1HMBOX1 negatively regulates NK cell functions by suppressing the NKG2D/DAP10 signaling pathway显示文摘HMBOX1 is a new member of the homeobox family.Homeobox members have been reported to participate in embryonic development and systemic metabolism,but the function of HMBOX1 remains unclear,especially in the hematopoietic system.Here,we show that HMBOX1is expressed at a high level in primary humanNKcells but is expressed at much lower levels inNKcell lines.Overexpression of HMBOX1 significantly inhibited NK cell activities,including natural cytotoxicity against tumor cells,the level of CD107a(a marker protein for degranulation)and the production of cytolytic proteins(perforin and granzymes).More interestingly,HMBOX1 negatively regulated the expression of NKG2D and the activation of the NKG2D/DAP10 signaling pathway in NK cells.This effect was reversed by knocking down HMBOX1.Taken together,these findings demonstrate that HMBOX1 may act as a negative regulator of NK cell functions via suppressing the NKG2D/DAP10 signaling pathway.Longyan Wu Cai Zhang Jian Zhang 2011Cellular & Molecular Immunology2011,8,5:10
2Geochemistry and Zircon U–Pb age of the Yao'an pseudoleucite porphyry,Yunnan Province,China显示文摘The Yao'an Pb–Ag deposit, located in the Chuxiong Basin, western Yangtze Block, is an important component of the Jinshajiang–Ailaoshan alkaline porphyry–related polymetallic intrusive belt. This complex suite of rock bodies includes a vein of pseudoleucite porphyry within deposits of syenite porphyry and trachyte.The pseudoleucite is characterized by a variable greyish,greyish-white, and greyish-green porphyritic texture. Phenocrysts are mainly pseudoleucite with small amounts of alkali feldspar and biotite. In an intense event, leucite phenocrysts altered to orthoclase, kaolinite, and quartz.Both the pseudoleucite porphyry and the syenite porphyry samples were typical alkali-rich, K-rich, al-rich rocks with high LaN/YbNratios; enriched in light rare earth elements and large-ion lithophile elements, and depleted in high field strength elements; and with strongly negative Ta, Nb, and Ti(TNT) anomalies and slightly negative Eu anomalies—all characteristics of subduction-zone mantle-derived rock.We obtained a LA-ICP-MS zircon U–Pb age of 34.1 ± 0.3 Ma(MSWD = 2.4), which is younger than the established age of the Indian and Eurasian Plate collision.The magma derived from a Type-II enriched mantle formed in a post-collisional plate tectonic setting. 4Identification of a new isoform of the murine Sh2d1a gene and its functional implications显示文摘Signaling lymphocytic activation molecule(SLAM)-associated protein(SAP)is a Src homology(SH)domain 2-containing intracellular adaptor protein that is predominantly expressed in the hematopoietic system by T lymphocytes and NK cells.SAP protein is encoded by the SH2D1A gene located on the X chromosome.Loss-of-function mutations in SAP cause the X-linked lymphoproliferative disease(XLP),a severe immunodeficiency characterized by heightened susceptibility to Epstein-Barr virus and impaired humoral immunity.Normal individuals express several functional and non-functional isoforms of SAP as a result of alternative splicing.In this study,we identify a cryptic exon in the murine Sh2d1a gene.At the mRNA level,the new isoform of SAP(SAP-2)that includes this new exon is widely expressed in lymphoid tissues by C57BL/6 and 129 strains of inbred mice.SAP-2 accounts for approximately 1%3%of total SAP transcripts,and it is dynamically regulated during lymphocyte activation.At the protein level,the SAP-2 isoform is a 144 amino-acid protein.Compared to the dominant 126 amino-acid SAP-1 isoform,the additional 18 amino acids are inserted into a structural region that is critical for phosphotyrosine binding.Our functional analysis in vitro indicates that SAP-2 is a non-functional isoform due to decreased protein stability.Thus,both human and mouse have multiple SAP splice isoforms that may or may not function.Modulation of relative proportions of these isoforms is potentially a mechanism whereby cells can regulate SAP-mediated biological activities.WU LongYan LU PeiWen MA WeiWei CHU CoCo XU HePing QI Hai 2014Science China(Life Sciences)2014,57,1:2

6Dose rate effects on shape memory epoxy resin during 1 Me V electron irradiation in air显示文摘The effects of 1 Me V electron irradiation in air at a fixed accumulated dose and dose rates of 393.8,196.9,78.8,and 39.4 Gy s^(-1)on a shape memory epoxy(SMEP)resin were studied.Under low-dose-rate irradiation,accelerated degradation of the shape memory performance was observed;specifically,the shape recovery ratio decreased exponentially with increasing irradiation time(that is,with decreasing dose rate).In addition,the glass transition temperature of the SMEP,as measured by dynamic mechanical analysis,decreased overall with decreasing dose rate.The dose rate effects of 1 Me V electron irradiation on the SMEP were confirmed by structural analysis using electron paramagnetic resonance(EPR)spectroscopy and Fourier transform infrared(FTIR)spectroscopy.The EPR spectra showed that the concentration of free radicals increased exponentially with increasing irradiation time.Moreover,the FTIR spectra showed higher intensities of the peaks at 1660 and 1720 cm^(-1),which are attributed to stretching vibrations of amide C=O and ketone/acid C=O,at lower dose rates.The intensities of the IR peaks at 1660 and 1720 cm^(-1) increased exponentially with increasing irradiation time,and the relative intensity of the IR peak at 2926 cm^(-1)decreased exponentially with increasing irradiation time.The solid-state13 C nuclear magnetic resonance(NMR)spectra of the SMEP before and after 1 Me V electron irradiation at a dose of 1970 k Gy and a dose rate of 78.8 Gy s^(-1) indicated damage to the CH_(2)–N groups and aliphatic isopropanol segment.This result is consistent with the detection of nitrogenous free radicals,a phenoxy-type free radical,and several types of pyrolytic carbon radicals by EPR.During the subsequent propagation process,the free radicals produced at lower dose rates were more likely to react with oxygen,which was present at higher concentrations,and form the more destructive peroxy free radicals and oxidation products such as acids,amides,and ketones.The increase in peroxy free radicals at lower dose rates was thought to accelerate the degradation of the macroscopic performance of the SMEP.Longyan Hou Yiyong Wu Debin Shan Bin Guo Yingying Zong 2021Journal of Materials Science & Technology2021,,8:1
