2Effect of N-desulfated heparin on hepatic/renal ischemia reperfusion injury in rats显示文摘AIM: To investigate the effect of N-desulfated heparin onhepatic/renal ischemia and reperfusion injury in rats.METHODS: Using rat models of 60 minutes hepatic or renalischemia followed by 1 h,3 h,6 h and 24 h reperfusion,animalswere randomly divided into following groups,the shamoperated controls,ischemic group receiving only normalsaline,and treated group receiving N-desulfated heparin ata dose of 12 mg/kg at 5 minutes before reperfusion. P-selectin expression was detected in bepatic/renal tissueswith immunohistochemistry methodRESULTS: P-selectin expression, serum ALT, AST, BUN andCr levels were significantly increased during 60 minuteischemia and 1 h, 3 h, 6 h and 24 h reperfusion,while theincrement was significantly inhibited,and hepatic/renalpathology observed by light microscopy was remarkablyimproved by treatment with the N-desulfated heparin.Furthermore,the heparin was found no effects on PT and KPTT.CONCLUSION: P-selectin might mediate neutrophilinfiltration and contribute to hepatic/renal ischemia andreperfusion. 4Symmetric-key cryptosystem with DNA technology显示文摘DNA cryptography is a new field which has emerged with progress in the research of DNA computing. In our study, a symmetric-key cryptosystem was designed by applying a modern DNA biotechnology, microarray, into cryptographic technologies. This is referred to as DNA symmetric-key cryptosystem (DNASC). In DNASC, both encryption and decryption keys are formed by DNA probes, while its ciphertext is embedded in a specially designed DNA chip (microarray). The security of this system is mainly rooted in difficult biology processes and problems, rather than conventional computing technology, thus it is unaffected by changes from the attack of the coming quantum computer. The encryption process is a fabrication of a specially designed DNA chip and the decryption process is the DNA hybridization. In DNASC, billions of DNA probes are hybridized and identified at the same time, thus the decryption process is conducted in a massive, parallel way. 5The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 6Metabolomics of gastric cancer metastasis detected by gas chromatography and mass spectrometry显示文摘AIM:To elucidate the underlying mechanisms of metastasis and to identify the metabolomic markers of gastric cancer metastasis.METHODS:Gastric tumors from metastatic and nonmetastatic groups were used in this study.Metabolites and different metabolic patterns were analyzed by gas chromatography,mass spectrometry and principal components analysis (PCA),respectively.Differentiation performance was validated by the area under the curve (AUC) of receiver operating characteristic curves.RESULTS:Twenty-nine metabolites were differentially expressed in animal models of human gastric cancer.Of the 29 metabolites,20 were up-regulated and 9 were down-regulated in metastasis group compared to non-metastasis group.PCA models from the metabolite profiles could differentiate the metastatic from the nonmetastatic specimens with an AUC value of 1.0.These metabolites were mainly involved in several metabolic pathways,including glycolysis (lactic acid,alaline),serine metabolism (serine,phosphoserine),proline metabolism (proline),glutamic acid metabolism,tricarboxylic acid cycle (succinate,malic acid),nucleotide metabolism (pyrimidine),fatty acid metabolism (docosanoic acid,and octadecanoic acid),and methylation(glycine).The serine and proline metabolisms were highlighted during the progression of metastasis.CONCLUSION:Proline and serine metabolisms play an important role in metastasis.The metabolic profiling of tumor tissue can provide new biomarkers for the treatment of gastric cancer metastasis.Jin-Lian Chen,Jun-Duo Hu,Jing Fan,Jing Hong,Department of Gastroenterology,Shanghai Sixth People’s Hospital,Shanghai Jiao Tong University,Shanghai 200233,China Hui-Qing Tang,Jian-Zhong Gu,Shanghai Laboratory Animal Center,Chinese Academy of Sciences,Shanghai 201615,China 2010World Journal of Gastroenterology2010,16,46:11

