Formation and destruction processes of upper Sinian oil-gas pools in the Dingshan-Lintanchang structural belt, southeast Sichuan Basin, China显示文摘The lower Cambrian Niutitang Formation hydrocarbon source rocks at the Dingshan-Lintanchang structure in the southeast Sichuan Basin were of medium-good quality with two excellent hydrocarbon-generating centers developed in the periphery areas, with a possibility of forming a medium to large-sized oil-gas field. Good reservoir rocks were the upper Sinian (Dengying Formation) dolomites. The mudstone in the lower Cambrian Niutitang Formation with a good sealing capacity was the cap rock. The widely occurring bitumen in the Dengying Formation indicates that a paleo oil pool was once formed in the study area. The first stage of paleo oil pool formation was maturation of the lower Cambrian source rocks during the late Ordovician. Hydrocarbon generation from the lower Cambrian source rocks stopped due to the Devonian-Carboniferous uplifting. The lower Cambrian source rocks then restarted generation of large quantities of hydrocarbons after deposition of the middle Permian sediments. This was the second stage of the paleo oil pool formation. The oil in the paleo oil pool began to crack during the late Triassic and a paleo gas pool was formed. This paleo gas pool was destroyed during the Yanshan-Himalayan folding, uplifting and denudation. Bitumen can be widely seen in the Dengying Formation in wells and outcrops in the Sichuan Basin and its periphery areas. This provides strong evidence that the Dengying Formation in the Sichuan Basin and its periphery areas was once an ultra-large structural-lithologic oil-gas field, which was damaged during the Yanshan-Himalayan period.Liu Shugen Zhang Zhijing Huang Wenming Wang Guozhi Sun Wei Xu Guosheng Yuan Haifeng Zhang Changjun Deng Bin 2010Petroleum Science2010,7,3:20
Multi-Stage Basin Development and Hydrocarbon Accumulations: A Review of the Sichuan Basin at Eastern Margin of the Tibetan Plateau显示文摘Sichuan Basin is one of the uppermost petroliferous basins in China. It experienced three evolutionary phases which were marine carbonate platform(Ediacaran to Late Triassic), Indosinian-Yanshanian orogeny foreland basin(Late Triassic to Late Cretaceous) and uplift and tectonic modification(Late Cretaceous to Quaternary). The present-day tectonics of the Sichuan Basin and its periphery are characterized by three basic elements which are topography, basement type and surface structure, and two settings(plate margin and interior). Therefore, be subdivided into five units which have different structure and tectonic history. The basin contains five different sets of source rocks with thickness up to 2 500 m. These source rocks were well preserved due to the presence of Middel–Lower Triassic evaporites(>~200 m) and thick terrestrial sediments filling in the Indosinian-Yanshanian foreland basin(>3 000 m). The uplift and erosion since Late Cretaceous has significant influence on cross-strata migration and accumulation of oil and gas. The multi-phase evolution of the basin and its superimposed tectonic elements, good petroleum geologic conditions and diverse petroleum systems reveal its bright exploration prospects.Shugen Liu Bin Deng Luba Jansa Zhiwu Li Wei Sun Guozhi Wang Zhili Luo Ziquan Yong 2018Journal of Earth Science2018,29,2:17

Hydrothermal Dolomite in the Upper Sinian (Upper Proterozoic) Dengying Formation, East Sichuan Basin, China显示文摘Hydrothermal Dolomite(HTD) is present in the Upper Sinian(Upper Proterozoic) Dengying Formation, east Sichuan Basin, China. The strata are comprised by primary dolomite. The HTD has various textures, including zebra dolomite, subhorizontal sheet-like cavities filled by saddle dolomite and breccias cemented by saddle dolomites as well occur as a fill of veins and fractures. Also co-occur MVT type lead-zinc ores in the study area. The δ13C and δ18O isotopes of HTD in the Upper Sinian Dengying Formation are lighter than those of the host rocks, while 87Sr/86 Sr is higher. The apparent difference in carbon, oxygen and strontium isotopes, especially the large difference in 87Sr/86 Sr isotopes ratio indicate crystallization from hot basinal and/or hydrothermal fluids. Saddle dolomite was precipitated at temperatures of 270-320°C. The diagenetic parasequences of mineral assemblage deposited in the Dengying Formation are:(1) dolomite host rock →sphalerite-galena-barite-fluorite;(2) dolomite