1WRKY transcription factors in plant responses to stresses显示文摘The WRKY gene family is among the largest families of transcription factors(TFs) in higher plants.By regulating the plant hormone signal transduction pathway,these TFs play critical roles in some plant processes in response to biotic and abiotic stress.Various bodies of research have demonstrated the important biological functions of WRKY TFs in plant response to different kinds of biotic and abiotic stresses and working mechanisms.However,very little summarization has been done to review their research progress.Not just important TFs function in plant response to biotic and abiotic stresses,WRKY also participates in carbohydrate synthesis,senescence,development,and secondary metabolites synthesis.WRKY proteins can bind to W-box(TGACC(A/T)) in the promoter of its target genes and activate or repress the expression of downstream genes to regulate their stress response.Moreover,WRKY proteins can interact with other TFs to regulate plant defensive responses.In the present review,we focus on the structural characteristics of WRKY TFs and the research progress on their functions in plant responses to a variety of stresses.Jingjing Jiang Shenghui Ma Nenghui Ye Ming Jiang Jiashu Cao Jianhua Zhang 2017Journal of Integrative Plant Biology2017,59,2:75
2MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple.Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen 2019Horticulture Research2019,6,1:16
3Malus sieversii: the origin, flavonoid synthesis mechanism, and breeding of red-skinned and red-fleshed apples显示文摘Flavonoids play essential roles in human health.Apple(Malus domestica Borkh.),one of the most widely produced and economically important fruit crops in temperate regions,is a significant source of flavonoids in the human diet and is among the top nutritionally rated and most widely consumed fruits worldwide.Epidemiological studies have shown that the consumption of apples,which are rich in a variety of free and easily absorbable flavonoids,is associated with a decreased risk of various diseases.However,apple production is challenged by serious inbreeding problems.The narrowing of the hereditary base has resulted in apples with poor nutritional quality and low flavonoid contents.Recently,there have been advances in our understanding of the roles that Malus sieversii(Ledeb.)M.Roem has played in the process of apple domestication and breeding.In this study,we review the origin of cultivated apples and red-fleshed apples,and discuss the genetic diversity and construction of the core collections of M.sieversii.We also discuss current research progress and breeding programs on red-skinned and red-fleshed apples and summarize the exploitation and utilization of M.sieversii in the breeding of high-flavonoid,and red-fleshed apples.This study highlights a valuable pattern of horticultural crop breeding using wild germplasm resources.The future challenges and directions of research on the molecular mechanisms of flavonoid accumulation and high-flavonoid apple breeding are discussed.Nan Wang Shenghui Jiang Zongying Zhang Hongcheng Fang Haifeng Xu Yicheng Wang Xuesen Chen 2018Horticulture Research2018,5,1:15
4Stratigraphy of the Triassic-Jurassic Boundary Successions of the Southern Margin of the Junggar Basin,Northwestern China显示文摘The Triassic-Jurassic(Tr-J) boundary marks a major extinction event,which(~200 Ma) resulted in global extinctions of fauna and flora both in the marine and terrestrial realms.There prevail great challenges in determining the exact location of the terrestrial Tr-J boundary,because of endemism of taxa and the scarcity of fossils in terrestrial settings leading to difficulties in linking marine and terrestrial sedimentary successions.Investigation based on palynology and bivalves has been carried out over a 1113 m thick section,which is subdivided into 132 beds,along the Haojiagou valley on the southern margin of the Junggar Basin of the northern Xinjiang,northwestern China.The terrestrial Lower Jurassic is conformably resting on the Upper Triassic strata.The Upper Triassic covers the Huangshanjie Formation overlaid by the Haojiagou Formation,while the Lower Jurassic comprises the Badaowan Formation followed by the Sangonghe Formation.Fifty six pollen and spore taxa and one algal taxon were identified from the sediments.Based on the key-species and abundance of spores and pollen,three zones were erected:the Late Triassic(Rhaetian) Aratrisporites-Alisporites Assemblage,the Early Jurassic(Hettangian) Perinopollenites-Pinuspollenites Assemblage,and the Sinemurian Perinopollenites-Cycadopites Assemblage.The Tr-J boundary is placed between bed 44 and 45 coincident with the boundary between the Haojiagou and Badaowan formations.Beds with Ferganoconcha(?),Unio-Ferganoconcha and Waagenoperna-Yananoconcha