A Genetic Pathway for Tapetum Development and Function in Arabidopsis显示文摘In anther development, tapetal cells take part in complex processes, including endomitosis and apoptosis (programmed cell death). The tapetum provides many of the proteins, lipids, polysaccharides and other molecules necessary for pollen development. Several transcription factors, including DYT1, TDF1, AMS, MS188 and MS1, have been reported to be essential for tapetum development and function in Arabidopsis thaliana. Here, we present a detailed cytological analysis of knockout mutants for these genes, along with an in situ RNA hybridization experiment and double mutant analysis showing that these transcription factors form a genetic pathway in tapetum development. DYT1, TDF1 and AMS function in early tapetum development, while MS188 and MS1 are important for late tapetum development. The genetic pathway revealed in this work facilitates further investigation of the function and molecular mechanisms of tapetum development in Arabidopsis. | Jun Zhu Yue Lou Xiaofeng Xu Zhong-Nan Yang | 2011 | Journal of Integrative Plant Biology2011,53,11: | 32
The crustal structures of the central Longmenshan along and its margins as related to the seismotectonics of the 2008 Wenchuan Earthquake显示文摘In 2010,a 500-km-long wide-angle reflection/refraction seismic profile was completed, running northwest from the central Sichuan Basin. This profile orthogonally crosses the meizoseismal area of great Wenchuan earthquake of 12 May 2008,which occurred in the central part of the Longmenshan. The profile also passes through the northwestern Sichuan Plateau, along which a new deep seismic sounding observation system was set up that was much improved over previous datasets and enabled abundant observations to be recorded.Seismic wave phase records that reflect the structural characteristics of different tectonic blocks, especially the complicated phase features associated with the Wenchuan earthquake, were calculated and analyzed in detail.A 2D crustal P-wave velocity model for the orogenic belt in the central Longmenshan and its margins was determined, and crustal structure differences between the stable Sichuan Basin and the thickened northwestern Sichuan Plateau were characterized.Lithological variations within the upper and lower crust in the interior of the plateau, especially a great velocity decrease and plastic rheological properties associated with strong lithologic weakening in lower crust, were detected. From west to east in the lower crust beneath the orogenic belt lying between the Sichuan Basin and the northwestern Sichuan Plateau,a giant shovel-like upwelling is observed that dips gently in the lower part and at higher angles in the upper part;this is inferred to be related to the fault systems in the central Longmenshan. An upwelling in the upper-middle crust along the eastern margin of the orogenic belt is associated with steeply dipping thrusts that strongly uplift the upper crust and crystalline basement beneath a central fault system in the Longmenshan. The data, combined with an understanding of the regional tectonic stress field and previous geological results, enable a discussion of basin-and-range coupling, orogenic tectonics, the crustal fault system, and the seismogenic tectonic environment of the central Longmenshan along the eastern margin of the Qinghai-Tibet Plateau. | JIA ShiXu LIU BaoJin XU ZhaoFan LIU Zhi FENG ShaoYing ZHANG JianShi LIN JieYan TIAN XiaoFeng LIU QiaoXia GUO WenBin | 2014 | Science China Earth Sciences2014,57,4: | 26
Curative effect of heat-sensitive moxibustion on chronic persistent asthma: a multicenter randomized controlled trial显示文摘OBJECTIVE:To compare the curative effects of heat-sensitive moxibustion with conventional drugs on chronic persistent asthma and seek a valuable therapy to replaceWestern Medicine.METHODS:The participants in this multi-center,randomized,and controlled study were randomly divided into two groups:group A(n=144),treated with heat-sensitive moxibustion(50 sessions)and group B(n=144),treated with Seretide(salmeterol50μg/fluticasone 250μg,twice a day). The scores of asthma control test(ACT),forced expiratory volume in 1 second(FEV1),peak expiratory flow(PEF),and attack frequency were measured after 15,30,60,and 90 days of treatment.Patients followed up3 and 6 months after treatment.RESULTS:There was a significant difference(P=0.0002)in the ACT score and lung function between the two groups after 3 months of treatment and(P=0.000 03)during the follow-up visits. In addition,heat-sensitive moxibustion reduced attack frequency in the period from inclusion to the6-month follow-up visit.CONCLUSION:This study shows that heat-sensitive moxibustion may have a comparable curative effect toSeretide(salmeterol/fluticasone)on asthma. | Rixin Chen Mingren Chen Jun Xiong Zhenhai Chi B0 Zhang Ning Tian Zhenhua Xu Tangfa Zhang Wanyao Li Wei Zhang Xiaofeng Rong Zhen Wang Gang Sun Baohe Ge Guoxiong Yu Nanchang Song | 2013 | Journal of Traditional Chinese Medicine2013,33,5: | 20
