| 1 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |
| 2 | Graphitic carbon nitride (g-C3N4)-based nanosized heteroarrays: Promising materials for photoelectrochemical water splitting显示文摘Photoelectrochemical(PEC)water splitting is recognized as a sustainable strategy for hydrogen generation due to its abundant hydrogen source,utilization of inexhaustible solar energy,high-purity product,and environment-friendly process.To actualize a practical PEC water splitting,it is paramount to develop efficient,stable,safe,and low-cost photoelectrode materials.Recently,graphitic carbon nitride(g-C3N4)has aroused a great interest in the new generation photoelectrode materials because of its unique features,such as suitable band structure for water splitting,a certain range of visible light absorption,nontoxicity,and good stability.Some inherent defects of g-C3N4,however,seriously impair further improvement on PEC performance,including low electronic conductivity,high recombination rate of photogenerated charges,and limited visible light absorption at long wavelength range.Construction of g-C3N4-based nanosized heteroarrays as photoelectrodes has been regarded as a promising strategy to circumvent these inherent limitations and achieve the high-performance PEC water splitting due to the accelerated exciton separation and the reduced combination of photogenerated electrons/holes.Herein,we summarize in detail the latest progress of g-C3N4-based nanosized heteroarrays in PEC water-splitting photoelectrodes.Firstly,the unique advantages of this type of photoelectrodes,including the highly ordered nanoarray architectures and the heterojunctions,are highlighted.Then,different g-C3N4-based nanosized heteroarrays are comprehensively discussed,in terms of their fabrication methods,PEC capacities,and mechanisms,etc.To conclude,the key challenges and possible solutions for future development on g-C3N4-based nanosized heteroarray photoelectrodes are discussed. | Liqun Wang Wenping Si Yueyu Tong Feng Hou Daniele Pergolesi Jungang Hou Thomas Lippert Shi Xue Dou Ji Liang | 2020 | Carbon Energy2020,2,2: | 10 |
| 3 | Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics. | Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng | 2018 | Genes & Diseases2018,5,2: | 8 |
| 4 | Effect of NH_4F and Nano-SiO_2 on Morphological Control of α-Al_2O_3 Platelets via Solid-state Reaction显示文摘Alpha-alumina(α-Al_2O_3) platelets were prepared via solid-state reaction using pseudo-boehmite as the starting material, while NH_4F and nano-SiO_2 were used as additives, and nitric acid was used as the binder. The effect of these additives on properties of the alumina platelets was determined via scanning electron microscopy, X-ray diffraction, and measurements of the surface area and silicon content. A mechanism governing their role in the synthesis process was proposed. The results indicated that NH_4F could promote the formation of highly crystalline platelets. Addition of nano-SiO_2 could lead to increase in the diameter and reduction in the thickness of the platelets, and could also prevent their transformation to α-Al_2O_3. | 6 | Pore evolution characteristic of shale in the Longmaxi Formation,Sichuan Basin显示文摘Through the field emission scanning electronic microscope(FESEM)and the nitrogen adsorption test,pore type and structure of shale reservoir in the Longmaxi Formation in the Sichuan Basin were well studied.Result showed that the pore type includes organic pore,intercrystalline pore,dissolution intracrystalline pore and interparticle pore,and the organic pore was one of major pore types;among the organic pore,the micropore had large pore volume and specific surface area,and was the main storage space of shale gas.Through study on effect of total organic carbon(TOC),organic matter maturity(Ro),diagenesis and tectonism on shale porosity,influence of TOC on porosity could be divided into four stages:the rapid increasing stage(TOC from 0 to 2%),the slow decreasing stage(TOC from 2 to 3%),the rapid increasing stage(TOC from 3 to 4%or 6%)and the rapid decreasing stage(TOC>4%or 6%);influence of the maturity on porosity of shale could be divided into three stages:the rapid decreasing stage(Ro from 1.5 to 2.2%),the rapid increasing stage(Ro from 2.2 to 2.7%)and the rapid decreasing stage(Ro>2.7%);during the high thermal evolution stage,the organic diagenesis was stronger than the inorganic diagenesis;the tectonism had a great impact on porosity,and the more intense the tectonism was,the smaller the porosity would be.The evolution of shale porosity of the Longmaxi Formation underwent five stages:the immature rapid compaction stage(Ro<0.7%),the mature hydrocarbon generation and dissolution stage(Ro from 0.7 to 1.3%),the high mature pore closed stage(Ro from 1.3 to 2.2%),the overmature secondary pyrolysis stage(Ro from 2.2 to 2.7%)and the overmature slow compaction stage(Ro>2.7%);among which the mature hydrocarbon generation and dissolution stage and the overmature secondary pyrolysis stage were the most favorable shale pore development stages。 | Wenping Liu Jun Liu Molun Cai Chao Luo Xuewen Shi Jian Zhang | 2017 | Petroleum Research2017,2,4: | 3 |
| 7 | Application of contrast-enhanced ultrasound in minimally invasive ablation of benign thyroid nodules显示文摘Objective:This study aimed to investigate the application value of contrast-enhanced ultrasound(CEUS)before and after minimally invasive ablation procedures for benign thyroid nodule(s)(BTN).Methods:This prospective study included patients with BTNs scheduled to undergo ultrasound-guided minimally invasive ablation treatment.CEUS was performed before and after ablation(at 1 day,and 1,6,and 12 months after ablation).Changes in microvascular perfusion and the volume of BTNs were noted and assessed.Results:Sixty-two patients(62 BTNs),who underwent ablation procedures between June 2016 and August 2020,were included.All lesions were confirmed by biopsy,and histopathological results were obtained before ablation treatment.On preoperative CEUS,the lesions exhibited hyperenhancement(53.23%)or iso-enhancement(46.77%)during the arterial phase,and all lesions exhibited iso-enhancement in the venous and late phases.One day after ablation,none of the BTNs exhibited obvious enhancement on CEUS.One(1.61%)lesion was retreated due to a nodule-like enhancement area detected by CEUS at the 6-month follow-up.The mean nodular volume reduction rate(VRR)at 1,6,and 12 months follow-up demonstrated no significant difference between the two ablation groups(microwave ablation versus radiofrequency ablation).Twelve months after ablation,the mean(±SD)VRR of all BTNs was 60.3±10.3%.Conclusion:CEUS helped guide treatment decisions for BTNs before ablation treatment.Moreover,it could also be used to accurately and noninvasively evaluate treatment efficacy. | Jiaying Cao Peili Fan Feihang Wang Shuainan Shi Lingxiao Liu Zhiping Yan Yi Dong Wenping Wang | 2022 | Journal of Interventional Medicine2022,5,1: | 3 |

The ICP results indicated that the high temperature calcination process resulted in a large loss of lithium species because of volatilization, but there was still a small amount of residual lithium species in the α-Al_2O_3 platelets. The SEM micrographs showed that lithium nitrate led to decrease in the thickness of α-Al_2O_3 platelets and irregular morphology of aggregates. Pore structures results exhibited that addition of lithium nitrate led to decrease in the pore size and increase in the specific surface area of aggregates of α-Al_2O_3 platelets. The XRD and IR patterns suggested that the residual lithium and aluminum oxide formed LiAl_5O_8.
