| 1 | Early Serum HBsAg Kinetics as Predictor of HBsAg Loss in Patients with HBeAg-Negative Chronic Hepatitis B after Treatment with Pegylated Interferonα-2a显示文摘Hepatitis B surface antigen(HBsAg)loss is an ideal treatment endpoint for patients with chronic hepatitis B(CHB).We investigated the predictive value of on-treatment HBsAg levels for HBsAg loss in hepatitis B e antigen(HBe Ag)-negative CHB patients who received 120-week PEG-IFNα-2a treatment.Serum HBV DNA,HBsAg,and anti-HBs levels were assayed at baseline and every 3 months during the treatment.Of 81 patients,12 achieved HBsAg loss,20 achieved HBsAg\100 IU/mL,and 49 maintained HBs Ag C 100 IU/mL.HBsAg loss rate was only 3.7%at 48 weeks,while it reached to 11.1%and 14.8%after treatment of 96 weeks and 120 weeks.The cutoff HBs Ag levels at 12 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 400 IU/mL and 750 IU/mL,with AUC 0.725 and 0.722,positive predictive value(PPV)29.41%and 30.56%,and negative predictive value(NPV)93.75%and 97.78%,respectively.The cutoff HBsAg levels at 24 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 174 IU/m L and 236 IU/mL respectively,with AUC 0.925 and 0.922,PPV 40.0%and 46.15%,and both NPV 100%.The predictive ability of the cutoff HBsAg levels at 24 weeks was better than that at 12 weeks for HBs Ag loss at either 96 or 120 weeks(χ~2=3.880,P=0.049 andχ~2=4.412,P=0.036).These results indicate that extended therapy is critical to HBsAg loss in HBe Ag-negative CHB patients during PEG-IFN treatment,and the HBsAg level at 24 weeks can be used to predict HBsAg loss during tailoring PEG-IFN therapy. | Minghui Li Lu Zhang Yao Lu Qiqi Chen Huihui Lu Fangfang Sun Zhan Zeng Gang Wan Linqing Zhao Yao Xie | 2021 | Virologica Sinica2021,36,2: | 15 |
| 2 | Effect of Sc and Zr additions on microstructures and corrosion behavior of Al-Cu-Mg-Sc-Zr alloys显示文摘The effects of adding the alloy element Sc to Al alloys on strengthening, recrystallization and modification of the grain microstructure have been investigated. The combination of Sc and Zr alloying not only produces a remarkable synergistic effect of inhibition of recrystallization and refinement of grain size but also substantially reduce the amount of high-cost additional Sc. In this work, the microstructures and corrosion behavior of a new type of Al-Cu-Mg-Sc-Zr alloy with Sc/Zr ratio of 1/2 were investigated.The experimental results showed that the Sc and Zr additions to Al-Cu-Mg alloy could strongly inhibit recrystallization, refine grain size, impede the segregation of Cu element along the grain boundary and increase the spacing of grain boundary precipitates. In addition, adding Sc and Zr to Al-Cu-Mg alloy effectively restricts the corrosion mechanism conversion associated with Al2 Cu Mg particles, which resulted in the change of the cross-section morphology of inter-granular corrosion from an undercutting to an elliptical shape. The susceptibility to inter-granular corrosion was significantly decreased with increasing Sc and Zr additions to the Al-Cu-Mg alloy. | 3 | sLe^(x) expression in invasive micropapillary breast carcinoma is associated with poor prognosis and can be combined with MUC1/EMA as a supplementary diagnostic indicator显示文摘Objective: Mucin 1(MUC1/EMA) and sialyl Lewis X(sLe^(x)) indicate polarity reversal in invasive micropapillary carcinoma(IMPC). The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to assess their diagnostic and prognostic value in patients with IMPC.Methods: The expression of sLe^(x) and MUC1/EMA in 100 patients with IMPC and a control group of 89 patients with invasive ductal carcinoma not otherwise specified(IDC-NOS) were analyzed with IHC. Fresh tumor tissues were collected from patients with IMPC or IDC-NOS for primary culture and immunofluorescence analysis.Results: The rate of nodal metastasis was higher in patients with IMPC than those with IDC-NOS, and IMPC cells tended to express more sLe^(x) and MUC1/EMA in the cytomembranes(the stroma-facing surfaces of the micropapillary clusters) than IDC-NOS cells. In IMPC, high cytomembrane expression of sLe^(x), but not MUC1/EMA, indicated poor prognosis. In addition, among the 100 patients with IMPC, 10 patients had sLe^(x)+/EMA-expression patterns, and 8 patients