| 2 | Apelin-13 inhibits apoptosis and excessive autophagy in cerebral ischemia/reperfusion injury显示文摘Apelin-13 is a novel endogenous ligand for an angiotensin-like orphan G-protein coupled receptor,and it may be neuroprotective against cerebral ischemia injury.However,the precise mechanisms of the effects of apelin-13 remain to be elucidated.To investigate the effects of apelin-13 on apoptosis and autophagy in models of cerebral ischemia/reperfusion injury,a rat model was established by middle cerebral artery occlusion.Apelin-13(50μg/kg)was injected into the right ventricle as a treatment.In addition,an SH-SY5Y cell model was established by oxygen-glucose deprivation/reperfusion,with cells first cultured in sugar-free medium with 95%N2 and 5%CO2 for 4 hours and then cultured in a normal environment with sugar-containing medium for 5 hours.This SH-SY5Y cell model was treated with 10-7 M apelin-13 for 5 hours.Results showed that apelin-13 protected against cerebral ischemia/reperfusion injury.Apelin-13 treatment alleviated neuronal apoptosis by increasing the ratio of Bcl-2/Bax and significantly decreasing cleaved caspase-3 expression.In addition,apelin-13 significantly inhibited excessive autophagy by regulating the expression of LC3B,p62,and Beclin1.Furthermore,the expression of Bcl-2 and the phosphatidylinositol-3-kinase(PI3K)/Akt/mammalian target of rapamycin(mTOR)pathway was markedly increased.Both LY294002(20μM)and rapamycin(500 nM),which are inhibitors of the PI3K/Akt/mTOR pathway,significantly attenuated the inhibition of autophagy and apoptosis caused by apelin-13.In conclusion,the findings of the present study suggest that Bcl-2 upregulation and mTOR signaling pathway activation lead to the inhibition of apoptosis and excessive autophagy.These effects are involved in apelin-13-induced neuroprotection against cerebral ischemia/reperfusion injury,both in vivo and in vitro.The study was approved by the Animal Ethical and Welfare Committee of Jining Medical University,China(approval No.2018-JS-001)in February 2018. | Zi-Qi Shao Shan-Shan Dou Jun-Ge Zhu Hui-Qing Wang Chun-Mei Wang Bao-Hua Cheng Bo Bai | 2021 | Neural Regeneration Research2021,16,6: | 35 |

| 7 | Ecosystem vulnerability of China under B2 climate scenario in the 21st century显示文摘This paper applies climate change scenarios in China based on the SRES assumptions with the help of RCMs projections by PRECIS (providing regional climates for impacts studies) system introduced to China from the Hadley Centre for Climate Prediction and Research at a high-resolution (50 km×50 km) over China. This research focuses on B2 scenario of SRES. A biogeochemical model 'Atmosphere Vegetation Integrated Model (AVIM2)' was applied to simulating ecosystem status in the 21st century. Then vulnerability of ecosystems was assessed based on a set of index of mainly net primary production (NPP) of vegetation. Results show that climate change would affect ecosystem of China severely and there would be a worse trend with the lapse of time. The regions where having vulnerable ecological background would have heavier impacts while some regions with better ecological background would also receive serious impacts. Extreme climate even would bring about worse impact on the ecosystems. Open shrub and desert steppe would be the two most affected types. When the extreme events happen, vulnerable ecosystem would extend to part of defoliate broad-leaved forest, woody grassland and evergreen conifer forest. Climate change would not always be negative. It could be of some benefit to cold region during the near-term. | 10 | Prophylaxis of chronic kidney disease after liver transplantation-experience from west China显示文摘AIM:To evaluate the prophylaxis of chronic kidney disease (CKD) after liver transplantation (LT) with low-dose calcineurin inhibitor (CNI) and mycophenolate mofetil (MMF).METHODS:From March 1999 to December 2009,a total of 572 patients (478 males and 94 females) underwent LT enrolled in the study.Initial immunosuppression was by triple-drug regimens that included a CNI,MMF,and prednisone.The initial dose of CNI was 0.05-0.10mg/kg per day for tacrolimus (TAC) and 5-10mg/kg per d for cyclosporine A (CSA) respectively,and was gradually reduced based on a stable graft function.The serum trough level of CNI was 6-8 ng/mL for TAC and 120-150 ng/mL for CSA 3-mo