| 3 | Clinical application of specific antibody against glypican-3 for hepatocellular carcinoma diagnosis显示文摘Glypican-3(GPC3) is a promising tumor marker for hepatocellular carcinoma(HCC) diagnosis with high sensitivity and specificity.The aim of this study was to establish an immunohistochemical detection method for GPC3 using the 7D11 monoclonal antibody(7D11 mAb) and evaluate its application for HCC diagnosis.The feasibility of the 7D11 mAb was evaluated by immunohistochemistry performed on adjacent normal liver and intrahepatic cholangiocarcinoma(ICC) samples,Furthermore,the serum GPC3 levels were evaluated in 40 HCC patients,7 ICC patients and 50 healthy donors.The results showed that GPC3 was expressed in 85% of HCC tissues(34/40),but was undetectable in ICC tissues and adjacent normal tissues.GPC3 was significantly increased in the serum of HCC patients(17/40,42.5%) but was undetectable in the serum of ICC patients(0/7,0%) and healthy donors(0/50,0%).This prospective study evaluated the clinical usefulness of 7D11 mAb for GPC3 detection in HCC patients.In conclusion,the use of 7D11 mAb might be good for GPC3 large-scale applications for clinical diagnosis of HCC. | YU JuanPing MA Qiang ZHANG Bin MA RuiJuan XU XiGuang LI MingSong XU WeiWen LI Ming | 2013 | Science China(Life Sciences)2013,56,3: | 18 |

| 6 | Transplantation of placenta-derived mesenchymal stem cells reduces hypoxic-ischemic brain damage in rats by ameliorating the inflammatory response显示文摘Hypoxic-ischemic brain damage(HIBD)is a common cause of infant death.The purpose of our research was to explore the immunoregulatory mechanism of placenta-derived mesenchymal stem cells(PD-MSCs)in HIBD treatment.Seven-day-old rat pups were randomly divided into HIBD,PD-MSC,fibroblast,and control groups.Forty-eight hours after HIBD induction,cells at a density of 5×104 cells/10µl were injected into the cerebral tissue in the PD-MSC and fibroblast groups.The TNF-α,interleukin-17(IL-17),interferon-γ(IFN-γ),and IL-10 levels were detected through quantitative real-time polymerase chain reaction(RT-PCR)and enzyme-linked immunosorbent assay(ELISA).Regulatory T cell(Tregs)populations were detected through flow cytometry,and forkhead box P3(Foxp3)was measured through western blot analysis.Behavioral tests and gross and pathological examinations showed that PD-MSC treatment exerted significantly stronger neuroprotective effects than the other treatments.The expression levels of pro-inflammatory cytokines were substantially upregulated after HI injury.Compared with fibroblast treatment,PD-MSC treatment inhibited the production of pro-inflammatory cytokines and increased the production of IL-10 in the ischemic hemispheres and peripheral blood serum(all P<0.01).Flow cytometry results showed a notable increase in the number of Tregs within the spleen of the HIBD group.Moreover,the number of Tregs and the Foxp3 expression levels were higher in the PD-MSC treatment group than in the HIBD and fibroblast groups(all P<0.01).Our research suggests that the mechanism of PD-MSC treatment for HIBD partially involves inflammatory response suppression. | Hongfang Ding Hui Zhang Huifang Ding Dong Li Xinhao Yi Xiaoxu Ma Ruijuan Li Mei Huang Xiuli Ju | 2017 | Cellular & Molecular Immunology2017,14,8: | 10 |

| 8 | High molecular karyotype variation revealed in indigenous Elymus nutans in the Qinghai Plateau显示文摘The karyotypes of 27 individuals of Elymus nutans from eight wild populations in the Qinghai Plateau were analyzed using sequential FISH and GISH. High FISH pattern polymorphism and karyotype variation were detected within and among populations. The chromosome variations were mainly characterized as repeat deletions and amplifications along with inter-genomic translocations. The chromosomes of the St and Y genomes demonstrated higher polymorphism than those of the H genome. Six different intergenomic translocations were identified in 33.3% of individuals; type Ⅰ and Ⅱ translocations were detected with higher frequency. Further analysis revealed that type Ⅰ and Ⅱ translocations were distributed in different geographic regions. | 9 | 5-Hydroxymethylome in Circulating Cell-free DNA as A Potential Biomarker for Non-small-cell Lung Cancer显示文摘Non-small-cell lung cancer(NSCLC), the most common type of lung cancer accounting for 85% of the cases, is often diagnosed at advanced stages owing to the lack of efficient early diagnostic tools. 