1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 3Effects of Cinnamon Acid on Respiratory Rate and Its Related Enzymes Activity in Roots of Seedlings of Malus hupehensis Rehd.显示文摘This paper studied the effects of cinnamon acid treatments on the respiratory rate and related enzymes activity in the seedling roots of Malus hupehensis Rehd.It would provide information for understanding the mechanisms of inhibition damage caused by continuous cultivation of apple tree.20 mL of solution containing different concentrations of cinnamon acid was added into container with the tested seedlings.After treatment,the samples were taken periodically and the respiratory rates were measured by OXY-LAB oxygen electrodes under 25°C stable temperature and then the activities of related enzymes were measured.The rates of total respiration and other 2 pathways [tricarboxylic acid cycle (TCA) and pentose phosphate pathway (PPP)] appeared initially an increasing treads and late (on the 3rd d) began to decline.However,they again appeared an increase trend at the end period,on the contrast,the respiratory rate of embden-meyer- hot-parnas (EMP) pathway appeared a stead decline tread but it had a recover on the last day.The respiratory rate of total and 3 pathways were decreased under 125 mg kg-1 (soil).The dynamic trends of the enzymes activities of pyrophosphate-dependent phosphofructokinase (PFK),glucose-6-phosphate dehydrogenase (G-6-PDH) and malate dehydrogenase (MDH) showed similarly.In conclusion,treatments of certain concentration of cinnamon acid would inhibit the respiratory rate and related enzymes activity of roots of M.hupehensis Rehd.And the inhibition degrees were positively related with concentration of cinnamon acid treatments.GAO Xiang-bin,ZHAO Feng-xia,SHEN Xiang,HU Yan-li,HAO Yun-hong,YANG Shu-quan,SU Li-tao and MAO Zhi-quan State Key Laboratory of Crop Biology/College of Horticultural Science and Engineering,Shandong Agricultural University,Tai’an 271018,P.R.China 2010Agricultural Sciences in China2010,9,6:9
4Arterial relaxation is coupled to inhibition of mitochondrial fission in arterial smooth muscle cells:comparison of vasorelaxant effects of verapamil and phentolamine显示文摘Mitochondria are morphologically dynamic organelles which undergo fission and fusion processes. Our previous study found that arterial constriction was always accompanied by increased mitochondrial fission in smooth muscle cells, whereas inhibition of mitochondrial fission in smooth muscle cells was associated with arterial relaxation. Here, we used the typical vasorelaxants, verapamil and phentolamine, to further confirm the coupling between arterial constriction and mitochondrial fission in rat aorta. Results showed that phentolamine but not verapamil induced vasorelaxation in phenylephrine(PE)-induced rat thoracic aorta constriction. Verapamil, but not phentolamine, induced vasorelaxation in high K^+(KPSS)-induced rat thoracic aorta constriction. Pre-treatment with phentolamine prevented PEbut not KPSS-induced aorta constriction and pre-treatment with verapamil prevented both PE-and KPSSinduced aorta constriction. Transmission electron microscopy(TEM) results showed that verapamil but not phentolamine inhibited KPSS-induced excessive mitochondrial fission in aortic smooth muscle cells,and verapamil prevented both PE-and KPSS-induced excessive mitochondrial fission in aortic smoothmuscle cells. Verapamil inhibited KPSS-induced excessive mitochondrial fission in cultured vascular smooth muscle cells(A10). 6Monitoring Value of Multimodal Magnetic Resonance Imaging in Disease Progression of Amyotrophic Lateral Sclerosis: A Prospective Observational Study显示文摘Background: Ongoing efforts have been made to identify new neuroimaging markers to track amyotrophic lateral sclerosis (ALS) progression. This study aimed to explore the monitoring value of multimodal magnetic resonance imaging (MRI) in the disease progression of ALS. Methods: From September 2015 to March 2017, ten patients diagnosed with ALS in Peking Union Medical College Hospital completed head MRI scans at baseline and during follow-up. Multimodal MRI analyses, including gray matter (GM) volume measured by voxel-based morphometry;cerebral blood flow (CBF) evaluated by arterial spin labeling;functional connectivity, including low-frequency fluctuation (fALFF) and regional homogeneity (ReHo), measured by resting-state functional MRI;and integrity of white-matter (WM) fiber tracts evaluated by diffusion tensor imaging, were performed in these patients. Comparisons of imaging metrics were made between baseline and follow-up using paired t-test. Results: In the longitudinal comparisons, the brain structure (GM volume of the right precentral gyri, left postcentral gyri, and right thalami) and perfusion (CBF of the bilateral temporal poles, left precentral gyri, postcentral gyri, and right middle temporal gyri) in both motor and extramotor areas at follow-up were impaired to different extents when compared with those at baseline (all P < 0.05, false discovery rate adjusted). Functional connectivity was increased in the motor areas (fALFF of the right precentral gyri and superior frontal gyri, and ReHo of right precentral gyri) and decreased in the extramotor areas (fALFF of the bilateral middle frontal gyri and ReHo of the right precuneus and cingulate gyri) (all P < 0.001, unadjusted). No significant changes were detected in terms of brain WM measures. 