1Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. 2Climate and litter C/N ratio constrain soil organic carbon accumulation显示文摘Soil organic carbon(SOC) plays critical roles in stabilizing atmospheric CO2 concentration, but the mechanistic controls on the amount and distribution of SOC on global scales are not well understood. In turn, this has hampered the ability to model global C budgets and to find measures to mitigate climate change. Here, based on the data from a large field survey campaign with 2600 plots across China’s forest ecosystems and a global collection of published data from forested land, we find that a low litter carbon-to-nitrogen ratio(C/N) and high wetness index(P/PET, precipitation-to-potentialevapotranspiration ratio) are the two factors that promote SOC accumulation, with only minor contributions of litter quantity and soil texture. The field survey data demonstrated that high plant diversity decreased litter C/N and thus indirectly promoted SOC accumulation by increasing the litter quality. We conclude that any changes in plant-community composition, plant-species richness and environmental factors that can reduce the litter C/N ratio, or climatic changes that increase wetness index, may promote SOC accumulation. 4Solidification Simulation of Investment Castings of Single Crystal Hollow Turbine Blade显示文摘The three-dimensional solidification simulation of the investment castings of single crystal hollow turbine blade at the withdrawal rates of 2 mm/min, 4.5 mm/min and 7 mm/min has been performed with the finite element thermal analysis. The calculated results are in accordance with the experimental ones. The results show that with the increase of withdrawal rate the concave curvature of the liquidus isotherm is larger and larger, and the temperature gradients of the blades increase. No effects of withdrawal rate on the distribution of the temperature gradients of the starter and helical grain selector of the blades are observed at withdrawal rates of 2 mm/min, 4.5 mm/min and 7 mm/min. 5Effects of Rhenium on Creep Rupture Life of a Single Crystal Superalloys显示文摘The slabs of a single crystal superalloy were poured with 2.2% Re, 3.2% Re and 4.2% Re and otherelement contents of the alloys were basicalIy kept no changed. The effects of Re on the ultimatetensile strength, creep rupture life and rafting of a single crystal superalloy were studied. 7Chimeric flagellin expressed by Salmonella typhimurium induces an ESAT-6-specific Th1-type immune response and CTL effects following intranasal immunization显示文摘The flagellin component FliC of Salmonella typhimurium is capable of activating the innate immune system via specific interactions with TLR5 and can also act as a carrier of foreign antigen to elicit antigen-specific immune responses.Thus,we constructed an attenuated Salmonella strain SL5928(fliC/esat)expressing chimeric flagellin that contained the ESAT-6 antigen coding sequence of Mycobacterium tuberculosis inserted into the highly variable region of the Salmonella flagellin coding gene fliCi.The chimeric flagellin functioned normally,as demonstrated using a flagella swarming assay and electron microscopy.To analyze the effects of chimeric flagellin,the cell-mediated immune response and cytotoxic T lymphocyte(CTL)effects specific for ESAT-6 antigen were tested after intranasal immunization of mice with flagellated Salmonella SL5928(fliC/esat).The results showed that SL5928(fliC/esat)intranasal immunization can strongly elicit an ESAT-6-specific T helper(Th)1-type immune response in mucosal lymphoid tissues,such as nasopharynx-associated lymph nodes,lung and Peyer’s patches,and a Th1/Th2 response was elicited in spleen and mesenteric lymph nodes.Furthermore,intranasal immunization of SL5928(fliC/esat)produced efficient CTL effects,as demonstrated using a 5-and 6-carboxyfluorescein diacetate succinimidyl ester(CFSE)assay.Thus,our study revealed that Salmonella flagellin acts as a carrier for foreign antigen and triggers strong Th1 and CTL responses during intranasal immunization.Chimeric flagellin is potentially an effective strategy for the development of novel vaccines against tuberculosis in humans and animals.Hui Zhang Liu Liu Ke Wen Jinlin Huang Shizhong Geng Junsong Shen Zhiming Pan Xinan Jiao 2011Cellular & Molecular Immunology2011,8,6:6