host rock →saddle dolomite →quartz;(3) dolomite host rock → saddle dolomite→bitumen;(4) dolomite host rock → saddle dolomite → barite. The mean chemical composition of the host dolomite matrix and HTD didn't change much during hydrothermal process. The fluids forming the HTDs in the Dengying Formation were mixtures of freshwater from the unconformity at the top of Sinian, fluids from diagenetic compaction and hydrocarbon generation & expulsion from the Lower Cambrian Niutitang Formation mudstones or the Doushantuo Formation silty mudstones, and hydrothermal fluids from the basement. The hydrocarbon reservoirs associated with the HTD were mostly controlled by the basement faults and fractures and karsting processes at the unconformity separating Sinian and Cambrian strata. The hydrocarbon storage spaces of HTD included dissolved cavities and intercrystalline pores. Dissolution cavities are extensive at the top of Dengying Formation, up to about 46 m below the unconformity between Sinian and Cambrian and were generated mainly during karstification. Hydrothermal alteration enhanced the reservoir property of the Dengying Formation dolomites with 3%-5% increase in porosity. No agreement has been reached why zebra dolomite occurs only in the Upper Sinian strata, which would indicate that HTD mineralization occurred during two different periods, each of them related to major extensional tectonic event. The early one related to the Xingkai taphrogenesis(Z2-?1)and the later one to the Emei taphrogenesis(D2-T2). But, all the data from saddle dolomite suggest that the predominant crystallization occurred during the latter event.LIU Shugen HUANG Wenming JANSA Luba F. 5Multi-Stage Hydrocarbon Accumulation and Formation Pressure Evolution in Sinian Dengying FormationCambrian Longwangmiao Formation, Gaoshiti-Moxi Structure, Sichuan Basin显示文摘Sichuan Basin is a typical superimposed basin, which experienced multi-phase tectonic movements, meanwhile Sinian–Cambrian underwent complex hydrocarbon accumulation processes, causing exploration difficulties in the past 60 years. Based on the microscopic evidence of fluid inclusions, combined with basin-modelling, this paper determines stages and time of hydrocarbon accumulation, reconstructs evolution of formation pressure and dynamic processes of hydrocarbon accumulation in Sinian Dengying Formation-Cambrian Longwangmiao Formation of Gaoshiti-Moxi structure. Three stages of inclusions are detected, including a stage of yellow-yellowgreen fluorescent oil inclusions, a stage of blue fluorescent oil-gas inclusions and a stage of non-fluorescent gas inclusions, reflecting the study area has experienced a series of complex hydrocarbon accumulation processes, such as formation of paleo-oil reservoirs, cracking of crude oil, formation of paleo-gas reservoirs and adjustment to present gas reservoirs, which occurred during 219–188, 192–146 and 168–0 Ma respectively. During the period of crude oil cracking, Dengying Formation-Longwangmiao Formation showed weak overpressure to overpressure characteristics, then after adjustment of paleo-gas reservoirs to present gas reservoirs, the pressure in Dengying Formation changed into overpressure but finally reduced to normal pressure system. However, due to excellent preservation conditions, the overpressure strength in Longwangmiao Formation only slightly decreased and was still kept to this day.Juan Wu Shugen Liu Guozhi Wang Yihua Zhao Wei Sun Jinming Song Yanhong Tian 2016Journal of Earth Science2016,27,5:8
Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes, downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. Finally, Kindlin-2 loss increases osteocyte apoptosis and impairs osteocyte spreading and dendrite formation. Thus, we demonstrate an important role of Kindlin-2 in the regulation of bone homeostasis and provide a potential target for the treatment of metabolic bone diseases.Huiling Cao Qinnan Yan Dong Wang Yumei Lai Bo Zhou Qi Zhang Wenfei Jin Simin Lin Yiming Lei Liting Ma Yuxi Guo Yishu Wang Yilin Wang Xiaochun Bai Chuanju Liu Jian QFeng Chuanyue Wu Di Chen Xu Cao Guozhi Xiao 2020Bone Research2020,8,1:8