bivalve assemblages are recognized.The Ferganoconcha(?) bed is limited to the upper Haojiagou Formation, Unio-Ferganoconcha and Waagenoperna-Yananoconcha assemblages are present in the middle and upper members of the Badaowan Formation.The sedimentary succession is interpreted as terrestrial with two mainly lake deposit intervals within Haojiagou and Badaowan formations,yielding fresh water algae and bivalves.However,the presence of brackish water algae Tasmanites and the marine-littoral facies bivalve Waagenoperna from the Badaowan Formation indicate that the Junggar Basin was influenced by sea water caused by transgressions from the northern Tethys,during the Sinemurian.SHA Jingeng Vivi VAJDA PAN Yanhong Linda LARSSON YAO Xiaogang ZHANG Xiaolin WANG Yaqiong CHENG Xiansheng JIANG Baoyu DENG Shenghui CHEN Siwei PENG Bo 2011Acta Geologica Sinica(English Edition)2011,85,2:7
5MdMYB6 regulates anthocyanin formation in apple both through direct inhibition of the biosynthesis pathway and through substrate removal显示文摘Anthocyanin biosynthesis and sugar metabolism are important processes during plant growth,but the molecular interactions underlying these pathways are still unclear.In this work,we analyzed the anthocyanin and soluble sugar contents,as well as the transcript levels of transcription factors that are known to be related to the biosynthesis of anthocyanin in‘Hongcui 1’apple flesh during fruit development.Overexpression of MdMYB6 in red-fleshed calli was found to reduce anthocyanin content and result in downregulated expression of the MdANS and MdGSTF12 proteins.Yeast one-hybrid and electrophoretic mobility shift analyses showed that MdMYB6 could directly bind to the promoters of MdANS and MdGSTF12,indicating that MdMYB6 could inhibit anthocyanin biosynthesis by regulating MdANS and MdGSTF12.Overexpression of MdTMT1 in the Arabidopsis tmt1 mutant restored the glucose and fructose contents to the wild-type levels,while overexpression of MdTMT1 in red-fleshed calli increased the contents of glucose and fructose but reduced the contents of UDP-glucose,UDP-galactose,and anthocyanin.Using a GUS reporter system,yeast one-hybrid,chromatin immunoprecipitation-PCR and electrophoretic mobility shift analyses,we found that MdMYB6 could bind to the promoter of MdTMT1,resulting in increased promoter activity.Overexpression of MdMYB6 in calli overexpressing MdTMT1 increased the expression of MdTMT1,which led to reduced contents of UDP-glucose and UDP-galactose and decreased anthocyanin content compared to those of the calli that overexpressed MdTMT1.This finding suggested that MdMYB6 could also inhibit anthocyanin biosynthesis by regulating MdTMT1 to decrease the contents of UDP-glucose and UDP-galactose.Taken together,these results showed that MdMYB6 and MdTMT1 play key roles in both anthocyanin biosynthesis and sugar transport.Haifeng Xu Qi Zou Guanxian Yang Shenghui Jiang Hongcheng Fang Yicheng Wang Jing Zhang Zongying Zhang Nan Wang Xuesen Chen 2020Horticulture Research2020,7,1:6
6Sediment Transport in the Nearshore Area of Phoenix Island显示文摘Based on the measured data, suspended sediment concentration, surface sediment grain size, current and waves, the sediment transport mechanisms and pathways in the Phoenix Island area were analyzed using methods of flux decomposition and Grain Size Trend Analysis(GSTA). The results show that net suspended sediment is mainly transported by average current, Stokes drift, and gravitational circulation. The transport direction of suspended sediment is varying and basically following the direction of residual tidal currents. Surface sediment transport pathways are primarily parallel to the coastline along with two convergent centers. Waves and longshore currents have a significant influence on sediment transport, but the influence is limited due to a steep and deep underwater bank. Tidal current is the main controlling factor for sediment transport, especially in the deep water area. Neither suspended nor surface sediment is transported towards the southwest. 7Beach Morphology and Coastline Evolution in the Southern Bohai Strait显示文摘The beach studied in this paper spans a length of 51 km and is one of several long sandy beaches in the southern Bohai Strait. Due to the obstruction of islands in the northeast and the influence of the underwater topography, the wave environment in the offshore area is complex; beach types and sediment transport characteristics vary along different coasts. The coastlines extracted from six aerial photographs in different years were compared to demonstrate the evolving features. Seven typical beach profiles were selected to study the lateral beach variation characteristics. Continuous wind and wave observation data from Beihuangcheng ocean station during 2009 were employed for the hindcast of the local wave environment