| 1 | Global methane and nitrous oxide emissions from terrestrial ecosystems due to multiple environmental changes显示文摘Greenhouse gas(GHG)-induced climate change is among the most pressing sustainability challenges facing humanity today,posing serious risks for ecosystem health.Methane(CH_(4))and nitrous oxide(N_(2)O)are the two most important GHGs after carbon dioxide(CO_(2)),but their regional and global budgets are not well known.In this study,we applied a process-based coupled biogeochemical model to concurrently estimate the magnitude and spatial and temporal patterns of CH_(4)and N_(2)O fluxes as driven by multiple environmental changes,including climate variability,rising atmospheric CO_(2),increasing nitrogen deposition,tropospheric ozone pollution,land use change,and nitrogen fertilizer use.The estimated CH_(4)and N_(2)O emissions from global land ecosystems during 1981-2010 were 144.39±12.90 Tg C/yr(mean 62 SE;1 Tg=1012 g)and 12.52±0.74 Tg N/yr,respectively.Our simulations indicated a significant(P,0.01)annually increasing trend for CH_(4)(0.43±0.06 Tg C/yr)and N_(2)O(0.14±0.02 Tg N/yr)in the study period.CH_(4)and N_(2)O emissions increased significantly in most climatic zones and continents,especially in the tropical regions and Asia.The most rapid increase in CH_(4)emission was found in natural wetlands and rice fields due to increased rice cultivation area and climate warming.N_(2)O emission increased substantially in all the biome types and the largest increase occurred in upland crops due to increasing air temperature and nitrogen fertilizer use.Clearly,the three major GHGs(CH_(4),N_(2)O,and CO_(2))should be simultaneously considered when evaluating if a policy is effective to mitigate climate change. | Hanqin Tian Guangsheng Chen Chaoqun Lu Xiaofeng Xu Wei Ren Bowen Zhang Kamaljit Banger Bo Tao Shufen Pan Mingliang Liu Chi Zhang Lori Bruhwiler Steven Wofsy | 2015 | Ecosystem Health and Sustainability2015,1,1: | 7 |
| 2 | Neonatal Maternal Deprivation Followed by Adult Stress Enhances Adrenergic Signaling to Advance Visceral Hypersensitivity显示文摘The pathophysiology of visceral pain in patients with irritable bowel syndrome remains largely unknown.Our previous study showed that neonatal maternal deprivation(NMD) does not induce visceral hypersensitivity at the age of 6 weeks in rats. The aim of this study was to determine whether NMD followed by adult stress at the age of 6 weeks induces visceral pain in rats and to investigate the roles of adrenergic signaling in visceral pain. Here we showed that NMD rats exhibited visceral hypersensitivity 6 h and 24 h after the termination of adult multiple stressors(AMSs). The plasma level of norepinephrine was significantly increased in NMD rats after AMSs. Whole-cell patch-clamp recording showed that the excitability of dorsal root ganglion(DRG) neurons from NMD rats with AMSs was remarkably increased. The expression of β_2 adrenergic receptors at the protein and mRNA levels was markedly higher in NMD rats with AMSs than in rats with NMD alone. Inhibition of β_2 adrenergic receptors with propranolol or butoxamine enhanced the colorectal distention threshold and application of butoxamine also reversed the enhanced hypersensitivity of DRG neurons. | 6 | Vitamin B6 regulates IL-33 homeostasis to alleviate type 2 inflammation显示文摘Interleukin-33(IL-33)is a crucial nuclear cytokine that induces the type 2 immune response and maintains immune homeostasis.The fine-tuned regulation of IL-33 in tissue cells is critical to control of the type 2 immune response in airway inflammation,but the mechanism is still unclear.Here,we found that healthy individuals had higher phosphate-pyridoxal(PLP,an active form of vitamin B6)concentrations in the serum than asthma patients.Lower serum PLP concentrations in asthma patients were strongly associated with worse lung function and inflammation.In a mouse model of lung inflammation,we revealed that PLP alleviated the type 2 immune response and that this inhibitory effect relied on the activity of IL-33.A mechanistic study showed that in vivo,pyridoxal(PL)needed to be converted into PLP,which inhibited the type 2 response by regulating IL-33 stability.In mice heterozygous for pyridoxal kinase(PDXK),the conversion of PL to PLP was limited,and IL-33 levels were increased in the lungs,aggravating type 2 inflammation.Furthermore,we found that the mouse double minute 2 homolog(MDM2)protein,an E3 ubiquitin-protein ligase,could ubiquitinate the N-terminus of IL-33 and sustain IL-33 stability in epithelial cells.PLP reduced MDM2-mediated IL-33 polyubiquitination and decreased the level of IL-33 through the proteasome pathway.In addition,inhalation of PLP alleviated asthma-related effects in mouse models.In summary,our data indicate that vitamin B6 regulates MDM2-mediated IL-33 stability to constrain the type 2 response,which might help develop a potential preventive and therapeutic agent for allergy-related diseases. | Songling Zhu Shufen Zhong Kebin Cheng Li-Sha Zhang Jiu-wu Bai Zu Cao Su Wang Wen Chen Shipeng Cheng Liyan Ma Zhiyang Ling Yuying Huang Wangpeng Gu Xiaoyu Sun Chunyan Yi Meng Zhao Shuo Liang Jin-Fu Xu Bing Sun Yaguang Zhang | 2023 | Cellular & Molecular Immunology2023,20,7: | 1 |

| 16 | Adult Stress Promotes Purinergic Signaling to Induce Visceral Pain in Rats with Neonatal Maternal Deprivation显示文摘Chronic visceral pain is one of the primary symptoms of patients with irritable bowel syndrome(IBS),which affects up to 15%of the population world-wide.The detailed mechanisms of visceral pain remain largely unclear.Our previous studies have shown that neonatal maternal deprivation(NMD)followed by adult multiple stress(AMS)advances the occurrence of visceral pain,likely due to enhanced norepinephrine(NE)-β2 adrenergic signaling.This study was designed to explore the roles of P2X3 receptors(P2X3Rs)in the chronic visceral pain induced by combined stress.Here,we showed that P2X3 Rs were co-expressed inβ2 adrenergic receptor(β2-AR)-positive dorsal root ganglion neurons and that NE significantly enhanced ATP-induced Ca2+signals.NMD and AMS not only significantly increased the protein expression of P2X3Rs,but also greatly enhanced the ATP-evoked current density,number of action potentials,and intracellular Ca2+concentration of colon-related DRG neurons.Intrathecal injection of the P2X3R inhibitor A317491 greatly attenuated the visceral pain and the ATP-induced Ca2+signals in NMD and AMS rats.Furthermore,theβ2-AR antagonist butoxamine significantly reversed the expression of P2X3Rs,the ATP-induced current density,and the number of action potentials of DRG neurons.Overall,our data demonstrate that NMD followed by AMS leads to P2X3R activation,which is most likely mediated by upregulation ofβ2 adrenergic signaling in primary sensory neurons,thus contributing to visceral hypersensitivity. | Shufen Hu Qian Sun Wan-Jie Du Jian Song Xin Li Ping-An Zhang Ji-Tian Xu Guang-Yin Xu | 2020 | Neuroscience Bulletin2020,36,11: | 1 |