Formation and evolution of Sinian oil and gas pools in typical structures, Sichuan Basin, China显示文摘The only major breakthrough in the exploration of the Sinian of the Sichuan Basin has been the Weiyuan gas field. Taking the typical structures in the Sichuan Basin as examples, an apatite fission track simulation was applied to constrain the thermal evolutionary history of the source rocks in this study. Combined with trap formation and evolution, the formation, destruction, and accumulation history of the Sinian reservoirs were analyzed from a dynamic perspective. The Sinian reservoirs underwent several primary stages: the paleo-reservoir formation and destruction in the late Silurian, hydrocarbon recharge in the Permian-Triassic, the cracking of oil to gas and dissolved-gas in the late Triassic-Mid-Late Jurassic, and the exsolution and accumulation of dissolved-gas as a result of episodic uplift since the Late Cretaceous. The exsolution process of dissolved-gas is of great significance to the accumulation of natural gas. The formation of the Weiyuan gas field is also related to this process. The Sinian in the Sichuan Basin has a broad exploration prospect, and exploration targets focused on the Leshan-Longnüsi Paleo-uplift tectonic zone with weak influences on potential hydrocarbon reservoirs from the late tectonism and transformation.Yuan Haifeng Liang Jiaju Gong Deyu Xu Guosheng Liu Shugen Wang Guozhi 2012Petroleum Science2012,9,2:7
Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture, leading to an increased risk of fractures.Recently, circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism. However, the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure. Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein, and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression, thereby stimulating the osteogenic differentiation of BMSCs.Furthermore, overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation, thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling, which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis.Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng 2022Bone Research2022,10,3:7
NUMERICAL STUDIES ON RESONANT AND ENHANCEMENT EFFECTS FOR COUPLING OF MICROWAVE PULSES INTO NARROW SLOTS显示文摘In this paper, modifications to the finite-difference time-domain(FD-TD) method for modeling microwave pulse coupling into a slot, which is much narrower than one conventional FD-TD cell, are discussed. The coupling process of microwave pulse into a slot is studied by using the modified FD-TD method, and the dependence of microwave coupling on slot sizes, the carrier frequencies and the polarization directions of the incident waves is analysed. Resonant and enhancement effects which occur in this process are observed. The condition at which the resonant effect takes place is also presented.Wang Jianguo Yu Hanqing Liu Guozhi Chen Yusheng Fan Ruyu (Northwest Institute of Nuclear Technology, Branch 15, P. O. 10CHIP regulates bone mass by targeting multiple TRAF family members in bone marrow stromal cells显示文摘Carboxyl terminus of Hsp70-interacting protein(CHIP or STUB1) is an E3 ligase and regulates the stability of several proteins which are involved in different cellular functions. Our previous studies demonstrated that Chip deficient mice display bone loss phenotype due to increased osteoclast formation through enhancing TRAF6 activity in osteoclasts. In this study we provide novel evidence about the function of CHIP. We found that osteoblast differentiation and bone formation were also decreased in Chip KO mice. In bone marrow stromal(BMS) cells derived from Chip^(-/-) mice, expression of a panel of osteoblast marker genes was significantly decreased. ALP activity and mineralized bone matrix formation were also reduced in Chip-deficient BMS cells. We also found that in addition to the regulation of TRAF6, CHIP also inhibits TNFα-induced NF-κB signaling through promoting TRAF2 and TRAF5 degradation. Specific deletion of Chip in BMS cells downregulated expression of osteoblast marker genes which could be reversed by the addition of NF-κB inhibitor. These results demonstrate that the osteopenic phenotype observed in Chip^(-/-) mice was due to the combination of increased osteoclast formation and decreased osteoblast differentiation. Taken together, our findings indicate a significant role of CHIP in bone remodeling.Tingyu Wang Shan Li Dan Yi Guang-Qian Zhou Zhijie Chang Peter X.Ma Guozhi Xiao Di Chen 2018Bone Research2018,6,2:5
