| 1 | Platinum catalysts supported on Nafion functionalized carbon black for fuel cell application显示文摘We report a high performance supported Pt catalyst, in which a perfluorosulfonic acid (Nafion) functionalized carbon black is used as support. The catalyst is characterized by infrared spectroscopy (IR), transmission electron microscopy (TEM) and X-ray diffraction (XRD). The TEM image shows that the active Pt component is in nanoparticles and highly dispersed on the carbon black with an average particle size of 1.9 nm. The catalyst shows improved activity towards the methanol anodic oxidation and oxygen reduction reaction (ORR), resulting from the high dispersion of active Pt component. It leads to increases in electrochemically accessible surface areas and ion channels, as well as easier chargetransfer at polymer/electrolyte interfaces. | 2 | Vapor phase fabrication of three-dimensional arrayed BiI3 nanosheets for cost-effective solar cells显示文摘Multilayered photovoltaic absorbers have triggered widespread attention for their unique structure and properties.However,multilayered materials in the randomly oriented polycrystalline thin-film lead to ineffective carrier transport and collection,which hinders the process of achieving high-performance solar cells.Herein,this issue is tackled by producing the three-dimensional(3D)heterojunction BiI3 nanosheets(NSs)solar cells,which embed vertically aligned monocrystalline BiI3 NSs into spiro-OMeTAD.The preferred orientation of BiI3 NSs and large p-n junction areas of 3D heterojunction structure enable a strong light absorption and effective carrier transport and collection,and thus a power conversion efficiency(PCE)of 1.45%was achieved.Moreover,this PCE is the highest ever reported for BiI3 based solar cells to our best knowledge.Moreover,the nonencapsulated device remained 96%of the initial PCE after 24 h continuous one sun illumination at^70%humidity condition,and 82%of the initial PCE after 1-month storage at^30%humidity condition. | Yiyi Zhu Qianpeng Zhang Matthew Kam Swapnadeep Poddar Leilei Gu Shijun Liang Pengfei Qi Feng Miao Zhiyong Fan | 2020 | InfoMat2020,2,5: | 2 |

| 4 | Correlation between pathogenesis of dampness syndrome and Interleukin-2,Interleukin-8 in rats显示文摘OBJECTIVE: To explore the pathogenesis of dampness syndrome by detecting the changes of interleukin-2 (IL-2) and interleukin-8 (IL-8) levels. METHODS: Female Sprague Dawley rats were divided randomly into five groups according to the random number table: a normal group (GroupⅠ), an external dampness group (GroupⅡ), an internal dampness group (Group Ⅲ), and an external and internal dampness group (GroupⅣ). Twenty days after the model made, IL-2 and IL-8 levels were detected by radioimmunoassay method. RESULTS: The IL-2 and IL-8 levels among groups were significant (F=3.102, P<0.05; F=2.657, P<0.05, respectively). The level of IL-2 in Group Ⅱ and Group Ⅲ were higher than that in GroupⅠ(P<0.05, P<0.01, respectively), especially higher in the Group Ⅲ compared with GroupⅡ (P<0.05). The lev- el of IL-8 in Group Ⅲ were higher than those inGroupⅠ, GroupⅡ and GroupⅣ (P<0.05, P<0.01, P< 0.05, respectively). In the GroupⅢ, the 24-hour wa ter and body weight were higher than that in the GroupⅣ (all P<0.05), and spontaneous movemen frequency was higher than those in GroupⅡ and GroupⅣ (P<0.05). | 5 | A Fragment Substitution in Promoter of MS92/PTC1 Causes Male Sterility in Rice显示文摘Persistent tapetal cell1(PTC1) plays a curial role in pollen development, and is thought to function as a transcriptional activator in rice. However, the molecular mechanism of PTC1 in regulating pollen development and its cis-elements are not well understood. We identified a novel weak male sterility mutant(ms92) which exhibited expanded tapetum and shrink pollen grains. Map-based cloning and allelic analysis suggested that the male sterility of ms92 was caused by a DNA fragment substitution in the promoter of PTC1. The decreased expression of MS92/PTC1 in ms92 and cis-element analysis indicated that the substituted sequence contained several potential binding cis-element of negative feedback. MS92/PTC1 was specifically expressed in tapetum and microspores at the young microspore stage, and its protein was localized in nucleus. We further found that MS92/PTC1 functions as a transcription activator by recognizing H3K4me3. Transcriptomic analysis revealed that a number of genes involved in tapetum degeneration and pollen wall formation were down-regulated in ms92, which are the potential targets of MS92/PTC1. | 8 | Glutamine mitigates murine burn sepsis by supporting macrophage M2 polarization through repressing the SIRT5-mediated desuccinylation of pyruvate dehydrogenase显示文摘Background:Alternative(M2)-activated macrophages drive the anti-inflammatory response against sepsis,a leading cause of death in patients suffering from burn injury.Macrophage M2 polarization is intrinsically linked with dominant oxidative phosphorylation(OXPHOS).Glutamine serves as a major anaplerotic