2MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple.Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen 2019Horticulture Research2019,6,1:16
3Experimental studies on the P-T stability conditions and influencing factors of gas hydrate in different systems显示文摘The P-T stability conditions of gas hydrate in different systems (i.e., solution, silica sand, and marine sediment) were studied using multi-step decomposition method with our experimental equipment. The effects of different ions with various concentrations and sediment grains on the P-T stability conditions of gas hydrate were investigated. The results show that different ions have different influences on the phase equilibrium of gas hydrate. However, the influence of ions is in a similar trend: the larger the concentration, the bigger the P-T curve shifts to the left. For the silica sand, the influence of pore capillarity of coarse particles (> 460 μm) can be negligible. The P-T curve measured in coarse silica is in agreement with that in pure water. However, the influence of pore capillarity of fine particles (< 35 μm) is significant. The maximum reduction value of temperature is 1.5 K for methane hydrate under stable state. The sediment from the South China Sea significantly affects the P-T stability conditions of methane hydrate, with an average reduction value of 1.9 K within the experimental conditions. This is mainly the result of both the pore water salinity and the pore capillarity of sediment. 4Load-sharing Characteristic of Multiple Pinions Driving in Tunneling Boring Machine显示文摘The failure of the key parts, such as gears, in cutter head driving system of tunneling boring machine has not been properly solved under the interaction of driving motors asynchronously and wave tunneling torque load. A dynamic model of multi-gear driving system is established considering the inertia effects of driving mechanism and cutter head as well as the bending-torsional coupling. By taking into account the nonlinear coupling factors between ring gear and multiple pinions, the influence for meshing angle by bending-torsional coupling and the dynamic load-sharing characteristic of multiple pinions driving are analyzed. Load-sharing coefficients at different rotating cutter head speeds and input torques are presented. Numerical results indicate that the load-sharing coefficients can reach up to 1.2-1.3. A simulated experimental platform of the multiple pinions driving is carried out and the torque distributions under the step load in driving shaft of pinions are measured. The imbalance of torque distribution of pinions is verified and the load-sharing coefficients in each pinion can reach 1.262. The results of simulation and test are similar, which shows the correctness of theoretical model. A loop coupling control method is put forward based on current torque master slave control method. The imbalance of the multiple pinions driving in cutter head driving system of tunneling boring machine can be greatly decreased and the load-sharing coefficients can be reduced to 1.051 by using the loop coupling control method. 6Primary structure of helix C to helix G of a new retinal protein in H.sp.xz515显示文摘The gene fragment encoding the retinal protein from helix C to helix G in a new strain of halobacteria, H.sp.xz515 has been amplified by PCR method. The nucleotide sequence of this fragment has been determined. The deduced amino acid sequence has been compared with halobium br and other two br-like proteins, ar-1 and ar-2. Results show that those amino acid residues in br, essential for proton pumping and binding to retinal, are conserved. 7Genome-wide identification of agronomically important genes in outcrossing crops using OutcrossSeq显示文摘Many important crops(e.g.,tuber,root,and tree crops)are cross-pollinating.For these crops,no inbred lines are available for genetic study and breeding because they are self-incompatible,clonally propagated,or have a long generation time,making the identification of agronomically important genes difficult,particularly in crops with a complex autopolyploid genome.In this study,we developed a method,OutcrossSeq,for mapping agronomically important loci in outcrossing crops based on whole-genome low-coverage resequencing of a large genetic population,and designed three computation algorithms in OutcrossSeq for different types of outcrossing populations.We applied OutcrossSeq to a tuberous root crop(sweet potato,autopolyploid),a tree crop(walnut tree,highly heterozygous diploid),and hybrid crops(double-cross populations)to generate high-density genotype maps for the outcrossing populations,which enable precise identification of genomic loci underlying important agronomic traits.Candidate causative genes at these loci were detected based on functional clues.Taken together,our results indicate that OutcrossSeq is a robust and powerful method for identifying agronomically important genes in heterozygous species,including polyploids,in a cost-efficient way.The OutcrossSeq software and its instruction manual are available for downloading at www.xhhuanglab.cn/tool/OutcrossSeq.html.Mengjiao Chen Weijuan Fan Feiyang Ji Hua Hua Jie Liu Mengxiao Yan Qingguo Ma Jiongjiong Fan Qin Wang Shufeng Zhang Guiling Liu Zhe Sun Changgeng Tian Fengling Zhao Jianli Zheng Qi Zhang Jiaxin Chen Jie Qiu Xin Wei Ziru Chen Peng Zhang Dong Pei Jun Yang Xuehui Huang 2021Molecular Plant2021,14,4:4