5-Hydroxymethylcytosine(5 hmC) signatures in circulating cell-free DNA(cfDNA) that carries the cancer-specific epigenetic patterns may represent the valuable biomarkers for discriminating tumor and healthy individuals, and thus could be potentially useful for NSCLC diagnosis. Here,we employed a sensitive and reliable method to map genome-wide 5 hmC in the cfDNA of Chinese NSCLC patients and detected a significant 5 hmC gain in both the gene bodies and promoter regions in the blood samples from tumor patients compared with healthy controls. Specifically, we identified six potential biomarkers from 66 patients and 67 healthy controls(mean decrease accuracy>3.2, P < 3.68E-19) using machine-learning-based tumor classifiers with high accuracy. | 12 | Pore Structure Regulation of Carboxyethylated Microfibrillated Cellulose Films显示文摘Carboxyethylation is a recent chemical pretreatment for preparation of microfibrillated cellulose(MFC).The carboxyethylated MFC film prepared by coating method has compact structure and high mechanical properties.In order to expand its application,three approaches including using organic solvents,different drying methods and cationic additives,have been adopted in this paper to enrich and regulate the pore structure of MFC film.The results show that all the approaches can improve the pore structure but decrease the mechanical properties of MFC film.When organic solvents such as ethanol,isopropanol and n-butanol were used to replace the water in MFC suspension or pre-dried MFC film,the pore structure of films were increased.Additionally,the film obtained by freeze-drying or air-drying after freezing in liquid nitrogen or freezer has high porosity but poor strength.The best drying process is to rewet dry MFC film,freeze in liquid nitrogen and then freeze-dry.Moreover,the addition of cationic polyelectrolytes or alkene ketone dimer(AKD)in MFC suspension can also significantly increase the film's porosity.Through the above approaches,the porosity of carboxyethylated MFC film can be regulated between 20% and 90%. | Meican Li Jinghuan Chen Jingang Liu Zehong Xu Yanqun Su Ruijuan Zhang Yanfen Du | 2020 | Paper And Biomaterials2020,5,2: | 5 |
| 13 | 键价和方法计算离子输运通道筛选锂离子无机固体电解质(英文)显示文摘Inorganic solid electrolytes have distinguished advantages in terms of safety and stability,and are promising to substitute for conventional organic liquid electrolytes.However,low ionic conductivity of typical candidates is the key problem.As connective diffusion path is the prerequisite for high performance,we screen for possible solid electrolytes from the 2004 International Centre for Diffraction Data(ICDD)database by calculating conduction pathways using Bond Valence(BV)method.There are 109846 inorganic crystals in the 2004 ICDD database,and 5295 of them contain lithium.Except for those with toxic,radioactive,rare,or variable valence elements,1380 materials are candidates for solid electrolytes.The rationality of the BV method is approved by comparing the existing solid electrolytes’conduction pathways we had calculated with those from experiments or first principle calculations.The implication for doping and substitution,two important ways to improve the conductivity,is also discussed.Among them Li2CO3 is selected for a detailed comparison,and the pathway is reproduced well with that based on the density functional studies.To reveal the correlation between connectivity of pathways and conductivity,/-LiAlO2 and Li2CO3 are investigated by the impedance spectrum as an example,and many experimental and theoretical studies are in process to indicate the relationship between property and structure.The BV method can calculate one material within a few minutes,providing an efficient way to lock onto targets from abundant data,and to investigate the structure-property relationship systematically. | GAO Jian CHU Geng HE Meng ZHANG Shu XIAO RuiJuan LI Hong CHEN LiQuan | 2014 | Science China(Physics,Mechanics & Astronomy)2014,57,8: | 5 |
| 14 | Eco-friendly PVA/Kaolin Clay Coating for Barrier Paper显示文摘The resistance of wood-fiber paper to water, grease, and water vapor is usually attained by immersing the base paper in hydrophobic oil, laminating with a plastic or metal film, or the application of a barrier coating. Oil impregnation and the addition of films may make the paper difficult to recycle or persistent in the environment owing to their strong binding force and nondegradability. Environmental concerns have attracted worldwide attention to eco-friendly barrier coatings. In this study, degradable polyvinyl alcohol(PVA) and kaolin clay pigment were used to prepare coatings that were applied to a base paper. By measuring the barrier properties of the coated paper, including the water absorptiveness(Cobb60 value), Hercules sizing degree, oil resistance(Kit rating), and water vapor transmission rate(WVTR), an optimal coating formulation and process were proposed. To examine the barrier mechanism of the PVA/kaolin clay coating, we characterized the coating microstructures using a scanning electron microscopy(SEM) and a mercury porosimeter. The results showed that the Cobb60 value and water vapor transmission rate of the coated paper decreased by 61.4% and 98.6%, respectively, compared with the base paper, for a pre-coating weight of 0.98 g/m^2 and a top-coating weight of about 3.23 g/m^2. Furthermore, the Hercules sizing degree rose by a factor of 337.2, while the oil resistance(Kit rating) increased from 0 to 12. The optimum drying temperature for a wet coating layer was found to be 170℃, and the optimum weight ratio of PVA to kaolin clay in the coating was determined to be 50∶50. It was assumed that the PVA/kaolin clay coating improved the smoothness of the paper considerably and decreased the pore size by filling the