Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. The X protein ofHBV can activate NF-κB to translocate into nucleiof hepatocellular carcinoma cells.Shuang Ping Guo~1 Wen Liang Wang~1 Yu Qiang Zhai~2 Yi Ling Zhao~1 ~1Department of Pathology,Xijing Hospital of the Fourth Military Medical University,Xi’an,China ~2Department of Urology, the Central Hospital of Xi’an,China 2001World Journal of Gastroenterology2001,7,3:55

The Genome Sequence Archive Family: Toward Explosive Data Growth and Diverse Data Types显示文摘The Genome Sequence Archive(GSA)is a data repository for archiving raw sequence data, which provides data storage and sharing services for worldwide scientific communities.Considering explosive data growth with diverse data types, here we present the GSA family by expanding into a set of resources for raw data archive with different purposes,namely,GSA(https://ngdc.cncb.ac.cn/gsa/),GSA for Human(GSA-Human,https://ngdc.cncb.ac.cn/gsa-human/),and Open Archive for Miscellaneous Data(OMIX,https://ngdc.cncb.ac.cn/omix/).Compared with the 2017 version,GSA has been significantly updated in data model, online functionalities, and web interfaces.GSA-Human,as a new partner of GSA,is a data repository specialized in human genetics-related data with controlled access and security.OMIX,as a critical complement to the two resources mentioned above,is an open archive for miscellaneous data.Together, all these resources form a family of resources dedicated to archiving explosive data with diverse types, accepting data submissions from all over the world, and providing free open access to all publicly available data in support of worldwide research activities.Tingting Chen Xu Chen Sisi Zhang Junwei Zhu Bixia Tang Anke Wang Lili Dong Zhewen Zhang Caixia Yu Yanling Sun Lianjiang Chi Huanxin Chen Shuang Zhai Yubin Sun Li Lan Xin Zhang Jingfa Xiao Yiming Bao Yanqing Wang Zhang Zhang Wenming Zhao 2021Genomics, Proteomics & Bioinformatics2021,19,4:38
GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://bigd.big.ac.cn/gsa or http://gsa.big.ac.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world, and makes all these data publicly available to worldwide scientific communities. In the era of big data, GSA is not only an important complement to existing INSDC members by alleviating the increasing burdens of handling sequence data deluge, but also takes the significant responsibility for global big data archive and provides free unrestricted access to all publicly available data in support of research activities throughout the world.Yanqing Wang Fuhai Song Junwei Zhu Sisi Zhang Yadong Yang Tingting Chen Bixia Tang Lili Dong Nan Ding Qian Zhang Zhouxian Bai Xunong Dong Huanxin Chen Mingyuan Sun Shuang Zhai Yubin Sun Lei Yu Li Lan Jingfa Xiao Xiangdong Fang Hongxing Lei Zhang Zhang Wenming Zhao 2017Genomics, Proteomics & Bioinformatics2017,15,1:13
Association between large artery stenosis, cerebral small vessel disease and risk of ischemic stroke显示文摘We aimed to assess the associations of large artery stenosis(LAS)and cerebral small vessel disease(CSVD)with the risk of ischemic stroke and to investigate their respective and combined contributions. In the prospective population-based Shunyi Study,1,082 stroke-free participants aged 55.9±9.1 years were included.Participants were followed for incident stroke throughout the study period(2013-2019).Total small vessel disease score was used to measure CSVD burden.Cervico-cerebral large artery stenosis was evaluated via brain magnetic resonance angiography and carotid ultrasound. We estimated the risk of ischemic stroke in relation to LAS and CSVD with Cox regression models.During a mean follow-up of 4.2 years,34 participants(3.1%)experienced at least one ischemic stroke.Severe LAS(≥50% stenosis versus no stenosis:HR=3.27(95%CI:1.31-8.18))and high CSVD burden(total small vessel disease score 2-4 versus 0 point:HR=12.73(4.83-33.53))were associated with increased stroke risk independently. In multivariate models,CSVD burden(7.72%)explained a larger portion of the variation in stroke risk than severity of LAS(3.49%).Our findings identified that both LAS and CSVD were associated with future ischemic stroke in asymptomatic subjects, while those with high CSVD burden deserve more attention in primary prevention of stroke.Fei Han Ding-Ding Zhang Fei-Fei Zhai Jun Xue Jiang-Tao Zhang Shuang Yan Li-Xin Zhou Jun Ni Ming Yao Meng Yang Ming-Li Li Zheng-Yu Jin Qing Dai Shu-Yang Zhang Li-Ying Cui Yi-Cheng Zhu 2021Science China(Life Sciences)2021,64,9:9
