1The current agonists and positive allosteric modulators ofα7 nAChR for CNS indications in clinical trials显示文摘The alpha-7 nicotinic acetylcholine receptor(α7 nAChR), consisting of homomeric α7 subunits, is a ligand-gated Ca^(2+)-permeable ion channel implicated in cognition and neuropsychiatric disorders. Enhancement of α7 nAChR function is considered to be a potential therapeutic strategy aiming at ameliorating cognitive deficits of neuropsychiatric disorders such as Alzheimer's disease(AD) and schizophrenia. Currently, a number of α7 nAChR modulators have been reported and several of them have advanced into clinical trials. 2A wireless, implantable optoelectrochemical probe for optogenetic stimulation and dopamine detection显示文摘Physical and chemical technologies have been continuously progressing advances in neuroscience research.The development of research tools for closed-loop control and monitoring neural activities in behaving animals is highly desirable.In this paper,we introduce a wirelessly operated,miniaturized microprobe system for optical interrogation and neurochemical sensing in the deep brain.Via epitaxial liftoff and transfer printing,microscale light-emitting diodes(micro-LEDs)as light sources and poly(3,4-ethylenedioxythiophene)polystyrene sulfonate(PEDOT:PSS)-coated diamond films as electrochemical sensors are vertically assembled to form implantable optoelectrochemical probes for real-time optogenetic stimulation and dopamine detection capabilities.A customized,lightweight circuit module is employed for untethered,remote signal control,and data acquisition.After the probe is injected into the ventral tegmental area(VTA)of freely behaving mice,in vivo experiments clearly demonstrate the utilities of the multifunctional optoelectrochemical microprobe system for optogenetic interference of place preferences and detection of dopamine release.The presented options for material and device integrations provide a practical route to simultaneous optical control and electrochemical sensing of complex nervous systems.Changbo Liu Yu Zhao Xue Cai Yang Xie Taoyi Wang Dali Cheng Lizhu Li Rongfeng Li Yuping Deng He Ding Guoqing Lv Guanlei Zhao Lei Liu Guisheng Zou Meixin Feng Qian Sun Lan Yin Xing Sheng 2020Microsystems & Nanoengineering2020,6,1:3
3Bioaccumulation and human health risk assessment of trace metals in the freshwater mussel Cristaria plicata in Dongting Lake, China显示文摘Trace metal contamination in water and bioaccumulation in aquatic organisms are human health risks of increasing concern. However, the bioaccumulation of trace metals in the organs of the mussel Cristaria plicata in Dongting Lake, China and the human health risks of mussel consumption are largely unknown. We investigated the concentrations of 15 trace metals and metalloids in surface water, sediments, and C. plicata organs(foot, gill, mantle, and visceral mass) and quantified the bioaccumulation and human health risk of these trace metals in specimens collected from Dongting Lake. The concentrations of most metals in surface water exceeded previously published background values. In contrast, the concentrations of most metals in sediments showed a decreasing trend. Overall, the metal concentrations in the gill and visceral masses of C. plicata were higher than those in the foot and mantle, and higher bioaccumulation capacities were observed for essential metals than for nonessential metals. The mean concentrations of the trace elements Zn, Pb, Cd, As, Cu, and Cr in C. plicata foot samples were lower than the threshold values established by international and Chinese organizations. The estimated daily intake(EDI) values of the essential metal Mn in C. plicata foot was higher than the recommended tolerable daily intake(TDI) values for juveniles. Only Mn for juveniles and As for both juveniles and adults may pose noncarcinogenic health risks through foot consumption. 6Core pluripotency factors promote glycolysis of human embryonic stem cells by activating GLUT1 enhancer显示文摘Human embryonic stem cells (hESCs) depend on glycolysis for energy and substrates for biosynthesis. To understand the mechanisms governing the metabolism of hESCs, we investigated the transcriptional regulation of glucose transporter 1 (GLUT1, SLC2A1), a key glycolytic gene to maintain pluripotency. By combining the genome-wide data of bin ding sites of the core pluripotency factors (SOX2, OCT4, NANOG, denoted SON), chromosomal interaction and histone modification in hESCs, we identified a potential enhancer of the GLUT1 gene in hESCs, de noted GLUT1 enhancer (GE) element GE interacts with the promoter of GLUT1, and the deletion of GE significantly reduces the expression of GLUT1, glucose uptake and glycolysis of hESCs, confirming that GE is an enhancer of GLUT1 in hESCs. In addition, the mutation of SON binding motifs within GE reduced the expression of GLUT1 as well as the interaction between GE and GLUT1 promoter, indicating that the binding of SON to GE is important for its activity. Water wave pump is a new type pump, which uses the particular quality of water wave and re-divides the inflow energy to increase the pressure of one part of the inflow water with the rest water flowing away freely. The research and development of such a pump is of importance and significant value and profitable social interest in that it can fully utilize the residual energy of natural source in industrial and civil water circle systems. 8Deep Reinforcement Learning Based Charging Scheduling for Household Electric Vehicles in Active Distribution Network显示文摘With the booming of electric vehicles(EVs) across the world, their increasing charging demands pose challenges to urban distribution networks. Particularly, due to the further implementation of time-of-use prices, the charging behaviors of household EVs are concentrated on low-cost periods, thus generating new load peaks and affecting the secure operation of the medium-and low-voltage grids. This problem is particularly acute in many old communities with relatively poor electricity infrastructure. In this paper, a novel two-stage charging scheduling scheme based on deep reinforcement learning is proposed to improve the power quality and achieve optimal charging scheduling of household EVs simultaneously in active distribution network(ADN) during valley period. In the first stage, the optimal charging profiles of charging stations are determined by solving the optimal power flow with the objective of eliminating peak-valley load differences. In the second stage, an intelligent agent based on proximal policy optimization algorithm is developed to dispatch the household EVs sequentially within the low-cost period considering their discrete nature of arrival. Through powerful approximation of neural network, the challenge of imperfect knowledge is tackled effectively during the charging scheduling process. 9Highly Stable Perovskite Solar Cells by Reducing Residual Water-Induced Decomposition of Perovskite显示文摘Residual water-induced decomposition is one of the dominant reasons for the decay of power conversion efficiency (PCE) in perovskite solar cells (Pero-SCs). To solve this problem, we introduce traces amount of sodium hyaluronate (SH) into the perovskite active layer to reduce the remaining water during the preparation of perovskite films. Unlike the traditionally adopted passive protection of perovskite from exterior water by low surface energy coatings, this study provides active control of the interior water by the addition of a water adsorbent into the perovskite films. The encapsulated Pero-SCs with SH retain approximately 70% of their initial PCE in 4000 h, while those without SH retain 32% of their initial PCE in 1000 h under the dark and ambient atmosphere. The unencapsulated Pero-SCs with SH stored in N2 atmosphere maintain over 94% of the initial PCE in 3000 h at room temperature away from light and remain over 88% of the initial PCE in 2000 h even the devices are heated to 70 °C. It has been proven that the improved stability is mainly due to the well-controlled residual water in perovskite films.