8CO_2 efflux from different forest soils and impact factors in Dinghu Mountain, China显示文摘CO2 fluxes from soils and related environmental factors were measured in three forest ecosystems of Dinghu Mountain using static chamber-gas chromatograph technique for one year. The seasonal pattern of CO2 flux, contribution of litter on total CO2 flux and the correlations of CO2 flux with soil temperature and soil water content were examined for each type of forest. The results were given as followings: (1) The seasonal patterns of CO2 flux from soil of the three types of forest were similar, with a higher CO2 flux in rainy season than in dry season. The comparative relations of mean annual CO2 fluxes between the three sites were expressed as:monsoon forest > mixed forest > pine forest. (2) CO2 fluxes from litter decomposition in monsoon forest, mixed forest and pine forest accounted for 24.43%, 41.75% and 29.23% of the corresponding total CO2 fluxes from forest floor, respectively. (3) Significant relationships were found between CO2 fluxes and soil temperatures at 5 cm depth for the three types of forest, which could be best described by exponential equations. The calculated Q10 values based on soil temperature at 5 cm depth ranged from 1.86 to 3.24. 9Kernel selection with spectral perturbation stability of kernel matrix显示文摘Kernel selection is one of the key issues both in recent research and application of kernel methods.This is usually done by minimizing either an estimate of generalization error or some other related performance measure. Use of notions of stability to estimate the generalization error has attracted much attention in recent years. Unfortunately, the existing notions of stability, proposed to derive the theoretical generalization error bounds, are difficult to be used for kernel selection in practice. It is well known that the kernel matrix contains most of the information needed by kernel methods, and the eigenvalues play an important role in the kernel matrix. Therefore, we aim at introducing a new notion of stability, called the spectral perturbation stability,to study the kernel selection problem. This proposed stability quantifies the spectral perturbation of the kernel matrix with respect to the changes in the training set. We establish the connection between the spectral perturbation stability and the generalization error. By minimizing the derived generalization error bound, we propose a new kernel selection criterion that can guarantee good generalization properties. In our criterion,the perturbation of the eigenvalues of the kernel matrix is efficiently computed by solving the derivative of a newly defined generalized kernel matrix. 10Effects of precipitation on soil organic carbon fractions in three subtropical forests in southern China显示文摘Aims The aim of this study was to investigate the effects of precipitation changes on soil organic carbon(SOC)fractions in subtropical forests where the precipitation pattern has been altered for decades.Methods We conducted field manipulations of precipitation,including ambient precipitation as a control(CK),double precipitation(DP)and no precipitation(NP),for 3 years in three forests with different stand ages(broadleaf forest[BF],mixed forest[MF]and pine forest[PF])in subtropical China.At the end of the experiment,soil samples were collected to assay SOC content,readily oxidizable organic carbon(ROC)and non-readily oxidizable organic carbon(NROC),as well as soil microbial biomass carbon(MBC),pH and total nitrogen content.Samples from the forest floors were also collected to analyze carbon(C)functional groups(i.e.alkyl C,aromatic C,O-alkyl C and carbonyl C).Furthermore,fine root biomass was measured periodically throughout the experiment.Important Findings Among the forests,ROC content did not exhibit any notable differences,while NROC content increased significantly with the stand age.This finding implied that the SOC accumulation observed in these forests resulted from the accumulation of NROC in the soil,a mechanism for SOC accumulation in the mature forests of southern China.Moreover,NP treatment led to significant reductions in both ROC and NROC content and therefore reduced the total SOC content in all of the studied forests.Such decreases may be due to the lower plant-derived C inputs(C quantity)and to the changes in SOC components(C quality)indicated by C functional groups analyses under NP treatment.DP treatment in all the forests also tended to decrease the SOC content,although the decreases were not statistically significant with the exception of SOC and ROC content in PF.This finding indicated that soils in MF and in BF may be more resistant to precipitation increases,possibly due to less water limitations under natural conditions in the two forests.Our results therefore highlight the different responses of SOC and its fractions to precipitation changes among the forests and suggest that further studies are needed to improve our understanding of SOC dynamics in such an important C sink region.Xiaomei Chen Deqiang Zhang Guohua Liang Qingyan Qiu Juxiu Liu Guoyi Zhou Shizhong Liu Guowei Chu Junhua Yan 2016Journal of Plant Ecology2016,9,1:4
11Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4

14Optical bandgap energy of CH3NH3PbI3 perovskite studied by photoconductivity and reflectance spectroscopy显示文摘Organic-inorganic hybrid perovskite materials have been receiving considerable attention due to their promising applications in many optoelectronic fields. However, the optical bandgap of CH_3NH_3PbI_3 perovskite is still disputed. In order to comprehend the intrinsic characteristics of these materials, we study the near-band-edge optical responses of the solution-processed CH_3NH_3PbI_3 perovskite crystals by photoconductivity and reflectance spectroscopy at room temperature. All these spectra of the CH_3NH_3PbI_3 polycrystals show only one significant absorption edge at 1.58 eV. However, we observe an extra absorption edge at 1.47 eV in CH_3NH_3PbI_3 single crystal. We establish a simple kinetic model of charge annihilation processes to explain why the low-energy structure of the photoconductivity and diffuse reflectance spectra of CH_3NH_3PbI_3 polycrystals disappear, which provides another possibility for understanding the difference in absorption edge. 16Low cycle fatigue properties and microstructure evolution at 760℃ of a single crystal superalloy显示文摘Low cycle fatigue(LCF) behavior of a single crystal superalloy was investigated at 760 ℃. Microstructure evolution and fracture mechanism were studied by scanning electron microscopy(SEM) and transmission electron microscopy(TEM), respectively. The results show that the fatigue data fluctuation was small and the fatigue parameters of the alloy had been determined. On increasing the cyclic number, the alloy initially showed slight cyclic softening at the early two or three cycles and slowly hardened to some extent afterwards, then kept stable for the most of the remaining fatigue life. The LCF of the alloy at 760 ℃ can be attributed to the main elastic damage in fatigue processing. The initiation site of fatigue crack was at or near the surface of the samples. Crack propagated perpendicularly to the loading direction at first and then along {111}octahedral slip planes. The fatigue fracture mechanism was quasi-cleavage fracture. The γ'phase morphology still maintained cubic shape after fracture. There were a number of slip bands shear the γ'precipitates and γ matrix near the fracture surface of the specimen. 18A Reconfigurable Microstrip Patch Antenna with Frequency and Circular Polarization Diversities显示文摘A novel microstrip patch antenna with both frequency and circular polarization diversities is presented. The proposed antenna consists of two parts, a patch with two orthogonal slots on the upper substrate and a3 d B hybrid coupler fed structure which printed on the lower substrate. Two groups of diodes placed across the orthogonal slots are utilized to achieve frequency reconfiguration. Another two diodes, placed between the input port of the antenna and the 3d B hybrid coupler, are utilized to switch between Left-hand circular polarization(LHCP)and Right-hand circular polarization(RHCP). Full wave simulation is used for the antenna analysis, and a prototype of the antenna with an integrated Direct current(DC) biasing circuit has been fabricated and measured. 19China’s bioenergy industry development roadmap显示文摘Positive development of renewable energy, saving and substitution of fossil energy, promotion of the energy structure adjustment are the inevitable strategy choices of China’s sustainable development. This paper discussed the China’s bioenergy resources status, development targets and technology development roadmaps. China has 136.140 million hm2 of marginal land, which distribute mainly in western and northern regions. There are 1 billion t of crop residues and forestry waste annually, and 300 million t can be used to produce different kinds of bioenergies. And organic waste and manure can generate 50 billion m3 of biogas. The discussed development target indicated that it can construct a biomass oilfield with the capacity of 100 million t/year and reduce 200 million t of CO_2 emission by 2020. The bioenergy technology development roadmap indicated that the bioethanol mainly uses non grain starch and hemicellulose products as raw materials in the near-term (2006-2010).