Current understanding of osteoarthritis pathogenesis and relevant new approaches显示文摘Osteoarthritis(OA)is the most common degenerative joint disease that causes painful swelling and permanent damage to the joints in the body. The molecular mechanisms of OA are currently unknown.OA is a heterogeneous disease that affects the entire joint, and multiple tissues are altered during OA development. To better understand the pathological mechanisms of OA,new approaches, methods, and techniques need to be used to understand OA pathogenesis. In this review,we first focus on the epigenetic regulation of OA,with a particular focus on DNA methylation, histone modification, and microRNA regulation, followed by a summary of several key mediators in OA-associated pain.We then introduce several innovative techniques that have been and will continue to be used in the fields of OA and OA-associated pain, such as CRISPR,scRNA sequencing, and lineage tracing.Next,we discuss the timely updates concerning cell death regulation in OA pathology, including pyroptosis, ferroptosis, and autophagy,as well as their individual roles in OA and potential molecular targets in treating OA.Finally, our review highlights new directions on the role of the synovial lymphatic system in OA.An improved understanding of OA pathogenesis will aid in the development of more specific and effective therapeutic interventions for OA.Liping Tong Huan Yu Xingyun Huang Jie Shen Guozhi Xiao Lin Chen Huaiyu Wang Lianping Xing Di Chen 2022Bone Research2022,10,4:4
Synthesis and characterization of wormhole-like mesoporous Ce_(0.6)Zr_(0.35)Y_(0.05)O_2 solid solutions显示文摘Nanoparticles of Ce0.6Zr0.35Y0.05O2 (CZY) solid solution have been prepared by the CTAB (hexadecyl-trimethyl ammonium bromide), CTAB-EG (ethylene glycol) templating, and CTAB-EG-NaCl (in which the pores of the precursor synthesized by the CTAB-EG method is filled by a certain amount of NaCl) method, respectively. The physical properties of these materials were characterized by means of tech-niques such as X-ray diffraction (XRD), high resolution scanning electron microscopy (HRSEM), transmission electron microscopy (TEM), selected area electron diffraction (SAED), and N2 adsorp-tion-desorption measurements. The CZY samples synthesized by the above three methods display wormhole-like mesoporous morphology and cubic crystal structures. The materials are narrow in pore size distribution (averaged pore diameter = 5.3―7.1 nm), high in surface areas (95―119 m2/g), and large in pore volumes (0.16―0.18 cm3/g). It has been demonstrated that the introduction of NaCl is capable of retaining the pore structures of solid nanomaterials at high-temperature calcination.WANG GuoZhi ZHANG Lei DENG JiGuang DAI HongXing HE Hong ZI XueHong 2007Chinese Science Bulletin2007,52,2:4
Research on paleo-fluid sources and hydrocarbon preservation conditions in marine carbonates inthe central Yangtze,China显示文摘87Sr/86Sr,δ18O andδ13C ratios of calcite, dolomite, gypsum filling vugs and fissures in marine carbonates and their host rocks from the Sinian to the middle Triassic, are employed to trace the possible source and migration path of key fluids related to development of hydrocarbon, hydrocarbon preservation condition are then discussed further.Comprehensive research, based on the paleo-fluid,the property of formation water and the deformation intensity etc.,indicates that the preservation conditionsin the Shizhu synclinorium zone and Fangdoushan anticlinorium zone are the most predominant in the central Yangtze.Three sets of fluid systems in the Shizhu synclinorium zone are identified.Little fluid exchange occurs between the upper-middle fluid system and the lower fluid system,so two independent preservation units have developed. Both the Permian and the Triassic in the upper preservation unitsand the Dengying Formation of the Sinian and the lower Cambrian in the lower preservation units havegood preservation conditions. The preservation condition in the lower association(Z-S)located in the Huaguoping synclinorium zone in the western Hubei and Hunan is better than that in other tectonic units, where the fluids in the lower association migrated vertically across strata when the fluid isolating intervalswere destroyed, the regional seals are absent and the conditions for the preservation of hydrocarbon accumulations are totally destroyed. The preservation condition is increasingly more favorable from the western Jiangxi and Hunan to the western Hubei&eastern Chongqing on the whole.Wang Guozhi Xu Guosheng Yuan Haifeng Hu Xiaofeng Zou Can 2011Petroleum Science2011,8,3:4

