| 1 | Chimeric flagellin expressed by Salmonella typhimurium induces an ESAT-6-specific Th1-type immune response and CTL effects following intranasal immunization显示文摘The flagellin component FliC of Salmonella typhimurium is capable of activating the innate immune system via specific interactions with TLR5 and can also act as a carrier of foreign antigen to elicit antigen-specific immune responses.Thus,we constructed an attenuated Salmonella strain SL5928(fliC/esat)expressing chimeric flagellin that contained the ESAT-6 antigen coding sequence of Mycobacterium tuberculosis inserted into the highly variable region of the Salmonella flagellin coding gene fliCi.The chimeric flagellin functioned normally,as demonstrated using a flagella swarming assay and electron microscopy.To analyze the effects of chimeric flagellin,the cell-mediated immune response and cytotoxic T lymphocyte(CTL)effects specific for ESAT-6 antigen were tested after intranasal immunization of mice with flagellated Salmonella SL5928(fliC/esat).The results showed that SL5928(fliC/esat)intranasal immunization can strongly elicit an ESAT-6-specific T helper(Th)1-type immune response in mucosal lymphoid tissues,such as nasopharynx-associated lymph nodes,lung and Peyer’s patches,and a Th1/Th2 response was elicited in spleen and mesenteric lymph nodes.Furthermore,intranasal immunization of SL5928(fliC/esat)produced efficient CTL effects,as demonstrated using a 5-and 6-carboxyfluorescein diacetate succinimidyl ester(CFSE)assay.Thus,our study revealed that Salmonella flagellin acts as a carrier for foreign antigen and triggers strong Th1 and CTL responses during intranasal immunization.Chimeric flagellin is potentially an effective strategy for the development of novel vaccines against tuberculosis in humans and animals. | Hui Zhang Liu Liu Ke Wen Jinlin Huang Shizhong Geng Junsong Shen Zhiming Pan Xinan Jiao | 2011 | Cellular & Molecular Immunology2011,8,6: | 6 |

| 9 | Identification of New Resistance Loci Against Sheath Blight Disease in Rice Through Genome-Wide Association Study显示文摘Sheath blight(SB) caused by the soil borne pathogen Rhizoctonia solani is one of the most serious global rice diseases. Breeding resistant cultivar is the most economical and effective strategy to control the disease. However, no rice varieties are completely resistant to SB, and only a few reliable quantitative trait loci(QTLs) linked with SB resistance have been identified to date. In this study, we conducted a genome-wide association study(GWAS) of SB resistance using 299 varieties from the rice diversity panel 1(RDP1) that were genotyped using 44 000 high-density single nucleotide polymorphism(SNP) markers. Through artificial inoculation, we found that only 36.5% of the tested varieties displayed resistance or moderate resistance to SB. In particular, the aromatic and aus sub-populations displayed higher SB resistance than the tropical japonica(TRJ), indica and temperate japonica sub-populations. Seven varieties showed similar resistance levels to the resistant control YSBR1. GWAS identified at least 11 SNP loci significantly associated with SB resistance in the three independent trials, leading to the identification of two reliable QTLs, qSB-3 and qSB-6, on chromosomes 3 and 6. Using favorable alleles or haplotypes of significantly associated SNP loci, we estimated that both QTLs had obvious effects on reducing SB disease severity and can be used for enhancing SB resistance, especially in improving SB resistance of TRJ sub-population rice varieties. | 10 | Self-supported hierarchical porous Li_(4)Ti_(5)O_(12)/carbon arrays for boosted lithium ion storage显示文摘The development of fast rechargeable lithium ion batteries(LIBs)is highly dependent on the innovation of advanced high-power electrode materials.In this work,for the first time,we report a sacrificial NiO arrays template method for controllable synthesis of self-supported hierarchical porous Li_(4)Ti_(5)O_(12)/C(LTO/C)nanoflakes arrays,for use as fast rechargeable anodes for LIBs.The ultrathin(2-3 nm)carbon layer was uniformly coated on the LTO forming arrays architecture.The hierarchical porous LTO/C nanoflakes consisted of primary cross-linked nanoparticles of 50-100 nm and showed large porosity.Because of the enhanced electrical conductivity and accelerated ion transfer channels,the well-designed binderfree porous LTO/C nanoflakes arrays exhibited notable high-rate lithium ion storage performance with smaller polarization,better electrochemical reactivity,higher specific capacity(157 mAh g^(-1) at the current density of 20C)and improved long-term cycling life(96.2% after 6000 cycles at 20C),superior to the unmodified porous LTO arrays counterpart(126 mAh g^(-1) at 20C and 88.0%after 6000 cycles at 20C).Our work provides a new template for the construction of high-performance high-rate electrodes for electrochemical energy storage. | Jun Liu Aixiang Wei Guoxiang Pan Shenghui Shen Zhiming Xiao Yu Zhao Xinhui Xia | 2021 | Journal of Energy Chemistry2021,30,3: | 2 |

| 17 | Detection of cotton waterlogging stress based on hyperspectral images and convolutional neural network显示文摘Waterlogging in the early stage of cotton will reduce the number of bolls and do harm to yield.Early detection of waterlogging will help farmers to adjust cotton management and save the loss.To evaluate the application of deep learning for the detection of early waterlogging,this study applied a convolutional neural network(CNN)to classify different durations of waterlogging stress(0,2,4,6,8,10 d)based on hyperspectral images(HSIs)of cotton leaves.An experiment was designed to simulate the situation of cotton under waterlogging stress and collect HSIs of visible and near-infrared(VNIR 450-950 nm)spectra with 126 bands 66 d after cotton sowing(66 DAS).It was found the spectral curve reflectance of waterlogging cotton was higher than that of non-waterlogging cotton.Especially near 550 nm and 750 nm,and the spectral curve increased with durations of waterlogging stress and there were‘blue shift’phenomena for the position of the red edge of the spectra.The first principal components of HSIs after band randomly discarding and principal component analysis(PCA)were used to build a dataset.GoogLeNet Inception-v3(GLNI-v3)and VGG-16 models were selected to detect cotton waterlogging stress with the dataset.The results showed that the average time for a round training for GLNI-v3 was 13.337 s,with a classification accuracy of 96.95%and a loss value of 0.09431.The average time for a round training for VGG-16 was 21.470 s,with a classification accuracy of 97.00%and a loss value of 0.17912.Though these two models had similar classification accuracy and loss value,GLNI-v3 achieved a high accuracy with fewer training iterations.The durations of waterlogging stress of cotton in a short-term can be detected by HSIs of cotton leaves and CNN models are suitable for the classification of HSIs,and this method can provide support for cotton yield estimation and loss assessment after waterlogging. | Jing Zhao Fangjiang Pan Zhiming Li Yubin Lan Liqun Lu Dongjian Yang Yuting Wen | 2021 | International Journal of Agricultural and Biological Engineering2021,14,2: | 1 |
