2Mechanism of Kinect-based virtual reality training for motor functional recovery of upper limbs after subacute stroke显示文摘The Kinect-based virtual reality system for the Xbox 360 enables users to control and interact with the game console without the need to touch a game controller,and provides rehabilitation training for stroke patients with lower limb dysfunctions.However,the underlying mechanism remains unclear.In this study,18 healthy subjects and five patients after subacute stroke were included.The five patients were scanned using functional MRI prior to training,3 weeks after training and at a12-week follow-up,and then compared with healthy subjects.The Fugl-Meyer Assessment and Wolf Motor Function Test scores of the hemiplegic upper limbs of stroke patients were significantly increased 3 weeks after training and at the 12-week follow-up.Functional MRI results showed that contralateral primary sensorimotor cortex was activated after Kinect-based virtual reality training in the stroke patients compared with the healthy subjects.Contralateral primary sensorimotor cortex,the bilateral supplementary motor area and the ipsilateral cerebellum were also activated during hand-clenching in all 18 healthy subjects.Our findings indicate that Kinect-based virtual reality training could promote the recovery of upper limb motor function in subacute stroke patients,and brain reorganization by Kinect-based virtual reality training may be linked to the contralateral sensorimotor cortex.Xiao Bao Yurong Mao Qiang Lin Yunhai Qiu Shaozhen Chen Le Li Ryan S.Cates Shufeng Zhou Dongfeng Huang 2013Neural Regeneration Research2013,8,31:15
3The Exhumation History of North Qaidam Thrust Belt Constrained by Apatite Fission Track Thermochronology:Implication for the Evolution of the Tibetan Plateau显示文摘Determining the spatio-temporal distribution of the deformation tied to the India-Eurasian convergence and the impact of pre-existing weaknesses on the Cenozoic crustal deformation is significant for understanding how the convergence between India and Eurasia contributed to the development of the Tibetan Plateau. The exhumation history of the northeastern Tibetan Plateau was addressed in this research using a new apatite fission track(AFT) study in the North Qaidam thrust belt(NQTB). Three granite samples collected from the Qaidam Shan pluton in the north tied to the Qaidam Shan thrust, with AFT ages clustering in the Eocene to Miocene. The other thirteen samples obtained from the Luliang Shan and Yuka plutons in the south related to the Luliang Shan thrust and they have showed predominantly the Cretaceous AFT ages. Related thermal history modeling based on grain ages and track lengths indicates rapid cooling events during the Eocene-early Oligocene and since late Miocene within the Qaidam Shan, in contrast to those in the Cretaceous and since the Oligocene-Miocene in the Luliang Shan and Yuka region. The results, combined with published the Cretaceous thermochronological ages in the Qaidam Shan region, suggest that the NQTB had undergo rapid exhumation during the accretions along the southern Asian Andean-type margin prior to the India-Eurasian collision. The Cenozoic deformation initially took place in the North Qaidam thrust belt by the Eocene, which is consistent with the recent claim that the deformation of the northeastern Tibetan Plateau initiated in the Eocene as a response to continental collision between India and Eurasia. 5Conceptual models for fracturing in fault related folds显示文摘Fault related folds and fractures, which always improve reservoirs and trap capacity, especially in the oil and gas fields in western China, are of extreme importance for oil and gas exploration. Based on four assumptions and fault related fold theory, we propose four conceptual models for fracturing in fault related folds, i.e., a simple-step parallel fault bend anticline, a simple-step parallel fault propagation anticline, a multiple-bend fault-bend fold and a break-forward imbricate. Fracture conditions depend on structural evolution and specific site conditions. 