| 2 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 4 | Single-cell atlas reveals a distinct immune profile fostered by T cell-B cell crosstalk in triple negative breast cancer显示文摘Background:Characterizing the unique immune microenvironment of each tumor is of great importance for better predicting prognosis and guiding cancer immunotherapy.However,the unique features of the immune microenvironment of triple negative breast cancer(TNBC)compared with other subtypes of breast cancer remain elusive.Therefore,we aimed to depict and compare the immune landscape among TNBC,human epidermal growth factor receptor 2-positive(HER2^(+))breast cancer,and luminal-like breast cancer.Methods:Single-cell RNA sequencing(scRNA-seq)was performed on CD45^(+)immune cells isolated from human normal breast tissues and primary breast tumors of various subtypes.By analyzing the scRNA-seq data,immune cell clusters were identified and their proportions as well as transcriptome features were compared among TNBC,human HER2^(+)breast cancer,and luminal-like breast cancer.Pseudotime and cell-cell communication analyses were also conducted to characterize the immune microenvironment.Results:ScRNA-seq data of 117,958 immune cells were obtained and 31 immune clusters were identified.A unique immunosuppressive microenvironment in TNBC was decoded as compared to that in HER2^(+)or luminal-like breast cancer,which was characterized by higher proportions of regulatory T cells(Tregs)and exhausted CD8+T cells and accompanied by more abundant plasma cells.Tregs and exhausted CD8+T cells in TNBC exhibited increased immunosuppression signature and dysfunctional scores.Pseudotime analyses showed that B cells tended to differentiate to plasma cells in TNBC.Cell-cell communication analyses indicated that these unique features are fostered by the diversified T cell-B cell crosstalk in TNBC.Based on the T cell-B cell crosstalk,a prognostic signaturewas established that could effectively predict the prognosis status for patients with TNBC.Additionally,it was found that TNBC had a higher proportion of cytotoxic natural killer(NK)cells,whereas HER2^(+)or luminal-like breast cancer lost this feature,suggesting thatHER2^(+)or luminal-like breast cancer,but not TNBC,may benefit from NK-based immunotherapy.Conclusions:This study identified a distinct immune feature fostered by T cell-B cell crosstalk in TNBC,which provides better prognostic information and effective therapeutic targets for breast cancer. | Shuning Ding Niu Qiao Qingchen Zhu Yiwei Tong Shengyue Wang Xiaosong Chen Qiang Tian Yichuan Xiao Kunwei Shen | 2023 | Cancer Communications2023,43,6: | 2 |
| 5 | Transfer of β-hydroxy-β-methylbutyrate from sows to their offspring and its impact on muscle fiber type transformation and performance in pigs显示文摘Background: Previous studies suggested that supplementation of lactating sows with β-hydroxy-β-methylbutyrate(HMB) could improve the performance of weaning pigs, but there were little information in the muscle fiber type transformation of the offspring and the subsequent performance in pigs from weaning through finishing in response to maternal HMB consumption. The purpose of this study was to determine the effect of supplementing lactating sows with HMB on skeletal muscle fiber type transformation and growth of the offspring during d 28 and180 after birth. A total of 20 sows according to their body weight were divided into the control(CON, n = 10) or HMB groups(HMB, n = 10). Sows in the HMB group were supplemented with β-hydroxy-β-methylbutyrate calcium(HMB-Ca) 2 g/kg feed during d 1 to 27 of lactation. After weaning, 48 mixed sex piglets were blocked by sow treatment and fed standard diets for post-weaning, growing, finishing periods. Growth performance was recorded during d 28 to 180 after birth. Pigs were slaughtered on d 28(n = 6/treatment) and 180(n = 6/treatment) postnatal and the longissimus dorsi(LD) was collected, respectively.Results: The HMB-fed sows during lactation showed increased HMB concentration(P < 0.05) in milk and LD of weaning piglets(P < 0.05). | 8 | Mechanism of pore pressure variation in multiple coal reservoirs,western Guizhou region,South China显示文摘Pore pressure is an important parameter in coalbed methane(CBM)exploration and development;however,the distribution pattern and mechanism for pore pressure differences in the Upper Permian CBM reservoirs are poorly understood in the western Guizhou region of South China.In this study,lateral and vertical variations and mechanisms for pore pressure differences are analyzed based on 126 injection-falloff and in-situ stress well test data measured in Permian coal reservoirs.Generally,based on the pore pressure gradient and coefficient in coal reservoirs of the western Guizhou region,five zones can be delineated laterally:the mining areas of Zhina,northem Liupanshui,northern Guizhou,northwestern Guizhou and southern Liupanshui.Vertically,there are two main typical patterns:i)the pore pressure gradient(or coefficient)is nearly unchanged in different coal reservoirs,and ii)the pore pressure gradient(or coefficient)has cyclic variations in a borehole profile with multiple coal seams,which suggests the existence of a'superimposed CBM system'.The mechanism analysis indicates that coal permeability,thermal evolution stage and hydrocarbon generation contribute little to pore pressure differences in coal reservoirs in the western Guizhou region.The present-day in-situ stress field,basement structure and tectonic activity may be the dominant factors affecting lateral pore pressure differences.The sealing capacity of caprocks and the present-day in-situ stress field are significant para-meters causing vertical pore pressure differences in coal reservoirs.These results are expected to provide new geological references for CBM exploration and develop-ment in the western Guizhou region. | Wei JU Zhaobiao YANG Yulin SHEN Hui YANG Geoff WANG Xiaoli ZHANG Shengyu WANG | 2021 | Frontiers of Earth Science2021,15,4: | 0 |