pores on the paper surface and forming an even film, thus enhancing the paper barrier performance. | 15 | Influence of organic amendments on the sorption of pentachlorophenol on soils显示文摘The influence of pH on the sorption of pentachlorophenol (PCP) onto three organic amendments, char, humic acid (HA) and peat, and the effect of organic amendments on PCP sorption to three kinds of soils were evaluated. The sorption of PCP on these sorbents fitted the Freundlich model well, suggesting that PCP sorption isotherms were nonlinear with exponential coefficient (N) value lower than 1. The PCP sorption capacity on three organic amendments and the N values increased with decreasing pH. The sorption capacities were in the order: char > HA > peat. Organic amendments resulted in an enhancement of sorption capacities of soils. The N values of PCP sorption on soils were decreased by char amendment and increased by HA and peat amendments. This study demonstrated that application of traditional organic amendments led to the enhancement of PCP sorption by soils. Tremendous efforts have been devoted to the optimization of the electrocatalytic performance of the catalysts under industrial-relevant current densities via rational structure design,which induces a preferential electron distribution that favors the adsorption/desorption behavior of the key intermediates, thus accelerating the reaction kinetics. In this review, a brief introduction of the current energy status will be first presented to necessitate the importance of green hydrogen.Followed by the basic concepts and fundamental understanding of the reaction mechanisms, we present efficient strategies for the enhancement of the electrocatalytic performance of the catalysts to meet the rigorous requirement under industrial conditions and the in-depth understanding behind the reinforcement will be briefly discussed next. Then the recent advances regarding the rational design of electrocatalysts operating at an industrial scale will be summarized. | 18 | Targeting lactate dehydrogenase A (LDHA) exerts antileukemic effects on T-cell acute lymphoblastic leukemia显示文摘Background:T-cell acute lymphoblastic leukemia(T-ALL)is an uncommon and aggressive subtype of acute lymphoblastic leukemia(ALL).In the serum of T-ALL patients,the activity of lactate dehydrogenase A(LDHA)is increased.We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes.The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.Methods:Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate.MTT,colony formation,apoptosis,and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells.Quantitative real-time PCR(qPCR)and Western blotting analyses were applied to determine the related signaling pathways.A mitochondrial reactive oxygen species(ROS)assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate.A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology,and then TUNEL,Western blotting,and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.Results:Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells.It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production(all P<0.001).Blocking LDHA significantly decreased the gene and protein expression of c-Myc,as well as the levels of phosphorylated serine/threonine kinase(AKT)and glycogen synthase kinase 3 beta(GSK-3β)in the phosphatidylinositol 3′-kinase(PI3K)signaling pathway.LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.Conclusion:Targeting LDHA exerted an antileukemic effect on T-ALL,representing a potential strategy for T-ALL treatment. | Haizhi Yu Yafei Yin Yifang Yi Zhao Cheng Wenyong Kuang Ruijuan Li Haiying Zhong Yajuan Cui Lingli Yuan Fanjie Gong Zhihua Wang Heng Li Hongling Peng Guangsen Zhang | 2020 | Cancer Communications2020,40,10: | 2 |

| 20 | Biochemical characterization of a polyethylene terephthalate hydrolase and design of high-throughput screening for its directed evolution显示文摘Polyethylene terephthalate(PET),one of the most widely used plastics in the world,causes serious environmental pollution.Recently,researchers have focused their efforts on enzymatic degradation of PET,which is an attractive way of degrading and recycling PET.In this work,PET hydrolase Sb PETase from Schlegelella brevitalea sp.nov.was biochemically characterized,and rational design was performed based on its sequence similarity with the previ-ously reported Is PETase from Ideonella sakaiensis,resulting in a triple mutant with increased activity.Furthermore,using a sec-dependent signal peptide PeIB and colicin release protein Kil,we set up a high-efficiency secretion system of PETase in Escherichia coli BL21(DE3),enabling higher PETase secretion.Utilizing this secretion system,we established a high-throughput screening method named SecHTS(sec retion-based h igh-throughput s creening)and performed directed evolution of Is PETase and Sb PETase through DNA shuffling.Finally,we generated a mutant Is PETase S139T with increased activity from the mutant library. | Xiaotong Wang Chaoyi Song Qingsheng Qi Youming Zhang Ruijuan Li Liujie Huo | 2022 | Engineering Microbiology2022,2,2: | 2 |