A Pilot Study of the Therapeutic Efficacy and Mechanism of Artesunate in the MRL/lpr Murine Model of Systemic Lupus Erythematosus显示文摘Recent evidence indicates that artesunate has immunomodulatory properties that might be useful for treating autoimmune disease. In this study,we conducted a pilot study and explored the effect and mechanism of artesunate on the treatment of systemic lupus erythematosus using an MRL/lpr murine model.MRL/lpr mice were divided into control, cyclophosphamide(CTX)and artesunate treatment groups.Blood was collected to measure serum levels of creatinine, antinuclear antibody(ANA)and anti-double-stranded DNA(anti-dsDNA)antibody.Twenty-four-hour urine was collected to measure levels of proteinuria. The concentration of monocyte chemotactic protein-1(MCP-1)in serum and urine was measured. The expression of MCP-1 in kidney was detected by Western blot and immunohistochemistry assay, respectively. The expression of B cell activating factor(BAFF)in spleen was determined by real time-PCR and immunoblotting. We found that artesunate significantly increased the survival rate, body weight and blood leukocyte counts, and reduced the serum levels of ANA and anti-dsDNA antibody titer,24 h urinary protein, and serum creatinine. Our results indicated that artesunate could decrease MCP-1,major pro-inflammation cytokine,in serum, urine and kidney. We also found that the level of BAFF,the major B cell activation factor, was decreased in artesunate treated MRL/lpr mice. Its efficacy was comparable with that of CTX in this study.Taken together,we have demonstrated that artesunate can inhibit the progression of disease and reverse the pathologic lesion of lupus nephritis. | Ouyang Jin Huayong Zhang Zhifeng Gu Shengnan Zhao Ting Xu Kangxing Zhou Bo Jiang Jie Wang Xiaofeng Zeng Lingyun Sun | 2009 | Cellular & Molecular Immunology2009,6,6: | 20
Search for the QCD critical point with fluctuations of conserved quantities in relativistic heavy-ion collisions at RHIC: an overview显示文摘Fluctuations of conserved quantities, such as baryon, electric charge, and strangeness number, are sensitive observables in relativistic heavy-ion collisions to probe the QCD phase transition and search for the QCD critical point. In this paper, we review the experimental measurements of the cumulants(up to fourth order) of event-byevent net-proton(proxy for net-baryon), net-charge and netkaon(proxy for net-strangeness) multiplicity distributions Au+Au collisions at sNN^(1/2) 7:7; 11:5; 14:5; 19:6; 27;39; 62:4; 200 Ge V from the first phase of beam energy scan program at the relativistic heavy-ion collider(RHIC). We also summarize the data analysis methods of suppressing the volume fluctuations, auto-correlations, and the unified description of efficiency correction and error estimation.Based on theoretical and model calculations, we will discuss the characteristic signatures of critical point as well as backgrounds for the fluctuation observables in heavy-ion collisions. The physics implications and the future secondphase of the beam energy scan(2019–2020) at RHIC will also be discussed. | Xiaofeng Luo Nu XU | 2017 | Nuclear Science and Techniques2017,28,8: | 19

Recent development of casing treatments for aeroengine compressors显示文摘Casing treatment is a mature stabilization technique which has been widely applied on aero-engines for modern aircrafts and turbo-chargers for automobiles. After the investigations of half century since the 1960 s, this technique has been well developed for various configurations with different effectiveness. From the perspective of stabilization mechanism, this paper roughly categorizes the configurations of casing treatment into two types: traditional ones which work by affecting the flow structure of blade tip region; a novel one named as Stall Precursor-Suppressed(SPS) casing treatment. The effectiveness of both types will be demonstrated for their applications on axial compressors and centrifugal compressors with uniform or distorted inlet. And the stabilization mechanism of casing treatments for regular types and SPS one will also be explained respectively. In addition, this review will summarize the methodologies of casing treatments with the numerical simulations for regular grooved configurations and the eigenvalue approach for SPS casing treatment.Looking forward to the future of compressor stabilization, casing treatment technique will still exist as a general and inexpensive option, and the exploration for its effectiveness and mechanism will be deeper with the development of computational fluid dynamics and advanced measurement techniques. | Xiaofeng SUN Xu DONG Dakun SUN | 2019 | Chinese Journal of Aeronautics2019,32,1: | 16

Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants, such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents, near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination, with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants, and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions. In contrast, the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite, indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants, mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents, since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations. Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together, these results suggest that the deployment of RBD-vaccines,through a third-dose boost, may be ideal for combating SARS-CoV-2 variants when necessary, especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12
