1Phytochrome B interacts with SWC6 and ARP6 to regulate H2A.Z deposition and photomorphogensis in Arabidopsis显示文摘Light serves as a crucial environmental cue which modulates plant growth and development, and which is controlled by multiple photoreceptors including the primary red light photoreceptor,phytochrome B(phyB). The signaling mechanism of phyB involves direct interactions with a group of basic helix-loop-helix(bHLH) transcription factors, PHYTOCHROME-INTERACTING FACTORS(PIFs), and the negative regulators of photomorphogenesis, COP1 and SPAs. H2 A.Z is an evolutionarily conserved H2 A variant which plays essential roles in transcriptional regulation. The replacement of H2 A with H2 A.Z is catalyzed by the SWR1 complex. Here, we show that the Pfr form of phyB physically interacts with the SWR1 complex subunits SWC6 and ARP6. phyB and ARP6 coregulate numerous genes in the same direction,some of which are associated with auxin biosynthesis and response including YUC9, which encodes a rate-limiting enzyme in the tryptophandependent auxin biosynthesis pathway. Moreover,phyB and HY5/HYH act to inhibit hypocotyl elongation partially through repression of auxin biosynthesis. 2Genetic Characteristics of 2009 Pandemic H1N1 Influenza A Viruses Isolated from China's Mainland显示文摘A total of 100 H1N1 flu real-time-PCR positive throat swabs collected from fever patients in Zhejiang, Hubei and Guangdong between June and November 2009, were provided by local CDC laboratories. After MDCK cell culture, 57 Influenza A Pandemic (H1N1) viruses were isolated and submitted for whole genome sequencing. A total of 39 HA sequences, 52 NA sequences, 36 PB2 sequences, 31 PB1 sequences, 40 PA sequences, 48 NP sequences, 51 MP sequences and 36 NS sequences were obtained, including 20 whole genome sequences. Sequence comparison revealed they shared a high degree of homology (96%~99%) with known epidemic strains (A/California/04/2009(H1N1). Phylogenetic analysis showed that although the sequences were highly conserved, they clustered into a small number of groups with only a few distinct strains. Site analysis revealed three substitutions at loop 220 (221-228) of the HA receptor binding site in the 39 HA sequences: A/Hubei/86/2009 PKVRDQEG→PKVRDQEA, A/Zhejiang/08/2009 PKVRDQEG→PKVRDQER, A/Hubei/75/2009 PKVRDQEG→PKVRDQGG, the A/Hubei/75/2009 was isolated from an acute case, while the other two were from patients with mild symptoms. Other key sites such as 119, 274, 292 and 294 amino acids of NA protein,627 of PB2 protein were conserved. Meanwhile, all the M2 protein sequences possessed the Ser32Asn mutation, suggesting that these viruses were resistant to adamantanes. 3Amyloid precursor protein and growth-associated protein 43 expression in brain white matter and spinal cord tissues in a rat model of experimental autoimmune encephalomyelitis显示文摘Studies have demonstrated that amyloid precursor protein(APP)expression increases in multiple sclerosis tissues during acutely and chronically active stages.To determine the relationship between axonal injury and regeneration in multiple sclerosis,an animal model of experimental autoimmune encephalomyelitis was induced using different doses of myelin basic protein peptide.APP and growth-associated protein 43(GAP-43),which is considered a specific marker of neural regeneration,were assessed by western blot analysis.Expression of APP and GAP-43,as well as the correlation between these two proteins,in brain white matter and spinal cord tissues of experimental autoimmune encephalomyelitis rats at different pathological stages was analyzed.Results showed that APP and GAP-43 expression increased during the acute stage and decreased during remission,with a positive correlation between APP and GAP-43 expression in brain white matter and spinal cord tissues.These results suggest that APP and GAP-43 could provide nutritional and protective effects on damaged neurons.Yizhou Wang Shuang Kou Jingcheng Tang Ping Zhang Qiuxia Zhang Yan Liu Qi Zheng Hui Zhao Lei Wang 2011Neural Regeneration Research2011,6,2:3