Fluid migration paths in the marine strata of typical structures in the western Hubei-eastern Chongqing area,China显示文摘The western Hubei-eastern Chongqing area is an important prospective zone for oil and gas exploration in the central Yangtze area. Three representative structures, the Xinchang structure, Longjuba gas-bearing structure and the Jiannan gas field, were selected to analyze biomarker parameters in marine strata and to examine various types of natural gas and hydrocarbon sources. Fluid inclusions; carbon, oxygen, and strontium isotopic characteristics; organic geochemical analysis and simulation of hydrocarbon generation and expulsion history of source rocks were used for tracing fluid migration paths in marine strata of the study area. The Carboniferous-Triassic reservoirs in three typical structures all experienced at least two stages of fluid accumulation. All marine strata above the early Permian were shown to have fluids originating in the Permian rocks, which differed from the late stage fluids. The fluids accumulated in the late Permian reservoirs of the Xinchang structure were Cambrian fluids, while those in the late Carboniferous reservoirs were sourced from a combination of Silurian and Cambrian fluids. A long-distance and large-scale cross-formational flow of fluids destroyed the preservation conditions of earlier accumulated hydrocarbons. A short-distance cross-formational accumulation of Silurian fluids was shown in the late Permian reservoirs of the Longjuba structure with favorable hydrocarbon preservation conditions. The fluid accumulation in the Carboniferous reservoirs of the Jiannan structure mainly originated from neighboring Silurian strata with a small amount from the Cambrian strata. As a result, the Jiannan structure was determined to have the best preservation conditions of the three. Comparative analysis of fluid migration paths in the three structures revealed that the zone with a weaker late tectonism and no superimposition and modification of the Upper and Lower Paleozoic fluids or the Upper Paleozoic zone with the fluid charging from the Lower Paleozoic in the western Hubei-eastern Chongqing area are important target areas for future exploration.Xu Guosheng Liang Jiaju Gong Deyu Wang Guozhi Yuan Haifeng Cao Junxing Zhang Chengfu 2013Petroleum Science2013,10,1:3
Resting-state connectivity in the default mode network and insula during experimental low back pain显示文摘Functional magnetic resonance imaging studies have shown that the insular cortex has a significant role in pain identification and information integration, while the default mode network is associated with cognitive and memory-related aspects of pain perception. However, changes in the functional connectivity between the default mode network and insula during pain remain unclear. This study used 3.0 T functional magnetic resonance imaging scans in 12 healthy subjects aged 24.8 ± 3.3 years to compare the differences in the functional activity and connectivity of the insula and default mode network between the baseline and pain condition induced by intramuscular injection of hypertonic saline. Compared with the baseline, the insula was more functionally connected with the medial prefrontal and lateral temporal cortices, whereas there was lower connectivity with the posterior cingulate cortex, precuneus and inferior parietal lobule in the pain condition. In addition, compared with baseline, the anterior cingulate cortex exhibited greater connectivity with the posterior insula, but lower connectivity with the anterior insula, during the pain condition. These data indicate that experimental low back pain led to dysfunction in the connectivity between the insula and default mode network resulting from an impairment of the regions of the brain related to cognition and emotion, suggesting the importance of the interaction between these regions in pain processing.Shanshan Zhang Wen Wu Guozhi Huang Ziping Liu Shigui Guo Jianming Yang Kangling Wang 2014Neural Regeneration Research2014,9,2:3
