels of ALT and AST of higher dose treatment groups were markedly increased.Blood glucose levels of treatment groups were higher than those of control group.There was also an induction in TSH,T3,and f T3.UDPGT,PROD,and EROD activities were found to have been increased significantly in the high dose group.Histopathologic liver changes were characterized by hepatocyte hypertrophy and cytoplasmic vacuolization.Our findings suggest that DBDPE can cause a certain degree of mouse liver damage and insufficiency.Conclusion DBDPE has the activity of endocrine disruptors in Bal/C mice,which may induce drug-metabolizing enzymes including CYPs and UDPGT,and interfere with thyroid hormone levels mediated by Ah R and CAR signaling pathways.Endocrine disrupting activity of DBDPE could also affect the glucose metabolism homeostasis. | SUN Ru Bao SHANG Shuai ZHANG Wei LIN Ben Cheng WANG Qiang SHI Yun XI Zhu Ge | 2018 | Biomedical and Environmental Sciences2018,31,1: | 7 |
| 4 | Cytotoxicity and Apoptosis Induction in Human HepG2 Hepatoma Cells by Decabromodiphenyl Ethane显示文摘Objective To investigate the toxic effects of decabromodiphenyl ethane (DBDPE), used as an alternative to decabromodiphenyl ether in vitro. Methods HepG2 cells were cultured in the presence of DBDPE at various concentrations (3.125-100.0 mg/L) for 24, 48, and 72 h respectively and the toxic effect of DBDPE was studied. Results As evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide and lactate dehydrogenase assays and nuclear morphological changes, DBDPE inhibited HepG2 viability in a time- and dose-dependent manner within a range of 12.5 mg/L to 100 mg/L and for 48 h and 72 h. Induction of apoptosis was detected at 12.5-100 mg/L at 48 h and 72 h by propidium iodide staining, accompanied with overproduction of reactive oxygen species (ROS). | 13 | Thrombin Generation Increasing with Age and Decreasing with Use of Heparin Indicated by Calibrated Automated Thrombogram Conducted in Chinese显示文摘Objective Calibrated Automated Thrombogram(CAT) is a test to monitor the generation of thrombin. It can be described by four parameters: lag time, peak thrombin, endogenous thrombin potential(ETP) and time to peak(ttPeak). This study aims to determine the normal ranges of CAT parameters in Chinese, and evaluate whether thrombin generation is correlated with the concentration of heparin/low molecular weight heparin. Methods Plasma from 120 healthy subjects were collected to determine the normal rangea of CAT parameters in Chinese. Normal plasma pool(NPP, n=25) spiked with different concentrations of heparin or enoxaparin were used to detecte CAT parameters. The overall and age specific normal ranges of CAT parameters were calculated using descriptive statistics method with mean±2SD. The correlation between CAT parameters and age or concentrations of heparin, enoxaparin were analyzed with linear regression model. Results The normal ranges for lag time, peak thrombin, ETP, ttPeak in the subjects were 3.648±2.465 min, 367.39±151.93 nmol/L, 2277±1030 nmol/L·min and 6.372±4.280 min respectively. Age was linearly correlated with lag time(r=-0.6583, P<0.0001), peak thrombin(r=0.4863, P<0.0001), ETP(r=0.3608, P<0.0014) and ttPeak(r=-0.6313, P<0.0001). The values of ETP/peak ratio were linearly correlated with concentrations of heparin. Conclusion The normal ranges of four CAT parameters for Chinese were determined. CAT parameters are associated with age. However, there are few relevant epidemiologic studies. We conducted a cross-sectional study in Songnan community, China in 2009 to investigate the association between serum fetuin-A concentrations and thyroid function. A total of 2,984 participants aged 40 years and older were analyzed. Multivariable linear regression analysis revealed that serum fetuin-A concentrations were positively associated with log(free triiodothyronine) and were inversely associated with log(thyroid peroxidase antibody) after adjustment(both P < 0.05). Compared with the participants in the lowest tertile of free triiodothyronine and free thyroxine level, those in the highest tertile had higher fetuin-A concentrations.