7Alteration of oncogenic IGF-II gene methylation status associates with hepatocyte malignant transformation显示文摘Background: Oncogenic insulin-like growth factor-II(IGF-II) is overexpressed in hepatocellular carcinoma(HCC). The present study aimed to analyze the dynamic alteration of IGF-II CpG site methylation status and its molecular mechanism in HCC progression. Methods: IGF-II alterations were observed in rat hepatocarcinogenesis models induced by 2-acetylaminofluorene. Liver IGF-II expression was compared by immunohistochemistry or tissue IGF-II specific concentration(nmol/mg protein). Status of human IGF-II promoter 3(P3) or rat IGF-II P2 CpG site methylation was amplified by methylation-specific polymerase chain reaction(MSP). Serum IGF-II levels were quantitatively detected by an enzyme-linked immunosorbent assay. Results: The levels of hepatic IGF-II expression were significantly elevated in the HCC group( P < 0.001). The unmethylation rate of IGF-II P3 CpG sites was 100% in the HCC-, 52.5% in the paracancerous-, and none(0%) in the distal noncancerous-tissues. Abnormal IGF-II expression was related to differentiation degree, tumor invasion, and positive HBV-DNA(all P < 0.001), with a negative correlation between P3 methylation degree and IGF-II expression. There was a positive correlation between liver IGF-II specific concentration and circulating IGF-II level( r = 0.97, P < 0.001). Significantly negative correlation was found between IGF-II P2 CpG site methylation and circulating IGF-II( r s =-0.89, P < 0.001) or liver IGF-II level( r s =-0.84, P < 0.001). 82D materials for conducting holes from grain boundaries in perovskite solar cells显示文摘Grain boundaries in organic-inorganic halide perovskite solar cells(PSCs)have been found to be detrimental to the photovoltaic performance of devices.Here,we develop a unique approach to overcome this problem by modifying the edges of perovskite grain boundaries with flakes of high-mobility two-dimensional(2D)materials via a convenient solution process.A synergistic effect between the 2D flakes and perovskite grain boundaries is observed for the first time,which can significantly enhance the performance of PSCs.We find that the 2D flakes can conduct holes from the grain boundaries to the hole transport layers in PSCs,thereby making hole channels in the grain boundaries of the devices.Hence,2D flakes with high carrier mobilities and short distances to grain boundaries can induce a more pronounced performance enhancement of the devices.This work presents a cost-effective strategy for improving the performance of PSCs by using high-mobility 2D materials.Peng You Guanqi Tang Jiupeng Cao Dong Shen Tsz-Wai Ng Zafer Hawash Naixiang Wang Chun-Ki Liu Wei Lu Qidong Tai Yabing Qi Chun-Sing Lee Feng Yan 2021Light(Science & Applications)2021,10,4:3

10Transcriptome analysis of the influence of CPPU application for fruit setting on melon volatile content显示文摘In fruit production,the application of the plant growth regulator 1-(2-chloro-4-pyridyl)-3-phenylurea(CPPU)dulls the fruit aroma.Gas chromatography-mass spectrometry and transcriptome analyses were performed on CPPU-treated and pollinated fruits to determine how CPPU affects the production of aroma in melon fruit.The results showed that the contents of two important esters(benzyl acetate and phenethyl acetate)in the CPPU-treated fruits were significantly lower than those in the pollinated fruits.Transcriptome sequencing data revealed that most differentially expressed genes were involved in“phenylalanine metabolism”pathway,and their expression was significantly decreased in the CPPU-treated fruits.Further analysis showed that the phenylalanine content in the CPPU-treated fruits was significantly higher than that in the pollinated fruits.In summary,CPPU application interferes with phenylalanine metabolism in melon fruits and affects the production of aromatic esters.CHENG Jin-tao CHEN Hai-wen DING Xiao-chen SHEN Tai PENG Zhao-wen KONG Qiu-sheng HUANG Yuan BIE Zhi-long 2021Journal of Integrative Agriculture2021,20,12:2