| 10 | Shell-formation mediated surface composition of uniform two-component microparticles fabricated by micro-fluidic spray drying: Effect of component size and solubility显示文摘This work is aimed to study the effects of component size and solubility on the surface composition of spray dried(SD)uniform two-component particles fabricated by micro-fluidic spray dryer.Various precursor liquid consisting of small molecular of methionine(Met,33 g/L)or lysine(Lys,739 g/L)and large-sized silica(12 nm)were prepared by adjusting the mass ratio of components.X-ray energy disper-sive results showed that the respective enrichment degree(De)of Met and Lys on the surface of SD-M1S9 and-L1S9 prepared at 150 C were 182±9% and 125±14%.The De of hydrophobic Met for SD-M1S1 and-M9S1 were 46±9% and 4±2%,respectively,whereas relative hydrophilic Lys mainly distributed internal of the particle meanwhile the De of silica on the surface for SD-L1S1 and-L9S1 were 17±4%and 12±1%,respectively.Drying temperature(120 and 180℃)showed more apparent effect on the De of amino acid for the particles of less amino acid.The possible formation mechanism of surface composition and the surface composition impact on the wettability of particles were explored.These results provide new guidance for manufacturing functional SD powders with various components. | Xiaoqian Ma Shen Yan Shengyu Zhang Quanyi Yin Xiaodong Chen Winston Duo Wu | 2022 | Particuology2022,20,8: | 0 |

| 14 | Designing TiO_(2)/Au/Prussian blue heterostructures nanorod arrays for ultra-stable cycle and ultra-fast response electrochromism显示文摘Prussian blue(PB)is an anodic coloring candidate in the wide area of electrochromic(EC)applications.However,the co-influence of weak adhesion and low electrical conductivity leads to the poor stability and slow switching speed.To tackle this bottleneck,a novel TiO_(2)/Au/PB nanorod array is designed through hydrothermal and electrodeposition approaches on fluorine-doped tin oxide(FTO)glass.Such a designed ternary array structure could not only increase reactive site and conductivity,but also improve ion storage capacity and promote charge transfer,attributed to the synergistic effect of TiO_(2)/Au/PB core–shell heterostructure and the localized surface plasmon resonance(LSPR)effect of Au nanoparticles.Besides,density functional theory(DFT)calculation confirms the strong interaction between rutile TiO_(2)and FTO substrate,which contributes to the improvement of EC cycle stability.Benefiting from these effects,the TiO_(2)/Au/PB film shows a fast coloration/bleaching response of 1.08/2.01 s(2.17/4.48 s,PB film)and ultra-stable EC performance of 86.8%after 20,000 cycles(50%after 600 cycles,PB film).Furthermore,the high-intensity light source can be shot clearly by the designed and assembled EC iris device(ECID)with TiO_(2)/Au/PB film as an EC layer,while the photograph without an ECID is blurry,confirming the feasibility of the material in portable digital products. | Miao Xu Kang Li Shen Wang Shengyu Zhou Hulin Zhang Hongbo Xu Jiupeng Zhao Yao Li | 2023 | Nano Research2023,16,2: | 0 |
| 15 | Environmental hypoxia induces apoptosis in large yellow croaker Larimichthys crocea via both intrinsic and extrinsic pathways显示文摘Hypoxia has become an unfavorable factor affecting the sustainable development of the large yellow croaker Larimichthys crocea,an economically important mariculture fish in China.Apoptosis is a consequence of hypoxia on fish.However,the effects of hypoxia stress on apoptosis in L.crocea remain largely unknown.We investigated the effect of environmental hypoxia on apoptosis in L.crocea.Results show that hypoxia induced apoptosis in L.crocea both in vivo and in vitro.The mitochondrial membrane potential was significantly reduced in large yellow croaker fry(LYCF)cells.The expression levels of Bcell lymphoma/leukemia-2(Bcl-2)m RNA and protein were also significantly decreased in the liver and LYCF cells during 96 h and 48 h of hypoxia stress,respectively,whereas the expression level of Bcl-2 associated X(Bax)mRNA,Casp3 mRNA,and activity of caspase-3/7/9 were significantly increased,indicating that hypoxia induced caspase-dependent intrinsic apoptosis in L.crocea.The expression level of the apoptosis-inducing factor(AIF)protein was significantly increased in the liver and LYCF cells.The level of AIF protein was significantly decreased in the cytoplasm but increased in the nuclei of L.crocea,demonstrating that hypoxia induced the AIF-mediated caspase-independent intrinsic apoptosis.In addition,the activity of caspase-8 was significantly increased,indicating that hypoxia stress induced extrinsic apoptosis in L.crocea.Therefore,hypoxia induced apoptosis in L.crocea through both the intrinsic and extrinsic pathways.The present study accumulated basic biological information to help elucidate the mechanism of hypoxia response in marine fish. | Shengyu LUO Cheng LIU Xinming GAO Jingqian WANG Yibo ZHANG Jie DING Congcong HOU Junquan ZHU Bao LOU Weiliang SHEN Xiongfei WU Chundan ZHANG | 2023 | Journal of Oceanology and Limnology2023,41,6: | 0 |