Virtual Machine-Based Task Scheduling Algorithm in a Cloud Computing Environment显示文摘Virtualization technology has been widely used to virtualize single server into multiple servers, which not only creates an operating environment for a virtual machine-based cloud computing platform but also potentially improves its efficiency. Currently, most task scheduling-based algorithms used in cloud computing environments are slow to convergence or easily fall into a local optimum. This paper introduces a Greedy Particle Swarm Optimization(G&PSO) based algorithm to solve the task scheduling problem. It uses a greedy algorithm to quickly solve the initial particle value of a particle swarm optimization algorithm derived from a virtual machine-based cloud platform. The archived experimental results show that the algorithm exhibits better performance such as a faster convergence rate, stronger local and global search capabilities, and a more balanced workload on each virtual machine. Therefore, the G&PSO algorithm demonstrates improved virtual machine efficiency and resource utilization compared with the traditional particle swarm optimization algorithm.Zhifeng Zhong Kun Chen Xiaojun Zhai Shuange Zhou 2016Tsinghua Science and Technology2016,21,6:5
Does persistence of showy calyces in Limonium leptolobum enhance pollinator attraction?显示文摘Aims The calyx, the outermost whorl of a flower(usually green),has been considered to function to protect flowers.in some species,how-ever,calyces are colorful and retained during seed development.Limonium species have been exploited as cut flower crops because the calyces persist for several months after the corolla has closed. To explore the adaptive significance of the persistent calyx in a desert plant Limonium leptolobum,we ask whether persistence of caly-ces can enhance pollinator attraction by enlarging floral displays, increasing reproductive success in this self-incompatible species. Methods The yellow flower of L.leptolobum lasted 1-2 days but its white, membranous calyx extended fully after the corolla closed, and per-sisted for over 2 months in the field, making hundreds of‘showy flowers’on one individual. To examine the ecological function of calyces,we test the pollinator attraction hypothesis.in an experi-mental population,we compared the difference in visit frequency and visitor behavior between intact inflorescences and inflores-cences with their calyces removed on the same individual plants.Important findingsin four experimental plots four types of floral visitors were observed including bees, butterflies, syrphid flies and day-flying moths. No significant preference was observed between calyx-free and intact inflorescences for both first arrivals and total visit frequency of all types of floral visitors, indicating that the persistence of calyces did not make plants more attractive to potential pollinators. The pollina-tor attraction hypothesis for the showy calyces was not supported by the current data.Whether the calyx in this desert plant helps seed development or has other functions needs further study.Ai-Qin Zhang Shuang He Ya-Xin Zhai Shuang-Quan Huang 2015Journal of Plant Ecology2015,8,2:5
Efficient structural seismic performance evaluation method using improved endurance time analysis显示文摘Evaluation of structural performance under seismic excitations from low intensity to high intensity is essential to verify the seismic resistant capacity of a structure, and usually carried out by the incremental dynamic analysis (IDA) method or pushover method. The recently developed endurance time (ET) method is another method that uses dynamic pushover excitations, i.e., endurance time acceleration function, to obtain results similar to those obtained by IDA or pushover methods with low computational cost and acceptable accuracy. This study proposes an improvement on the ET method by considering more restrictions for both the elastic and inelastic response spectra in the generation procedure, and by specifying a target duration. Four reinforced concrete frame structures with 4, 8, 12, and 16 stories are adopted to verify the accuracy of the improved method. Comparison of the results obtained by the proposed method, the ET method and the IDA method shows that the improved method has a higher accuracy than the ET method. For evaluation of structural responses under specifi c ground motion intensity, which is typically required in seismic design codes, the results obtained by the proposed method are compared with fi ve commonly used ground motion selection methods, and shows the proposed method provides acceptable accuracy for engineering applications.Li Shuang Liu Kun Liu Xiangyang Zhai Changhai Xie Fengwei 2019Earthquake Engineering and Engineering Vibration2019,18,4:3