using a regional spectral wave model. Then the results of the wave hindcast were incorporated into the LITDRIFT model to compute the sediment transport rates and directions along the coasts and analyze the longshore sand movement. The results show that the coastline evolution of sand beaches in the southern Bohai Strait has spatial and temporal variations and the coast can be divided into four typical regions. Region(I), the north coast of Qimudao, is a slightly eroded and dissipative beach with a large sediment transport rate; Region(II), the southwest coast of Gangluan Port, is a slightly deposited and dissipative beach with moderate sediment transport rate; Region(III), in the central area, is a beach that is gradually transformed from a slightly eroded dissipative beach to a moderately or slightly strong eroded bar-trough beach from west to east with a relatively moderate sediment transport rate. Region(IV), on the east coast, is a strongly eroded and reflective beach with a weak sediment transport rate. The wave conditions exhibit an increasing trend from west to east in the offshore area. The distribution of the wave-induced current inside the wave breaking region and the littoral sediment transport in the nearshore region exhibit a gradual weakening tendency from west to east, which is opposite to the trend of the wave conditions outside the breaking region. The presence of submerged shoal(Dengzhou Shoal), deep trough(Dengzhou Channel), islands and irregular topography influnces the wave climate, beach types, wave-induced current features, littoral sediment transport trends and coastline evolution patterns in the southern Bohai Strait. 8Formation and evolution of intraplatform basin from the late Sinian to early Cambrian in Sichuan Basin,China显示文摘Based on the drilling,logging,seismic and outcrop data,the formation and evolution of the late Sinian to early Cambrian intraplatform basin in Sichuan Basin was studied.Through the analysis of the total residual thickness of Member 3+Member 4 of Dengying Formation,the total thickness of Maidiping Formation+Qiongzhusi Formation,and lithofacies characteristics of Maidiping Formation,the distribution of the intraplatform basin was defined and its controlling effect on natural gas distribution of Dengying Formation was revealed.The intraplatform basin was characterized by small total residual thickness of Member 3 and Member 4 of Dengying Formation,the large total thickness of Maidiping Formation and Qiongzhusi Formation,and the deep-water phosphorous-carbonaceous-siliceous mudstone in Maidiping Formation.Horizontally,the intraplatform basin was mainly distributed in Qingchuan-Ziyang-Yibin area,and experienced four evolution periods.During the period of Member 1 and Member 2 of Dengying Formation,the Sichuan Basinwas a rimmed carbonate platform with the well developed mounds and shoals on the platform margin and its interior.The intraplatform basin in western Sichuan Basin was initially formed due to the first episode of Tongwan movement.During the period of Member 3 and Member 4 of Dengying Formation,the depositional center was formed in Moxi,Changning and Zhengxiong area due to extension and rifting of the Upper Yangtze Craton,thereafter,the Sichuan Basin evolved into a rimmed carbonate platform with an intraplatform basin.The second episode of Tongwan movement between Sinian and Cambrian led to the overall uplift and exposure of Sichuan Basin characterized by weathering denudation at high position and downward fluid erosion at low position.During the period of Maidiping Formation,three lithofacies zones were developed horizontally,and the third episode of the Tongwan movement between Maidiping Formation and Qiongzhusi Formation modified the sedimentary geomorphology locally.During the period of Qiongzhusi Formation and Canglangpu Formation,continuous deposition made the Sichuan Basin evolve into a gentle siliciclastic ramp.The evolution of the intraplatform basin not only promoted formation of high-quality platform-margin dolomite reservoirs of Member 4 of Dengying Formation,but also controlled distribution of the high-quality source rocks of Maidiping Formation and Qiongzhusi Formation.The dolomite of Dengying Formation in the surrounding area of the intraplatform basin was the most favorable hydrocarbon exploration area because of its good source-reservoir configuration.Hui Zhou Wei Li Baomin Zhang Jingjiang Liu Shenghui Deng Shiben Zhang Xiuqin Shan Jing Zhang Xiaobo Wang Hua Jiang 2017Petroleum Research2017,2,1:2

