1Inactivation of PTEN is associated with increased angiogenesis and VEGF overexpression in gastric cancer显示文摘AIM: To investigate the expression of PTEN/MMAC1/TEP1 and vascular endothelial growth factor (VEGF), their roles in biologic behavior and angiogenesis and their association in gastric cancer.METHODS: Immunohistochemical staining was used to evaluate the expression of PTEN, VEGF and microvascular density (MVD) on paraffin-embedded sections in 70 patients with primary gastric cancer and 24 patients with chronic superficial gastritis (CSG). Expression of PTEN, VEGF and MVD were compared with clinicopathological features of gastric cancer. The relationship between expression of PTEN, VEGF and MVD as well as the relationship between PTEN and VEGF expression in caner cells were investigated. RESULTS: PTEN expression significantly decreased (t= 3.98, P<0.01) whereas both VEGF expression and MVD significant increased (t = 4.29 and 4.41, respectively, both P<0.01) in gastric cancer group compared with CSG group. PTEN expression was significantly down-regulated (t=1.95, P<0.05) whereas VEGF expression (t = 2.37, P<0.05) and MVD (t= 3.28, P<0.01) was significantly up-regulated in advanced gastric cancer compared with early-stage gastric cancer. PTEN expression in gastric cancer showed a negative association with lymph node metastasis (t= 3.91, P<0.01), invasion depth (t= 1.95, P<0.05) and age (t= 4.69, P<0.01). MVD in PTEN-negative gastric cancer was significantly higher than that in PTEN-positive gastric cancer (t=3.69, P<0.01), and there was a negative correlation betweenPTEN expression and MVD (γ=-0.363, P<0.05). VEGF expression was positively associated with invasion depth (especially with serosa invasion, t = 4.69, P<0.01), lymph node metastasis (t= 2.31, P<0.05) and TNM stage (t= 3.04, P<0.01). MVD in VEGF-positive gaslyic cancer was significantly higher than that in VEGF-negative gastric cancer (t=4.62, P<0.01), and there was a positive correlation between VEGF expression of and MVD (y = 0.512, P<0.05). VEGF expression in PTEN-negative gaslyic cancer was significantly stronger than that in PTEN-positive gastric cancer (t=2.61, P<0.05), and there was a significantly negative correlation between the expression of VEGF and PTEN (γ=-0.403, P<0.05).CONCLUSION: Our results imply that inactivation of PTEN gene and over-expression of VEGF contribute to the neovascularization and progression of gastric cancer. PTEN-related angiogenesis might be attributed to its up-regulation of VEGF expression. 2Role of adhesion molecules and dendritic cells in rat hepatic/renal ischemia-reperfusion injury and anti-adhesive intervention with anti-P-selectin lectin-EGF domain monoclonal antibody显示文摘AIM: To investigate the role of P-selectin, intercellular adhesion molecule-1 (ICAM-1) and dendritic cells (DCs)in liver/kidney of rats with hepatic/renal ischemiareperfusion injury and the preventive effect of anti-Pselectin lectin-EGF domain monoclonal antibody (anti-PsLEGFmAb) on the injury.METHODS: Rat models of hepatic and renal ischemiareperfusion were established. The rats were then divided into two groups, one group treated with anti-PsL-EGFmAb(n = 20) and control treated with saline (n = 20). Both groups were subdivided into four groups according to reperfusion time (1, 3, 6 and 24 h). The sham-operated group (n = 5) served as a control group. DCs were observed by the microscopic image method, while P-selectin and ICAM-1 were analyzed by immunohistochemistry.RESULTS: P-selectin increased significantly in hepatic sinusoidal endothelial cells and renal tubular epithelial cells 1 h after ischemia-reperfusion, and the expression of ICAM-1 was up-regulated in hepatic sinusoid and renal vessels after 6 h. CD1a+CD80+DCs gradually increased in hepatic sinusoidal endothelium and renal tubules and interstitium 1 h after ischemia-reperfusion, and there was the most number of DCs in 24-h group. 3Effect of tetramethylpyrazine on P-selectin and hepatic/renal ischemia and reperfusion injury in rats显示文摘AIM: To investigate the effect of tetramethylpyrazine on hepatic/renal ischemia and reperfusion injury in rats. METHODS: Hepatic/renal funclJon, histopathotogical changes, and hepatic/renal P-selectin expression were studied with biochemical measurement and immunohistochemistry in hepatic/renal ischemia and reperfusion injury in rat models. RESULTS: Hepatic/renal insufficiency and histopathological damage were much less in the tetramethylpyrazine-treated group than those in the saline-treated groups. Hepatic/renal P-selectin expression was down regulated in the tetramethylpyrazine-treated group. CONCLUSION: P-selectin might mediate neutrophil infiltration and contribute to hepatic/renal ischemia and reperfusion injury. 4Effect of P-selectin monoclonal antibody on metastasis of gastric cancer and immune function显示文摘AIM: To investigate the effect of cell adhesion molecule Pselectin monoclonal antibody (Mab) on metastasis and immune function of mice orthototopically implanted with human gastric cancer tissue.METHODS: SCID mice were implanted orthotopically with SGC-7901 human gastric carcinoma tissue. Starting from day 3 after operation, animals were given intravenously PBS or P-selectin Mab (100 μg/injection) (for both normal mice and tumor-implanted mice with tumors), twice weekly for 3weeks. Two animals in each group were sacrificed randomly at the 1st, 2nd, 4th week and 6th week. While T cell and B cell transformation indices were determined with the 3H TdR infiltration method, the NK cell activity was detected by the LDH release method.RESULTS: The metastatic rate in the P-selectin Mab treated group was lower than that in the PBS treated group (with tumors). 