2Localization of NOS-like protein in guard cells of Vicia faba L.and its possible function显示文摘Using the immuno-fluorescence and immuno-gold electron microscope technology, localization of ni- tric oxide synthase (NOS)-like proteins was determined in guard cells of Vicia faba L. NOS is mainly localized in nucleus, cytoplasm, chloroplast, mitochondria and the cell wall of guard cells. Scorch and exogenous JA can enhance the level of nitric oxide (NO) and increase NOS activity in both leaf and epidermis, and the changing pattern of NOS activity was consistent with the change of NO. NOS in- hibitor, L-NAME, inhibited JA-induced NO generation. From the results, we presumed that NO genera- tion from NOS pathway is the main pathway in the stress and JA responses. The pharmacological ex- periment showed that increasing the Ca2+ at a suitable concentration promoted leaf NOS activity and the NO level, indicating that NOS activity together with the distribution of NO is Ca2+-dependent. 3Effect of Ganoderma lucidum spore on expression of insulin-like growth factor-1, nuclear factor-kappa B, and neuronal apoptosis in the epileptic rat brain显示文摘BACKGROUND:It has been reported that Ganoderma lucidum spore powder, a very well known Chinese traditional medicine, can affect immunoregulation, free radical scavenging, and anti-hypoxia responses. OBJECTIVE: To investigate the effect of Ganoderma lucidum spore powder on expression of insulin-like growth factor-1 (IGF-1), nuclear factor-κB (NF-κB) and neuronal apoptosis in rats with pentylenetetrazol (PTZ)-induced epilepsy. DESIGN, TIME AND SETTING: A cellular and molecular biology experiment with randomized controlled study design was performed at the Central Laboratory of Basic Medical College of Jiamusi University from June to August 2005. MATERIALS: Thirty healthy, adult, male, Wistar rats were selected and randomly divided into 3 groups (10 rats per group): control, epilepsy model, and Ganoderma lucidum spore powder. A sub-eclampsia PTZ dose (35 mg/kg) was intraperitoneally injected to induce epilepsy in the latter two groups. Wild Ganoderma lucidum spore powder (30 g/L) was provided by the wild Ganoderma lucidum plant nursery at Jiamusi, China. Immunohistochemical detection and terminal deoxynucleotidyl transferase-mediate dUTP nick end-labeling (TUNEL) kits were purchased from Wuhan Boster Biological Technology Co., Ltd., China. METHODS: Ganoderma lucidum spore powder was intragastrically administered at a dose of 10.0 mL/kg, once a day for 28 days. In the epilepsy and control groups, an equivalent volume of normal saline was intragastrically administered. MAIN OUTCOME MEASURES: Immunoreactivity for IGF-1 and NF-κB/P65 were detected by immunohistochemical staining. Neuronal apoptosis was detected using TUNEL methods. RESULTS: The hippocampus and cerebral cortex of rats with PTZ-induced epilepsy exhibited a higher number of apoptotic cells at high magnification (×400), compared with the control group. Expression of IGF-1 and NF-κB were higher in the epilepsy group, compared with the control group (P < 0.01). In Ganoderma lucidum spore-treated rats, fewer apoptotic cells were observed in the hippocampus and cerebral cortex, expression of NF-κB/P65 was lower, and immunoreactivity to IGF-1 increased more distinctly, compared with the epilepsy group. In addition, seizure latency was longer on 17, 21, and 25 days post-PTZ treatment in the Ganoderma lucidum spore powder group, compared with the epilepsy group (P < 0.05-0.01). CONCLUSION: Ganoderma lucidum spore powder down-regulated expression of NF-κB in brain tissues of rats with PTZ-induced epilepsy, increased immunoreactivity to IGF-1, and inhibited neuronal apoptosis. 4Influence of Ganoderma lucidum spores on the levels of neuropeptide Y and somatostatin in brains of seizure rats显示文摘BACKGROUND: Previous research has revealed that somatostatin can induce epilepsy, and that the levels of neuropeptide Y may increase and become more active in brain areas with epileptic seizures. OBJECTIVE: To observe the effect of Ganoderma lucidum spore powder on the neuropeptide Y and somatostatin content in the cerebral cortex and hippocampal regions of seizure rats induced by pentylenetetrazol (PTZ). Furthermore, to verify any effect of Ganoderma lucidum spore powder on inhibition of epileptic seizures. DESIGN, TIME AND SETTING: A randomized group animal study was performed in August 2007 in the School of Basic Medical Sciences, Jiamusi University (Jiamusi, Heilongjiang, China). MATERIALS: Thirty healthy, male, Wistar rats, aged 12 weeks and weighing 180–220 g, were taken as the experimental animals. PTZ (Sigma Company, United States) was used to induce epilepsy. Ganoderma lucidum spores (Leyss, ex Fr variety) were