1Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 2Emerging roles of transforming growth factor β signaling in wet age-related macular degeneration显示文摘Age-related macular degeneration (AMD) is one of the major causes of irreversible blindness among aging populations in developed countries and can be classified as dry or wet according to its progression.Wet AMD,which is characterized by angiogenesis on the choroidal membrane,is uncommonly seen but more severe.Controlling or completely inhibiting the factors that contribute to the progression of events that lead to angiogenesis may be an effective strategy for treating wet AMD.Emerging evidence has shown that transforming growth factor-β(TGF-β) signaling plays a significant role in the progression of wet AMD.In this review,we described the roles of and changes in TGF-β signaling in the development of AMD and discussed the mechanisms of the TGF-β superfamily in choroidal neovascularization (CNV) and wet AMD,including the modulation of angiogenesis-related factors,inflammation,vascular fibrosis,and immune responses,as well as cross-talk with other signaling pathways.These remarkable findings indicate that TGF-β signaling is a potential target for wet AMD treatment.Kai Wang Haoran Li Ruipu Sun Chaxian Liu Yunfei Luo Shuhua Fu Ying Ying 2019Acta Biochimica et Biophysica Sinica2019,51,1:5
3Effect of Preheating Temperature on Surface Cracking of High Nitrogen CrMn Austenitic Stainless Steel显示文摘18Mn18Cr0.5N steel specimens were preheated at the temperatures from 1100 to 1250°C for 5 min,and then cooled to 950°C and compressed.The cracking behaviors were investigated using optical microscopy and scanning electron microscopy.The results showed that the hot workability of 18Mn18Cr0.5N steel gradually decreased with increasing preheating temperature between 1100 and 1200°C,and quickly deteriorated up to 1250°C.Above 1200°C,delta ferrite particles appeared in 18Mn18Cr0.5N steel,promoted cavity coalescence on grain boundary,and accelerated surface crack formation during the hot working process.Zhenhua Wang Wantang Fu Shuhua Sun Zhiqing Lv Wenhui Zhang 2010Journal of Materials Science & Technology2010,26,9:4
4The direct targets of CBFs:In cold stress response and beyond显示文摘Cold acclimation in Arabidopsis thaliana triggers a significant transcriptional reprogramming altering the expression patterns of thousands of cold-responsive(COR) genes. Essential to this process is the C-repeat binding factor(CBF)-dependent pathway, involving the activity of AP2/ERF(APETALA2/ethylene-responsive factor)-type CBF transcription factors required for plant cold acclimation. In this study, we performed chromatin immunoprecipitation assays followed by deep sequencing(ChIP-seq) to determine the genomewide binding sites of the CBF transcription factors. Cold-induced CBF proteins specifically bind to the conserved C-repeat(CRT)/dehydrationresponsive elements(CRT/DRE;G/ACCGAC) of their target genes. A Gene Ontology enrichment analysis showed that 1,012 genes are targeted by all three CBFs. Combined with a transcriptional analysis of the cbf1,2,3 triple mutant, we define 146 CBF regulons as direct CBF targets. In addition, the CBF-target genes are significantly enriched in functions associated with hormone, light,and circadian rhythm signaling, suggesting that the CBFs act as key integrators of endogenous and external environmental cues. 5Levels of Crotonaldehyde and 4-hydroxy-(E)-2-nonenal and Expression of Genes Encoding Carbonyl-Scavenging Enzyme at Critical Node During Rice Seed Aging显示文摘The critical node(CN) is an important stage during seed aging, which is related to effective genebank conservation. Previous studies have demonstrated that proteins undergo carbonylated modification at the CN in rice, indicating oxidative damage. However, the levels of reactive carbonyl species(RCS) and the associated scavenging system at the CN are largely unknown. In this study, we optimized methods for the extraction and analysis of RCS from dry rice embryos. In order to acquire seeds at the CN, rice seeds were subjected to natural conditions for 7, 9, 11 and 13 months, and the seed germination rates were reduced to 90%, 82%, 71% and 57%, respectively. We chose the stage with seed germination rate of 82% as the CN according to the rice seed vigor loss curve. The levels of crotonaldehyde and 4-hydroxy-(E)-2-nonenal(HNE) were significantly increased at the CN. In addition, genes encoding carbonyl-scavenging enzyme, including Os ALDHs and Os AKRs, were significantly down-regulated at the CN, and reductions in the expression of Os ALDH2-2, Os ALDH2-5, Os ALDH3-4, Os ALDH7, Os AKR1 and Os AKR2 in particular could be responsible for RCS accumulation. 