A sophisticated simulation for the fracture behavior of concrete material using XFEM显示文摘The development of a powerful numerical model to simulate the fracture behavior of concrete material has long been one of the dominant research areas in earthquake engineering.A reliable model should be able to adequately represent the discontinuous characteristics of cracks and simulate various failure behaviors under complicated loading conditions. In this paper,a numerical formulation, which incorporates a sophisticated rigid-plastic interface constitutive model coupling cohesion softening, contact, friction and shear dilatation into the XFEM,is proposed to describe various crack behaviors of concrete material. An effective numerical integration scheme for accurately assembling the contribution to the weak form on both sides of the discontinuity is introduced. The effectiveness of the proposed method has been assessed by simulating several well-known experimental tests. It is concluded that the numerical method can successfully capture the crack paths and accurately predict the fracture behavior of concrete structures. The influence of mode-Ⅱ parameters on the mixed-mode fracture behavior is further investigated to better determine these parameters.Zhai Changhai Wang Xiaomin Kong Jingchang Li Shuang Xie Lili 2017Earthquake Engineering and Engineering Vibration2017,16,4:3

Dimensional analysis of earthquake-induced pounding between adjacent inelastic MDOF buildings显示文摘In this study the seismic pounding response of adjacent multi-degree-of-freedom(MDOF) buildings with bilinear inter-story resistance characteristics is investigated through dimensional analysis. The application of dimensional analysis leads to a condensed presentation of the response, and the remarkable self-similarity property for bilinear MDOF buildings with inelastic collision is uncovered. It is shown that when the response is expressed in the appropriate dimensionless form, response spectra for any intensity of the excitation collapse to a single master curve. The reduced Π set explicitly describes the interaction between the colliding structures. The effect of pounding on the MDOF building's response is illustrated using three well-divided spectral regions(amplifi ed, de-amplifi ed and unaffected regions). Parametric studies are conducted to investigate the effects of the story stiffness of structures, the story stiffness ratio and mass ratio of adjacent buildings, the structural inelastic characteristics and the gap size values. Results show that(i) the infl uence of system stiffness ratio to the lighter and more fl exible building is more signifi cant in the fi rst spectral region, where the maximum response of the building is amplifi ed because of pounding; and(ii) the velocity and pounding force of the heavier and stiffer building is unexpectedly sensitive to the mass ratio of adjacent buildings.Zhai Changhai Jiang Shan Li Shuang Xie Lili 2015Earthquake Engineering and Engineering Vibration2015,14,2:2
QNE1 is a key flowering regulator determining the length of the vegetative period in soybean cultivars显示文摘The soybean E1 gene is a major regulator that plays an important role in flowering time and maturity. However,it remains unclear how cultivars carrying the dominant E1 allele adapt to the higher latitudinal areas of northern China. We mapped the novel quantitative trait locus QNE1(QTL near E1) for flowering time to the region proximal to E1 on chromosome 6 in two mapping populations.Positional cloning revealed Glyma.06G204300,encoding a TCP-type transcription factor,as a strong candidate gene for QNE1.Association analysis further confirmed that functional single nucleotide polymorphisms(SNPs) at nucleotides 686 and 1,063 in the coding region of Glyma.06G204300 were significantly associated with flowering time. The protein encoded by the candidate gene is localized primarily to the nucleus.Furthermore, soybean and Brassica napus plants overexpressing Glyma.06G204300 exhibited early flowering. We conclude that despite their similar effects on flowering time,QNE1 and E4 may control flowering time through different regulatory mechanisms, based on expression studies and weighted gene co-expression network analysis of flowering time-related genes.Deciphering the molecular basis of QNE1 control of flowering time enriches our knowledge of flowering gene networks in soybean and will facilitate breeding soybean cultivars with broader latitudinal adaptation.Zhengjun Xia Hong Zhai Yanfeng Zhang Yaying Wang Lu Wang Kun Xu Hongyan Wu Jinglong Zhu Shuang Jiao Zhao Wan Xiaobin Zhu Yi Gao Yingxiang Liu Rong Fan Shihao Wu Xin Chen Jinyu Liu Jiayin Yang Qijian Song Zhixi Tian 2022Science China(Life Sciences)2022,65,12:1