Construction of a host range-expanded hybrid baculovirus of BmNPV and AcNPV, and knockout of cysteinase gene for more efficient expression显示文摘AcNPV (Autographa californica nuclear polyhedrosis virus) and BmNPV(Bombyx mori nuclear polyhedrosis virus) are two principal insect-baculovirus expression systems, each having different characteristics. AcNPV has a wider host range and can infect a series of cell lines thus making it suitable for cell suspension culture expression, but the small size of the host insect, A. californica, makes AcNPV less suitable for large scale protein synthesis. In contrast, BmNPV can only infect the silkworm, Bombyx mori, which is well-known for its easy rearing and large size. These characteristics make the BmNPV system especially suitable for large-scale industrial ex-pression. To utilize the advantages of both AcNPV and BmNPV, we tried to expand their host range through homologous recombination and successfully constructed a hybrid baculovirus of AcNPV and BmNPV, designated as HyNPV. The hybrid baculovirus can infect the hosts of both AcNPV and BmNPV. Taking the human basic fibroblast growth factor (bFGF) gene as an appli-cation example, we constructed a recombinant, HyNPV-bFGF. This construct is able to express the bFGF protein both in silkworm larvae and in common-use cell lines, sf21, sf9 and High-five. Moreover, to reduce the loss of recombinant protein due to degradation by proteases that are simultaneously expressed by the baculovirus, we knocked out the cysteinase gene coding for one of the most important baculovirus proteases. This knockout mutation improves the produc-tion efficiency of the bFGF recombinant protein. | WU Xiaofeng, CAO Cuiping, XU Yaxiang & LU Xingmeng College of Animal Sciences, Zhejiang University, Huajiachi Campus, Hangzhou 310029, China | 2004 | Science China(Life Sciences)2004,47,5: | 12
Low-mature gases and typical low-mature gas fields in China显示文摘No natural gas pool of industrial importance could be formed at the low-evolution stage of organic matter. In the 1980s, on the basis of the development in exploration practice, the hypotheses of bio-thermo-catalytic transitional zone gases and early thermogenic gases were proposed. The lower-limit Ro values for the formation and accumulation of natural gases of industrial importance have been expanded to 0.3%―0.4%. In the light of the two-stage model established on the basis of carbon isotope fractionation in coal-type natural gases, the upper-limit Ro values have been set at 0.8%―1.0%. In terms of the geological practice in the low-mature gas zones and China's main coal-type gas fields, it is feasible and proper to set the upper-limit Ro value of low-mature gases at 0.8%. Supper-large gas fields such as the Urengoy gas field in western Siberian Basin should belong to low-mature gas fields, of which the natural gas reserves account for more than 20% of the global proven reserves, providing strong evidence for the significance of such a type of resources. The proven natural gas reserves in the Turpan-Hami Basin of China have almost reached 1000 × 108 m3. The main source rocks in this area are the Jurassic Xishanyao Formation, which occurs as a suite of coal series strata. The corresponding thermal evolution indices (Ro ) are mainly within the range of about 0.4%―0.8%, the δ 13C1 values of methane vary between-44‰ and-39‰ (correspondingly Ro =0.6%―0.8%), and those of ethane are within the range of-29‰―-26‰, indicating that natural gases in the Turpan-Hami Basin should be designated to coal-type low-mature gases. The light hydrocarbon evolution indices of natural gases also fall within the area of low evolution while the precursor type of light hydrocarbons also shows the characteristics of the coal-type. The geological background, carbon isotopic composition and light hydrocarbon index all provide strong evidence suggesting that the proven natural gases in the Turpan-Hami Basin are low-mature gases. In China a gas field with the gas reserves reaching 300 ×108 m3 can be defined as a large gas field, and thus the proven low-mature gases in the Turpan-Hami Basin are equivalent to the reserves of three large gas fields. Its existence is of great significance in research on and exploration of low-mature gases in China. | XU YongChang WANG ZhiYong WANG XiaoFeng ZHENG JianJing DU HongYu | 2008 | Science China Earth Sciences2008,51,2: | 11