6Simultaneous determination of Repaglinide and Irbesartan in biological plasmas using micellar enhanced excitation-emission matrix fluorescence coupled with ATLD method显示文摘A highly sensitive and selective 3D excitation-emission fluorescence method has been proposed to rapidly quantify the combined antidiabetics Repaglinide(Re) and Irbesartan(Ir) in rat and human plasmas with the aid of second-order calibration method based on alternating trilinear decomposition(ATLD) method. Re and Ir with weak fluorescence can be endowed with strong fluorescent property by changing the microenvironment in samples and improving the fluorescence quantum yield by using an appropriate micellar enhanced surfactant. The enhanced excitation-emission matrix fluorescence of Re and Ir can be accurately resolved and can simultaneously attain the optimal concentration even in the presence of a potentially strong intrinsic fluorescence from complex biological matrices, such as rat and human plasmas, by using the ATLD method, which completely exploits the 'second-order advantage'. The average recoveries of Re and Ir obtained from ATLD with the factor number of 3(N=3) were 101.0%±4.3% and 99.1%±4.1% for rat plasma and 100.5%±5.4% and 97.1%±3.6% for human plasma. Several statistical methods, including Student's t-test, figures of merit, and elliptical joint confidence region, have been utilized to evaluate the accuracy of the proposed method. 14In-situ imaging of strain-induced enhancement of hydrogen evolution activity on the extruded MoO_(2)sheets显示文摘Strain engineering is a useful strategy for modifying the catalytic activity of electrocatalysts.However,in-situ visual characterization of the strain effect on the catalytic activity at nanoscale remains a huge challenge.Herein,we performed in-situ electrochemical scanning tunneling microscopy(EC-STM)imaging measurements at the local strained regions of extruded singlecrystal molybdenum dioxide(MoO_(2))sheets with combination of current noise analysis(n-EC-STM).The intensity-enhanced noise was observed at the local strained region compared to the unstrained regions in the same frame,which reveals the positive effect of compressive strain on the hydrogen evolution reaction(HER)activity of MoO_(2)provided that the intensity of noise is positively correlated with catalytic HER Faradic current.Therefore,we clearly“see”the strain-induced enhancement of HER activity of MoO_(2)at nanoscale by means of noise visualization.This work extends the visual characterization of strain engineering in electrocatalysis and related fields.Mansheng Liao Qiaomei Zhu Shuhua Li Qiongqiong Li Ziting Tao Yongchun Fu 2023Nano Research2023,16,4:0

16Plasma-inducedε-MnO_(2)based aqueous zinc-ion batteries and their dissolution-deposition mechanism显示文摘MnO_(2)has attracted great interest in working as the cathode of zinc ion batteries.However,the development of high-capacity,high-energy-density,and durable manganese-based cathodes with an easy synthesis strategy and proper energy storage mechanism remains an ongoing challenge.Herein,a facile plasmainduced strategy was demonstrated to introduce oxygen vacancies into theε-MnO_(2),and the obtained oxygen vacancies-richε-MnO_(2)nanosheets(ε-MnO_(2-x))show satisfactory electrochemical performances.Furthermore,an appropriate energy storage mechanism for dissolution/deposition was proposed.Thanks to a synergistic effect of the oxygen vacancies inε-MnO_(2)nanosheets and the exposed free-standing collector for Mn^(2+) dissolution/deposition,theε-MnO_(2-x) nanosheets electrode delivers a remarkable capacity(337 mAh g^(-1)at 0.1 A g^(-1))and exhibits an ultrahigh energy density of 462 Wh kg^(-1)(based on the weights of the cathode active material).Furthermore,impressive durability with 85.9%capacity retention after 1000 cycles was obtained.The superior electrochemical performance makes the plasma-induced strategy promising for designing advanced metal oxide electrode materials for high-performance aqueous zinc ion batteries.Le Zhang Shuhua Yang Wenqing Fu Yanwei Cui Jieqiang Wang Degang Zhao Chao Yang Xiutong Wang Bingqiang Cao 2022Journal of Materials Science & Technology2022,,32:0