purchased from Jiamusi City Wild Growing Case of the Ganoderma Lucidum (China). Rabbit anti-somatostatin antibodies and secondary antibodies were purchased from Wuhan Boster Company (China). Neuropeptide Y radioimmunoassay kit was purchased from Beijing Furui Biotechnology Company (China). METHODS: Thirty rats were randomly divided into three groups: a control group, an epilepsy model group and a Ganoderma lucidum spore-treated group. Each group contained 10 animals. Rats in the epilepsy model group were treated with intraperitoneal injections of PTZ and gastric perfusion of physiologic saline. In the Ganoderma lucidum spore-treated group, intraperitoneal injection of PTZ and gastric perfusion of Ganoderma lucidum spore powder were administered. The blank control group was only administered with the physiological saline by intraperitoneal injection and gastric perfusion. MAIN OUTCOME MEASURES: Immunohistochemical staining and radioimmunoassay methods were used to observe the changes of somatostatin and neuropeptide Y content in brain tissue of epileptic rats, as well as the morphology of neurons. RESULTS: All 30 rats were involved in the result analysis, without any loss. The number of somatostatin immunoreacted positive cells in the cerebral cortex and hippocampus was significantly increased in the epilepsy model group compared to the blank control group (P < 0.01). The number of somatostatin immunoreacted positive cells in cerebral cortex and hippocampus was significantly decreased in the Ganoderma lucidum spore-treated group compared to the epilepsy model group (P < 0.01). The contents of neuropeptide Y in cerebral cortex and hippocampus were significantly increased in the epilepsy model group compared to the blank control group (P < 0.01). The contents of neuropeptide Y in the cerebral cortex and hippocampus were significantly decreased in the Ganoderma lucidum spore-treated group compared to the epilepsy model group (P < 0.05). The epilepsy seizures in the Ganoderma lucidum spore-treated group were obviously reduced compared to the epilepsy model group. 5Muscarinic acetylcholine receptor is involved in acetylcholine regulating stomatal movement显示文摘In animal cells, action of acetylcholine depends on its binding with its two specific receptors on the plasma membrane: the nicotinic and muscarinic respectively. The present investigation has shown that agonists of muscarinic receptor (muscarine) could induce stomatal opening, while the antagonists (atropine) could block stomatal opening induced by acetylcholine. Their effects can only be realized in medium containing Ca2+, but not in medium containing K+. 6Association between hsa-miR-34b/c rs4938723 T>C promoter polymorphism and cancer risk: a meta-analysis based on 6,036 cases and 6,204 controls显示文摘Objective: Emerging evidence shows that microRNAs(miRNAs) function as tumor suppressors or oncogenes in human carcinogenesis. A single nucleotide polymorphism(SNP) located in the pri-miRNA promoter may affect the processing and expression of mature miRNA. However, previous studies showed conflicting results regarding the association of hsa-miR-34b/c rs4938723 T > C promoter polymorphism with cancer. Therefore, we conducted a meta-analysis to determine the association of polymorphism with cancer risk.Methods: A computerized search of PubMed, Web of Science, and Chinese National Knowledge Infrastructure(CNKI) for publications on hsa-miR-34b/c rs4938723 T > C promoter polymorphism and cancer risk was performed and the genotype data were analyzed in a meta-analysis. Odds ratios(ORs) with 95% confidence intervals(CIs) were estimated to assess the association. Test of heterogeneity, cumulative meta-analysis, sensitivity analysis and assessment of bias were performed in our meta-analysis by STATA software 12.0.Results: There was no significant association between hsa-miR-34b/c rs4938723 polymorphism and overall cancer risk in the comparison models. Moreover, subgroup analysis revealed that the variant CT(OR =1.19, 95% CI: 1.03-1.37) and CC/CT(OR =1.18, 95% CI: 1.03-2.35) genotypes were associated with an increased risk of hepatocellular carcinoma(HCC) compared with wild-type TT genotype. However, a decreased risk of colorectal cancer(CRC) was found in the genetic model of CC/TT(OR =0.66, 95% CI: 0.47-0.92) and CC/CTTT(OR =0.67, 95% CI: 0.49-0.93). Conclusions: The results suggest that hsa-miR-34b/c rs4938723 polymorphism may play an opposite role in different types of cancer based on current studies, which is the main origin of heterogeneity in this metaanalysis. 