1Chemical genomics reveals inhibition of breast cancer lung metastasis by Ponatinib via c-Jun显示文摘Metastasis is the leading cause of human cancer deaths.Unfortunately,no approved drugs are available for antimetastatic treatment.In our study,high-throughput sequencing-based high-throughput screening(HTS^2)and a breast cancer lung metastasis(BCLM)-associated gene signature were combined to discover anti-metastatic drugs.After screening of thousands of compounds,we identified Ponatinib as a BCLM inhibitor.Ponatinib significantly inhibited the migration and mammosphere formation of breast cancer cells in vitro and blocked BCLM in multiple mouse models.Mechanistically,Ponatinib represses the expression of BCLM-associated genes mainly through the ERK/c-Jun signaling pathway by inhibiting the transcription of JUN and accelerating the degradation of c-Jun protein.Notably,JUN expression levels were positively correlated with BCLM-associated gene expression and lung metastases in breast cancer patients.Collectively,we established a novel approach for the discovery of anti-metastatic drugs,identified Ponatinib as a new drug to inhibit BCLM and revealed c-Jun as a crucial factor and potential drug target for BCLM.Our study may facilitate the therapeutic treatment of BCLM as well as other metastases.Wei Shao Shasha Li Lu Li Kequan Lin Xinhong Liu Haiyan Wang Huili Wang Dong Wang 2019Protein & Cell2019,10,3:7
2N_2O and NH_3 emissions from a bioreactor landfill operated under limited aerobic degradation conditions显示文摘The combination of leachate recirculation and aeration to landfill may be an efficient way for in-situ nitrogen removal.However,nitrogenous substances contained in the landfill layer are concomitantly transformed into N2O and NH3,leading to increased emissions into the atmosphere.In the present study,the emissions of N2O and NH3 were measured under conditions of fresh or partially stabilized refuse with or without leachate recirculation or intermittent aeration.The results showed that the largest N2O emission(12.4 mg-N/L of the column) was observed in the aerated column loaded with partially stabilized refuse and recycled with the leachate of low C/N ratio;while less than 0.33 mg-N/L of the column was produced in the other columns.N2O production was positively correlated with the prolonged aerobic time and negatively related with the C/N ratio in the recycled leachate.NH3 volatilization increased with enhanced gas flow and concentration of free ammonia in the leachate,and the highest cumulative volatilization quantity was 1.7 mg-N/L of the column.Pinjing He Na Yang Huili Gu Hua Zhang Liming Shao 2011Journal of Environmental Sciences2011,23,6:5
3Silk materials for medical, electronic and optical applications显示文摘Silk derived from the silkworm Bombyx mori is among the most important fibrous protein biomaterials due to large-scale production from natural sources, excellent biocompatibility, unique mechanical properties and controllable degradation. Silk fibroin can be processed into a variety of formats to match different applications, such as tissue engineering, drug delivery or as the passive substrate of a bio-device. Advances in fabrication technologies provide new possibilities for the combination of silk fibroin with other nanomaterials to functionalize silk fibroin for specialized purposes, including sensing, cell visualization,resistance to ultraviolet light and provision of antibacterial properties. As the requirement for wearable and intelligent devices has become increasingly important over recent years, silk fibroin has been utilized as the active element in electronic and optical instruments. 4Role of humidity on the structures and properties of regenerated silk fibers显示文摘Silk fiber was processed from highly concentrated spinning dope to solid fibers along with water removal. To understand the mechanism of water removal during silk fiber spinning process, a microfluidic chip was designed and applied to investigate the structures and mechanical properties of two kinds of regenerated silk fibroin fibers dry-spun at different relative humidity. The experimental results showed that the diameters of the fibers spun at 40% RH are always larger than the fibers spun at 50% RH due to different removal rates of water. The fibers spun at low humidity contain more β-sheet structure and lower degree of chain orientation and crystalline orientation. 5Primary cilia mediate Klf2-dependant Notch activation in regenerating heart显示文摘Unlike adult mammalian heart,zebrafish heart has a remarkable capacity to regenerate after injury.Previous study has shown Notch signaling activation in the endocardium is essential for regeneration of the myocardium and this activation is mediated by hemodynamic alteration after injury,however,the molecular mechanism has not been fully explored.In this study we demonstrated that blood flow change could be perceived and transmitted in a primary cilia dependent manner to control the hemodynamic responsive klf2 gene expression and subsequent activation of Notch signaling in the endocardium.First we showed that both homologues of human gene KLF2 in zebrafish,klf2a and klf2b,could respond to hemodynamic alteration and both were required for Notch signaling activation and heart regeneration.Further experiments indicated that the upregulation of klf2 gene expression was mediated by endocardial primary cilia.Overall,our findings reveal a novel aspect of mechanical shear stress signal in activating Notch pathway and regulating cardiac regeneration.Xueyu Li Qiang Lu Yuanyuan Peng Fang Geng Xuelian Shao Huili Zhou Ying Cao Ruilin Zhang 2020Protein & Cell2020,11,6:3
