10The biochemical behavior and mechanism of uranium(Ⅵ)bioreduction induced by natural Bacillus thuringiensis显示文摘For a broader understanding of uranium migration affected by microorganisms in natural anaerobic environment,the bioreduction of uranium(Ⅵ)(U(Ⅵ))was revealed in Bacillus thuringiensis,a dominant bacterium strain with potential of uranium-tolerant isolated from uranium contaminated soil.The reduction behavior was systematically investigated by the quantitative analysis of U(Ⅳ)in bacteria,and mechanism was inferred from the pathway of electron transmission.Under anaerobic conditions,appropriate biomass and sodium lactate as electron donor,reduction behavior of U(Ⅵ)induced by B.thuringiensis was restricted by the activity of lactate dehydrogenase,which was directly affected by the initial pH,temperature and initial U(Ⅵ)concentration of bioreduction system.Bioreduction of U(Ⅵ)was driven by the generation of nicotinamide adenine dinucleotide(NADH)from enzymatic reaction of sodium lactate with various dehydrogenase.The transmission of the electrons from bacteria to U(Ⅵ)was mainly supported by the intracellular NADH dehydrogenase-ubiquinone system,this process could maintain the biological activity of cells.Shunzhang Chen Junyuan Gong Yanxia Cheng Yuqi Guo Feize Li Tu Lan Yuanyou Yang Jijun Yang Ning Liu Jiali Liao 2024Journal of Environmental Sciences2024,,2:0
11High expression of EPO gene using un-dhfr negative cell显示文摘Erythropoietin (EPO) genomic gene was cloned and its expression vector pOP13/EPO was constructed. CHOK12 cell was transfected by this vector using lipofectin method. A stable expression cell strain C10 cell with the EPO production at 160IU/d in 10\+6 cells were obtained at 400 μg/mL G418. Based on the C10 cell, another vector pHY/dhfr (dihydrofolate reductase) that carries a dhfr gene and a selecting marker of hygromycin B resistant gene was transferred to this cell. Several cell clones were obtained at 200 μg/mL hygromycin B. These cell clones that can express both EPO gene and exogenous dhfr gene were selected under the progressively increased concentration to 1 μmol methotrexate(MTX). Some high EPO expression cell clones were obtained, the highest expression was 2 400 IU/d in 10\+6 cells, 15 times higher than that without MTX pressure. Then, a method of EPO high expression by using undhfr negative cell was primarily established. 12Serologic study on the outbreak of acute upper respiratory tract infections caused by adenovirus 3显示文摘From April to June,2004,an outbreak of acute upper respiratory tract infections(AURTI)occurred in the north area of Jiangsu Province,China.Twenty throat swabs were collected with 13 of them presenting an adenovirus(Ad)-like cytopathogenic effect on HEp-2.These were verified as Ad by the electron microscope,direct immunofluorescence assay and Ad primer-mediated PCR.Moreover,they were identified as adenovirus type 3(Ad3)by type-specific PCR and sequencing of the amplification products.Subsequent serologic studies were carried out to finally diagnose and document the outbreak.The neutralization test of paired serum of six in nine cases show obviously increased antibodies titers.The positive rate of IgM,IgG and recovery phase neutralization antibodies of the cases were 3.7%,44.4%and 59.5%respectively while those of the controls were 0%,8.3%and 33.3%respectively.The P values of Chi-Square were 0.510,0.018 and 0.226 respectively.The concordance between IgG detected by ELISA and neutralization anti bodies detected by the neutralization test was 61.4%and the P value of Kappa was 0.070.By the serologic study,we can definitively diagnose that this outbreak of acute respiratory infections was caused by Adenovirus 3.JIANG Lufang JU Liwen JIANG Renjie LIN Yuzun ZHOU Liandi YU Shunzhang JIANG Qingwu 2007Frontiers of Medicine2007,1,2:0
13Unusual uranium biomineralization induced by green algae: Behavior investigation and mechanism probe显示文摘As a biosorbent,algae are frequently used for the biotreatment or bioremediation of water contaminated by heavymetal or radionuclides.However,it is unclear that whether or not the biomineralization of these metal or radionuclides can be induced by algae in the process of bioremediation and what the mechanism is.In this work,Ankistrodsemus sp.has been used to treat the uranium-contaminated water,and more than 98%of uranium in the solution can be removed by the alga,when the initial uranium concentration ranges from 10 to 80 mg/L.Especially,an unusual phenomenon of algae-induced uranium biomineralization has been found in the process of uranium bioremediation and its mineralization mechanism has been explored bymultiple approaches.It is worth noticing that the biomineralization of uranium induced by Ankistrodsemus sp.is significantly affected by contact time and pH.Uranium is captured rapidly on the cell surface via complexation with the carboxylate radical,amino and amide groups of themicroalgae cells,which provides nucleation sites for the precipitation of insolubleminerals.Uranium stimulates Ankistrodsemus sp.to metabolize potassium ions(K+),which may endow algae with the ability to biomineralize uranium into the rose-like compreignacite(K_(2)[(UO_(2))6O_(4)(OH)_(6)]•8H_(2)O).As the time increased,the amorphous gradually converted into compreignacite crystals and a large number of crystals would expand over both inside and outside the cells.To the best of our knowledge,this is the first investigated microalgae with a time-dependent uranium biomineralization ability and superior tolerance to uranium.This work validates that Ankistrodsemus sp.is a promising alga for the treatment of uranium-contaminated wastewater.Yanxia Cheng Ting Zhang Shunzhang Chen Feize Li Renwei Qing Tu Lan Yuanyou Yang Jiali Liao Ning Liu 2023Journal of Environmental Sciences2023,,2:0
14Using RAPD in Neisseria gonorrhoeae genotyping and transmission detection显示文摘The aim of this paper is to develop an applic-able Random Amplified Polymorphic DNAs(RAPD)method for genotyping Neisseria gonorrhoeae strain,and discuss the possibility of using the RAPD method to trace N.gonorrhoeae strain transmission route.Four different pretreatment methods were used on the N.gonorrhoeae genomic DNA component,and the best adaptive extract method was selected for RAPD.Different RAPD primers sequence was used for amplification and their differenti-ating capabilities for N.gonorrhoeae strains were com-pared.Applicable RAPD primer was selected for N.gonorrhoeae genotyping and then applied into transmis-sion detection.The results show that the so called cetyl-trimethylammonium bromide(CTAB)method for extracting genomic DNA could give integrated genomic DNA and give out relatively better RAPD fingerprint maps,subsequently,using selected RAPD primer could give out a group of amplification polymerase chain reac-tion bands.The fingerprint maps from different N.gonor-rhoeae strains were distinctive.Some main segments were common to all the N.gonorrhoeae strains tested.Some segments were different among the N.gonorrhoeae strains.According to the fingerprint maps and similarity index of different N.gonorrhoeae isolates,isolates from a pair of sex-partners were very similar.Based on these findings,the best extracting method and suitable RAPD primer were chosen.The RAPD fingerprint maps could type N.gonorrhoeae effectively and could be used as an additional approach in molecular epidemiology for tracing infection sources.Tiejun ZHANG Xiaoming ZHOU Tao ZHANG Yue CHEN Shunzhang YU Qingwu JIANG 2008Frontiers of Medicine2008,2,3:0