post-operation,4-6 ng/mL for TAC and 80-120 ng/mL for CSA 1-year after transplantation was expected with stable liver function.MMF was personalized between 1.0-1.5 g/d.Glomerular filtration rate (GFR) was estimated by an abbreviated Modification of Diet in Renal Disease formula.Risk factors of CKD were examined by univariate and multivariate logistic regression.RESULTS:With a definition of GFR<60 mL/min per 1.73 m 2,the incidence of CKD was 17.3% 5-year after LT.There were 68.3% (293 of 429 cases) patients managed to control their TAC trough concentrations within 8 ng/mL and 58.0% (83 of 143 cases) patients' CSA trough concentrations within 150 ng/mL.Of the 450 recipients followed-up over 1 year,55.5% (183 of 330 cases) of which were treated with TAC had a trough concentration ≤ 6 ng/mL while 65.8% (79 of 120 cases) of which were treated with CSA had a concentration ≤ 120 ng/mL.The incidence of CKD in the groups of lower CNI trough concentrations was significantly lower than the groups with CNI concentrations above the ideal range.Patients with CKD had much higher CNI trough concentrations than that of patients without CKD.MMF was adopted in 359 patients (62.8%).Patients administrated with MMF had a relatively low CNI trough concentrations but with no significant difference.The graft function remained stable during follow-up.No difference was found between different groups of CNI trough concentrations.Pre-LT renal dysfunction,ages,acute kidney injury,high blood trough concentrations of CNI in 3 mo (TAC>8 ng/mL,CSA>150 ng/mL) and hypertension after operation were associated with CKD progression,while male gender and adoption of MMF were protection factors.CONCLUSION:Low dose of CNI combined with MMF managed to prevent CKD after LT with stable graft function. | Zhen-Yong Shao Lu-Nan Yan Wen-Tao Wang Bo Li Tian-Fu Wen Jia-Yin Yang Ming-Qing Xu Ji-Chun Zhao Yong-Gang Wei | 2012 | World Journal of Gastroenterology2012,18,9: | 10 |
| 11 | Enhancement of strength and electrical conductivity for a dilute Al-Sc-Zr alloy via heat treatments and cold drawing显示文摘Developing heat-resistant conductors with high strength and high electrical conductivity is a key issue in the electrical conductor industries, as the ever-increasing power transmission poses higher requirement on the thermal stability of electrical conductor wires. Dilute Al-Sc-Zr alloys are considered as promising candidates due to the excellent heat resistance and high electrical conductivity, but the low strength always limits their application on electrical wires. Yet, few efforts on process design have been made in dilute Al-Sc-Zr alloys to enhance the strength. Here, various kinds of processing paths via combination of cold drawing, ageing and/or annealing were conducted to improve the strength and electrical conductivity of a dilute Al-Sc-Zr alloy. Results show that enhanced strength and electrical conductivity were obtained after cold drawing + ageing or pre-ageing + cold drawing + annealing treatments processes.Optimal properties(194 MPa in ultimate tensile strength and 61% IACS in electrical conductivity) were obtained through cold drawing followed by ageing. Microstructure evolution which affects strength and electrical conductivity was systematically investigated using TEM and 3 DAP. The enhanced strength was mainly attributed to the suitable interactions between strain strengthening and precipitation strengthening. The enhancement in electrical conductivity was caused by precipitation of solute atoms and recovery of defects. | 14 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 15 | Development and validation of an ocean wave retrieval algorithm for VV-polarization Sentinel-1 SAR data显示文摘The purpose is to study the accuracy of ocean wave parameters retrieved from C-band VV-polarization Sentinel-1Synthetic Aperture Radar(SAR) images, including both significant wave height(SWH) and mean wave period(MWP), which are both calculated from a SAR-derived wave spectrum. The wind direction from in situ buoys is used and then the wind speed is retrieved by using a new C-band geophysical model function(GMF) model,denoted as C-SARMOD. Continuously, an algorithm parameterized first-guess spectra method(PFSM) is employed to retrieve the SWH and the MWP by using the SAR-derived wind speed. Forty–five VV-polarization Sentinel-1 SAR images are