| 1 | Incidence and mortality of colorectal cancer in China, 2011显示文摘Objective: Colorectal cancer is the third most common type of cancer and the fourth leading cause of cancer-related death in the world. This article provides the most up-to-date overview of colorectal cancer burden in China.Methods: Totally 234 cancer registries submitted data of 2011 to the National Central Cancer Registry(NCCR). Qualified data from 177 registries was pooled and analyzed. The crude incidence and mortality rates of colorectal cancer were calculated by age, gender and geographic area. The numbers of new cases and deaths were estimated using the 5-year age-specific cancer incidence/mortality rates and the corresponding populations. China census in 2000 and Segi's world population were applied for age standardized rates.Results: The estimate of new cases diagnosed with colorectal cancer of China in 2011 was 310,244(178,404 for males and 131,840 for females, 195,117 in urban areas and 115,128 in rural areas), accounting for 9.20% of overall new cancer cases. The crude incidence of colorectal cancer ranked fourth in all cancer sites with rate of 23.03/100,000(25.83/100,000 for males and 20.08/100,000 for female, 28.25/100,000 in urban areas and 17.54/100,000 in rural areas). The age-standardized rates by China population and by World population were 16.79/100,000 and 16.52/100,000, respectively. The estimated number of colorectal cancer deaths of China in 2011 was 149,722(86,427 for males and 63,295 for females, 91,682 in urban areas and 58,040 in rural areas), accounting for 7.09% of overall cancer deaths. The crude mortality rate for colorectal cancer ranked fifth leading cause of cancer-related death in all cancer sites with rate of 11.11/100,000(12.51/100,000 for males and 9.64/100,000 for female, 13.27/100,000 in urban areas and 8.84/100,000 in rural areas). The age-standardized rates by China population and by World population for mortality were 7.77/100,000 and 7.66/100,000, respectively. For both of incidence and mortality, the rates of colorectal cancer were much higher in males than in females, and in rural areas than in urban areas. The rate of colorectal cancer increased greatly with age, especially after 40 or 45 years old.Conclusions: Colorectal cancer is a relative common cancer in China, especially for males in urban areas. | 2 | Primary adenocarcinomas of lower esophagus,esophagogastric junction and gastric cardia:in special reference to China显示文摘Gastric cardia adenocarcinoma (GCA) is an under-studied subject. The pathogenesis, molecular changes in the early stage of carcinogenesis and related risk factors have not been well characterized. There is evidence, however, that GCA differs from cancer of the rest of the stomach in terms of natural history and histopathogenesis. Adenocarcinomas of the lower esophagus, esophagogastric junction (EGJ)and gastric cardia have been given much attention because of their increasing incidences in the past decades, which is in striking contrast with the steady decrease in distal stomach adenocarcinoma. In China, epidemiologically, GCA shares very similar geographic distribution with esophageal squamous cell carcinoma (SCC), especially in Linzhou (formerly Linxian County), Henan Province, North China,the highest incidence area of esophageal SCC in the world.Historically, both GCA and SCC in these areas were referred to as esophageal cancer (EC) by the public because of the common syndrome of dysphagia. In Western countries,Barrett's esophagus is very common and has been considered as an important precancerous lesion of adenocarcinoma at EGJ. Because of the low incidence of Barrett's esophagus in China, it is unlikely to be an important factor in early stage of EGJ adenocarcinoma development.However, Z line up-growth into lower esophagus may be one of the characteristic changes in these areas in early stage of GCA development. Whether intestinal metaplasia (IM) is a premalignant lesion for GCA is still not clear. Higher frequency of IM observed at adjacent GCA tissues in Henan suggests the possibility of IM as a precancerous lesion for GCA in these areas. Molecular information on GCA,especially in early stage, is very limited. The accumulated data about the changes of tumor suppressor gene, such as p53 mutation, and ontogeny, such as C-erbB2, especially the similar alterations in GCA and SCC in the same patient,indicated that there might be some similar risk