4Low phosphorus promotes NSP1–NSP2 heterodimerization to enhance strigolactone biosynthesis and regulate shoot and root architecture in rice显示文摘Phosphorus is an essential macronutrient for plant development and metabolism,and plants have evolved ingenious mechanisms to overcome phosphate(Pi)starvation.However,the molecular mechanisms underlying the regulation of shoot and root architecture by low phosphorus conditions and the coordinated utilization of Pi and nitrogen remain largely unclear.Here,we show that Nodulation Signaling Pathway 1(NSP1)and NSP2 regulate rice tiller number by promoting the biosynthesis of strigolactones(SLs),a class of phytohormones with fundamental effects on plant architecture and environmental responses.We found that NSP1 and NSP2 are induced by Oryza sativa PHOSPHATE STARVATION RESPONSE2(OsPHR2)in response to low-Pi stress and form a complex to directly bind the promoters of SL biosynthesis genes,thus markedly increasing SL biosynthesis in rice.Interestingly,the NSP1/2–SL signaling module represses the expression of CROWN ROOTLESS 1(CRL1),a newly identified early SL-responsive gene in roots,to restrain lateral root density under Pi deficiency.We also demonstrated that GR24^(4DO) treatment under normal conditions inhibits the expression of OsNRTs and OsAMTs to suppress nitrogen absorption but enhances the expression of OsPTs to promote Pi absorption,thus facilitating the balance between nitrogen and phosphorus uptake in rice.Importantly,we found that NSP1p:NSP1 and NSP2p:NSP2 transgenic plants show improved agronomic traits and grain yield under low-and medium-phosphorus conditions.Taken together,these results revealed a novel regulatory mechanism of SL biosynthesis and signaling in response to Pi starvation,providing genetic resources for improving plant architecture and nutrient-use efficiency in low-Pi environments.Kun Yuan Hao Zhang Chaoji Yu Nan Luo Jijun Yan Shuang Zheng Qingliang Hu Dahan Zhang Liquan Kou Xiangbing Meng Yanhui Jing Mingjiang Chen Xinwei Ban Zongyun Yan Zefu Lu Jian Wu Yu Zhao Yan Liang Yonghong Wang Guosheng Xiong Jinfang Chu Ertao Wang Jiayang Li Bing Wang 2023Molecular Plant2023,16,11:2
5Molecular characterization of avian influenza virus (H7N8) isolated from poultry in Central China in the mid-1980s显示文摘The molecular and pathogenic properties of avian influenza virus (A/duck/Hubei/216/1985/H7N8) isolated from Hubei Province of China in 1985 were characterized.The hemagglutinin gene (HA) of Dk/Hb/216/85/H7N8 had the multiple amino acid se-quences (-PEIPKGRG-) at the connecting peptide between HA1 and HA2, which is considered to be a distinguishing molecular characteristic of low pathogenicity.The key sites of host markers among the genes (M, NP, NS, PA and PB2) of Dk/Hb/ 216/85/H7N8 were similar to those of H5N1 viruses.Phylogenetic analysis showed that the genes (M, NS and PB2) of Dk/Hb/ 216/85/H7N8 clustered closely with those of Gs/Guangdong/1996/H5N1 (Gs/GD/96), implying that three ancient genes of Gs/ GD/96-like viruses had been circulating in Central China during the 1980s.In experimental infection, Dk/Hb/216/85/H7N8 was lowly pathogenic to chickens and mice, consistent with the molecular feature of HA gene.YANG JiaLin XIA Han ZHAO JiuRu HE XiaoBin PAN LiMin TANG Shuang ZHANG Zhong KOU Zheng LI TianXian 2010Chinese Science Bulletin2010,55,16:1
