| 4 | RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy,thus calling for the development of safe and effective vaccines.The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines.Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator,elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections.The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge.These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV. | Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7 |
| 5 | Parallel ADR Detection Based on Spark and BCPNN显示文摘Adverse Drug Reaction(ADR) is one of the major challenges to the evaluation of drug safety in the medical field. The Bayesian Confidence Propagation Neural Network(BCPNN) algorithm is the main algorithm used by the World Health Organization to monitor ADRs. Currently, ADR reports are collected through the spontaneous reporting system. However, with the continuous increase in ADR reports and possible use scenarios, the efficiency of the stand-alone ADR detection algorithm will encounter considerable challenges. Meanwhile, the BCPNN algorithm requires a certain number of disk I/O, which leads to considerable time consumption. In this study,we propose a Spark-based parallel BCPNN algorithm, which speeds up data processing and reduces the number of disk I/O in BCPNN, and two optimization strategies. Then, the ADR data collected from the FDA Adverse Event Reporting System are used to verify the performance of the proposed algorithm and its optimization strategies.Experiments show that the parallel BCPNN can significantly accelerate data processing and the optimized algorithm has a high acceleration rate and can effectively prevent memory overflow. Finally, we apply the proposed algorithm to a dataset provided by a real medical consortium. | 12 | Evaluation of spatial epitope computational tools based on experimentally-confirmed dataset for protein antigens显示文摘Antibody molecules interact with antigen proteins through the epitope area,where the epitope residues are found to be discontinuous or spatial or conformational rather than linear on the protein surface.There are various computational algorithms to predict the spatial epitopes,and each of them have an outstanding performance based on their individual testing dataset.In this work,an independent dataset was created through collection of the epitope residual sites which have been confirmed by experiments. | 16 | Traffic light detection and recognition for autonomous vehicles显示文摘Traffic light detection and recognition is essential for autonomous driving in urban environments. A camera based algorithm for real-time robust traffic light detection and recognition was proposed, and especially designed for autonomous vehicles. Although the current reliable traffic light recognition algorithms operate well under way, most of them are mainly designed for detection at a fixed position and the effect on autonomous vehicles under real-world conditions is still limited. Some methods achieve high accuracy on autonomous vehicle, but they can't work normally without the aid of high-precision priori map. The authors presented a camera-based algorithm for the problem. The image processing flow can be divided into three steps, including pre-processing, detection and recognition. Firstly, red-green-blue(RGB) color space is converted to hue-saturation-value(HSV) as main content of pre-processing. In detection step, the transcendental color threshold method is used for initial filterings, meanwhile, the prior knowledge is performed to scan the scene in order to quickly establish candidate regions. For recognition, this article use histogram of oriented gradients(HOG) features and support vector machine(SVM) as well to recognize the state of traffic light. The proposed system on our autonomous vehicle was evaluated. The qualitative and quantitative analysis results show that the roll gap of the first two rewinding rolls and the angle between the line that connects the center points of the first two rewinding rolls and the vertical line play an important role in determining the ideal log diameter, which is related to the production capacity of the rewinder.