factors,such as nitrosamine, involved in both GCA and SCC in Henan population. | 3 | CYP1A1,GSTs and mEH polymorphisms and susceptibility to esophageal carcinoma:Study of population from a high-incidence area in north China显示文摘AIM: To characterize cytochrome P4501A1 (CYPIA1), glutathione S-transferases (GSTs) and microsomal epoxide hydrolase (mEH) polymorphisms in Chinese esophageal cancer patients. METHODS: Multiplex polymerase chain reaction (PCR) and PCR based restriction fragment length polymorphisms (PCRRFLP) were used to detect polymorphism changes of CYP,GSTs and mEH on esophageal cancerous and precancerous lesions as well as in case control group. All the examination samples were obtained from Linzhou (formerly Linxian), Henan Province, the highest incidence area for esophageal. RESULTS: The frequency of CYP1A1 3'' polymorphism in case control group (26/38, 68 %) was significantly higher than in esophageal squamous cell carcinoma^roup (ESCC) (29/62, 47 %) (P<0.05). A significant difference in the incidence of mEH slow allele variant was observed between case control group (15/38, 39 %) and esophageal dysplasiagroup (22/32, 69 %) or ESCC group (39/62, 63 %) (P<0.05). However, no significant difference was observed among different groups in the polymorphisms of CYPIA1 exon 7, GSTM1, GSTT1, GSTP1 and mEH fast allele. | 4 | An integrated approach to the detection of colorectal cancer utilizing proteomics and bioinformatics显示文摘AIM: To find new potential biomarkers and to establish patterns for early detection of colorectal cancer.METHODS: One hundred and eighty-two serum samples including 55 from colorectal cancer (CRC) patients, 35 from colorectal adenoma (CRA) patients and 92 from healthy persons (HP) were detected by surface-enhanced laser desorption/ionization mass spectrometry (SELDI-MS). The data of spectra were analyzed by bioinformatics tools like artificial neural network (ANN) and support vector machine (SVM).RESULTS: The diagnostic pattern combined with 7 potential biomarkers could differentiate CRC palJents from CRA patients with a specificity of 83%, sensitivity of 89% and positive predictive value of 89%. | 5 | Honokiol induces apoptosis through p53-independent pathway in human colorectal cell line RKO显示文摘AIM: To investigate the signal pathway of honokiol-induced apoptosis on human colorectal carcinoma RKO cells and to evaluate whether p53 and p53-related genes were involved in honokiol-treated RKO cells.METHODS: Cell cycle distribution and subdiploid peak were analyzed with a flow cytometer and DNA fragment with electrophoresis on agarose gels. Transcriptional level of Bax, Bcl-2, Bid and Bcl-xl was accessed by RT-PCR.Western blotting was used to measure p53 protein expression and other factors related to apoptosis.Proliferation inhibition of two cell lines (RKO, SW480) with high expression of p53 and one cell line with p53 negative expression (LS180) was monitored by MTr assay.RESULTS: Honokiol induced RKO cell apoptosis in a dosedependent manner. The mRNA expression level and proteinlevel of Bid were up-regulated while that of Bcl-xl wasdown-regulated, but no changes in Bax and Bcl-2 were observed. Western blotting showed p53 expression had no remarkable changes in honokiol-induced RKO cell apoptosis. | 6 | Honokiol:A potent chemotherapy candidate for human colorectal carcinoma显示文摘AIM: To investigate the anticancer activity of Honokiol on RKO,a human colorectal carcinoma cell line in vitro and in vivo,and to evaluate its possible use in clinic.METHODS: In vitro anticancer activity of honokiol was demonstrated by its induction of apoptosis in tumor cells.We analyzed cell proliferation with MTT assay, cell cycle with flow cytosmeter, DNA fragment with electrophoresis on agarose gels. To test the mechanism of honokiol-induced apoptosis, Westem blotting was used to investigate the factors involved in this process. The pharmacokinetics study of honokiol was tested by high phase liquid chromatography.In in vivo study, Balb/c nude mice were incubated with RKO cells. Honokiol was injected intraperitoneally every other day into tumor bearing Balb/c nude mice.RESULTS: Our results showed that honokiol induced apoptosis of RKO cells in a time- and dose-dependent manner. At 5-10 ug/mL for 48 h, honokiol induced apoptosis through activating Caspase cascades. Pharmacokinetics study demonstrated that, honokiol could be absorbed quickly by intraperitoneal injection, and maintained in plasma for