1Expression of the Phycoerythrin Gene of Gracilaria lemaneiformis (Rhodophyta) in E. coli and Evaluation of the Bioactivity of Recombinant PE显示文摘Phycoerythrin (PE) is one of the most important proteins involved in light capturing during photosynthesis in red algae. Its potential biological activities had gained wide concerns. In the present study, tumor cytotoxic and hydroxyl radical assay were preformed to detect the bioactivity of recombinant PE. Recombinant plasmids pGEX-PE and pBGL were transformed into E.coli BL21 to make two recombinant strains BEX (pGEX-PE) and BGL (pBGL). PE expressing in BEX (pGEX-PE) was validated by SDS-PAGE and Western blotting analysis. SDS-PAGE analysis indicated that the PE-GST fusion protein was mostly inclusion bod- ies. Specific expression of PE was confirmed by Western blotting analysis. The recombinant E. coli BEX (pGEX-PE) cells were col- lected and sonicated. The supernatants were reserved for the tumor cytotoxic experiments. The result of tumor cytotoxic assay indi- cated that the supernatants containing PE had the activity of inhibiting the growth of Hela cells and with the increase of protein con- centration, the inhibiting rate increased from 37.31% to 63.26%, which showed significant difference from the control. Hydroxyl radical scavenging effect was tested with supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates treated with sonication and heating. For the sonication samples, the scavenging rates of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were significantly higher than the negative control BL21(pGEX-4T) (P<0.02), and the scavenging rates increased slowly following the increase of the protein content. For the heating samples, except for the 0.2 mg mL-1 BGL (pBGL) products, the scavenging effects of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were stronger than that of negative control BL21(pGEX-4T). However, the effect intensity was not positively correlated with the increase of the protein concentration. Though a partially de- creased hydroxyl radical scavenging activity was led by heating, the biological activity was still retained and conspicuous. 2Mode transition induced by back-breakdown of the gliding arc and its influence factors显示文摘Gliding arc is a mostly used non-equilibrium plasma generation method whose characteristic is affected by the backbreakdown phenomenon.Mode transition induced by back-breakdown of the gliding arc is studied in this work and effects of gas flow rate,applied voltage,electrode parameters on mode transition were studied.Experimental results show that there are two typical modes during the development of the gliding arc.Mode A comprises periodicitical development of the arc from the minimum gap breakdown to the longest extinguishment,while the arc in mode B continues to produce a back-breakdown at the ends of the electrodes.As the flow rate descends,the gliding arc gradually changes from mode A to B.It is the decrease of the arc velocity caused by lower flow rate leads to the occurrence of back-breakdown,which generates mode B.Smaller electrode opening angle,shorter length and wider minimum gap reduce the gliding speed,so that arc is more likely to enter mode B.As the applied voltage is increased,enhancing of the electric field strength on the breakdown path of the back-breakdown and thickening of the arc's diameter allow the gliding arc to enter mode B at a higher arc speed.Ruobing Zhang Haochen Huang Tianshu Yang 2020High Voltage2020,5,3:3
3Ultrasound-guided percutaneous microwave ablationfor small liver cancers adjacent to large vessels:long-term outcomes and strategies显示文摘Objective The aim of the study was to evaluate the long-term efficacy and safety of percutaneous microwave ablation(MWA) for small hepatic cancers adjacent to large vessels and to investigate the treatment strategies.Methods From March 2009 to July 2015,a total of 86 patients with 94 tumors underwent ultrasound(US)-guided percutaneous MWA,with pathologically proven or clinically diagnosed liver cancers measuring ≤ 3 cm in diameter and located ≤ 10 mm from a major vessel(n = 94).Regular follow-up after MWA was performed to assess treatment efficacy and perioperative complications.Results The complete ablation rate at 1 month after MWA was 93.3%(84/90).The 6-,9-,12-,24-,36-,48-,60-,72-,and 84-month local recurrence rates were 2.4%,2.4%,3.7%,6.6%,8.4%,8.4%,8.4%,8.4%,and 8.4%,respectively.There were no major complications.The perioperative special complication rate was 5.32%(5/94),including 3 cases of moderate liver function damage and 2 cases of limited sub-capsular hematoma.Conclusion Percutaneous MWA for small hepatic cancers adjacent to large vessels is feasible,effective,and safe with an acceptable rate of complications.The key point is to strictly follow operative indications and adopt proper treatment strategies.Ruobing Liu Kaiyan Li Hongchang Luo Wei Zhang Tingting Zhang Meng Gao Wenhui Zha Xinwu Cui Youbin Deng 2017Oncology and Translational Medicine2017,3,2:3