6Arabidopsis cryptochrome 1 promotes stomatal development through repression of AGB1 inhibition of SPEECHLESS DNA-binding activity显示文摘Cryptochromes are blue light photoreceptors that mediate various light responses in plants and mammals. The heterotrimeric G-protein is known to regulate various physiological processes in plants and mammals. In Arabidopsis, cryptochrome 1(CRY1) and the G-protein β subunit AGB1 act antagonistically to regulate stomatal development.The molecular mechanism by which CRY1 and AGB1 regulate this process remains unknown.Here, we show that Arabidopsis CRY1 acts partially through AGB1, and AGB1 acts through SPEECHLESS(SPCH), a master transcription factor that drives stomatal initiation and proliferation, to regulate stomatal development. We demonstrate that AGB1 physically interacts with SPCH to block the b HLH DNA-binding domain of SPCH and inhibit its DNA-binding activity. Moreover, we demonstrate that photoexcited CRY1 represses the interaction of AGB1 with SPCH to release AGB1 inhibition of SPCH DNA-binding activity, leading to the expression of SPCH-target genes promoting stomatal development. Taken together, our results suggest that the mechanism by which CRY1 promotes stomatal development involves positive regulation of the DNA-binding activity of SPCH mediated by CRY1 inhibition of the AGB1-SPCH interaction. 7The involvement of the N-terminal PHR domain of Arabidopsis cryptochromes in mediating light signaling显示文摘Light is a key environmental cue that fundamentally regulates all aspects of plant growth and development,which is mediated by the multiple photoreceptors including the blue light photoreceptors cryptochromes(CRYs).In Arabidopsis,there are two well-characterized homologous CRYs,CRY1 and CRY2.Whereas CRYs are flavoproteins,they lack photolyase activity and are characterized by an Nterminal photolyase-homologous region(PHR)domain and a C-terminal extension domain.It has been established that the C-terminal extension domain of CRYs is involved in mediating light signaling through direct interactions with the master negative regulator of photomorphogenesis,COP1.Recent studies have revealed that the N-terminal PHR domain of CRYs is also involved in mediating light signaling.In this review,we mainly summarize and discuss the recent advances in CRYs signaling mediated by the N-terminal PHR domain,which involves the N-terminal PHR domain-mediated dimerization/oligomerization of CRYs and physical interactions with the pivotal transcription regulators in light and phytohormone signaling.Wenxiu Wang Zhilei Mao Tongtong Guo Shuang Kou Hong-Quan Yang 2021aBIOTECH2021,2,2:0
8Molecular characterization of H3N2 and H4N6 subtypes avian influenza viruses isolated from mallards in Poyang Lake,China in 2010显示文摘Poyang Lake is the largest inland freshwater lake in China and contains many species of wild birds and waterfowls.We conducted a survey of avian influenza viruses in nine semi-artificial waterfowl farms in Poyang Lake during January to March of 2010.Out of 1036 cloacal swabs collected,three H3N2 and one H4N6 influenza viruses were isolated from healthy mallards.All the isolates were genetically and phylogenetically characterized.The analysis of putative HA cleavage sites showed that all the four isolates possessed the molecular characteristics(QTRGL for H3N2 viruses,PEKASR for H4N6 virus) of lowly pathogenic avian influenza(LPAI) virus.The phylogenetic analysis of the viral genomes showed that all four virus isolates clustered in the Eurasian clade of influenza viruses.The M gene of the viruses possessed the highest homology with highly pathogenic H5N1 influenza viruses.In addition,co-infection of H3N2 and H4N6 in the same farm was observed.And interestingly,we isolated two subtypes viruses(H3N2 and H4N6) and their progeny virus(H3N2) with evidence of genome reassortment from the same farm,in which the PB1 and PB2 gene segments of H4N6 replaced those of the H3N2 strain.The results of animal infection experiments showed that all the four isolated viruses were lowly pathogenic to chickens and not pathogenic to mice,which was consistent with the results of genetic analysis.ZHU Na ZHAO JiuRu LI YongDong DING ChangQing XIA Han TANG Shuang ZHANG Zhong YU JingFeng CHEN Jing FAN ZhaoJun KOU Zheng LI TianXian 2012Chinese Science Bulletin2012,57,27:0