5Ni_(2)P/NiMoP heterostructure as a bifunctional electrocatalyst for energy-saving hydrogen production显示文摘Electrochemical water splitting is a sustainable and feasible strategy for hydrogen production but is hampered by the sluggish anodic oxygen evolution reaction(OER).Herein,an effective approach is introduced to significantly decrease the cell voltage by replacing the anodic OER with a urea oxidation reaction(UOR).A Ni_(2)P/NiMoP nanosheet catalyst with a hierarchical architecture is uniformly grown on a nickel foam(NF)substrate through a simple hydrothermal and phosphorization method.The Ni_(2)P/NiMoP achieves impressive HER activity,with a low overpotential of only 22 mV at 10 mA cm^(-2)and a low Tafel slope of 34.5 mV dec^(−1).In addition,the oxidation voltage is significantly reduced from 1.49 V to 1.33 V after the introduction of 0.33 M urea.Notably,a two-electrode electrolyzer employing Ni_(2)P/NiMoP as a bifunctional catalyst exhibits a current density of 10 mA cm^(-2)at a cell voltage of 1.35 V and excellent long-term durability after 80 h.Tongzhou Wang Xuejie Cao Lifang Jiao 2021eScience2021,1,1:10
6Historical retrospect and future prospect of research on effective teaching in China——Knowledge mapping analysis based on the research over the past three decades (1986-2016)显示文摘This paper takes China National Knowledge Infrastructure (CNKI) as its literature data sources, and uses Bicomb and SPSS software to draw knowledge mapping for the hot spots of 290 papers about effective teaching in China from 1986 to 2016, so as to visually show the hot issues and development trend of effective teaching over the past three decades in China. The research shows that hot issues in effective teaching research over the past 30 years mainly fall into the following three categories: First, effective teaching studies on classroom teaching activities (from the perspective of teachers);Second, effective teaching studies under the background of new curriculum reform (perspective of curriculum reform);Third, effective teaching studies on students' self-learning (from the perspective of students). Effective teaching research in China in the future should grasp the following trends: Firstly, expansion of concept from static definition to dynamic understanding. Secondly, change of research perspective from focusing on teachers' teaching behavior to students' learning process and learning situation. Thirdly, deepening of research content from macro expound based on new curriculum reform to medium and micro research that combines core competence development of the students, the discipline core quality formation, and the implementation of specific curriculum standards. 8PEM water electrolysis for hydrogen production:fundamentals,advances,and prospects显示文摘Hydrogen,as a clean energy carrier,is of great potential to be an alternative fuel in the future.Proton exchange membrane(PEM)water electrolysis is hailed as the most desired technology for high purity hydrogen production and self-consistent with volatility of renewable energies,has ignited much attention in the past decades based on the high current density,greater energy efficiency,small mass-volume characteristic,easy handling and maintenance.To date,substantial efforts have been devoted to the development of advanced electrocatalysts to improve electrolytic efficiency and reduce the cost of PEM electrolyser.In this review,we firstly compare the alkaline water electrolysis(AWE),solid oxide electrolysis(SOE),and PEM water electrolysis and highlight the advantages of PEM water electrolysis.Furthermore,we summarize the recent progress in PEM water electrolysis including hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)electrocatalysts in the acidic electrolyte.We also introduce other PEM cell components(including membrane electrode assembly,current collector,and bipolar plate).Finally,the current challenges and an outlook for the future development of PEM water electrolysis technology for application in future hydrogen production are provided.Tongzhou Wang Xuejie Cao Lifang Jiao 2022Carbon Neutrality2022,1,1:2
9Crystalline-amorphous interfaces of NiO-CrO_(x)electrocatalysts for boosting the urea oxidation reaction显示文摘The overall energy efficiency of electrochemical systems is severely hindered by the traditional anodic oxygen evolution reaction(OER).Utilizing urea oxidation reaction(UOR)with lower thermodynamic potential to replace OER provides a promising strategy to enhance the energy efficiency.Amorphous and heterojunctions electrocatalysts have been aroused extensive studies owing to their unique physicochemical properties and outperformed activity.Herein,we report a simple method to construct a novel crystalline-amorphous NiO-CrO_(x)heterojunction grown on Ni foam for UOR electrocatalyst.The NiO-CrO_(x)electrocatalyst displays excellent UOR performance with an ultralow working potential of 1.32 V at 10 mA·cm^(−2)and ultra-long stability about 5 days even at 100 mA·cm^(−2).In-situ Raman analysis and temperature-programmed desorption(TPD)measurement verify that the presence of the amorphous CrO_(x)phase can boost the reconstruction from NiO to active NiOOH species and enhance adsorption ability of urea molecule.Besides,the unique crystalline-amorphous interfaces are also benefit to improving the UOR performance.Xuejie Cao Tongzhou Wang Hongye Qin Guangliang Lin Lihua Zhao Lifang Jiao 2023Nano Research2023,16,3:1