more than 10 h. | 7 | Annual cost of illness of stomach and esophageal cancer patientsin urban and rural areas in China: A multi-center study显示文摘Objective: Stomach and esophageal cancer are imposing huge threats to the health of Chinese people whereas there were few studies on the financial burden of the two cancers.Methods: Costs per hospitalization of all patients with stomach or esophageal cancer discharged between September 2015 and August 2016 in seven cities/counties in China were collected, together with their demographic information and clinical details. Former patients in the same hospitals were sampled to collect information on annual direct non-medical cost, indirect costs and annual number of hospitalization. Annual direct medical cost was obtained by multiplying cost per hospitalization by annual number of hospitalization. Annual cost of illness(ACI)was obtained by adding the average value of annual direct medical cost, direct non-medical cost and indirect cost,stratified by sex, age, clinical stage, therapy and pathologic type in urban and rural areas. Costs per hospitalization were itemized into eight parts to calculate the proportion of each part. All costs were converted to 2016 US dollars(1 USD=6.6423 RMB).Results: Totally 19,986 cases were included, predominately male. Mean ages of stomach cancer and urban patients were lower than that of esophageal cancer and rural patients. ACI of stomach and esophageal cancer patients were $10,449 and $13,029 in urban areas, and $2,927 and $3,504 in rural areas, respectively. Greater ACI was associated with male, non-elderly patients as well as those who were in stage I and underwent surgeries.Western medicine fee took the largest proportion of cost per hospitalization.Conclusions: The ACI of stomach and esophageal cancer was tremendous and varied substantially among the population in China. | 8 | Protective effect of periplaneta americana extract on intestinal mucosal barrier function in patients with sepsis显示文摘OBJECTIVE: To investigate the effect of the periplaneta americana extract on the intestinal mucosal barrier and prognostic implications in patients with sepsis. METHODS: Sixty and six patients with sepsis were assigned randomly to treatment group (32 cases) and control group (32 cases). The extractfrom periplaneta americana plus conventional medication for sepsis was administered to the treatment group, while the control group only received conventional treatment. The gastrointestinal function scores and acute physiology and chronic health evaluation II (APACHEⅡ) scores of all subjects were documented at baseline, at days 1, 3 and 7 after treatment respectively and their blood endotoxin was tested at the same time points as well.The incidence of death was recorded for both groups throughout the trial. RESULTS: At days 3 and 7 after treatment, gastroin-testinal function score, APACHE II, and endotoxin level in treatment group wasbetter than that in control group and the difference between them was significant (both P<0.05). Although the incidence of death in treatment group was less than that in control group, the difference between the two groups was not significant (P> 0.05). | 9 | Intra-tumor injection of H101,a recombinant adenovirus,in combination with chemotherapy in patients with advanced cancers:A pilot phase Ⅱ clinical trial显示文摘AIM: HI01, an EIB 55 kD gene deleted adenovirus, has been shown to possess oncolysis activity experimentally and proved to be safe in preliminary phase I study. The current study was designed to evaluate its anti-tumor activity and toxicity in combination with chemotherapy in patients with late stage cancers.METHODS: H101 5.0×10^11 virus particles were given by intra-tumor injection daily for five consecutive days at every three-week cycle, combined with routine chemotherapy,to one of the tumor lesions of 50 patients with different malignant tumors. Tumor lesions without H101 injection in the same individuals were used as controls. The efficacy and toxicity were recorded.RESULTS: Forty-six patients were evaluable with a 30.4% response rate. H101 injection in combination with chemotherapy induced three complete response (CR) and 11 partial response (PR), giving an overall response rate of 28.0% (14/50) among intention-to-treat patients. The response rate for the control lesions was 13.0%, including one case with CR and five cases with PR, which was significantly lower than that for the injected lesions (P<0.05).Main side effects were fever (30.2%) and pain at the injected sites (26.9%). Grade 1 hepatic dysfunction was found in four patients, grade 2 in one patient, and grade 4 in one patient. Hematological toxicity (grade 4) was found in four patients.CONCLUSION: Intra-tumor injection of the genetically engineered adenovirus H101 exhibits potential anti-tumor activity to refractory malignant tumors in combination with chemotherapy. | 10 | Incidence and mortality of lung cancer in China, 2008-2012显示文摘Objective: To analyze the incidence and mortality rates of lung cancer in China from 2008 to 2012.Methods: Incident and death cases of lung cancer were retrieved from the National Central Cancer Registry(NCCR) database collecting from 135 cancer registries in China during 2008-2012.The crude incidence and mortality rates of lung cancer were calculated by area(urban/rural),region(eastern,middle,western),gender and age group(0,1-4,5-9,…,85+).China census in 2000 and Segi's world population were applied for agestandardized rates.JoinPoint(Version 4.5.0.1) model was used for time trend analysis.Results: The crude incidence rate of lung cancer was 54.66/100,000 which ranked the first in overall cancers.The age-standardized incidence rates by China population(ASIRC) and by World population(ASIRW) were35.13/100,000 and 34.86/100,000,respectively.The crude mortality of lung cancer in China was 45.60/100,000 and it was the first cause of cancer-related death in overall cancers.The age-standardized mortality rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 28.57/100,000 and 28.22/100,000,respectively.Incidence and mortality rates of lung cancer were higher in males than in females and higher in urban areas than in rural areas.Eastern areas had the highest incidence and mortality rates followed by middle and western areas.Incidence and mortality rates of lung cancer retained low level in age groups before 40 years old but increased greatly after and peaked in age group of 80-84.During 2003-2012,the temporal trend of the incidence rate of lung cancer in both sexes in China was general stable(P<0.05).The lung cancer incidence rate increased by 0.71% per year in females(P<0.05) and 2.26% per year in rural areas(P<0.05).The mortality rate of lung cancer decreased slightly annually during 2003-2012 in China(P>0.05).In urban areas,it declined by 0.76%per year(P<0.05),but rose by 2.09% per year(P<0.05) in rural areas.Conclusions: Appropriate targeted prevention,early detection and treatment programs should be carried out to control the local burden of lung cancer. | Shuzheng Liu Qiong Chen Lanwei Guo Xiaoqin Cao Xibin Sun Wanqing Chen Jie He | 2018 | Chinese Journal of Cancer Research2018,30,6: | 19 |
| 11 | Nested case-control study on the risk factors of colorectal cancer显示文摘AIM: To investigate the risk factors of colon cancer andrectal cancer.METHODS: A nested case-control study was conducted ina cohort of 64 693 subjects who participated in a colorectalcancer screening program from 1989 to 1998 in Jiashancounty, Zhejiang, China. 196 cases of colorectal cancer weredetected from 1990 to 1998 as the case group and 980non-colorectal cancer subjects, matched with factors of age,gender, resident location, were randomly selected from the64 693 cohort as controls. By using univariate analysis andmutivariate conditional logistic regression analysis, the oddsratio (OR) and its 95 % confidence interval (95 %CI) werecalculated between colorectal cancer and personal habits,dietary factors, as well as intestinal related symptoms.RESULTS: The mutivariate analysis results showed that aftermatched with age, sex and resident location, mucous bloodstool history and mixed sources of drinking water were closelyassociated with colon cancer and rectal cancer, OR values forthe mucous blood stool history were 3.508 (95 %CI: 1.370-8.985) and 2.139 (95 %CI: 1.040-4.402) respectively; for themixed drinking water sources, 2.387 (95 %CI: 1.243-4.587)and 1.951 (95 %CI: 1.086-3.506) respectively. All reachedthe significant level with a P-value less than 0.05.CONCLUSION: The study suggested that mucous bloodstool history and mixed sources of drinking water were therisk factors of colon cancer and rectal cancer. | 12 | Induction of HSF1 expression is associated with sporadic colorectal cancer显示文摘AIM: To explore the activation of signal b-ansduction pathways related with the carcinogenesis of sporadic colon cancers. METHODS: A gene