had sLe^(x)-/EMA+ expression patterns. The primary IMPC cells were suspended, non-adherent tumor cell clusters, whereas the primary IDC cells were adherent tumor cells. | 7 | Effects of copper mining on heavy metal contamination in a rice agrosystem in the Xiaojiang River Basin, southwest China显示文摘Metallic ore mining causes heavy metal pollution worldwide.However,the fate of heavy metals in agrosystems with long-term contamination has been poorly studied.Dongchuan District(Yunnan,southwest China),located at the middle reaches of the Xiaojiang River,is a well-known 2000-year-old copper mining site.In this work,a survey on soil heavy metal contents was conducted using a handheld X-ray fluorescence instrument to understand the general contamination of heavy metals in the Xiaojiang River Basin.Furthermore,river water,soil,and rice samples at six sites along the fluvial/alluvial fans of the river were collected and analyzed to implement an environmental assessment and an evaluation of irrigated agrosystem.V,Zn,and Cu soil levels(1724,1047,and 696mg·kg-1,respectively)far exceeded background levels.The geo-accumulation indexes(Igeo)showed that cultivated soils near the mining sites were polluted by Cd and Cu,followed by Zn,V,Pb,Cr,Ni,and U.The pollution index(Pi)indicated that rice in the area was heavily polluted with Pb,Cr,Cd,Ni,Zn,and Cu.The difference in orders of metal concentrations between the soil and rice heavy metal contamination was related to the proportion of bioavailable heavy metals in the soil.The crop consumption risk assessment showed that the hazard quotient exceeded the safe threshold,indicating a potential carcinogenic risk to consumers.The Nemerow integrated pollution index and health index indicated that the middle of the river(near the mining area)was the heaviest polluted site. | Wanqiu Pu Jiaqi Sun Fangfang Zhang Xingyue Wen Wenhu Liu Chengmin Huang | 2019 | Acta Geochimica2019,38,5: | 4 |
| 8 | Concentrations of heavy metals and polycyclic aromatic hydrocarbons in needles of Masson pine (Pinus massoniana L.) growing nearby different industrial sources显示文摘Emissions from industrial activities pose a serious threat to human health and impose the need for monitoring both inorganic and organic pollutants in industrial areas. We selected Masson pine (Pinus massoniana L.) as potential biomonitor and collected the current (C) and previous year (C+1) needles from three industrial sites dominated by petrochemical, ceramics manufacturing, and iron and steel smelting plants and one remote site to determine heavy metals (Cu, Cd, Pb, Zn, Cr, Ni and Co) and polycyclic aromatic hydrocarbons (PAHs) in unwashed and water-washed needles. Both unwashed and washed C+1 needles showed generally higher concentrations of heavy metals and PAHs than C needles, although the washed needles more clearly spotlighted the accumulation effect of PAHs over exposure time. Water-washing resulted in a significant decrease in needle PAH concentrations with more significant effects shown in C needles. By contrast, needle heavy metal concentrations were much less affected by washing. Although heavy metals and PAHs might differ in adsorption and uptake strategies, their higher concentrations in the needles at the industrial sites indicated conspicuous contamination due to industrial emissions there. | 9 | Establishment of a new acute-on-chronic liver failure model显示文摘To establish an animal model of acute-on-chronic liver failure(ACLF) that would replicate the pathological process of ACLF in humans, rats were administered porcine serum(PS) for 11 weeks. Liver fibrosis was determined by pathological and biochemical assessments. The animals then were injected with D-galactosamine(D-gal) and lipopolysaccharide(LPS). The survival times of animals with cirrhosis and ACLF were determined over 48 h. Other animals were killed at 0, 4, 8 and 12 h after administration of D-gal/LPS. Liver injury was assessed by histopathological analysis and biochemical indices, and apoptosis was detected by Western blot and TUNEL analysis. After PS administration for 11 weeks the serum levels of hyaluronic acid and N-procollagen type III peptide increased significantly, and serious fibrosis and cirrhosis was observed at weeks 10 and 