11The Phototropism of Jurassic Petrified Wood in North China Plate显示文摘Trees on the side directly exposed to sunlight generally grow faster than on the opposite side,a phenomenon termed plant phototropism.There are in situ vertical trunks of silicified wood in the Xiadelongwan area of Yanqing County,north Beijing,where the first National Geologic Park of Petrified Wood of China has been built since 2002.A few trunks have well-preserved growth rings.One petrified stump from the formation shows a positive phototropism direction of SW23O0.As compared with the modern normal growth stumps in Beijing plain area,which have a positive phototropism direction of SW210 °± 5°,the evidence of wood phototropism supports the conclusion of previous palaeomagnetic studies that the North China Plate has rotated clockwise since the Late Jurassic.The known petrified wood stumps in the Yanshan-Liaoning area are mainly found from the strata of 165-136 Ma,which corresponds to the main stage of the Yanshanian Movement.JIANG Zikun LIU Benper HUANG Min DENG Shenghui LU Yuanzheng LIU Lu LI Mingming DONG Shuxin WANG Yongdong 2014Acta Geologica Sinica(English Edition)2014,88,5:1
12Cr^(3+)/Y^(3+)co-doped persistent luminescence nanoparticles with biological window activation for in vivo repeatable imaging显示文摘The near-infrared(NIR)persistent luminescence materials(PLMs)can remain long-lasting luminescence after removal of the excitation light,which permits bioimaging with high sensitivity owing to the absence of background fluorescence interference from in situ excitation.Recently,the NIR PLMs have aroused intensive research interest in bioimaging.However,the optimal excitation wavelength of current NIR PLMs is located in the ultraviolet region with shallow tissue penetration,making it difficult to activate effectively in vivo,and seriously hindering their further application in bioimaging.Herein,we report a novel kind of Cr^(3+)ions and Y^(3+)ions co-doped NIR PLM,Zn_(1.3)Ga_(1.4)Sn_(0.3)O_(4):Cr^(3+),Y^(3+)(ZGSCY),which emits NIR persistent luminescence at 696 nm.Compared with Zn_(1.3)Ga_(1.4)Sn_(0.3)O_(4):Cr^(3+)(ZGSC)excited by the light with a wavelength in the biological window(>650 nm),after being co-doped with Y^(3+)ions,the NIR persistent luminescence performance of ZGSCY is significantly improved because of the increase of trap concentration in the matrix.In addition,we synthesized ZGSCY nanoparticles(NPs)by the combustion method,which exhibit excellent optical properties after being excited by the light with a wavelength in the biological window.After surface modification with PEG,the ZGSCY NPs present low cytotoxicity.Notably,due to the co-doping of Y^(3+)ions,the signal-to-noise ratio(SNR)of ZGSCY NPs in vivo imaging is about 1.8 times higher than that of the ZGSC NPs.Furthermore,the rechargeable in vivo imaging and passive tumor-targeted imaging are successfully achieved by activating with a lightemitting diode(LED,659 nm)after intravenous injection of ZGSCY.Thus,this kind of NIR PLM with high excitation efficiency performance in the biological window is expected to promote its biomedical application in deep tissues.Huimin Jiang Lin Liu Kexin Yu Xianggui Yin Shenghui Zheng Liang Song Junpeng Shi Yun Zhang 2022Journal of Rare Earths2022,40,9:1
13Crystal structures of main proteases of SARS-CoV-2 variants bound to a benzothiazole-based inhibitor显示文摘Main protease(Mpro)serves as an indispensable factor in the life cycle of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)as well as its constantly emerging variants and is therefore considered an attractive target for antiviral drug development.Benzothiazole-based inhibitors targeting Mpro have recently been investigated by several groups and proven to be promising leads for coronaviral drug development.In the present study,we determine the crystal structures of a benzothiazole-based inhibitor,YH-53,bound to Mpro mutants from SARS-CoV-2 variants of concern(VOCs)or variants of interest(VOIs),including K90R(Beta,B.1.351),G15S(Lambda,C.37),Y54C(Delta,AY.4),M49I(Omicron,BA.5)and P132H(Omicron,B.1.1.529).The structures show that the benzothiazole group in YH-53 forms a C-S covalent bond with the sulfur atom of catalytic residue Cys145 in SARSCoV-2 Mpro mutants.Structural analysis reveals the key molecular determinants necessary for interaction and illustrates the binding mode of YH-53 to these mutant Mpros.In conclusion,structural insights from this study offer more information to develop benzothiazole-based drugs that are broader spectrum,more effective and safer.Jiqing Luo Weiwei Wang Haihai Jiang Wenwen Li Pei Zeng Jie Wang Xuelan Zhou Xiaofang Zou Shenghui Chen Qisheng Wang Jin Zhang Jian Li 2023Acta Biochimica et Biophysica Sinica2023,55,8:0