source to fuel OXPHOS,but it remains unknown whether glutamine can modulate metabolic checkpoints in OXPHOS that favour M2 polarization.The study aims to explore whether glutamine essentially supports M2 polarization in IL-4-stimulated murine macrophages by sustaining the activity of PDH and whether glutamine augments macrophage M2 polarization and thus alleviates inflammation and organ injury in a murine burn sepsis model.Methods:To understand how glutamine promotes M2 activation in interleukin(IL-4)-treated murine macrophages,we detected glutamine-dependent M2 polarization and its relationship with the pyruvate dehydrogenase(PDH)complex by RT-PCR,flow cytometry and western blot.To explore how glutamine modulates PDH activity and thus supports M2 polarization,we compared the expression,phosphorylation and succinylation status of PDHA1 and then examined sirtuin SIRT5-dependent desuccinylation of PDHA1 and the effects of SIRT5 overexpression on M2 polarization by RT-PCR,flow cytometry and western blot.To determine whether glutamine or its metabolites affect M2 polarization,macrophages were cocultured with metabolic inhibitors,and then SIRT5 expression and M2 phenotype markers were examined by RT-PCR,flow cytometry and western blot.Finally,to confirm the in vivo effect of glutamine,we established a burn sepsis model by injecting Pseudomonas aeruginosa into burn wounds and observing whether glutamine alleviated proinflammatory injuries by RT-PCR,flow cytometry,western blot,immunofluorescent staining,hematoxylin-eosin staining and enzyme-linked immuno sorbent assay.Results:We showed that consumption of glutamine supported M2 activation in IL-4-treated murine macrophages by upregulating the activity of PDH.Mechanistically,glutamine did not affect the expression or alter the phosphorylation status of PDHA1 but instead downregulated the expression of SIRT5 and repressed SIRT5-dependent desuccinylation on PDHA1,which in turn recovered PDH activity and supported M2 polarization.This effect was implemented by its secondary metaboliteα-ketoglutarate(αKG)rather than glutamine itself.Finally,we demonstrated that glutamine pro-moted macrophage M2 polarization in a murine burn sepsis model,thereby repressing excessive inflammation and alleviating organ injury in model mice.Conclusions:Glutamine mitigates murine burn sepsis by essentially supporting macrophage M2 polarization,with a mechanism involving the repression of the SIRT5-mediated desuccinylation of pyruvate dehydrogenase that replenishes OXPHOS and sustains M2 macrophages. | Yuanfeng Zhu Xiaoli Chen Yongling Lu Lin Xia Shijun Fan Qianying Huang Xin Liu Xi Peng | 2022 | Burns & Trauma2022,10,1: | 1 |
| 9 | Origin and chromatin remodeling of young X/Y sex chromosomes in catfish with sexual plasticity显示文摘Assembly of a complete Y chromosome is a significant challenge in animals with an XX/XY sex-determination system.Re cently,we created YY-supermale yellow catfish by crossing XY males with sex-reversed XY females,providing a valuable model for Y-chromosome assembly and evolution.Here,we assembled highly homomorphic Y and X chromosomes by sequencing genomes of the YY supermale and XX female in yellow catfish,reve aling their nucleotide divergences with only less than 1%and with the same gene compositions.The sex-determining region(SDR)was identified to locate within a physical distance of0.3 Mb by FSTscanning.Strikingly,the incipient sex chromosomes were revealed to originate via autosome-autosome fusion and were characterized by a highly rearranged region with an SDR downstream of the fusion site.We found that the Y chromosome was at a very early stage of differentiation,as no clear evidence of evolutionary strata and classical structure features of recombination suppression for a rather late stage of Y-chromosome evolution were observed.Significantly,a number of sex-antagonistic mutations and the accumulation of repetitive elements were discovered in the SDR,which might be the main driver of the initial establishment of recombination suppression between young X and Y chromosomes.Moreover,distinct three-dimensional chromatin organizations of the Y and X chromosomes were identified in the YY supermales and XX females,as the X chromosome exhibited denser chromatin structure than the Y chromosome,while they respectively have significantly spatial interactions with female-and male-related genes compared with other autosomes.The chromatin configuration of the sex chromosomes as well as the nucleus spatial organization of the XX neomale were remodeled after sex reversal and similar to those in YY supermales,and a male-specific loop containing the SDR was found in the open chromatin region.Our results elucidate the origin of young sex chromosomes and the chromatin remodeling configuration in the catfish sexual plasticity. | Gaorui Gong Yang Xiong Shijun Xiao Xi-Yin Li Peipei Huang Qian Liao Qingqing Han Qiaohong Lin Cheng Dan Li Zhou Fan Ren Qi Zhou Jian-Fang Gui Jie Mei | 2023 | National Science Review2023,10,2: | 0 |