collected, which cover the in situ buoys around US coastal waters. A total of 52 subscenes are selected from those images. The retrieval results are compared with the measurements from in situ buoys. The comparison performs good for a wind retrieval, showing a 1.6 m/s standard deviation(STD) of the wind speed, while a 0.54 m STD of the SWH and a 2.14 s STD of the MWP are exhibited with an acceptable error.Additional 50 images taken in China's seas were also implemented by using the algorithm PFSM, showing a 0.67 m STD of the SWH and a 2.21 s STD of the MWP compared with European Centre for Medium-range Weather Forecasts(ECMWF) reanalysis grids wave data. | 16 | Real-time Microwave Exposure Induces Calcium Efflux in Primary Hippocampal Neurons and Primary Cardiomyocytes显示文摘Objective To detect the effects of microwave on calcium levels in primary hippocampal neurons and primary cardiomyocytes by the real-time microwave exposure combined with laser scanning confocal microscopy. Methods The primary hippocampal neurons and primary cardiomyocytes were cultured and labeled with probes, including Fluo-4 AM, Mag-Fluo-AM, and Rhod-2, to reflect the levels of whole calcium [Ca^(2+)], endoplasmic reticulum calcium [Ca^(2+)]ER, and mitochondrial calcium [Ca^(2+)]MIT, respectively. Then, the cells were exposed to a pulsed microwave of 2.856 GHz with specific absorption rate(SAR) values of 0, 4, and 40 W/kg for 6 min to observe the changes in calcium levels. Results The results showed that the 4 and 40 W/kg microwave radiation caused a significant decrease in the levels of [Ca^(2+)], [Ca^(2+)]ER, and [Ca^(2+)]MIT in primary hippocampal neurons. In the primary cardiomyocytes, only the 40 W/kg microwave radiation caused the decrease in the levels of [Ca^(2+)], [Ca^(2+)]ER, and [Ca^(2+)]MIT. Primary hippocampal neurons were more sensitive to microwave exposure than primary cardiomyocytes. The mitochondria were more sensitive to microwave exposure than the endoplasmic reticulum. Conclusion The calcium efflux was occurred during microwave exposure in primary hippocampal neurons and primary cardiomyocytes. | 18 | Accumulation characteristic of protein bodies in different regions of wheat endosperm under drought stress显示文摘The structural characteristics of protein body accumulation in different endosperm regions of hard wheat cultivar(XM33) and soft wheat cultivar(NM13) under drought stress were investigated. Drought stress treatment was implemented from plant regreening to the caryopsis mature stage. Microscope images of endosperm cells were obtained using resin semithin slice technology to observe the distribution and relative area of protein body(PB). Compared with NM13, relative PB area of XM33 was significantly higher in sub-aleurone endosperm region. The amount of accumulation, including the size and relative area of PB, in two wheat cultivars was higher in sub-aleurone region than that in central region at 18 days post anthesis(DPA). Drought stress significantly enhanced the sizes and relative areas of PBs in the dorsal and abdominal endosperms in two wheat cultivars. Particularly for dorsal endosperm, drought stress enhanced the relative PB area at 18 DPA and NM13(5.0% vs. 6.73%) showed less enhancement than XM33(5.49% vs. 8.96%). However, NM13(9.58% vs. 12.02%) showed greater enhancement than XM33(10.25% vs. 11.7%) at 28 DPA. The protein content in the dorsal and abdominal endosperms of the two wheat cultivars decreased at 12 DPA and then increased until 38 DPA. Drought stress significantly increased the protein contents in the two main regions. From 12 to 38 DPA, the amount of PB accumulation and the protein content were higher in XM33 than those in NM13. | 20 | Therapeutic effects of mesenchymal stem cells administered at later phase of recurrent experimental autoimmune uveitis显示文摘AIM:To test the therapeutic effects of delayed treatment of mesenchymal stem cells(MSCs) in recurrent experimental autoimmune uveitis(r EAU).METHODS: The efficacy of different regimens of MSC administration in r EAU were tested by evaluation of clinical and pathological intraocular inflammation,as well as retinal structural and functional integrity using optical coherence tomography(OCT) and electroretinogram(ERG).The retinal sections were also immunostained with antibodies to glial fibrillary acidic protein(GFAP)and rhodopsin(RHO).