8Germline mutation analysis of hPMS2 gene in Chinese families with hereditary nonpolyposis colorectal cancer显示文摘AIM: To study the germline mutation of hPMS2 gene in 26 unrelated Chinese hereditary nonpolyposis colorectal cancer (HNPCC) probands and to fulfill the screening strategy for HNPCC in Chinese. METHODS: Genomic DNA was extracted from the peripheral blood. To avoid the interference of pseudogene in detection of the remaining 11 exons (exon 1-5, 9, 11-15), long-range polymerase chain reaction (PCR) was conducted to amplify the complete coding region of hPMS2 gene firstly. Then 1/8 of the PCR productswere used as template to amplify the individual exon respectively and DNA sequencing was done. Direct DNA sequencing of the conventional PCR products of exon 6, 7, 8 and 10 of hPMS2 gene was performed. The same analysis was made in 130 healthy persons without family histories of HNPCC to further investigate the pathological effects of the detected missense mutation. RESULTS: One HNPCC proband fulf illed Bethesda guidelines and was found to carry the germline mutation of hPMS2 gene, which has not been reported in Chinese HNPCC families. It was a missense mutation at c.1532C>T of exon 11. It was detected in three controls as well with an occurrence rate of 2.3% (3/130). Since it could not be found in the PMS2-single nucleotide polymorphism (SNP) database, this missense mutation is a new SNP unreported up to date. Meanwhile, 260 reported SNPs of hPMS2 gene were detected in the 26 HNPCC probands. The 2nd and 5th exons were probably the hot SNP regions of hPMS2 gene in Chinese HNPCC families involving 53.1% of all reported SNP. CONCLUSION: The germline mutation of hPMS2 gene may be rare in Chinese HNPCC families. 9HBsAg loading on dendritic cells in patients with chronic hepatitis B: expressions of phenotypic molecules显示文摘BACKGROUND: The antigen reducing ability of dentritic cells (DCs), a kind of antigen presenting cells (APCs) initiating immune response, is associated with the specific immune tolerance of chronic hepatitis B(CHB) patients. However, the dysfunction of DCs can be possibly reversed by the stimulation of antigen peptides. In this study, DCs were cultured from peripheral blood monocytes (PBMCs) in patients with CHB in vitro, and the expression of phenotypic molecules on DCs loaded by different concentrations of HBsAg was observed. METHODS: Forty patients with CHB were divided randomly into 4 groups(10 patients in each group). PBMCs were isolated, and DCs were cultured after addition of granulocyte/macrophage colony-stimulating factor (GMCSF) and interleukin 4(IL-4). On the 9th day, DCs of the experimental groups were loaded at HBsAg concentrations of 2.5mg/L, 5mg/L and 10mg/L for 24 hours, whereas those of the control group were not loaded. An electron microscope was used to analyze the morphological changes of the DCs. The expression of phenotypic molecules on DCs in different groups was detected with flow cytometry. RESULTS: A combination of GM-CSF and IL-4 produced DCs from PBMCs in patients with CHB after being cultured for 9 days, whose morphological changes were tested by an electron microscope. The expression of phenotypic molecules on DCs in the control group was as low as CD83 (8.02±3.99)%, CD80(8.77±2.06)%, and MHC-DR (14.05±2.66)%. Loaded by different concentrations of HBsAg, the up-regulation of phenotypic molecules on DCs was found, with CD83(18.35±2.93)%, CD80(42.63±7.15)% and MHC-DR(47.49±6.59)% in 2.5mg/L HBsAg loading group, CD83(17.88±3.12)%, CD80(45.24± 10.93)% and MHC-DR(47.07±8.52)% in 5mg/L HBsAg loading group and CD83(16.74±2.86)%, CD80(44.59±6.99)% and MHC-DR(48.59±7.42)% in 10mg/L HBsAg loading group, respectively. Compared with the control group, the phenotypic molecules in the experimental groups were all different significantly (P<0.01), but among them, there were no differences (P>0.05). CONCLUSIONS: DCs cultured from PBMCs in the patients with CHB under the conditions of GM-CSF and IL-4 present on the typical dendritic morphology but are immature for expressing low phenotypic molecules. 10A New Type of Polymer-Supported Metallocene Catalyst for Ethylene Polymerization显示文摘A new polymer-supported metallocene catalyst has been prepared. The polymer- supported metallocene displayed considerably high activity in ethylene polymerization, the highest being 3.62(107gPE/molZr(h, the molecular weight of the polyethylene produced was Mn = 1.29(105, about 3-4 times those of corresponding homogeneous zirconocenes. 