7Macrophage-mediated tumor-targeted delivery of engineered Salmonella typhimurium VNP20009 in anti-PD1 therapy against melanoma显示文摘Bacterial antitumor therapy has great application potential given its unique characteristics,including genetic manipulation, tumor targeting specificity and immune system modulation. However,the nonnegligible side effects and limited efficacy of clinical treatment limit their biomedical applications. Engineered bacteria for therapeutic applications ideally need to avoid their accumulation in normal organs and possess potent antitumor activity. Here, we show that macrophage-mediated tumor-targeted delivery of Salmonella typhimurium VNP20009 can effectively reduce the toxicity caused by administrating VNP20009 alone in a melanoma mouse model. This benefits from tumor-induced chemotaxis for macrophages combined with their slow release of loaded strains. Inspired by changes in the tumor microenvironment, including a decrease in intratumoral dysfunctional CD8+T cells and an increase in PDL1 on the tumor cell surface, macrophages were loaded with the engineered strain VNP-PD1nb, which can express and secrete anti-PD1 nanoantibodies after they are released from macrophages. This novel triple-combined immunotherapy significantly inhibited melanoma tumors by reactivating the tumor microenvironment by increasing immune cell infiltration, inhibiting tumor cell proliferation, remodeling TAMs to an M1-like phenotype and prominently activating CD8+T cells. 14Continuous-wave terahertz in-line holographic diffraction tomography with the scattering fields reconstructed by a physics-enhanced deep neural network显示文摘Diffraction tomography is a promising,quantitative,and nondestructive three-dimensional(3D)imaging method that enables us to obtain the complex refractive index distribution of a sample.The acquisition of the scattered fields under the different illumination angles is a key issue,where the complex scattered fields need to be retrieved.Presently,in order to develop terahertz(THz)diffraction tomography,the advanced acquisition of the scattered fields is desired.In this paper,a THz in-line digital holographic diffraction tomography(THz-IDHDT)is proposed with an extremely compact optical configuration and implemented for the first time,to the best of our knowledge.A learning-based phase retrieval algorithm by combining the physical model and the convolution neural networks,named the physics-enhanced deep neural network(PhysenNet),is applied to reconstruct the THz in-line digital hologram,and obtain the complex amplitude distribution of the sample with high fidelity.The advantages of the PhysenNet are that there is no need for pretraining by using a large set of labeled data,and it can also work for thick samples.Experimentally with a continuous-wave THz laser,the PhysenNet is first demonstrated by using the thin samples and exhibits superiority in terms of imaging quality.More importantly,with regard to the thick samples,PhysenNet still works well,and can offer 2D complex scattered fields for diffraction tomography.Furthermore,the 3D refractive index maps of two types of foam sphere samples are successfully reconstructed by the proposed method.For a single foam sphere,the relative error of the average refractive index value is only 0.17%,compared to the commercial THz time-domain spectroscopy system.This demonstrates the feasibility and high accuracy of the THz-IDHDT,and the idea can be applied to other wavebands as well.XIAOYU JIN JIE ZHAO DAYONG WANG JOHN JHEALY LU RONG YUNXIN WANG SHUFENG LIN 2023Photonics Research2023,11,12:0

17Microfluidic chip of concentration gradient and fluid shear stress on a single cell level显示文摘Concentration gradient and fluid shear stress(FSS)for cell microenvironment were investigated through microfluidic technology.The Darcy–Weisbach equation combined with computational fluid dynamics modeling was exploited to design the microfluidic chip,and the FSS distribution on the cell model with varying micro-channels(triangular,conical,and elliptical).The diffusion with the incompressible laminar flow model by solving the time-dependent diffusion–convection equation was applied to simulate the gradient profiles of concentration in the micro-channels.For the study of single cell in-depth,the FSS was investigated by the usage of polystyrene particles and the concentration diffusion distribution was studied by the usage of different colors of dyes.A successful agreement between model simulations and experimental data was obtained.Finally,based on the established method,the communication between individual cells was envisaged and modeled.The developed method provides valuable insights and allows to continuously improve the design of microfluidic devices for the study of single cell,the occurrence and development of tumors,and therapeutic applications.Xuexia Lin Jianlong Su Shufeng Zhou 2022Chinese Chemical Letters2022,33,6:0
