1Folate receptor-mediated boron-10 containing carbon nanoparticles as potential delivery vehicles for boron neutron capture therapy of nonfunctional pituitary adenomas显示文摘Invasive nonfunctional pituitary adenomas(NFPAs) are difficult to completely resect and often develop tumor recurrence after initial surgery.Currently,no medications are clinically effective in the control of NFPA.Although radiation therapy and radiosurgery are useful to prevent tumor regrowth,they are frequently withheld because of severe complications.Boron neutron capture therapy(BNCT) is a binary radiotherapy that selectively and maximally damages tumor cells without harming the surrounding normal tissue.Folate receptor(FR)-targeted boron-10 containing carbon nanoparticles is a novel boron delivery agent that can be selectively taken up by FR-expressing cells via FR-mediated endocytosis.In this study,FR-targeted boron-10 containing carbon nanoparticles were selectively taken up by NFPAs cells expressing FR but not other types of non-FR expressing pituitary adenomas.After incubation with boron-10 containing carbon nanoparticles and following irradiation with thermal neutrons,the cell viability of NFPAs was significantly decreased,while apoptotic cells were simultaneously increased.However,cells administered the same dose of FR-targeted boron-10 containing carbon nanoparticles without neutron irradiation or received the same neutron irradiation alone did not show significant decrease in cell viability or increase in apoptotic cells.The expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated in NFPAs after treatment with FR-mediated BNCT.In conclusion,FR-targeted boron-10 containing carbon nanoparticles may be an ideal delivery system of boron to NFPAs cells for BNCT.Furthermore,our study also provides a novel insight into therapeutic strategies for invasive NFPA refractory to conventional therapy,while exploring these new applications of BNCT for tumors,especially benign tumors.DAI CongXin CAI Feng HWANG Kuo Chu ZHOU YongMao ZHANG ZiZhu LIU XiaoHai MA SiHai YANG YaKun YAO Yong FENG Ming BAO XinJie LI GuiLin WEI JunJi JIAO YongHui WEI ZhenQing MA WenBin WANG RenZhi 2013Science China(Life Sciences)2013,56,2:8
2The RNA-binding protein Musashi2 governs osteoblast-adipocyte lineage commitment by suppressing PPARγsignaling显示文摘Osteoporosis caused by aging is characterized by reduced bone mass and accumulated adipocytes in the bone marrow cavity. How the balance between osteoblastogenesis and adipogenesis from bone marrow mesenchymal stem cells(BMSCs) is lost upon aging is still unclear. Here, we found that the RNA-binding protein Musashi2(Msi2) regulates BMSC lineage commitment. Msi2 is commonly enriched in stem cells and tumor cells. We found that its expression was downregulated during adipogenic differentiation and upregulated during osteogenic differentiation of BMSCs. Msi2 knockout mice exhibited decreased bone mass with substantial accumulation of marrow adipocytes, similar to aging-induced osteoporosis. Depletion of Msi2 in BMSCs led to increased adipocyte commitment. Transcriptional profiling analysis revealed that Msi2 deficiency led to increased PPARγ signaling.RNA-interacting protein immunoprecipitation assays demonstrated that Msi2 could inhibit the translation of the key adipogenic factor Cebpα, thereby inhibiting PPAR signaling. Furthermore, the expression of Msi2 decreased significantly during the aging process of mice, indicating that decreased Msi2 function during aging contributes to abnormal accumulation of adipocytes in bone marrow and osteoporosis. 4Cooperative wireless networks for better green services显示文摘In order to guarantee the overall return on investment (ROI), improve user experience and quality of service (QoS), save energy, reduce electra magnetic interference (EMI) and radiation pollution, and enable the sustainable deployment of new profitable applications and services in heterogeneous wireless networks coexistence reality, this paper proposes a cross-network cooperation mechanism to effectively share network resources and infrastructures, and then adaptively control and match multi-network energy distribution characteristics according to actual user/service requirements in different geographic areas. Some idle or lightly-loaded