Subduction tectonics vs.plume tectonics——Discussion on driving forces for plate motion显示文摘Plate tectonics describes the horizontal motions of lithospheric plates, the Earths outer shell, and interactions among them across the Earths surface.Since the establishment of the theory of plate tectonics about half a century ago, considerable debates have remained regarding the driving forces for plate motion. The early'Bottom up'view,i.e.,the convecting mantledriven mechanism, states that mantle plumes originating from the core-mantle boundary act at the base of plates, accelerating continental breakup and driving plate motion.Toward the present, however, the'Top down'idea is more widely accepted, according to which the negative buoyancy of oceanic plates is the dominant driving force for plate motion, and the subducting slabs control surface tectonics and mantle convection. In this regard, plate tectonics is also known as subduction tectonics.'Top down'tectonics has received wide supports from numerous geological and geophysical observations. On the other hand, recent studies indicate that the acceleration/deceleration of individual plates over the million-year timescale may reflect the effects of mantle plumes. It is also suggested that surface uplift and subsidence within stable cratonic areas are correlated with plumerelated magmatic activities over the hundred-million-year timescale. On the global scale, the cyclical supercontinent assembly and breakup seem to be coupled with superplume activities during the past two billion years. These correlations over various spatial and temporal scales indicate the close relationship and intensive interactions between plate tectonics and plume tectonics throughout the history of the Earth and the considerable influence of plumes on plate motion.Indeed,we can acquire a comprehensive understanding of the driving forces for plate motion and operation mechanism of the Earth's dynamic system only through joint analyses and integrated studies on plate tectonics and plume tectonics. | Ling CHEN Xu WANG Xiaofeng LIANG Bo WAN Lijun LIU | 2020 | Science China Earth Sciences2020,63,3: | 10
Land-surface processes and summer-cloud-precipitation characteristics in the Tibetan Plateau and their effects on downstream weather:a review and perspective显示文摘Correct understanding of the land-surface processes and cloud-precipitation processes in the Tibetan Plateau(TP)is an important prerequisite for the study and forecast of the downstream activities of weather systems and one of the key points for understanding the global atmospheric movement. In order to show the achievements that have been made, this paper reviews the progress on the observations for the atmospheric boundary layer,land-surface heat fluxes,cloud-precipitation distributions and vertical structures by using ground-and space-based multiplatform, multisensor instruments and the effect of the cloud system in the TP on the downstream weather. The results show that the form drag related to the topography,land-atmosphere momentum and scalar fluxes is an important part of the parameterization process. The sensible heat flux decreased especially in the central and northern TP caused by the decrease in wind speeds and the differences in the ground-air temperatures.Observations show that the cloud and precipitation over the TP have a strong diurnal variation.Studies also show the compressed-air column in the troposphere by the higher-altitude terrain of the TP makes particles inside clouds vary at a shorter distance in the vertical direction than those in the non-plateau area so that precipitation intensity over the TP is usually small with short duration, and the vertical structure of the convective precipitation over the TP is obviously different from that in other regions. In addition, the influence of the TP on severe weather downstream is preliminarily understood from the mechanism. It is necessary to use model simulations and observation techniques to reveal the difference between cloud precipitation in the TP and non-plateau areas in order to understand the cloud microphysical parameters over the TP and the processes of the land boundary layer affecting cloud, precipitation and weather in the downstream regions. | Yunfei Fu Yaoming Ma Lei Zhong Yuanjian Yang Xueliang Guo Chenghai Wang Xiaofeng Xu Kun Yang Xiangde Xu Liping Liu Guangzhou Fan Yueqing Li Donghai Wang | 2020 | National Science Review2020,7,3: | 10
Antenatal taurine reduces cerebral cell apoptosis in fetal rats with intrauterine growth restriction显示文摘From pregnancy to parturition, Sprague-Dawley rats were daily administered a low protein diet to establish a model of intrauterine growth restriction. From the 12 th day of pregnancy, 300 mg/kg taurine was daily added to food until spontaneous delivery occurred. Brain tissues from normal neonatal rats at 6 hours after delivery, neonatal rats with intrauterine growth restriction, and neo-natal rats with intrauterine growth restriction undergoing taurine supplement were obtained for fur-ther experiments. The terminal deoxyribonucleotidyl transferase (TdT)-mediated biotin-16-dUTP nick-end labeling assay revealed that the number of apoptotic cells in the brain tissue of neonatal rats with intrauterine growth restriction significantly increased. Taurine supplement in pregnant rats reduced cell apoptosis in brain tissue from neonatal rats with intrauterine growth restriction. Immu-nohistochemical staining revealed that taurine supplement increased glial cell line-derived neuro-trophic factor expression and decreased caspase-3 expression in the cerebral cortex of intrauterine growth-restricted fetal rats. These results indicate that taurine supplement reduces cell apoptosis through the glial cell line-derived neurotrophic factor-caspase-3 signaling pathway, resulting in a protective effect on the intrauterine growth-restricted fetal rat brain. | Jing Liu Xiaofeng Wang Ying Liu Na Yang Jing Xu Xiaotun Ren | 2013 | Neural Regeneration Research2013,8,23: | 9
