| 3 | Mycoplasma pneumoniae Macrolide Resistance and MLVA Typing in Children in Beijing,China,in 2016:Is It Relevant?显示文摘Objective The aim of this study is to investigate the macrolide resistance rate and molecular type with multiple-locus variable-number tandem-repeat analysis(MLVA)of Mycoplasma pneumoniae of Beijing in 2016 in pediatric patients.Methods Real-time quantitative polymerase chain reaction(PCR)was used to identify M.pneumoniae,and MLVA was performed.The domain V of the 23 S rRNA was sequenced to detect macrolide-resistant point mutations.We also investigated the activities of antibiotics against M.pneumoniae isolates in vitro.Results The PCR detection rate of M.pneumoniae in children in Beijing was 40%,and the macrolide resistance rate was 66%.The A2063 G mutation in the 23 S rRNA V region is the dominant mutation(137/146,93.84%),whereas the A2064 G mutation is rare(9/146,6.16%).Seventy-three samples were typed successfully by MLVA typing,including 86.3%(63/73)were MLVA type 4-5-7-2,and 13.7%(10/73)were MLVA type 3-5-6-2.No other types were found.No strains were resistant to levofloxacin or tetracycline.Conclusion In 2016,a specific decrease in the macrolide resistance rate occurred in Beijing.The detection rate and macrolide resistance rate of outpatients are lower than those of inpatients.The A2063 G mutants M.pneumoniae have high levels of resistance to erythromycin and azithromycin.The primary MLVA type is 4-5-7-2,followed by 3-5-6-2.No other MLVA types were detected.No strains resistant to tetracycline or levofloxacin were found in vitro. | DOU Hai Wei TIAN Xiu Jun XIN De Li WEI Ran ZHOU Wei WANG Hong QIN Xuan Guang SHAO Jun Yan XU Bao Ping GE Li Xia SHI Da Wei | 2020 | Biomedical and Environmental Sciences2020,33,12: | 9 |
| 4 | H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak显示文摘AIM To investigate the protective efficacy ofH2 strain attenuated live hepatitis A vaccines(H2-strain vaccines)in hepatitis A(HA)outbreaks.METHODS With the permission of theirparents,5551 pre-school and grade 1-3 primaryschool children were inoculated with 1 dose(106.5TCID50)of H2-strain vaccines in anonrandomized,controlled trial conducted inFucheng County,Hebei Province in May 1997.Another 6485 children in the same grades andcompatible in gender and age were enrolled ascontrols.Epidemiological and serological surveywas conducted to evaluate the protectiveefficacy of the vaccines.ELISA was used todetect serum IgM anti-HAV.RESULTS HA outbreak started in early May1998,peaked in the middle of the same month,and lasted about 80 days.Overall 302 HA caseswere found,192(53.58%)were 5-9 years old.One vaccinee and 25 control cases were found tohave hepatitis A,which account for 0.28%(1/356)and 5.92%(25/422)of all vaccinees andcontrols in the 14 villages,respectively.The protective efficacy of vaccines was 95.27%(95% Cl:85.83%-104.72%).In subjects testedfor anti-HAV IgM from 13 villages,1(0.40%)overt and 11(4.06%)asymptomatic HAV caseswere found in 271 vaccinees,but 21(6.69%)ofovert and asymptomatic ones were found in 314controls.CONCLUSION H2 strain vaccines were excellent in preventing overt hepatitis A, but not so effective in preventing asymptomatic hepatitis A virus infection. | 7 | Higher sea surface temperature in the northern South China Sea during the natural warm periods of late Holocene than recent decades显示文摘The large-scale syntheses of global mean temperatures in IPCC fourth report suggested that the Northern Hemisphere temperature in the second half of the 20th century was likely the highest in at least the past1,300 years and the 1990s was likely the warmest decade.However,this remains debated and the controversy is centered on whether temperatures during the recent half century were higher than those during the Medieval Climate Anomaly(MCA,AD 800–1300)and the Roman Warm Period(RWP,BC 200–AD 400),the most recent two natural warm periods of the late Holocene.Here the high resolution sea surface temperatures(SSTs)of two time windows around AD 990(±40)and AD 50(±40),which located in the MCA and RWP respectively,were reconstructed by the Sr/Ca ratio and d18O of Tradacna gigas shells from the northern South China Sea.The results suggested that the mean SSTs around AD 990(±40)and AD 50(±40)were 28.1°C and 28.7°C,0.8°C and 1.4°C higher than that during AD 1994–2005,respectively.These records,together with the tree ring,lake sediment and literature records from the eastern China and northwest China,imply that the temperatures in recent decades do not seem to exceed the natural changes in MCA,at least in eastern Asia from northwest China to northern SCS. | Hong Yan Liguang Sun Da Shao Yuhong Wang Gangjian Wei | 2014 | Chinese Science Bulletin2014,59,31: | 4 |

