| 1 | Establishment of a Highly Efficient Regeneration System for the Mature Embryo Culture of Wheat显示文摘Establishment of a highly efficient regeneration system for the mature embryo of wheat will provide a convenient tool for wheat tissue culture and transformation, thereby facilitating the transformation of foreign genes into wheat. By using the mature embryos derived from 20 different wheat lines including Shi 4185, Yumai 66, Lunxuan 987, CB037, Yangmai 6, Xinchun 9, Bobwhite, Han 6172, Zheng 9023, Jimai 20, Ningchun 4, and Jing 411, the effects of some factors including inoculation methods, initiating culture media, organic additives, antioxidants, and auxins on the regeneration from the explants were evaluated. The results indicated that the scraping embryo culture was better than the whole embryo culture, the Aa medium was better than the SD2 medium and dicamba was better than 2,4-D in increasing the regeneration frequency. An Adi medium was established in this study by adding silver nitrate, cysteine, ascorbic acid, dicamba, glutamine into the Aa medium at the concentration of 4 , 40, 100, 2, and 5 mg L-1, respectively. By using the Adi medium and the scraping technique, the regeneration frequencies of the mature embryos of CB037, Lunxuan 987, Han 6172, Yangmai 6, Bobwhite, Zheng 9023, Shi 4185, and Jimai 20 became 85.6, 60.1, 46.0, 42.1, 42.0, 34.0, 33.0, and 32.0%, respectively, which were about 5-8 times higher than that obtained from the conventional culture mediums and techniques. | 2 | Improvement of Plant Regeneration from Immature Embryos of Wheat Infected by Agrobacterium tumefaciens显示文摘Wheat, one of the most important food crops, has been extensively studied with respects to plant regeneration and transformation employing the immature embryos as recipient tissues. However, the transformed tissues often become severely necrotic after co-cultivation with Agrobacterium, which is one of the major obstacles in gene delivery. In this study, wheat varieties CB037, Kenong 199, Xinchun 9, Lunxuan 987, and Shi 4185 showed desirable culture potential or high infection ability in Agrobacterium-mediated transformation. Similarly, optimal regeneration conditions were determined by testing their ability to inhibit the cell necrosis and cell death phenotype. Two auxins of 2,4-dichlorophenoxyacetic acid (2,4-D) and 3,6-dichloro-o-anisic acid (dicamba) were evaluated for highly significant effect on both callus and plantlet production, although they were genotype-dependent in wheat. Substitution of 2,4-D by dicamba enhanced the growth and regeneration ability of callus from the immature embryos of most genotypes tested. The callus growth state couldn’t be modified by adding antioxidants such as ascorbic acid, cysteine, and silver nitrate or organic additives such as thiamine HCl and asparagine to the media. On the contrary, the best tissue statement and plant regeneration was achieved by employing the media containing the simplest MS (Murashige and Skoog) components and dicamba without organic components and vitamins. | 3 | Cloning and Characterization of Genes Coding for Fructan Biosynthesis Enzymes (FBEs) in Triticeae Plants显示文摘Fructan is not only a carbon source for storage but also plays an important role as anti-stress agents in many plant species. Complex fructans having both β-(2,1)- and β-(2,6)-linked fructosyl units accumulate in Triticeae plants commonly. Three enzymes (sucrose: sucrose 1-fructosyltransferase, 1-SST, EC: 2.4.1.99; sucrose: fructan 6-fructosyltransferase, 6- SFT, EC: 2.4.1.10; and fructan: fructan 1-fructosyltransferase, 1-FFT, EC: 2.4.1.100) were involved in fructan biosynthesis in Triticeae plant species. We successfully isolated these genes from tetraploid wheat (Triticum turgidum, genotype: AABB), common wheat (Triticum aestivum L., genotype: AABBDD) and three wild relatives of common wheat, Triticum urartu Thum. (the origin of the AA genome), Aegilops speltoides (Tausch) Gren. (the putative source of the SS genome) and Aegilops tauschii Coss. (the source of the DD genome). Sequence analysis revealed that all the FBEs (fructan biosynthetic enzymes) had three highly conserved functional motifs except 1-SST (EU981912) from tetraploid wheat species only with conserved DPNG. Low pI (isoelectric point) and potential N-glycosylation sites were predicted, which were crucial for protein compartmentation and post-translational process. Analysis on subcelluar localization signals showed that