array monitoring the activation of 8 signal transduction pathways (PathwayFinder GEArray) was used to screen the differentially expressed genes between colorectal cancer and normal colon tissues. The differentially expressed genes were further analyzed by RT-PCR, using RNA derived from colorectal cancer and normal colon tissue of 35 patients.RESULTS: The expression of HSF1, HSF27, HSP90 and iNOS was increased in colon cancer tissues compared to normal colon tissue using PathwayFinder GEArray. The RT-PCR results showed that the expression of HSF1 was increased in 86% (30/35) patients and the expression of iNOS was increased in 63% (22/35) patients.CONCLUSION: The induction of HSF1 gene expression is associated with sporadic colon cancer. | 14 | Expression of ST13 in colorectal cancer and adjacent normal tissues显示文摘AIM: To investigate the in situ expression of suppressionof tumorigenecity 13 (ST13) mRNA in both colorectal cancer and adjacent normal tissues.METHODS: Colorectal cancer cell lines SW1116, SW620and CoLo205 were enrolled to confirm the feasibility of the in situ hybridization procedure. Seven colorectal cancer and adjacent normal tissues were included for RNA-RNA in situ hybridization.RESULTS: The expression of ST13 in the seven normal colon tissues was positive and the positive signals appeared in mucosal cells. Only three of the seven colorectal cancer tissues had positive hybridization signals that appeared in adenocarcinoma cells.CONCLUSION: The expression of ST13 decreases in colorectal cancer tissue compared with that in adjacent normal tissue. | 15 | Protective effect of Xuebijing injection on myocardial injury in patients with sepsis: a randomized clinical trial显示文摘OBJECTIVE: To investigate the protective effect and possible mechanism of Xuebijing Injection on myocardial injury in patients with sepsis, and to evaluate its prognostic implications.METHODS: Patients with septic myocardial injury were recruited, and were randomly divided into two groups: treatment group and control group. All patients in two groups received conventional cluster treatment, the patients in treatment group additional received Xuebijing injection dissolved in0.9% sodium chloride injection, and the patients in control group received the same amount of 0.9%sodium chloride injection. At the beginning of treatment and 3, 7 and 10-day after treatment, lab-oratory indicators of cardiac troponin Ⅰ(cTnI),N-terminal pro B-type natriuretic peptide(NT-pro BNP) and procalcitonin(PCT) were respectively tested in venous blood. The patient's length of stay in Intensive Care Unit(ICU) and the mortality in 28 days were recorded.RESULTS: At 3, 7 and 10-day after treatment, the improvements of c Tn I, NT-pro BNP and PCT in treatment group were better than those in control group, and the differences were statistically significant(P < 0.05). The mortality of treatment group in28 days was not significantly different from that of control group(P > 0.05). The numbers of new cancer cases and cancer deaths in Henan province with compiled cancer incidence and mortality rates were estimated.Methods: In 2015, all registries' data in Henan province were qualified for the national cancer registry annual report in 2012. The pooled data were stratified by area(urban/rural), gender, age group(0, 1-4, 5-9, 10-14, …, 85+) and cancer type. New cancer cases and deaths were estimated using age-specific rates and corresponding population of Henan province in 2012. The Chinese census data in 2000 and Segi's population were applied for age-standardized rates. All the rates were expressed per 100,000 person-years.Results: Qualified 19 cancer registries(4 urban and 15 rural registries) covered 16,082,688 populations of Henan province in 2012. The percentage of cases with morphologically verified(MV%) and death certificateonly cases(DCO%) were 69.84% and 2.30%, respectively, and the mortality to incidence rate ratio(M/I) was 0.64. It was estimated that there were 248,510 new cancer cases and 158,630 cancer deaths in Henan province in 2012. The incidence rate was 266.17/100,000(288.61/100,000 in males and 241.86/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASIRC) and by world standard population(ASIRW) were 208.95/100,000 and 206.41/100,000 with the cumulative incidence rate(0-74 years old) of 24.30%. The crude incidence rate in urban areas was higher than that in rural areas. However, after adjusted by age, the cancer incidence rate in