11Short-segment Barrett's esophagus and cardia intestinal metaplasia:A comparative analysis显示文摘AIM:To investigate the endoscopy and histology of short-segment Barrett's esophagus (SSBE) and cardia intestinal metaplasia (CIM),and their correlation with Helicobacter pylori (H. pylori) gastritis and gastroesophageal reflux disease (GERD). METHODS:Biopsy specimens were taken from 32 SSBE patients and 41 CIM patients with normal appearance of the esophagogastric junction. Eight biopsy specimens from the lower esophagus,cardia,and gastric antrum were stained with hematoxylin/eosin,Alcian blue/periodic acid-Schiff,Alcian blue/high iron diamine and Gimenez dye. Results were graded independently by one pathologist. RESULTS:The SSBE patients were younger than the CIM patients (P < 0.01). The incidence of dysplasia and incomplete intestinal metaplasia subtype was higher in SSBE patients than in CIM patients (P < 0.01). H. pylori infection was correlated with antral intestinal metaplasia (P < 0.05),but not with reflux symptomatic,endoscopic,or histological markers of GERD in CIM patients. SSBE was correlated with reflux symptomatic and endoscopic esophagitis (P < 0.01),but not with H. pylori infection and antral intestinal metaplasia. CONCLUSION:Dysplasia risk is significantly greater in SSBE patients than in CIM patients. 12世界家庭医生组织(WONCA)研究论文摘要汇编——居住在“冷点社区”与健康欠佳、享有卫生服务质量欠佳间的关系显示文摘目的患者居住的社区情况与其所享有的卫生服务质量之间的关系尚未完全明确。本研究旨在调查居住在'冷点社区'(经普查证实的低收入、低文化程度以及不享有多种福利的社区)与享有的卫生服务质量间的关系。方法在12个全科诊所中选取152 962例患者进行调查。将患者所住社区(根据普查所得结果)按照全科诊所服务范围进行编码,并根据低收入(收入低于联邦贫困线水平的2倍被认定为低收入)人群比例、低于高中文化人群比例以及社会剥夺指数水平的四分位数进行分类,具有最低收入、低学历人群比例或最高社会剥夺指数的社区被认定为'冷点社区'。采用双变量回归和Logistic回归分析患者所住'冷点社区'情况与患者慢性病患病情况(肥胖、未得到有效控制的糖尿病)、享有的卫生服务情况[肺炎疫苗接种、癌症(结肠癌、宫颈癌、前列腺癌)筛查以及应用阿司匹林防治心血管疾病情况]间的关系。结果各'冷点社区'患者的分布比例不同,最高比例是最低比例的10倍(29.1%比2.6%)。除了应用阿司匹林之外,享受其他卫生服务情况以及慢性病患病情况均受到了'冷点社区'状况的影响。居住在低学历'冷点社区'的患者患有未得到有效控制的糖尿病的比例高于居住在非'冷点社区'的患者(15%比13%,P<0.05)。回归分析表明,与居住在非'冷点社区'的患者相比,居住在贫困'冷点社区'、低学历'冷点社区'、社会剥夺指数高'冷点社区'的患者享有的宫颈癌筛查服务比例稍高,但享有的结肠癌筛查服务比例较低[OR(95%CI)分别为0.88(0.83,0.93)、0.87(0.82,0.92)、0.89(0.83,0.95)]。结论居住在冷点社区的患者拥有较高的慢性病患病率,且其享有的部分筛查服务质量较差。全科医生可应用领域数据来分配资源,主动积极筛查出此类社区中的慢性病患者以及应该享有却未能享受到筛查服务的患者。LIAW W KRIST A H TONG S T 本刊编辑部 2018中国全科医学2018,21,19:3

19Endothelin-1 activation in pediatric patients undergoing surgical coarctation of the aorta repair显示文摘AIM To determine endothelin-1(ET-1)concentration before and after surgical coarctectomy and evaluate its association with left ventricular geometric change.METHODSA prospective,cohort study of 24 patients aged 2 d to 10years with coarctation of the aorta undergoing surgical repair.A sub-cohort of patients with age<1 mo was classified as'neonates'.Echocardiograms were performed just prior to surgery and in the immediate post-op period to assess left ventricle mass index and relative wall thickness(RWT).Plasma ET-1 levels were assessed at both time points.Association between ET-1 levels and ventricular remodeling was assessed.RESULTSPatients<1 year demonstrated higher pre-op ET-1 than post-op(2.8 pg/m L vs 1.9 pg/m L,P=0.02).Conversely,patients>1 year had no change in ET-1 concentration before and after surgery(1.1 vs 1.4,NS).Pre-op,patients<1 year demonstrated significantly higher ET-1 than older children(2.8 vs 1.1,P=0.001).Post-op there was no difference between the age groups(1.9 vs 1.4,NS).Neither RWT nor left ventricle mass index(LVMI)varied from pre-op to post-op.The subset of neonates showed a strong positive correlation between pre-op ET-1 and RWT(r=0.92,P=0.001).Patients with ET-1>2 pg/m L preop demonstrated higher LVMI(65.7 g/m^(2.7) vs 38.5 g/m^(2.7),P=0.004)and a trend towards higher RWT(45%vs39%,P=0.07)prior to repair than those with lower ET-1concentration.CONCLUSIONET-1 concentration is significantly variable in the perioperative period surrounding coarctectomy.Older children and infants have different responses to surgical repair suggesting different mechanisms of activation.Benjamin Steven Frank Tracy T Urban Suhong Tong Courtney Cassidy Max B Mitchell Christopher S Nichols Jesse A Davidson 2017World Journal of Cardiology2017,9,12:2