| 12 | Hepatic encephalopathy complicated with hyponatremia and acid-base disturbance and its prognosis显示文摘Objective:To improve the diagnosis and therapeutic effect of occurrence and development of hyponatremia and disorder of acid-base balance among patients with hepatic encephalopathy(HE) by elucidating the regularity and mechanism,as well as its influence on prognosis.Methods:327 HE patients admitted to our hospital from January 1990 to June 2010 were enrolled.Meanwhile 316 patients hospitalized in the medical department of the same hospital were chosen as the control group.Patients in both groups were given the same methods to measure arterial blood gas parameters(pH value,PaCO2,[HCO3-],TCO2,BE and SaO2),blood biochemistry([Na+],[K+],[Cl-]),liver function,kidney function and blood glucose,serum sodium,and thereupon tocalculate the anion gap(AG) and the potential [HCO3-],and acid-base balance disorder.Results:Among the 327 HE patients,hyponatremia was found in 188 cases(57.4%),of whom 132 patients died(70.2%).While among the 316 patients in control group,68 presented with hyponatremia(21.5%),and 19 died(27.9%).The incidence and mortality were significantly different between the two groups(P<0.001).All the 327 patients presented with different degrees of acid-base balance disorder and 178 died(54.4%),in whom 164(50.2%) belonged to simple acid-base balance disorder and 74(45.1%) died,136(41.6%) were dual acid-base balance disorder and 80(58.8%) died,27(8.2%) were triple acid-base disturbance and 24(88.9%) died.Whereas in the control group only 83 patients(26.2%) were recognized as simple and dual acid-base balance disorder,and 18(21.7%) died.There was higher incidence of acid-base balance disorder and mortality rate in HE group than control one(P<0.001).Conclusion:Hyponatremia is valuable to judge HE patients' prognosis.The key parameters in the judgment and evaluation on acid-base balance disorder among HE patients are the change of pH values and serum electrolyte values.When pH value ≤ 7.30 or > 7.55,it generally suggests a poor prognosis. | Ren Chengshan Wang Lei Zhao Xiaoyan Yang Shiming Zhang Yanqi Wang Xiwen Bo Jianying Zhang Pengbin Guo Hong Ling Xianlong Li Yihui Da Siping Xie Xia Ren Wei Li Chunhua Qian Guisheng | 2012 | Journal of Medical Colleges of PLA(China)2012,27,3: | 3 |

| 14 | Effects of IκBα and its mutants on NF-κB and p53 signaling pathways显示文摘AIM: To study the effects of IκBα and its mutants (IκBαM, IκBα243N, IκBαM244C) on NF-κB, p53 and their downstream target genes. The relationship of NF- κB, p53, and IκBα was further discussed.METHODS: pECFP-IκBα, pECFP-IκBαM (amino acides 1-317, Ser32, 36A), pECFP-IκBα243N (amino acides 1-243), pECFP-IκBα244C (amino acides 244-317), pEYFP-p65 and pp53-DsRed were constructed and transfected to ASTC-α-1 cells. Cells were transfected with pECFP-C1 as a control. 30 h after the transfection, location patterns of NF-κB, p53 and IκBα (IκBαM, IκBα243N, IκBα244C) were observed by a laser scanning microscope (LSM510/ConfoCor2, Zeiss). RNA extraction and reverse transcription were performed in cells transfected or co-transfected with different plasmids. Effects of IκBα and its mutants on the transprition level of NF-κB, NF-κB downstream target gene TNF-α, p53 and p53 downstream target gene Bax were observed by real time QT-PCR. In all experiments β-actin was reference. Results are expressed as the target/reference ratio of the sample divided by the target/reference ratio of the control. Different transfected cells were incubated with CCK-8 for 2 h in the incubator. Then the absorbance at 450 nm was measured by using a microplate reader. RESULTS: Cells that were transfected with p53- DsRed revealed a predominant nuclear localization. YFP-p65 mainly existed in the cytoplasm. Cells were transfected with CFP-IκBα, CFP-IκBαM, and CFP- IκBα243N respectively and revealed a predominant cytosolic localization. However, cells transfected of CFP- IκBα244C revealed a predominant nuclear localization. The mRNA levels of p65, TNF-α, p53 and Bax in CFP-IκBα transfected cells did not change significantly, while in YFP-p65/CFP-IκBα co-transfected cells, IκBα decreased the transcription of p65 downstream gene