8Twirling reinforcing-reducing manipulation--central mechanism underlying antihypertensive effect on spontaneous hypertension in rats显示文摘OBJECTIVE: To investigate antihypertensive effect in rats in order to confirm that twirling reinforcing-reducing manipulation(TRRM) might be the central mechanism underlying the action.METHODS: In the study,^(18) F-2-fluoro-deoxy-D-glucose positron emission tomography(18 F-FDG-PET)was employed. Fifity-six spontaneous hypertensive rats(SHRs) were randomly divided into a model group, a single-needle acupuncture(SNA) group, a twirling reinforcing group(SNA + TRF) and a twirling reducing(SNA + TRD) group. Fourteen Wistar rats were assigned to the control group. The acupuncture intervention at Taichong(LR 3) acupoint was administered once daily in the SNA, SNA + TRF and SNA + TRD groups for 14 days, with 1 d interval between the two weeks. The blood pressure(BP) of all rats was measured repeatedly and18 F-FDG-PET scans were conducted on the 14 th day. PET images were processed with Statistical Parametric Mapping 8.0.RESULTS: After the intervention, systolic BP showed a significant decrease in the SNA, SNA+TRF and SNA + TRD versus the model groups(all P <0.01) and in the SNA + TRF and SNA + TRD versus the SNA groups(both P < 0.01), with the SNA+TRD group exhibited the best antihypertensive effect(P < 0.01). The key brain regions activated by TRRM were mainly concentrated in the cerebellum, hippocampus, hypothalamus, medulla oblongata, insular cortex, midbrain, thalamus and visual cortex.CONCLUTION: TRRM could significantly lower the BP of SHRs by improving the cerebral glucose metabolism of the activated key brain regions and the underlying central mechanism may be related to the central rennin-angiotensin system and neurotransmission.Guo Qiulei Liu Qingguo Sun Dongmei Nie Binbin 2018Journal of Traditional Chinese Medicine2018,38,3:4

10Imaging bacteriorhodopsin-like molecules of claret-membranes from Tibet halobacteria xz515 by atomic force microscope显示文摘Halobacteria H.sp.xz 515 was isolated from a salt lake in Tibet. Although proton release-and-uptake across claret membrane is in reverse order compared to bacteri-orhodopsin in purple membrane from Halobacterium Sali-narum, and its efficiency of proton pump is much lower, AFM image shows that the molecules are still arranged in a two-dimensional hexagonal lattice of trimers. Primary structure of C- to G-helix of the archaerhodopsin shows that it has only 56% homology with bacteriorhodopsin. But the interactive amino acid residues at the interface between B-and D-helixes are conserved. 11Effect of Methane Gas on Acoustic Characteristics of Hydrate-Bearing Sediment–Model Analysis and Experimental Verification显示文摘Gas leakage is an important consideration in natural systems that experience gas hydrate accumulation.A number of velocity models have been created to study hydrate-bearing sediments,including the BGTL theory,the weighted equation,the Wood equation,the K-T equation,and the effective medium theory.In previous work,we regarded water as the pore fluid,which meant its density and bulk modulus values were those of water.This approach ignores the presence of gas,which results in a biased calculation of the pore fluid's bulk modulus and density.To take into account the effect of gas on the elastic wave velocity,it is necessary to recalculate the bulk modulus and density of an equivalent medium.Thus,a high-pressure reactor device for simulating leakage systems was developed to establish the relationship between wave velocity and hydrate saturation in methane-flux mode.A comparison of the values calculated by the velocity model with the experimental data obtained in this study indicates that the effective medium theory(EMT,which considers gas effects)is more applicable than other models.For hydrate saturations of 10%–30%,the result ranges between EMT-B(homogenous gas distribution)and EMT-B(patchy gas distribution).For hydrate saturations of 30%–60%,the results are similar to those of the EMT-B(homogenous gas distribution)mode,whereas hydrate saturations of 60%–70%yield results similar to those of the EMT-A mode.For hydrate saturations greater than 80%,the experimental results are similar to those of the EMT-B mode.These results have significance for hydrate exploitation in the South China Sea.BU Qingtao HU Gaowei LIU Changling DONG Jie XING Tongju SUN Jianye LI Chengfeng MENG Qingguo 2021Journal of Ocean University of China2021,20,1:2