11. Cirrhotic rats were injected with D-gal/LPS to induced ACLF; the rate of mortality over 48 h was 80%. ALT and AST levels increased markedly at 4 h, but decreased significantly at8 and 12 h post-treatment. The total bilirubin, direct bilirubin, and total bile acids levels increased markedly at 8 and 12 h. Clotting times, TNF-α and IL-6 levels increased significantly, except for 12 h post-treatment.Apoptosis, inflammation and necrosis were elevated as determined by hematoxylin-eosin staining and TUNEL assays. BCL-2 levels decreased significantly, While BAX levels increased significantly.Cytochrome c expression peaked at 8 h postD-gal/LPS treatment. | 11 | Comparison of the interface reaction behaviors of CaO–V_(2)O_(5) and MnO_(2)–V_(2)O_(5) solid-state systems based on the diffusion couple method显示文摘The formation mechanism of calcium vanadate and manganese vanadate and the difference between calcium and manganese in the reaction with vanadium are basic issues in the calcification roasting and manganese roasting process with vanadium slag.In this work,CaO–V_(2)O_(5) and MnO_(2)–V_(2)O_(5) diffusion couples were prepared and roasted for different time periods to illustrate and compare the diffusion reaction mechanisms.Then,the changes in the diffusion product and diffusion coefficient were investigated and calculated based on scanning electron microscopy (SEM) with energy dispersive X-ray spectroscopy (EDS) analysis.Results show that with the extension of the roasting time,the diffusion reaction gradually proceeds among the CaO–V_(2)O_(5) and MnO_(2)–V_(2)O_(5) diffusion couples.The regional boundaries of calcium and vanadium are easily identifiable for the CaO–V_(2)O_(5) diffusion couple.Meanwhile,for the MnO_(2)–V_(2)O_(5) diffusion couple,MnO_(2) gradually decomposes to form Mn_(2)O_(3),and vanadium diffuses into the interior of Mn_(2)O_(3).Only a part of vanadium combines with manganese to form the diffusion production layer.CaV_(2)O_(6) and MnV_(2)O_(6) are the interfacial reaction products of the CaO–V_(2)O_(5) and MnO_(2)–V_(2)O_(5) diffusion couples,respectively,whose thicknesses are 39.85 and 32.13μm when roasted for 16 h.After 16 h,both diffusion couples reach the reaction equilibrium due to the limitation of diffusion.The diffusion coefficient of the CaO–V_(2)O_(5) diffusion couple is higher than that of the MnO_(2)–V_(2)O_(5) diffusion couple for the same roasting time,and the diffusion reaction between vanadium and calcium is easier than that between vanadium and manganese. | Jing Wen Hongyan Sun Tao Jiang Bojian Chen Fangfang Li Mengxia Liu | 2023 | International Journal of Minerals,Metallurgy and Materials2023,30,5: | 2 |
| 12 | The partitioning behavior of trace elements in subduction zones:Advances and prospects显示文摘The partitioning behavior of trace elements is of key importance for understanding the geochemical process and material cycle mechanism in subduction zones.This paper focuses on the advances and prospects on the studies of trace element partitioning in subduction zones from the following four aspects.(1)The properties of fluids derived from subducting slabs and their ability in element transport.How slab-derived solute-rich fluids and supercritical fluids are formed and what the roles and key control factors of these fluids are in transferring of elements(especially the high field strength elements)from slab to wedge are discussed.We point out that the detailed investigations of supercritical fluids may provide a new perspective for the element migration mechanism,material cycle process,arc magma genesis and so on.(2)The behavior of transition elements during mantle wedge melting.The behavior of the first row transition elements(Sc,Ti,V,Cr,Mn,Fe,Co,Ni,Cu,Zn)in the mantle partial melting process is compatible or incompatible,depending on residual mineral assemblage and physicochemical conditions.The partitioning behavior of the elements such as Sc,Ti,Co,Ni and Zn whose valence states do not change in the melting process mainly depends on the residual mineral assemblage and temperature,whereas the partitioning behavior of the multivalent elements such as V and Fe is also the function of oxygen fugacity(fO_(2))in addition to mineral assemblage and temperature.Therefore,the partitioning behavior of transition elements has important applications in tracing lithologic inhomogeneity and fO_(2)of the mantle wedges.