17Expression of GDF-5 during Limb Skeletal Development of Mice and the effect of GDF-5 on bone marrow mesenchymal stem cells in vitro显示文摘Objective: To investigate the expression of growth differentiation factor 5(GDF-5) during limb skeletal development of mice and the effect of GDF-5 on bone marrow mesenchymal stem cells in vitro. Methods: The expression of GDF-5 mRNA and protein in mouse fetal limb buds were detected in embryonic day 11.5-15.5(E11.5-15.5) by RT-PCR and Western blotting respectively. Type Ⅱ collagen protein was examined with immunocytochemistry and the sulfate glycosaminoglycan was measured by Alcian blue. Results: During early stage of developmental skeletogenesis, the expression of GDF-5mRNA was constant and began with embryos E11.5, highlighted at embryos E12.5 and E13.5, subsequently dropped at embryos E14.5 and E15.5. There was very significant difference(P < 0.01) in average light density ratio of GDF-5/β-actin between E12.5-13.5 and the other three days. The expression of GDF-5 protein had a similar change with mRNA during limb skeletogenesis. Immunocytochemistry showed that GDF-5 could promote expression of Type Ⅱ collagen protein and histological staining of proteoglycan with Alcian blue revealed the deposition of typical cartilage extracellular matrix components. 18Modulating the electronic spin state by constructing dual-metal atomic pairs for activating the dynamic site of oxygen reduction reaction显示文摘In this study,dual-metal atomic pairs of manganese(Mn)-iron(Fe)binuclear sites(BNSs)with two conjoint MnN4 and FeN4 moieties(MnFeN8)anchored onto a graphite-like structure(GLS)(Mn-Fe BNSs/GLS)were constructed.The binuclear MnFeN8 structure was verified experimentally and theoretically.Magnetic measurements and Gaussian calculations reveal that this unique Mn-Fe BNSs exhibit strong short-range electronic interaction between Mn and Fe sites,which decouples two paired d electrons in Fe sites,thereby transforming Fe sites from an intermediate to a high spin state.The optimal electronic configuration of Fe sites and their binuclear structure facilitate an oxygen reduction reaction(ORR)thermodynamically and dynamically,respectively,endowing Mn-Fe BNSs with improved ORR performance.Shenghua Ye Shuhua Xie Yaqi Lei Xiuyuan Yang Jing Hu Lirong Zheng Zhida Chen Yonghuan Fu Xiangzhong Ren Yongliang Li Xiaoping Ouyang Qianling Zhang Jianhong Liu Xueliang Sun 2023Nano Research2023,16,2:0

20Micro-coevolution of host genetics with gut microbiome in three Chinese ethnic groups显示文摘Understanding the micro-coevolution of the human gut microbiome with host genetics is challenging but essential in both evolutionary and medical studies.To gain insight into the interactions between host genetic variation and the gut microbiome,we analyzed both the human genome and gut microbiome collected from a cohort of 190 students in the same boarding college and representing 3 ethnic groups,Uyghur,Kazakh,and Han Chinese.We found that differences in gut microbiome were greater between genetically distinct ethnic groups than those genetically closely related ones in taxonomic composition,functional composition,enterotype stratification,and microbiome genetic differentiation.We also observed considerable correlations between host genetic variants and the abundance of a subset of gut microbial species.Notably,interactions between gut microbiome species and host genetic variants might have coordinated effects on specific human phenotypes.Bacteroides ovatus,previously reported to modulate intestinal immunity,is significantly correlated with the host genetic variant rs12899811(meta-P=5.55×10^(-5)),which regulates the VPS33B expression in the colon,acting as a tumor suppressor of colorectal cancer.These results advance our understanding of the micro-coevolution of the human gut microbiome and their interactive effects with host genetic variation on phenotypic diversity.Mingyue Cheng Xueling Ge Chaofang Zhong Ruiqing Fu Kang Ning Shuhua Xu 2021Journal of Genetics and Genomics2021,48,11:0