18Prevalence of familial hypercholesterolemia and its association with coronary artery disease:A Chinese cohort study显示文摘Background:Familial hypercholesterolemia(FH)is underrecognized,and its association with coronary artery disease(CAD)remains limited,especially in China.We aimed to investigate the prevalence of FH and its relationship with CAD in a large Chinese cohort.Methods:FH was defined using the Make Early Diagnosis to Prevent Early Death(MEDPED)criteria.The crude and age-sex standardized prevalence of FH were calculated based on surveys of the Prediction for Atherosclerotic Cardiovascular Disease Risk in China(China-PAR)project during 2007−2008.The associations of FH with incident CAD and its major subtypes were estimated with the cohort-stratified multivariate Cox proportional hazard models based on the data from the baseline to the last follow-up(2018−2020).Results:Among 98,885 included participants,190 participants were defined as FH.Crude and age−sex standardized prevalence and 95%confidence interval(CI)of FH were 0.19%(0.17%-0.22%)and 0.13%(0.10%-0.16%),respectively.The prevalence varied across age groups and peaked in the group of 60-<70 years(0.28%),and the peak prevalence(0.18%)in males was earlier,yet lower than the peak crude prevalence in females(0.41%).During a mean follow-up of 10.7 years,2493 cases of incident CAD were identified.After multivariate adjustment,FH patients had a 2.03-fold greater risk of developing CAD compared to non-FH participants.Conclusions:The prevalence of FH was estimated to be 0.19%in the participants,and it was associated with an elevated risk of incident CAD.Our study suggests that early screening of FH has certain public health significance for the prevention of CAD.Xiapikatijiang Aihaiti Shufeng Chen Jianxin Li Zhennan Lin Qingmei Cui Xue Xia Fangchao Liu Chong Shen Dongsheng Hu Keyong Huang Yingxin Zhao Fanghong Lu Xiaoqing Liu Jie Cao Ling Yu Ying Li Huan Zhang Zhenyan Fu Liancheng Zhao Jianfeng Huang Dongfeng Gu Xiangfeng Lu 2023Chronic Diseases and Translational Medicine2023,9,2:0

20Quantitative proteomics provides insight into the response of the marine dinoflagellate Prorocentrum donghaiense to changes in ambient phosphorus显示文摘Dinofl agellates are the major causative agents of harmful algal blooms in the global ocean and they usually form blooms under conditions of very low dissolved inorganic phosphorus(DIP).However,the mechanisms underpinning the dinofl agellate blooms remain unclear.Here,we quantitatively compared protein expression profi les of a marine dinofl agellate,Prorocentrum donghaiense,grown in inorganic P-replete,P-defi cient,and DIP-and dissolved organic phosphorus(DOP)-resupplied conditions by employing a Tandem Mass Tag(TMT)-based quantitative proteomic approach.Proteins involved in intracellular P reallocation,organic P,and non-P lipid utilization were up-regulated under the P-defi cient condition,while inorganic phosphate transporters varied insignifi cantly.In response to the P resupplementation,nitrogen metabolism,ribosome,porphyrin,and chlorophyll metabolism were up-regulated,while lysosome,and starch and sucrose metabolism were down-regulated.Notably,photosynthesis was up-regulated and secondary metabolism was down-regulated only in the DIP-resupplied cells,whereas amino acid metabolism and vitamin B6 metabolism were up-regulated in the DOP-resupplied cells,indicating diff erential response mechanisms of P.donghaiense to DIP or DOP resupplementation.Our results indicated that P.donghaiense initiated multiple strategies in response to an ambient inorganic P-defi ciency,and its efficient DOP assimilation by providing both P and carbon sources might be a key factor driving bloom formations of P.donghaiense in a low DIP environment.Shufeng ZHANG Chunjuan YUAN Ying CHEN Lin LIN Dazhi WANG 2022Journal of Oceanology and Limnology2022,40,2:0