(3)The specificity of element partitioning behavior during arc magma evolution.Garnet has Dy/Yb partitioning behavior different from amphibole,and rutile has Nb/Ta partitioning behavior different from amphibole.Dy/Yb and Nb/Ta partitioning differences for these minerals enable to distinguish the specific evolution process of arc magmas.The Dy/Yb and Nb/Ta generally decrease with the increase of SiO_(2)in arc magmas,indicating that amphibole fractionation should be the most important during arc magma differentiation.(4)The behavior of sulfur and chalcophile elements and porphyry metallogeny.In subduction zones,the behavior of chalcophile elements such as Cu and Au is controlled by sulfide and fluid.Therefore,the stability of sulfide,the time at which the fluid exsolves from the melt relative to sulfide saturation,the fluid/sulfide mass ratio and fluid/melt Cu and Au partition coefficients in intermediate-felsic magma-H_(2)O systems are especially important in understanding Cu and Au enrichment in magma-hydrothermal processes.Intermediate-felsic magmas mainly originate from the differentiation of arc magmas at lower crustal reservoirs,and thus the fluid exsolution from the lower crustal reservoirs and the fluid/melt and fluid/sulfide partition coefficients of Cu and Au should be the keys to understanding quantitatively how Cu and Au are migrated from the deep crust to the shallow site of mineralization. | Xiaolin Xiong Xingcheng Liu Li Li Jintuan Wang Wei Chen Mengfei Ruan Ting Xu Zhongxing Sun Fangfang Huang Jianping Li&Lei Zhang | 2020 | Science China Earth Sciences2020,63,12: | 2 |

| 14 | The Scorpion Venom Peptide Smp76 Inhibits Viral Infection by Regulating Type-Ⅰ Interferon Response显示文摘Dengue virus(DENV) and Zika virus(ZIKV) have spread throughout many countries in the developing world and infect millions of people every year, causing severe harm to human health and the economy. Unfortunately, there are few effective vaccines and therapies available against these viruses. Therefore, the discovery of new antiviral agents is critical.Herein, a scorpion venom peptide(Smp76) characterized from Scorpio maurus palmatus was successfully expressed and purified in Escherichia coli BL21(DE3). The recombinant Smp76(rSmp76) was found to effectively inhibit DENV and ZIKV infections in a dose-dependent manner in both cultured cell lines and primary mouse macrophages. Interestingly,rSmp76 did not inactivate the viral particles directly but suppressed the established viral infection, similar to the effect of interferon(IFN)-b. Mechanistically, rSmp76 was revealed to upregulate the expression of IFN-b by activating interferon regulatory transcription factor 3(IRF3) phosphorylation, enhancing the type-Ⅰ IFN response and inhibiting viral infection.This mechanism is significantly different from traditional virucidal antimicrobial peptides(AMPs). | 16 | LILRB4 ITIMs mediate the T cell suppression and infiltration of acute myeloid leukemia cells显示文摘We recently demonstrated that leukocyte Ig-like receptor 4(LILRB4)expressed by monocytic acute myeloid leukemia(AML)cells mediates T-cell inhibition and leukemia cell infiltration via its intracellular domain.The cytoplasmic domain of LILRB4 contains three immunoreceptor tyrosine-based inhibitory motifs(ITIMs);the tyrosines at positions 360,412,and 442 are phosphorylation sites.Here,we analyzed how the ITIMs of LILRB4 in AML cells mediate its function.Our in vitro and in vivo data show that Y412 and Y442,but not Y360,of LILRB4 are required for T-cell inhibition,and all three ITIMs are needed for leukemia cell infiltration.We constructed chimeric proteins containing the extracellular domain of LILRB4 and the intracellular domain of LILRB1 and vice versa.The intracellular domain of LILRB4,but not that of LILRB1,mediates T-cell suppression and AML cell migration.Our studies thus defined the unique signaling roles of LILRB4 ITIMs in AML cells. | Zunling Li Mi Deng Fangfang Huang Changzhu Jin Shuang Sun Heyu Chen Xiaoye Liu Licai He Ali HSadek Cheng Cheng Zhang | 2020 | Cellular & Molecular Immunology2020,17,3: | 1 |