8Ganoderma lucidum spore powder modulates Bcl-2 and Bax expression in the hippocampus and cerebral cortex,and improves learning and memory in pentylenetetrazole-kindled rats显示文摘We studied the effects of Ganoderma lucidum spore powder on Bax and Bcl-2 expression and neuronal apoptosis in pentylenetetrazole-kindled epileptic rats.Sixty adult rats were randomly divided into a control group,an epileptic group (kindled) and three medication groups (150,300,450 mg/kg given to kindled rats).Bax and Bcl-2 immunohistochemistry and TUNEL labeling showed that the number of Bax-and TUNEL-positive cells in the hippocampus and cerebral cortex decreased significantly in the high-dose medication group,while the number of Bcl-2 immunoreactive cells increased.The Morris water maze test showed that high-dose treatment significantly shortened escape latency and increased spatial probe trial performance.Our findings indicate that a high dose of Ganoderma lucidum spore powder upregulates the expression of antiapoptotic Bcl-2 protein in the hippocampus and cerebral cortex,inhibits proapoptotic Bax expression,and decreases seizure-induced neuronal apoptosis.Further,Ganoderma lucidum appears to protect against epilepsy-related learning and memory impairments.Shuang Zhao Shengchang Zhang Shuqiu Wang 2011Neural Regeneration Research2011,6,30:2
9Effects of Ganoderma lucidum spore powder on astrocyte expression and glutamine synthetase activity in the hippocampal region of epileptic rats显示文摘BACKGROUND: Recent studies have demonstrated that astrocyte dysfunction plays a central role in inhibiting epileptic seizures and that regulation of astrocyte function may be a new target for treatment of epilepsy. OBJECTIVE: To observe the effects of Ganoderma lucidum spore powder (GLSP) on astrocyte morphology and glutamine synthetase (GS) activity in the hippocampal region of epileptic rats. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Function Laboratory, College of Basic Medicine, Jiamusi University between October and December 2006. MATERIALS: A total of 30 Sprague Dawley (SD) rats were randomized to three groups (n = 10): control, model, and GLSP. GLSP was sourced from Jiamusi Wild Ganoderma Lucidum Planting Base and prepared to 30 g/L with physiological saline before use. Pentylenetetrazol (PTZ) (10 g/L) was provided by Sigma Company, USA. METHODS: The control group received intraperitoneal (i.p.) and intragastric (i.g.) physiological saline. Following epilepsy induction by i.p. administration of PTZ (35 mg/kg), rats from the model and GLSP groups were ig injected with physiological saline and GLSP (300 mg/kg), respectively. Each compound was administered once per day, for a total of 28 successive days. Epileptic seizure convulsions were graded 0–5. A higher grade indicated more severe epilepsy. Only those rats showing stage 2 or higher convulsions at least 5 times successively were included in further experiments. MAIN OUTCOME MEASURES: Immediately after injection, seizure activity was monitored for 30 minutes to determine the latent period and seizure duration; simultaneously, astrocyte numbers and GS activity in the hippocampal region of rats with epilepsy were detected by immunohistochemistry. RESULTS: All 30 rats were included in the final analysis. On day 28, following PTZ administration epileptic seizures were not found in the control group. In the GLSP group, rats exhibited rhythmic head nodding or facial spasms, and the latent period was significantly longer than that of the model group (P < 0.05). In the model group, the majority of rats exhibited myoclonus or generalized convulsions, and epileptic seizure duration was slightly (but not significantly) longer than that in the GLSP group (P > 0.05). Within 2–3 minutes of PTZ injection, the model group exhibited grade 4 or 5 epileptic seizures, and the GLSP group mostly showed grade 2 or 3, and occasionally grade 4 or 5, epileptic seizures. All rats recovered within 30 minutes. The model group exhibited significantly increased astrocytes (P < 0.05), with thicker cellular processes and a higher number of cellular processes, and significantly decreased GS activity (P < 0.05) than the control group. The astrocyte count was significantly decreased but GS activity was significantly increased in the GLSP group when compared with the model group (P < 0.05); however, astrocyte appearance was similar in both groups (P < 0.05). CONCLUSION: GLSP can effectively inhibit astrocyte numbers and elevate GS activity in the hippocampal region of rats with epilepsy, thereby reducing epileptic seizures.Shiling Zhang Shuqiu Wang 2008Neural Regeneration Research2008,3,12:2