only 6-SFT had vacuolar-directed signal. Sequences alignment result showed that 1-SST and 1-FFT were more conservative and had closer relationship each other, while 6-SFT was more active during the evolution processing. According to the syntenic relationship between wheat and rice genome, FBEs were predicated to be located on the homeologous group 6 and group 2 chromosomes. Expression profile confirmed that expression of all the three FBEs were drought-stress induced. | 4 | Pedicled greater omentum flap for preventing bile leak in liver transplantation patients with poor biliary tract conditions显示文摘BACKGROUND:Bile leak remains a main complication in liver transplantation patients with poor biliary tract conditions,mainly caused by an insufficient blood supply or dysplasia of the biliary tract.Although Roux-en-Y modus operandi can be adopted,the risk of other complications of the biliary tract such as infection increases.Using pedicled greater omentum flaps to wrap the anastomotic stoma, which increases the biliary tract blood supply,may reduce the incidence of bile leak. METHODS:Fourteen patients undergoing piggy-back liver transplantation and having poor biliary tract conditions were treated with pedicled greater omentum flaps to wrap the anastomotic stoma of the biliary tract. Their clinical data were analyzed retrospectively. RESULTS:Of the 14 patients,only one(7.1%)had a mild bile leak on the 8th day post-operation and fully recovered after symptomatic treatment.The other patients had no biliary complications. | 5 | Hepatic veins anatomy and piggy-back liver transplantation显示文摘BACKGROUND:The piggy-back caval anastomosis technique is widely used in orthotopic liver transplantation although it carries an increased risk of complications,including outflow obstruction and Budd-Chiari syndrome.The aim of this study is to clarify the anatomy and variations of hepatic veins(HVs) draining into the inferior vena cava(IVC),and to classify the surgical techniques of piggy-back liver transplantation(PBLT) based on the anatomy of HVs which can reduce the occurrence of complications.METHODS:PBLT was performed in 248 consecutive cases at our hospital from January 2004 to August 2011.The anatomy of recipients' HVs was determined when removing the native diseased livers.Both anatomy of HVs and short HVs draining into the IVC were recorded.These data were collected and analyzed.RESULTS:We classified anatomic variations of HVs in the 248 livers into five types according to the way of drainage into the IVC:type I(trunk type of left and middle HVs),142(57.3%) patients;type II(trunk type of right and middle HVs),54(21.8%);type III(trunk type of left,middle and right HVs),14(5.6%);type IV(non-trunk type of left,middle and right HVs),of which,type IVa,16(6.5%),in the same horizontal plane;type IVb,18(7.3%),in different horizontal planes;and type V(segment type),4(1.6%).The patients whose HVs anatomy belonged to types I,II and III underwent classical piggy-back liver transplantation.Type IVa patients had classical PBLT via HV venoplasty prior to piggy-back anastomosis,while type IVb patients and type V patients could only have modified PBLT.CONCLUSION:This study demonstrates that HVs can be classified according to the anatomy of their drainage into the IVC and we can use this classification to choose the best operative approach to PBLT. | Ying-Zi Ming,Ying Niu,Ming-Jie Shao,Xing-Guo She and Qi-Fa Ye Research Center of Chinese Health Ministry on Transplantation Medicine Engineering and Technology,The Third Xiangya Hospital,Central South University,Changsha 410013,China | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,4: | 4 |

Brain metastasis(BM) is a common complication of NSCLC, with 25%–40% of patients developing BM during the course of the disease. A significant strategy of local disease control in the central nervous system is radiation therapy. With the development of precision medicine,the concept of treating lung cancer BM has gradually changed. In this case, we performed a surgical procedure to obtain enough tumor tissue for the detection of the target gene and other related experiments after the patient was informed. Finally, we found that the patient had both hepatocyte growth factor receptor(MET) gene amplification and kinesin light chain 1-anaplastic lymphoma kinase fusion(KLC1-ALK) through next-generation sequencing and showed sensitivity to the targeted therapy of crizotinib. The patient exhibited good response.