12A Review of Co-processing of Kitchen Waste and Municipal Residual Sludge显示文摘In this paper,the methods of kitchen waste disposal in recent years were summed up,and the advantages and disadvantages of the current treatment methods were analyzed. At the same time,it has been found that the co-processing of kitchen waste and residual sludge not only has dealt with both kitchen waste and residual sludge,but also the carbon and nitrogen ratio and moisture content of the mixture are balanced,and increases the processing efficiency significantly. It has a certain processing advantage. At present,most research is about the anaerobic digestion of kitchen waste and residual sludge,and the process is more mature,but the study of VFAs accumulation is still insufficient. Aerobic composting is still in the early stage of development,and there are few studies on it at home and abroad. 13Melatonin antagonizes ovarian aging via YTHDF2-MAPK-NF-κB pathway显示文摘Cellular senescence is closely associated with age-related diseases.Ovarian aging,a special type of organ senescence,is the pathophysiological foundation of the diseases of the reproductive system.It is characterized by the loss of integrity of the surface epithelium and a gradual decrease in the number of human ovarian surface epithelial cells(HOSEpiCs).To contribute to the research on delaying ovarian aging,we aimed to investigate the novel epigenetic mechanism of melatonin in protecting HOSEpiCs.We discovered that melatonin has antagonistic effects against the oncogene-induced senescence(OIS)of HOSEpiCs.Mechanistically,the oncogene Ras decreased the expression of YTHDF2,which is the reader of RNA-m6A,by stimulating the generation of reactive oxygen species(ROS).Moreover,we found that the suppression of YTHDF2 increased the expression of MAP2K4 and MAP4K4 by enhancing the stability of the transcription of their mRNAs,thereby upregulating the expression of the senescence-associated secretory phenotype(SASP)through the activation of the MAP2K4 and MAP4K4-dependent nuclear factor-κB(NF-κB)signaling pathways.We further determined that melatonin has antagonistic effects against the OIS of HOSEpiCs by inhibiting the ROS-YTHDF2-MAPK-NF-κB pathway.These findings provide key insights into the potential avenues for preventing and treating ovarian aging.Ruigong Zhu Xian Ji Xuan Wu Jiajing Chen Xuesong Li Hong Jiang Haiping Fu Hui Wang Zhe Lin Xin Tang Shixiu Sun Qingguo Li Bingjian Wang Hongshan Chen 2022Genes & Diseases2022,9,2:2

15Synthesis, structure, and magnetism of three manganese-organic framework with PtS topology显示文摘Three 3D manganese-organic frameworks, Mn2(BPTC)(DMF)2(H2O)·DMF·3H2O(1), Mn2(BPTC)(bipy)(DMF)·DMF·H2O(2), and Mn2(BPTC)(phen)(DMF)·EtOH(3), have been solvothermally synthesized using 3,3′,5,5′-biphenyltetracarboxylic acid(H4BPTC). All complexes are characterized by PXRD, EA, IR and TG. The results show that they all bear the PtS topology with(42.84)(42.84) for the vertex symbols of the planar and tetrahedral nodes, in which the BPTC ligand is considered as a square-planar 4-connected linker, and every binuclear SBU connected to the four BPTC ligands is simplified into tetrahedral 4-connected nodes. Because the three coordination sites of one metal center of SBU are occupied by the coordinated solvent molecules, complex 1 exhibits low stability. After substituting 2,2′-bipy or 1,10-phen for two coordinated solvent molecules, complexes 2 and 3 display evidently higher structure stability. 16Dynamic Response of Shallow-Buried Tunnels Traversing High Loess Slopes显示文摘Considering the existence of numerous shallow-buried tunnels traversing high slopes in the loess area in western China and the fact of high seismic intensity there,we investigate the dynamic response rules of a shallow-buried loess tunnel and its slope under the action of seismic waves with different intensities.Through large-scale shaking table model tests,we successfully analyze the characteristics and process of the destabilization of tunnels and slopes,and propose valuable suggestions regarding the reinforcement parts of a tunnel for reducing seismic damage.The results show that the main seismic damage on a slope include the failure of the sliding surface between the top and foot and the stripping of the soil around the tunnel entrance,while the damage on a tunnel is mainly manifested as the seismic-induced subsidence at the portal section and the cracking deformation at the joint areas.Finally,we propose that the“staggered peak distribution”phenomenon of the maximum acceleration values at the vault and inverted arch area can be considered as a criterion indicating that the tunnel enters into the threshold of dynamic failure.SUN Wen LIANG Qingguo WANG Lili CAO Xiaoping 2020Earthquake Research in China2020,34,4:1

19Effects of Phosphorus Concentration on the Treatment of Domestic Wastewater by a Combination of Hydrolytic Acidification and Biological Aerated Filter Filled with Mussel Shells显示文摘The combination of hydrolytic acidification and biological aerated filter( BAF) filled with mussel shells was used to treat domestic wastewater,and the removal rates of chemical oxygen demand( COD),ammonia nitrogen( NH_3-N) and total phosphorus( TP) by the system were analyzed under different TP concentrations. When TP concentration ranged from 12. 39 to 14. 69 mg / L,the removal rate of COD was the best,over 90. 92%; as TP concentration varied from 2. 26 to 2. 61 mg / L,the removal rates of NH_3-N and TP were the best,up to 100. 00% and76. 38% respectively.