10Effect of Fanbaicao(Herba Potentillae Discoloris) oil on the expression of p21 and CDK4 in HepG2 cells显示文摘OBJECTIVE:To research the anti-cancer mechanism of the Traditional Chinese Medicine Fanbaicao(Herba Potentillae Discoloris) oil in the human hepatoma cell line Hep G2.METHODS:Gas chromatography was used to analyze the components of Fanbaicao(Herba Potentillae Discoloris).We tested the inhibitory effect of Fanbaicao(Herba Potentillae Discoloris) oil on the human hepatoma cell line Hep G2 in vitro using 3-(4,5-Dimet hylt hiazol-2-yl)-2,5-dip henyltetrazoliumbromide assays.Fluorescence activating cell sorter analysis was used to examine the levels of apoptosis,and western blot and immunofluorescence were used to detect the expression of p21,p-p21 and CDK4 proteins.RESULTS:Fanbaicao(Herba Potentillae Discoloris)oil contains 45 ingredients,and L-ascorbic acid 2,6-bispalmitate was the main component and accounted for 44.96% of total drive-off peak area.Other components included(Z)-14-met hyl-8-exadecenal-acetal(8.56%),phytol(7.74%) and lauric acid(6.31%).Fanbaicao(Herba Potentillae Discoloris)oil treatment reduced the proliferation of Hep G2 cells and the half growth inhibition concentration(IC50) was 2.03 mg/m L.Furthermore,we also observed significantly increased Hep G2 cell apoptosis in a dose-dependent manner(P < 0.05).Fanbaicao(Herba Potentillae Discoloris) oil significantly increased the expression of p21 and p-p21 and significantly decreased the expression of CDK4 in Hep G2 cells compared with controls(P < 0.01).CONCLUSION:Our results showed that Fanbaicao(Herba Potentillae Discoloris) oil has anti-cancer activities in Hep G2 cells,which is probably related to the upregulation of p21 and p-p21 and downregulation of CDK4 expression.Liu Lei Chen Guang Wang Baixin Chen Liping Wang Shuqiu Liu Zhixin Ma Xiaoru Wang Fangfang Liang Yanfeng Wu Jiamei Yang Zhiwei 2016Journal of Traditional Chinese Medicine2016,36,4:1
11Changes of biochemical indices in brain, liver tissue and serum in mice with Alzheimer disease after Chinese medicine treatment显示文摘BACKGROUND: Alzheimer disease is a main type of dementia, and the important clinical characteristic is the rapid declines of memory and cognitive ability. OBJECTIVE: To study changes of biochemical indices in brain, liver tissue and serum, as well as memory of mice with Alzheimer disease after Chinese medicine treatment. DESIGN: A comparative animal experimental observation. SETTING: Haierfu Research Center of Youjiang Medical College for Nationalities. MATERIALS: Forty-eight healthy Kunming mice (24 males and 24 females), 3 months old, were provided by the animal room of Youjiang Medical College for Nationalities. The animals were divided into four groups according to sex and body mass: control group, model group, Wuyuan Buxue treated group, Haierfu treated group, and 12 mice in each group. Wuyuan Buxue oral liquid was extracted from Polygonum multiflorum Thunb (red, radix) and longan meat (country medicine quasi- word B20020828). Haierfu oral liquid was extracted from Yinhua, poriacocos, licorice, etc (Q/452600RYYLC01-92). METHODS: The experiment was completed in Haierfu Research Center and Institute of Heavy Metal and Fluorosis-Arsenism of Youjiang Medical College for Nationalities from May 2006 to December 2006. ① All animals except those in the control group were given feed which was mixed with AlCl3 (12 g/L), and they could freely drink 3 g/L Al(NO3)3. The mice in the control group were given normal feed. Wuyuan Buxue oral liquid and Haierfu oral liquid were distilled by distilled water for one time respectively. Five months after model establishment, mice in the Wuyuan Buxue treated group and Haierfu treated group were given intrapastric perfusion of Wuyuan Buxue oral liquid and Haierfu oral liquid respectively, and those in the model group and control group were given intrapastric perfusion of distilled water of the same volume. All the mice were treated for 45 days. ② The swimming time (s) and error times were determined with Y-shape water maze before and after experiment; Hemoglobin was determined by method of HICN. After treatment, serum was isolated from eyeballs to determine the activities of glutathione peroxidase (GSH-PX), urea and cholesterol. The animals were executed to remove brain and liver, then 10% homogenate was prepared to determine the activity of acetylcholinesterase (AChE), clearance of O2–·, content of malondialdehyde (MDA), activities of superoxide dismutase (SOD) and GSH-PX. MAIN OUTCOME MEASURES: ① Swimming time and error times in the water maze, and the content of hemoglobin; ② alanine aminotransferase (ALT) activity and contents of urea and cholesterol in serum after treatment; ③ Activities of GSH-PX, AChE and SOD, and MDA content in liver, and AChE activity in brain after treatment. RESULTS: Totally 48 mice were used, and some of them died due to unknown reasons, finally 32 mice (8 in each group) were involved in the analysis of results. ① Content of hemoglobin before and after