1CD38 deficiency alleviates Ang II-induced vascular remodeling by inhibiting small extracellular vesicle-mediated vascular smooth muscle cell senescence in mice显示文摘CD38 is the main enzyme for nicotinamide adenine dinucleotide(NAD)degradation in mammalian cells.Decreased NAD levels are closely related to metabolic syndromes and aging-related diseases.Our study showed that CD38 deficiency significantly alleviated angiotensin II(Ang ll)-induced vascular remodeling in mice,as shown by decreased blood pressures;reduced vascular media thickness,media-to-lumen ratio,and collagen deposition;and restored elastin expression.However,our bone marrow transplantation assay showed that CD38 deficiency in lymphocytes led to lack of protection against Ang ll-induced vascular remodeling,suggesting that the effects of CD38 on Ang ll-induced vascular remodeling might rely primarily on vascular smooth muscle cells(VSMCs),not lymphocytes.In addition,we observed that CD38 deficiency or NAD supplementation remarkably mitigated Ang ll-induced vascular senescence by suppressing the biogenesis,secretion,and internalization of senescence-associated small extracellular vesicles(SA-sEVs),which facilitated the senescence of neighboring non-damaged VSMCs.Furthermore,we found that the protective effects of CD38 deficiency on VSMC senescence were related to restoration of lysosome dysfunction,particularly with respect to the maintenance of sirtuin-mediated mitochondrial homeostasis and activation of the mitochondria-lysosomal axis in VSMCs.In conclusion,our findings demonstrated that CD38 and its associated intracellular NAD decline are critical for Ang ll-induced VSMC senescence and vascular remodeling.Lu Gan Demin Liu Jing Liu Erya Chen Chan Chen Lian Liu Hang Hu Xiaohui Guan Wen Ma Yanzi Zhang Yarong He Bofu Liu Songling Tang Wei Jiang Jianxin Xue Hongbo Xin 2021Signal Transduction and Targeted Therapy2021,6,7:2
2RNF152, a novel lysosome localized E3 ligase with pro-apoptotic activities显示文摘RING finger protein 152(RNF152)is a novel RING finger protein and has not been well characterized.We report here that RNF152 is a canonical RING finger protein and has E3 ligase activity.It is polyubiqitinated partly through Lys-48-linked ubiquitin chains in vivo and this phenomenon is dependent on its RING finger domain and transmembrane domain.RNF152 is localized in lysosomes and co-localized with LAMP3,a lysosome marker.Moreover,over-expression of RNF152 in Hela cells induces apoptosis.These results suggest that RNF152 is a lysosome localized E3 ligase with pro-apoptotic activities.It is the first E3 ligase identified so far that is involved in lysosome-related apoptosis.Songling Zhang Wei Wu Yanfang Wu Jiyan Zheng Talin Suo Hong Tang Jie Tang 2010Protein & Cell2010,1,7:1

4Study on the Phenotypic Mutation Generated from ^(60)Coγ-ray Irradiated Oxlias triangularis Purpurea Regeneration System显示文摘Mutations induced from tissue culture are easily to be separated,which might be propagated in the same medium,especially for the color-leaf ornamental grass,Oxalis triaggularis purpurea.Mutations of the ornamental traits in the tissue culture bottle could be investigated easily.The 35 d regeneration system group showed the lowest adventitious bud number and adventitious root number among 4 inoculation dates by 50 Gy dose of ^(60)Coγ radiation.More studies were carried out based on the 35d differentiation state of O.triangularis purpurea regeneration system.The 35 d regeneration system was then irradiated by 10,25 and 50 Gy doses of ^(60)Coγ rays.Numbers of adventitious buds and roots induced from the regeneration system were cut down with the increment of radiation doses.Seedling length was not distinctly reduced at the absorbed doses of 10 and 25 Gy,but reduced distinctly under 50 Gy of ^(60)Coγ irradiation.The optimal irradiation dose for 35 d O.triangularis regeneration system survival and mutation induction was approximately 25 Gy.The M_2 phenotypic mutation rate was 2.9%,especially,and the leaf number mutation accounted for 76%of the total mutation.The phenotypic mutations,especially in the 10 Gy group,on 0.1 m M Vc containing MS medium were recovered,which indicated that ROS plays a key role in the phenotypic mutation induced by ^(60)Coγ -rays.Jie REN Qi ZHU Yanqiong MENG Fei TANG Yun XUAN Dan SHI Jingjing WANG Yongjin DENG Songling FU 2017Agricultural Science & Technology2017,18,5:0