8Interfacial bonding enhancement of the RTV recoating with sandwiched contaminant by plasma jet显示文摘Room temperature vulcanised(RTV)silicone rubber is widely used in electrical power system,applied to the glass or porcelain insulator surface to enhance insulation.RTV coatings get contaminated after long-term operation and need to be recoated.Contaminants accumulate and embed into the surface of base RTV coating,forming a hard-to-remove inorganic transition layer.Such sandwiched contaminant layer destroys the adhesion between original and reapplied silicone rubber coating greatly.Authors’study innovatively reports a method to enhance interfacial bond properties of the RTV recoating with sandwiched contaminant layer,by using an atmospheric-pulsed discharge plasma jet(APPJ)for surface treatment.Water boiling test and 180-degree peel test were conducted to evaluate interfacial bond properties macroscopically.Scanning electron microscopy(SEM)was applied to observe the cross-section of recoating interface,while Fourier-transform infrared spectroscopy(FTIR)to detect chemical changes of contaminant layer,giving out microscopic and theoretical explanation.Shanshan Wang Shuang Li Jianjun Li Ruobing Zhang Hui Liu 2019High Voltage2019,4,4:1

15Interaction between plasma jet and silicone rubber covered by porous inorganic contaminants:Surface hydrophobicity or hydrophilicity?显示文摘The variation of surface hydrophobicity of silicone rubber materials covered with porous inorganic pollution layer under the action of atmospheric pressure plasma jet presents some new features,which have important engineering applications in the field of outdoor insulation of power systems.Interaction of the plasma with the polluted silicone rubber is studied and surface hydrophobicity is measured.Experimental results show that the exposure of plasma greatly accelerates the transfer of low molecular weight siloxanes(LMWs)in the pollution layer.However,the surface hydrophobicity is determined by the LMWs contents and the exposure time.Two acceleration effects of plasma during and after plasma exposure are proposed,their influence on surface hydrophobicity is analysed and related mechanisms are clarified.In addition,the competing influence of plasmainduced acceleration of the transfer of LMWs towards the surface and plasmaoxidation of LMWs on surface hydrophobicity is discussed.Shuang Li Jianjun Li Yongqiang Fu Ruobing Zhang 2022High Voltage2022,7,6:1
16Cryo-EM structures of PAC1 receptor reveal ligand binding mechanism显示文摘The pituitary adenylate cyclase-activating polypeptide type I receptor(PAC1R)belongs to the secretin receptor family and is widely distributed in the central neural system and peripheral organs.Abnormal activation of the receptor mediates trigeminovascular activation and sensitization,which is highly related to migraine,making PAC1R a potential therapeutic target.Elucidation of PAC1R activation mechanism would benefit discovery of therapeutic drugs for neuronal disorders.PAC1R activity is governed by pituitary adenylate cyclase-activating polypeptide(PACAP),known as a major vasodilator neuropeptide,and maxadilan,a native peptide from the sand fly,which is also capable of activating the receptor with similar potency.These peptide ligands have divergent sequences yet initiate convergent PAC1R activity.It is of interest to understand the mechanism of PAC1R ligand recognition and receptor activity regulation through structural biology.Here we report two near-atomic resolution cryo-EM structures of PAC1R activated by PACAP38 or maxadilan,providing structural insights into two distinct ligand binding modes.The structures illustrate flexibility of the extracellular domain(ECD)for ligands with distinct conformations,where ECD accommodates ligands in different orientations while extracellular loop 1(ECU)protrudes to further anchor the ligand bound in the orthosteric site.By structureguided molecular modeling and mutagenesis,we tested residues in the ligand-binding pockets and identified clusters of residues that are critical for receptor activity.The structures reported here for the first time elucidate the mechanism of specificity and flexibility of ligand recognition and binding for PACT R,and provide insights toward the design of therapeutic molecules targeting PAC1R.Jia Wang Xianqiang Song Dandan Zhang Xiaoqing Chen Xun Li Yaping Sun Cui Li Yunpeng Song Yao Ding Ruobing Ren Essa Hu Harrington Liaoyuan AHu Wenge Zhong Cen Xu Xin Huang Hong-Wei Wang Yingli Ma 2020Cell Research2020,30,5:1

19Study on the interaction mechanism of virgin olive oil polyphenols with mucin and α-amylase显示文摘The interaction mechanism of mucin and α-amylase with virgin olive oil (VOO) polyphenols (oleuropein (OL),tyrosol (TY),hydroxytyrosol (HT)) was analyzed by fluorescence,ultraviolet (UV) absorption,attenuated total reflection Fourier transform infrared (ATR-FTIR),circular dichroic (CD) spectroscopy and molecular docking.A total of 17 polyphenols have been identified in the selected VOO,and the TY,HT and OL were the main compounds.The quenching mechanism between mucin/α-amylase and three VOO polyphenols was static,mainly through van der Waals forces and hydrogen bonds.The results of UV absorption,ATR-FTIR,and CD spectroscopy revealed that the conformation of mucin changed after combining with the three polyphenols,while that of the α-amylase changed little.Molecular docking predicted the interaction sites of the three polyphenols with human salivary mucin and α-amylase.The present study could provide the theoretical foundation for further research on the interaction between human salivary protein and phenolic compounds in VOO.Gangcheng Wu Ruobing Mao Yiren Zhang Ling Zhu Emad Karrar Hui Zhang Qingzhe Jin Xingguo Wang 2022Food Bioscience2022,47,3:1