Inhibition of Axinl in osteoblast precursor cells leads to defects in postnatal bone growth through suppressing osteoclast formation显示文摘Axinl is a negative regulator of β-catenin signaling and its role in osteoblast precursor cells remains undefined. In the present studies,we determined changes in postnatal bone growth by deletion of Axinl in osteoblast precursor cells and analyzed bone growth in newborn and postnatal Axin1 O5X mice and found that hypertrophic cartilage area was largely expanded in AxinlOSX KO mice.A larger number of chondrocytes and unabsorbed cartilage matrix were found in the bone marrow cavity of Axin1OSX KO mice.Osteoclast formation in metaphyseal and subchondral bone areas was significantly decreased, demonstrated by decreased TRAPpositive cell numbers, associated with reduction of MMP9-and cathepsin K-positive cell numbers in Axin1 O5X KO mice.OPG expression and the ratio of O p g to Rankl were significantly increased in osteoblasts of Axinl O5X KO mice.Osteoclast formation in primary bone marrow derived microphage(BMM)cells was significantly decreased when BMM cells were cultured with conditioned media(CM)collected from osteoblasts derived from Axin1OSX mice compared with BMM cells cultured with CM derived from WT mice. Thus, the loss of Axinl in osteoblast precursor cells caused increased OPG and the decrease in osteoclast formation, leading to delayed bone growth in postnatal Axin1°sx KO mice.Bing Shu Yongjian Zhao Shitian Zhao Haobo Pan Rong Xie Dan Yi Ke Lu Junjie Yang Chunchun Xue Jian Huang Jing Wang Dongfeng Zhao Guozhi Xiao Yongjun Wang Di Chen 2020Bone Research2020,8,3:3
The coupling of fluid flow and uplift since Late Cretaceous in Zoige area, western Sichuan显示文摘The uplift indicated by five AFT (apatite fission track) samples is more than 3400 m by multi-episodic uplift since Late Cretaceous in Zoige area; especially the processes of fast uplift in Late Cretaceous and Neogene have important influences on the stress of paleo-fluid. Based on field geology, macroscopic features of fracture, and geochemistry of fluid inclusions, we decipher the paleo-fluid process of episodic migration. In early uplift stage, the temperature of inclusions increased with the constant salinity, whilst both of them proportionally decreased in the mid-late stage, indicating the different tendency of heat-fluid warming and freshwater contamination at different time. Of particular importance are the features of episodic fluid flow, such as ESR ages, and features of multi-episodic migration that correspond well with the process of multi-episodic uplift. Thus, concerning the rock stress-strain behavior responding to uplift, we further discuss the spatio-temporal coupling effect of episodic migration and decompression in multi-episodic uplift, thereby to better understand petroleum geology in the region.DENG Bin LIU ShuGen LI JuChu WANG GuoZhi LIU Shun LI ZhiWu LI XiangHui 2011Science China Earth Sciences2011,54,7:2
LIM domain proteins Pinch1/2 regulate chondrogenesis and bone mass in mice显示文摘The LIM domain-containing proteins Pinch1/2 regulate integrin activation and cell–extracellular matrix interaction and adhesion. Here,we report that deleting Pinch1 in limb mesenchymal stem cells(MSCs)and Pinch2 globally(double knockout;dKO)in mice causes severe chondrodysplasia, while single mutant mice do not display marked defects.Pinch deletion decreases chondrocyte proliferation, accelerates cell differentiation and disrupts column formation.Pinch loss drastically reduces Smad2/3 protein expression in proliferative zone(PZ)chondrocytes and increases Runx2 and Col10a1 expression in both PZ and hypertrophic zone(HZ)chondrocytes.Pinch loss increases sclerostin and Rankl expression in HZ chondrocytes, reduces bone formation, and increases bone resorption, leading to low bone mass.In vitro studies revealed that Pinch1 and Smad2/3 colocalize in the nuclei of chondrocytes.Through its C-terminal region,Pinch1 interacts with Smad2/3 proteins.Pinch loss increases Smad2/3 ubiquitination and degradation in primary bone marrow stromal cells(BMSCs).Pinch loss reduces TGF-β-induced Smad2/3 phosphorylation and nuclear localization in primary BMSCs.Interestingly, compared to those from single mutant mice,BMSCs from dKO mice express dramatically lower protein levels ofβ-catenin and Yap1/Taz and display reduced osteogenic but increased adipogenic differentiation capacity. Finally, ablating Pinch1 in chondrocytes and Pinch2 globally causes severe osteopenia with subtle limb shortening.Collectively, our findings demonstrate critical roles for Pinch1/2 and a functional redundancy of both factors in the control of chondrogenesis and bone mass through distinct mechanisms.Yiming Lei Xuekun Fu Pengyu Li Sixiong Lin Qinnan Yan Yumei Lai Xin Liu Yishu Wang Xiaochun Bai Chuanju Liu Di Chen Xuenong Zou Xu Cao Huiling Cao Guozhi Xiao 2020Bone Research2020,8,4:2