14Heterostructured NiS_(2)@SnS_(2) hollow spheres as superior high-rate and durable anodes for sodium-ion batteries显示文摘Tin-based sulfides have attracted increasing attention as anodes for sodium-ion batteries(SIBs) owing to their high theoretical capacity;however, the poor rate capability and inferior cycling stability caused by the low electrical conductivity, sluggish kinetics and drastic volume variations during cycling have greatly hampered their practical applications. Herein, heterostructured NiS_(2)@SnS_(2) hybrid spheres were delicately designed and constructed by anchoring interconnected SnS_(2) nanosheets on metalorganic frameworks(MOFs)-derived Ni S_(2) hollow spheres coupled with N-doped carbon skeleton through facile solvothermal and sulfurization/carbonization processes. The unique hollow heterostructure with highly conductive carbon matrix can effectively facilitate the charge transfer kinetics and ensure the desired buffer space while endowing more active sites and enhanced structural integrity, as demonstrated by the experimental and density functional theory(DFT) results. Benefitting from these merits, the NiS_(2)@SnS_(2) hybrid composite displays a high reversible capacity of 820 m Ah g^(-1) after 250 cycles at 1 A g^(-1), and retains a value of 673 m Ah g^(-1)after 1,300 cycles at 5 A g^(-1), manifesting the excellent high-rate and durable sodium storage behaviors when applied in SIBs. 16Study on the Sediment Transport Flux and Mechanism in the Bohai Strait at the Tidal and Monthly Scales in Summer显示文摘Based on the data of tidal currents and suspended sediment concentrations observed synchronously at 11 stations in the Bohai Strait lasting for 25 hours,the temporal and spatial variations of currents and suspended sediment concentrations in the Bohai Strait in summer were analyzed.The Study preliminarily discussed the transport mechanism,transport trend and transport flux of suspended sediments in summer,using flux-mechanism decomposition method and numerical simulation.The suspended sediment transport was mainly controlled by advection and next influenced by vertical net circulation,while resuspension is relatively weak in Bohai Strait.The single-width fluxes of investigation stations varied from 3.8 to 89.1 gm^(−1) s^(−1),with the maximum value in Miaodao Strait.The suspended sediment transport trends in Laotieshan channel along the vertical section are obviously distinct.The waters mainly flow out of the Bohai Sea in surface layer,while into the Bohai Sea in bottom layer.However,the transport trends of other channels in the centre and south are consistent vertically.The sediments in the Bohai Strait follows the transport pattern of moving outward from the south and inward from the north in summer,i.e.,the sediments are carried out of the Bohai Sea through the Laotieshan channel,while into the Bohai Sea through other channels.And the outflow flux exceeds the inflow flux in August with the net water flux of 1.4×10^(10)m^(3),basically same as the deliveries of the rivers into the Bohai Sea.Moreover,the suspended sediment flux is 0.33 Mt under the action of tidal residual currents in the Yellow Sea in August.YUAN Xiaodong FENG Xiuli HU Rijun JIANG Shenghui ZHONG Wei 2023Journal of Ocean University of China2023,22,1:0
17A reference-guided TILLING by amplicon-sequencing platform supports forward and reverse genetics in barley显示文摘Barley is a diploid species with a genome smaller than those of other members of the Triticeae tribe,making it an attractive model for genetic studies in Triticeae crops.The recent development of barley genomics has created a need for a high-throughput platform to identify genetically uniform mutants for gene function investigations.In this study,we report an ethyl methanesulfonate(EMS)-mutagenized population consisting of 8525M_(3) lines in the barley landrace“Hatiexi”(HTX),which we complement with a high-quality de novo assembly of a reference genome for this genotype.The mutation rate within the population ranged from 1.51 to 4.09 mutations per megabase,depending on the treatment dosage of EMS and the mutation discrimination platform used for genotype analysis.We implemented a three-dimensional DNA pooling strategy combined with multiplexed amplicon sequencing to create a highly efficient and cost-effective TILLING(targeting induced locus lesion in genomes)platform in barley.Mutations were successfully identified from 72 mixed amplicons within a DNA pool containing 64 individual mutants and from 56 mixed amplicons within a pool containing 144 individuals.We discovered abundant allelic mutants for dozens of genes,including the barley Green Revolution contributor gene Brassinosteroid insensitive 1(BRI1).As a proof of concept,we rapidly determined the causal gene responsible for a chlorotic mutant by following the MutMap strategy,demonstrating the value of this resource to support forward and reverse genetic studies in barley.Congcong Jiang Miaomiao Lei Yu Guo Guangqi Gao Lijie Shi Yanlong Jin Yu Cai Axel Himmelbach Shenghui Zhou Qiang He Xuefeng Yao Jinhong Kan Georg Haberer Fengying Duan Lihui Li Jun Liu Jing Zhang Manuel Spannagl Chunming Liu Nils Stein Zongyun Feng Martin Mascher Ping Yang 2022Plant Communications2022,3,4:0