base stations (BS or BSs) will be temporally turned off for saving energy and reducing EMI. 8Oleic acid reduces steroidogenesis by changing the lipid type stored in lipid droplets of ovarian granulosa cells显示文摘Background:Oleic acid is an abundant free fatty acid present in livestock that are in a negative energy-balance state,and it may have detrimental effects on female reproduction and fertility.Oleic acid induces lipid accumulation in bovine granulosa cells,which leads to a foam cell-like morphology and reduced steroidogenesis.However,why oleic acid increases lipid accumulation but decreases steroidogenesis remains unclear.This study focused on oleic acid’s effects on lipid type and steroidogenesis.Results:Oleic acid increased the lipid accumulation in a concentration-dependent manner and mainly increased the triglyceride level and decreased the cholesterol ester level.Oleic acid also led to a decline in estradiol and progesterone production in porcine granulosa cells in vitro.In addition,oleic acid up-regulated the expression of CD36 and diacylglycerol acyltransferase 2,but down-regulated the expression of 3-hydroxy-3-methylglutarylcoenzyme A reductase,scavenger receptor class B member 1 and acetyl-Coenzyme A acetyltransferase 2,as well as steroidogenesis-related genes,including cytochrome P450 family 11 subfamily A member 1,cytochrome P450family 19 subfamily A member 1 and 3 as well as steroidogenic acute regulatory protein at the mRNA and protein levels.An oleic acid-rich diet also enhanced the triglyceride levels and reduced the cholesterol levels in ovarian tissues of female mice,which resulted in lower estradiol levels than in control-fed mice.Compared with the control,decreases in estrus days and the numbers of antral follicles and corpora lutea,as well as an increase in the numbers of the atretic follicles,were found in the oleic acid-fed female mice.Conclusions:Oleic acid changed the lipid type stored in lipid droplets of ovarian granulosa cells,and led to a decrease in steroidogenesis.These results improve our understanding of fertility decline in livestock that are in a negative energy-balance state.Xiaoge Zhou Zhaoyi Mo Yankun Li Liang Huang Sihai Yu Lan Ge Yamei Hu Shengjie Shi Lutong Zhang Liguang Wang Lei Gao Gongshe Yang Guiyan Chu 2022Journal of Animal Science and Biotechnology2022,13,3:1

11Triton X-100辅助合成多级孔道结构ZSM-5沸石催化剂(英文)显示文摘ZSM-5 zeolite with a hexagonal cubic morphology was synthesized by a hydrothermal method using Triton X-100, a nonionic surfactant. The samples prepared with and without the surfactant were characterized by X-ray diffraction(XRD), Fourier transform infrared spectroscopy, N2 adsorption, high resolution transmission electron microscopy(TEM), high resolution scanning electron mi-croscopy, energy dispersive X-ray analysis, and NH3 temperature-programmed desorption. The XRD patterns confirmed the formation of a pure ZSM-5 crystalline phase without secondary phases. TEM images revealed that the hexagonal cubes were made of peanut-shaped nanoparticles with voids.The catalytic activity of the zeolite samples was evaluated using the selective oxidation of benzyl alcohol with tertiary-butyl hydrogen peroxide as the oxidant at 90 °C. The surfactant-assisted prep-aration yielded a zeolite that gave a higher conversion than the one prepared in the absence of the surfactant. 14Development of a colorimetric assay for rapid quantitative measurement of clavulanic acid in microbial samples显示文摘We developed a colorimetric assay to quantify clavulanic acid (CA) in culture broth of Streptomyces clavuligerus, to facilitate screening of a large number of S. clavuligerus mutants. The assay is based on a β-lactamase-catalyzed reaction, in which the yellow substrate nitrocefin (λmax =390 nm) is converted to a red product (λmax =486 nm). Since CA can irreversibly inhibit β-lactamase activity, the level of CA in a sample can be measured as a function of the A 390 /A 486 ratio in the assay mixture. The sensitivity and detection window of the assay were determined to be 50 g L-1 and 50 g L-1 to 10 mg L-1 , respectively. The reliability of the assay was confirmed by comparing assay results with those obtained by HPLC. The assay was used to screen a pool of 65 S. clavuligerus mutants and was reliable for identifying CA over-producing mutants. Therefore, the assay saves time and labor in large-scale mutant screening and evaluation tasks. The detection window and the reliability of this assay are markedly better than those of previously reported CA assays. 