| 1 | Molecular regulation of sucrose catabolism and sugar transport for development~ defence and phloem function显示文摘Sucrose(Suc) is the major end product of photosynthesis in mesophyll cells of most vascular plants. It is loaded into phloem of mature leaves for long-distance translocation to non-photosynthetic organs where it is unloaded for diverse uses. Clearly,Suc transport and metabolism is central to plant growth and development and the functionality of the entire vascular system. Despite vast information in the literature about the physiological roles of individual sugar metabolic enzymes and transporters,there is a lack of systematic evaluation about their molecular regulation from transcriptional to post-translational levels.Knowledge on this topic is essential for understanding and improving plant development,optimizing resource distribution and increasing crop productivity. We therefore focused our analyses on molecular control of key players in Suc metabolism and transport,including:(i) the identification of promoter elements responsive to sugars and hormones or targeted by transcription factors and microRNAs degrading transcripts of target genes; and(ii)modulation of enzyme and transporter activities through protein-protein interactions and other post-translational modifications. | 4 | Acoustofluidic separation enables early diagnosis of traumatic brain injury based on circulating exosomes显示文摘Traumatic brain injury(TBI)is a global cause of morbidity and mortality.Initial management and risk stratification of patients with TBI is made difficult by the relative insensitivity of screening radiographic studies as well as by the absence of a widely available,noninvasive diagnostic biomarker.In particular,a blood-based biomarker assay could provide a quick and minimally invasive process to stratify risk and guide early management strategies in patients with mild TBI(mTBI).Analysis of circulating exosomes allows the potential for rapid and specific identification of tissue injury.By applying acoustofluidic exosome separation—which uses a combination of microfluidics and acoustics to separate bioparticles based on differences in size and acoustic properties—we successfully isolated exosomes from plasma samples obtained from mice after TBI.Acoustofluidic isolation eliminated interference from other blood components,making it possible to detect exosomal biomarkers for TBI via flow cytometry.Flow cytometry analysis indicated that exosomal biomarkers for TBI increase in the first 24 h following head trauma,indicating the potential of using circulating exosomes for the rapid diagnosis of TBI.Elevated levels of TBI biomarkers were only detected in the samples separated via acoustofluidics;no changes were observed in the analysis of the raw plasma sample.This finding demonstrated the necessity of sample purification prior to exosomal biomarker analysis.Since acoustofluidic exosome separation can easily be integrated with downstream analysis methods,it shows great potential for improving early diagnosis and treatment decisions associated with TBI. | Zeyu Wang Haichen Wang Ryan Becker Joseph Rufo Shujie Yang Brian E.Mace Mengxi Wu Jun Zou Daniel T.Laskowitz Tony Jun Huang | 2021 | Microsystems & Nanoengineering2021,7,2: | 3 |
| 5 | Autonomic responses to blast overpressure can be elicited by exclusively exposing the ear in rats显示文摘Blast overpressure has become an increasing cause of brain injuries in both military and civilian populations. Though blast's direct effects on the cochlea and vestibular organs are active areas of study, little attention has been given to the ear's contribution to the overall spectrum of blast injury. Acute autonomic responses to blast exposure, including bradycardia and hypotension, can cause hypoxia and contribute to blast-induced neurotrauma. Existing literature suggests that these autonomic responses are elicited through blast impacting the thorax and lungs. We hypothesize that the unprotected ear also provides a vulnerable locus for blast to cause autonomic responses. We designed a blast generator that delivers controlled overpressure waves into the ear canal without impacting surrounding tissues in order to study the ear's specific contribution to blast injury. Anesthetized adult rats' left ears were exposed to a single blast wave ranging from 0 to 110 PSI(0-758 kPa). Blast exposed rats exhibited decreased heart rates and blood pressures with increased blast intensity, similar to results gathered using shock tubes and whole-body exposure in the literature. While rats exposed to blasts below 50 PSI(345 kPa) exhibited increased respiratory rate with increased blast intensity, some rats exposed to blasts higher than 50 PSI(345 kPa) stopped breathing immediately and ultimately died. These autonomic responses were significantly reduced in vagally denervated rats, again similar to whole-body exposure literature.