63D Printed Gelatin Scaffold with Improved Shape Fidelity and Cytocompatibility by Using Antheraea pernyi Silk Fibroin Nanofibers显示文摘Gelatin(G)is a commonly used natural biomaterial owing to its good biocompatibility and easy availability.However,using pure gelatin as a bioink can barely achieve an ideal shape fidelity in 3D printing.In this study,Antheraea pernyi silk fibroin nanofibers(ASFNFs)with arginine-glycine-aspartic acid(RGD)peptide and partial natural silk structure are extracted and combined with pure gelatin bioink to simultaneously improve the shape fidelity and cytocompatibility of corresponding 3D printed scaffold.Results show that the optimum printing temperature is 30℃ for these bioinks.The printed filaments using 16G/4ASFNFs bioink(16wt%gelatin and 4wt%ASFNFs)demonstrate better morphology and larger pore size than those printed by pure gelatin bioink(20G,20wt%gelatin),thus successfully improve the shape fidelity and porosity of the 3D printed scaffold.The 16G/4ASFNFs scaffold also demonstrate higher swelling ratio and faster degradation rate than the pure gelatin scaffold.Moreover,the cell viability and proliferation ability of Schwann cells cultured on the 16G/4ASFNFs scaffold are significantly superior than those cultured on the pure 20G scaffold.The ASFNFs enhanced 16G/4ASFNFs scaffold reported here are expected to be a candidate with excellent potential for biomedical applications.Shengzhi Zou Suna Fan Ana L.Oliveira Xiang Yao Yaopeng Zhang Huili Shao 2022Advanced Fiber Materials2022,4,4:2
7Thermal simulation experiment on the hydrocarbon regeneration of marine carbonate source rock显示文摘Hydrocarbon regeneration of marine carbonate source rock was simulated with thermal experiments in a laboratory. The results reveal that hydrocarbon regeneration does not simply continue the primary hydrocarbon generation process, and that, for marine carbonate source rock, discontinuous hydrocarbon generation differs greatly from the continuous generation. Several different features of hydrocarbon regeneration were observed in the experiments. First, the liquid hydrocarbon generation peak was always observed no matter what the initial maturity of the sample was. Moreover, the maturity and the liquid hydrocarbon yield corresponding to the peak varied with the sample’s initial maturity. Second, the hydrocarbon regeneration started earlier than the continuous one. In the experiments, the hydrocarbon could be re-generated when the sample maturity did not rise to any great extent. Third, the accumulative hydrocarbon-generating quantity during discontinuous generation was always more than that during continuous generation. And the hydrocarbon-generating quantity varied with the discontinuous generation history. 8CUL4B facilitates HBV replication by promoting HBx stabilization显示文摘Objective:Hepatitis B virus(HBV)infection is a major public health problem worldwide.However,the regulatory mechanisms underlying HBV replication remain unclear.Cullin 4 B-RING ubiquitin E3 ligase(CRL4 B)is involved in regulating diverse physiological and pathophysiological processes.In our study,we aimed to explain the role of CUL4 B in HBV infection.Methods:Cul4 b transgenic mice or conditional knockout mice,as well as liver cell lines with CUL4 B overexpression or knockdown,were used to assess the role of CUL4 B in HBV replication.Immunoprecipitation assays and immunofluorescence staining were performed to study the interaction between CUL4 B and HBx.Cycloheximide chase assays and in vivo ubiquitination assays were performed to evaluate the half-life and the ubiquitination status of HBx.Results:The hydrodynamics-based hepatitis B model in Cul4 b transgenic or conditional knockout mice indicated that CUL4 B promoted HBV replication(P<0.05).Moreover,the overexpression or knockdown system in human liver cell lines validated that CUL4 B increased HBV replication in an HBx-dependent manner.Importantly,immunoprecipitation assays and immunofluorescence staining showed an interaction between CUL4 B and HBx.Furthermore,CUL4 B upregulated HBx protein levels by inhibiting HBx ubiquitination and proteasomal degradation(P<0.05).Finally,a positive correlation between CUL4 B expression and HBV pg RNA level was observed in liver tissues from HBV-positive patients and HBV transgenic mice.Conclusions:CUL4 B enhances HBV replication by interacting with HBx and disrupting its ubiquitin-dependent proteasomal degradation.CUL4 B may therefore be a potential target for anti-HBV therapy.Haixia Shan Bo Wang Xiaodong Zhang Hui Song Xi Li Yongxin Zou Baichun Jiang Huili Hu Hao Dou Changshun Shao Lifen Gao Chunhong Ma Xiaoyun Yang Xiaohong Liang Yaoqin Gong 2022Cancer Biology & Medicine2022,19,1:2