15Dynamic-static coupled sensing of trace biomarkers by molecularly imprinted metal-organic frameworks显示文摘Accommodating target analytes within the pores of metal-organic frameworks(MOFs)to improve the sensing performance is an important but challenging task.Here,we report a novel molecular imprinting strategy to create target recognition sites in a tailored multicomponent MOF with the inter-ligand synergistic antenna effect to lanthanide ions,enabling selective recognition of trace biomarkers,which is critical to the early diagnosis of age-related diseases in blood samples with high sensitivity and ultralow limit of detection(LOD)of 69 nmol L^(-1).Compared with MOF-based sensors without imprinted recognition sites,the significantly enhanced sensing performance(both sensitivity and LOD)was attributed to a dynamic-static coupled sensing mechanism:the dynamic interactions involve concentrating the trace biomarkers at the imprinted recognition sites to enhance the sensing performance at ultralow concentration,and the static interactions are derived from electron/energy exchange between the molecularly imprinted MOF and the biomarker to govern the sensing performance.This work establishes a new molecular imprinting strategy to attain advanced materials for sensing trace bio-analytes.Hui Min Zongsu Han Tiankai Sun Kunyu Wang Jun Xu Peiyu Yao Sihai Yang Peng Cheng Wei Shi 2023Science China Chemistry2023,66,12:0

17Observation of oxygen evolution over a {Ni12}-cluster-based metal-organic framework显示文摘The development of efficient electrocatalysts based on non-noble metals for oxygen evolution reaction(OER) remains an important and challenging task. Multinuclear transition-metal clusters with high structural stability are promising OER catalysts but their catalytic role is poorly understood. Here we report the crystallographic observation of OER activity over robust {Ni12}-clusters immobilised in a porous metal-organic framework, NKU-100, by single-crystal X-ray diffraction as a function of external applied potential. We observed the aggregation of confined oxygen species around the {Ni12}-cluster as a function of applied potential during the electrocatalytic process. The refined occupancy of these oxygen species shows a strong correlation with the variation of current density. 20Tolerance and dependence of edomorphin-1 in rats and possible mechanisms显示文摘Objective: To observe the tolerance and the dependence of endomorphin-1 ( EM-1) in rats and the possible mechanisms.Methods: Sixty Sprague-Dawley rats were randomly allocated into saline, acute EM-1-treated and chronic EM-1-treated groups.The rats were intracerebroventricularly injected with saline, acute EM-1 10 !g/kg 30 min prior to sacrifice, and chronic EM-1 bydaily administration at 8: 00 A.M. and 15: 00 P.M. from 10 !g/kg on the 1st day to 50 !g/kg on the 9th day, respectively. Inchronic EM-1-treated group, the median antinociceptive dose (AD50) and the catatonic median effective dose (ED50) were deter-mined by the improved Dixon's method. Natural withdrawl test was used to assess the dependence of EM-1. Maximal binding ca-pacity(Bmax) and dissociation constant(Kd) of 3H-DAMGO, binding to mu-opioid receptor(MOR) in brain tissue, was mea-sured by Scatchard analysis. Gene expression of MOR was measured by reverse transcription-polymerase chain reaction(RT-PCR).Results: Tolerance of the antinociceptic and catatonic effects on the 3rd day(3.1-fold and 1.9-fold) and the 9th day(28.4-fold and8.5-fold) were observed in chronic EM-1-treated group (P < 0.05). Jumping times and withdrawal scores of rats were significantlyhigher in the chronic EM-1-treated group than those in saline group on the 9th day (P < 0.05). Bmax and mRNA expression ofMOR in cortex, midbrain and striatum were lower in chronic EM-1-treated group on the 9th day than the other two groups(P < 0.05),but Kd had no significant difference (P > 0.05). AD50, ED50, Bmax, Kd and gene expression of MOR were recorded. Conclusion:EM-1 possesses the tolerance and the dependence.