rural was higher than that in urban areas. The crude mortality of all cancers in Henan province was 169.90/100,000(201.23/100,000 in males and 135.95/100,000 in females). The age-standardized mortality rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 131.20/100,000 and 130.80/100,000, respectively. Among the patients aged 0-74 years, the cumulative mortality rate was 15.03%. The crude cancer mortality rate in urban areas was higher than that of rural areas. However, the age-standardized rate in rural areas was higher than that of urban areas. Cancers of lung, stomach, esophagus, liver, female breast, colorectum, cervix, brain, uterus and ovary were the most common cancers, accounting for about 82.80% of all cancer new cases. Lung cancer, stomach cancer, esophageal cancer, liver cancer, colorectal cancer, female breast cancer, brain cancer, leukemia, pancreatic cancer and cervix cancer were the leading causes of cancer deaths, accounting for about 88.50% of all cancer deaths. The burden between urban and rural, males and females were different.Conclusions: Registration data of Henan province was qualified to provide basic information on population-based cancer incidence, mortality for cancer prevention and control. The upper digestive tract cancer burden in Henan province, especially for males in rural areas, was higher. The incidence rate of female breast cancer was higher in urban areas. | 18 | Expression of a novel immunoglobulin gene SNC73 in human cancer and non-cancerous tissues显示文摘AIM: To investigate the expression of immunoglobulin gene SNC73 in malignant tumors and non-cancerous normal tissues.METHODS: Expression level of SNC73 in tumors and non-cancerous tissues from the same patient was determined by reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay (RT-PCR-ELISA) in 90 cases of malignant tumors, including colorectal cancer, gastric cancer, breast cancer, lung cancer and liver cancer, Analysis on the correlation of SNC73 expression with sex, age, site,grade of differentiation, depth of invasion, and metastases in colorectal cancer patients was made.RESULTS: Expression level of SNC73 in non-cancerous colorectal mucosa and colorectal cancerous tissues was 1.234±0.842 and 0.737±0.731, respectively (P<0.01), with the mean ratio of 7.134±14.092 (range, 0.36-59.54).Expression of SNC73 showed no significant difference among gastric cancer, breast cancer, lung cancer and liver cancer when compared with non-cancerous tissues (P>0.05). No correlation was found between SNC73 expression level and various clinicopathological factors, including sex, age, site,grade of differentiation, depth of invasion and metastases of CRC patients.CONCLUSION: Down-regulation of SNC73 expression may be a relatively specific phenomenon in colorectal cancer.SNC73 is a potential genetic marker for the carcinongenesis of colorectal cancer. | 19 | Characterization of CagA variable region of Helicobacter pylori isolates from Chinese patients显示文摘AIM: To characterize the CagA variable region of Helicobacter pylori isolates from Chinese patients.METHODS: DNA fragments in CagA variable region were amplified and sequenced respectively from genomic DNA of 19 isolates from patients with gastric cancer and 20isolates from patients with chronic gastritis. The tendency of phosphorylation in tyrosine(s) of CagA proteins was evaluated subsequently by phosphorylation assay in vivo and in vitro respectively.RESULTS: About 97.44% (38/39) H pylori isolates possessed CagA gene. CagA+ strains contained 2-4tandem five-amino-acid motifs EPIYA but only one EPIYA had repeated sequence in CagA variable region in different isolates. There was no significant difference between the number of EPIYA motifs in H pylori from patients with different diseases. Coal geology, as a practical science, has an access to gradual development with the growth and prosperity of China. From the meaning of the coalfield geology, this paper introduces the research progress and status of coalfield geology, including the research on coal basin, stratigraphy of coal-bearing stratigraphic sequences, coal-forming process, structural geology of coalfields, and exploration and development status of coal resources. The paper specially focuses on coal-forming process, including coal formation in terrestrial facies and transgressive / regression events, and introduces new technology which has been applied to research progress in structural geology of coalfield.