treatment: There were obvious changes in the groups (P < 0.05). ② Comparison of ALT, cholesterol and urea in serum: The contents of urea in the Wuyuan Buxue treated group and Haierfu treated group were obviously higher than those in the control group and model group (P < 0.05, 0.01); ALT and cholesterol had no significant differences among the groups (P > 0.05). ③ Comparisons of the activities of AChE, GSH-PX and SOD in brain homogenate: The activity of AChE was significantly higher in the control group than the model group and Haierfu treated group, also higher in the Wuyuan Buxue treated group than in the model group (P < 0.05). The activity of GSH-PX in the model group was significantly higher than those in the treated groups (P < 0.01). The activity of SOD in the control group was significantly higher than those in the treated groups (P < 0.05, P < 0.01). ④ Comparison of activities of GSH-PX and SOD, and MDA content in liver homogenate: The activities of GSH-PX and SOD in the model group were obviously lower than those in the Wuyuan Buxue treated group (P < 0.05). MDA content had no obvious differencesamong the groups (P > 0.05). ⑤ Comparison of MDA content, and clearance of O2–· in brain and liver: The MDA content in brain in the model group was significantly higher than those in the treated groups; The clearances of O2–· in liver in the treated groups were significantly higher than those in the control group and model group (P < 0.01). ⑥ Swimming time and error rate in water maze: Before treatment, the swimming speed had no obvious difference among the groups, while the error rate was obviously different. After treatment, there was no obvious difference in the control group; The swimming time was prolonged as compared with that before treatment in the other three groups, and there were no obvious differences among the three group; The error rate was the highest in the model group (25%), and the lowest in the Wuyuan Buxue treated group (8.3%). 12Effects of nanometer particles and water decoction of the Chinese medicine mixture of pinellia ternate and scorpion on P53 protein contents and apoptosis in the cerebral cortex and hippocampus of epileptic rats显示文摘BACKGROUND: Water decoction of the Chinese traditional medicine mixture of pinellia ternate and scorpion is an effective treatment for epilepsy. OBJECTIVE: To compare nanometer particles and effects of water decoction of Chinese traditional medicine mixture on P53 protein levels and apoptosis in the cerebral cortex and hippocampus of epileptic rats. DESIGN, TIME AND SETTING: This randomized, controlled molecular biology study was performed at the Key Laboratory of Child Neural Rehabilitation of Jiamusi University from October to December 2007. MATERIALS: Forty healthy male Wistar rats were used in this study. Convulsion rat models were established by intraperitoneal infusion of 35 mg/kg pentylenetetrazol, once a day, for 28 successive days. The Chinese traditional medicine mixture, comprising pinellia ternate, scorpion, centipede, bupleurum, peach pit and glycyrrhiza, was purchased from Beijing Tongrentang, China. The mixture was made into nanometer particles and water decoction. The apoptosis determination kit and P53 immunohistochemistry kit were bought from Boster, China. METHODS: Forty Wistar rats were randomly divided into four groups of ten rats per group, control, nanometer particle, water decoction and epileptic model groups. Rats in the nanometer particle and water decoction groups were respectively treated with 300 mg/kg Chinese herb nanometer particle suspension and water decoction by gavage, once a day, for 28 days. Rats in the epileptic model group were fed an equal volume of saline by gavage. Rats in the control group were only administered with the same volume of saline by ga- vage. MAIN OUTCOME MEASURES: Terminal deoxynucleotidyl Transferase Biotin-dUTP Nick End Labeling (TUNEL) and immunohistochemistry were used to respectively detect neuronal apoptosis and P53 protein expression in the rat brain cortex and hippocampus at 28 days following administration. RESULTS: The number of apoptotic neurons was lower in the nanometer particle and water decoction groups compared with the epileptic model group. The number of apoptotic cells and P53 protein expression in the rat cerebral cortex and hippocampus was greater in the epileptic model group compared with the control group (P < 0.05), but lower in the nanometer particle and water decoction groups compared with the epileptic model group (P < 0.05), and lower in the nanometer particle group compared with the water decoction group (P < 0.05). CONCLUSION: Chinese traditional medicine mixture decreases neuronal apoptosis and P53 protein expression in epileptic rats.