TNF-α (2.24 ± 0.503) compared with the YFP-p65/ CFP-C1 co-transfected cells (5.08 ± 0.891) (P < 0.05). Phosphorylation defective IκBα (IκBαM) decreased the transcription levels of all the four genes compared with the control (P < 0.05). The N terminus of IκBα (IκBα243N) increased the transcription of NF-κB (1.84 ± 0.176) and TNF-α (1.51 ± 0.203) a little bit. However, the C terminus of IκBα (IκBα244C) increased the transcription of NF-κB, TNF-α, p53 and Bax significantly (8.29 ± 1.662, 14.16 ± 2.121, 10.2 ± 0.621, 3.72 ± 0.346) (P < 0.05). The CCK-8 experiment also showed that IκBα244C and p53 synergistically mediate apoptosis. CONCLUSIONS: IκBα and its mutants (IκBαM, IκBα243N, IκBαM244C) have different effects on NF- κB and p53 signaling pathways, according to their different structures. IκBαM bounds with NF-κB and p53 in cytoplasm steadily, and inhibits both of the two signaling pathways. p53 and IκBα244C may be co-factor in inducing apoptosis. | 15 | Photo-promoted carbonylation of chloroalkanes with carbon monoxide by non-precious metal catalysts显示文摘The photo-promoted carbonylation of chloroalkanes with carbon monoxide was carried out under ambient conditions with copper and cadmium salts catalysts. The results indicated that the corresponding esters were produced with three salt catalysts, i.e. CuBr2, CuCl2 and CdI2. Among these catalysts, CdI2 was the most efficient in terms of ester yield and selectivity, particularly, 60% yield and 75% selectivity in the carbonylation of chlorocyclohexane were achieved. Furthermore, the yield and selectivity of the ester can be improved greatly by adding tri-n-butylamine in the CuBr2 and CuCl2 catalyst systems. On the other hand, the carbonylation did not proceed with single CdCl2, however, the catalytic activity of CdCl2 was increased greatly with NaIá2H2O as additive. | 18 | Genomic analyses reveal distinct genetic architectures and selective pressures in Chinese donkeys显示文摘Donkey(Equus asinus)is an important livestock animal in China because of its draft and medicinal value.After a long period of natural and artificial selection,the variety and phenotype of donkeys have become abundant.We clarified the genetic and demographic characteristics of Chinese domestic donkeys and the selection pressures by analyzing 78 whole genomes from 12 breeds.According to population structure,most Chinese domestic donkeys showed a dominant ancestral type.However,the Chinese donkeys still represented a significant geographical distribution trend.In the selective sweep,gene annotation,functional enrichment,and differential expression analyses between large and small donkey groups,we identified selective signals,including NCAPG and LCORL,which are related to rapid growth and large body size.Our findings elucidate the evolutionary history and formation of different donkey breeds and provide theoretical insights into the genetic mechanism underlying breed characteristics and molecular breeding programs of donkey clades. | Jiafei Shen Jie Yu Xuelei Da Mei Li Gang Wang Ningbo Chen Hong Chen Chuzhao Lei Ruihua Dang | 2021 | Journal of Genetics and Genomics2021,48,8: | 2 |

| 20 | Optimization and temperature effects on sandwich betavoltaic microbattery显示文摘The design scheme of a sandwich-structure betavoltaic microbattery based on silicon using63Ni is presented in this paper.This structure differs from a monolayer energy conversion unit.The optimization of various physical parameters and the effects of temperature on the microbattery were studied through MCNP.For the proposed optimization design,P-type silicon was used as the substrate for the betavoltaic microbattery.Based on the proposed theory,a sandwich microbattery with a shallow junction was fabricated.The temperature dependence of the device was also measured.The open-circuit voltaic(Voc)temperature dependence of the optimized sandwich betavoltaic microbattery was linear.However,the Voc of the betavoltaic microbattery with a high-resistance substrate exponentially decreased over the range of room temperature in the experiment and simulation.In addition,the sandwich betavoltaic microbattery offered higher power than the monolayer betavoltaic one.The results of this paper provide a significant technical reference for optimizing the design and studying temperature effects on betavoltaics of the same type. | LIU YunPeng TANG XiaoBin XU ZhiHeng HONG Liang WANG Peng CHEN Da | 2014 | Science China(Technological Sciences)2014,57,1: | 2 |