Research on power-supply cost of regional power system under carbon-peak target显示文摘With the establishment of the carbon-peak target by 2030,the direction of carbon emission reduction in China’s energy system has been further clarified. As the industry with the largest proportion of carbon emissions in China, the lowcarbon transformation of the electric power industry is critical to realize the carbon-peak target.Current research mostly focuses on technical analysis or system cost accounting of the carbon-peak realization path at the national level. There is a lack of targeted research on regional power systems with complex inter-regional power flow exchange and limited energy resource development.Simultaneously, the calculation of the system cost lacks the perspective of the life cycle and ignores the inertia of the stock and change inertia of incremental disturbance. From the perspective of the life cycle, this study proposes a calculation model of power supply cost for regional power systems according to the carbon-peak target, analyzes the realization path of the carbon target from an economic perspective, and provides references for the path selection and policy formulation of system transformation.Jingyi Wang Min Cang Xiaomeng Zhai Shuang Wu Xi Cheng Lei Zhu 2022Global Energy Interconnection2022,5,1:1

Effects of hanging wall and footwall on demand of structural input energy during the 2008 Wenchuan earthquake显示文摘Systematic differences in the duration and frequency content of ground motions from the hanging wall and footwall during the 2008 Wenchuan earthquake are investigated, focusing on the influence of these differences on structural input energy based on the elastic and inelastic energy responses of structures.A comparison of the input energy spectra between the hanging wall and the footwall reveal that the structural input energy on the hanging wall is not amplified due to the short duration and low peak ground velocity to acceleration ratio(V/A).However,the larger demand of structural input energy on the footwall in the range of medium and long periods is observed and the demand increases up to 50% relative to the average level of structural input energy for rupture distances larger than 30 km.The importance of considering the footwall effect on structural input energy when comparing ground motions in the range of medium and long periods is recognized.Lu Shunan Li Shuang Zhai Changhai Xie Lili 2013Earthquake Engineering and Engineering Vibration2013,12,1:1

Aquaporin 4 deficiency eliminates the beneficial effects of voluntary exercise in a mouse model of Alzheimer’s disease显示文摘Regular exercise has been shown to reduce the risk of Alzheimer’s disease(AD).Our previous study showed that the protein aquaporin 4(AQP4),which is specifically expressed on the paravascular processes of astrocytes,is necessary for glymphatic clearance of extracellular amyloid beta(Aβ)from the brain, which can delay the progression of Alzheimer’s disease. However,it is not known whether AQP4-regulated glymphatic clearance of extracellular Aβis involved in beneficial effects of exercise in AD patients. Our results showed that after 2 months of voluntary wheel exercise,APP/PS1 mice that were 3 months old at the start of the intervention exhibited a decrease in Aβburden, glial activation, perivascular AQP4 mislocalization, impaired glymphatic transport, synapse protein loss, and learning and memory defects compared with mice not subjected to the exercise intervention. In contrast,APP/PS1 mice that were 7 months old at the start of the intervention exhibited impaired AQP4 polarity and reduced glymphatic clearance of extracellular Aβ,and the above-mentioned impairments were not alleviated after the 2-month exercise intervention.Compared with age-matched APP/PS1 mice,AQP4 knockout APP/PS1 mice had more serious defects in glymphatic function,Aβplaque deposition, and cognitive impairment, which could not be alleviated after the exercise intervention. These findings suggest that AQP4-dependent glymphatic transport is the neurobiological basis for the beneficial effects of voluntary exercises that protect against the onset of AD.Yun Liu Pan-Pan Hu Shuang Zhai Wei-Xi Feng Rui Zhang Qian Li Charles Marshall Ming Xiao Ting Wu 2022Neural Regeneration Research2022,17,9:1
