| 2 | Expression of miR-125b in the new,highly invasive glioma stem cell and progenitor cell line SU3显示文摘MicroRNA (miR)-125b has been shown to play a potential role in the development of glioma stem cells.However,the relationship between miRNA and glioma stem cells is still elusive.This study was designed to elucidate this potential relationship.We established a highly invasive glioma stem cell and progenitor (GSCP) cell line SU3.SU3 cell suspensions were injected into nude mice brains in situ,and the invasiveness of graft tumors was analyzed using hematoxylin and eosin staining as well as immunohistochemistry.Real-time polymerase chain reaction (PCR) was used to measure the expression levels of miR-125b in SU3 and other cells.In vitro,SU3 cells expressed CD133 and nestin as well as differentiation markers glial fibrillary acidic protein (GFAP) and β-tubulin III,which were consistent with the characteristics of glioma stem cells.Scratch assays indicated that the migration ability of SU3 cells was stronger than that of U251 stem cells (U251s).In vivo,SU3 cells invaded into each part of the mouse brain from the caudate nucleus in a diffuse pattern and highly expressed invasive and proliferative cell markers matrix metalloprotease 2 (MMP2),MMP9,and Ki-67.Real-time PCR results revealed that the levels of miR-125b and MMP9 were significantly higher in SU3 and SU2,also a highly invasive GSCP cell line we established before,than in U251s.High expression of miR-125b both in newly established GSCPs,SU3,and long-term cultured GSCPs,SU2 suggests that miR-125b exhibits oncogene-like behavior.This behavior should be considered in further studies of miR-125b in cancer stem cells.Furthermore,MMP9,which plays a role in cancer stem cell invasion,may be a target gene of miR-125b. | Yi Wan Xi-Feng Fei Zhi-Min Wang Dong-Yi Jiang Han-Chun Chen Jian Yang Lei Shi Qiang Huang | 2012 | Chinese Journal of Cancer2012,31,4: | 25 |

| 4 | Nomogram to predict overall survival after gallbladder cancer resection in China显示文摘AIM To integrate clinically significant variables related to prognosis after curative resection for gallbladder carcinoma(GBC) into a predictive nomogram.METHODS One hundred and forty-two GBC patients who underwent curative intent surgical resection at Peking Union Medical College Hospital(PUMCH) were included. This retrospective case study was conducted at PUMCH of the Chinese Academy of Medical Sciences and Peking Union Medical College(CAMS & PUMC) in China from January 1, 2003 to January 1, 2018. The continuous variable carbohydrate antigen 19-9(CA19-9) was converted into a categorical variable(cCA19-9) based on the normal reference range. Stages 0 to IIIA were merged into one category, while the remaining stages were grouped into another category. Pathological grade X(GX) was treated as a missing value. A multivariate Cox proportional hazards model was used to select variables to construct a nomogram. Discrimination and calibration of the nomogram were performed via the concordance index(C-index) and calibration plots. The performance of the nomogram was estimated using the calibration curve. Receiver operating characteristic(ROC) curve analysis and decision curve analysis(DCA) were performed to evaluate the predictive accuracy and net benefit of the nomogram, respectively.RESULTS Of these 142 GBC patients, 55(38.7%) were male, and the median and mean age were 64 and 63.9 years, respectively. Forty-eight(33.8%) patients in this cohort were censored in the survival analysis. The median survival time was 20 months. A series of methods, including the likelihood ratio test and Akaike information criterion(AIC) as well as stepwise, forward, and backward analyses, were used to select the model, and all yielded identical results. Jaundice [hazard ratio(HR) = 2.9; 95% confidence interval(CI): 1.60-5.27], cCA19-9(HR = 3.2; 95%CI: 1.91-5.39), stage(HR = 1.89; 95%CI: 1.16-3.09), and resection(R)(HR = 2.82; 95%CI: 1.54-5.16) were selected as significant predictors and combined into a survival time predictive nomogram(C-index = 0.803; 95%CI: 0.766-0.839). High prediction accuracy(adjusted C-index = 0.797) was further verified via bootstrap validation. The calibration plot demonstrated good performance of the nomogram. ROC curve analysis revealed a high sensitivity and specificity. | 9 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. | 11 | Seroepidemiological Investigation of Lyme Disease and Human Granulocytic Anaplasmosis among People Living in Forest Areas of Eight Provinces in China显示文摘Objective Lyme disease and Human granulocytic anaplasmosis are tick‐borne diseases caused by Borrelia burgdorferi and Anaplasma phagocytophilum respectively. We have investigated infection and co-infection of the two diseases in the population of forest areas of eight provinces in China by measuring seroprevalence of antibodies against B. burgdorferi and A. phagocytophilum. Methods Forest areas in 8 provinces were chosen for investigation using whole sampling and questionnaire survey methods. 3 669 serum samples from people in the forest areas were tested for the presence of antibodies by indirect immunofluorescent assay (IFA). Results Seroprevalence against B. burgdorferi was 3% to 15% and against A. phagocytophilum was 2% to 18% in the study sites in the 8 provinces in China. We also found co-infection of B. burgdorferi and A. phagocytophilum in 7 of the 8 provinces (the exception being the Miyun area in Beijing). The seroprevalence for both B. burgdorferi and A. phagocytophilum was significantly higher among people exposed to ticks than among people who were not exposed to ticks. In this study, one-step pyrolysis method was introduced to prepare porous biochar from kapok fibers, and effect of pyrolysis temperature on structure and electrochemical performance of the porous biochar was investigated. It was found that pyrolysis temperature played an important role in determining microstructure of the biochar. At the pyrolysis temperature of 750℃, the as-prepared biochar(C_(KF)-750) represented a largest specific surface area of 1125.7 m2 g^(-1) and pore volume of 0.7130 m^3 g^(-1), and hence brings C_(KF)-750 a highest specific capacitance of 283 Fg^(-1) at a current density of 1 A g^(-1) in a 6 mol L^(-1) KOH electrolyte. Furthermore, the cycle stability of C_(KF)-750 was wonderful,and the specific capacitance retained almost constant after 10000 cycles. | 16 | Using approximate dynamic programming for multi-ESM scheduling to track ground moving targets显示文摘This paper researches the adaptive scheduling problem of multiple electronic support measures(multi-ESM) in a ground moving radar targets tracking application. It is a sequential decision-making problem in uncertain environment. For adaptive selection of appropriate ESMs, we generalize an approximate dynamic programming(ADP) framework to the dynamic case. We define the environment model and agent model, respectively. To handle the partially observable challenge, we apply the unsented Kalman filter(UKF) algorithm for belief state estimation. To reduce the computational burden, a simulation-based approach rollout with a redesigned base policy is proposed to approximate the long-term cumulative reward. Meanwhile, Monte Carlo sampling is combined into the rollout to estimate the expectation of the rewards. | 18 | Human adenovirus(HAdV)infection in children with acute respiratory tract infections in Guangzhou,China,2010–2021:a molecular epidemiology study显示文摘Background Human adenovirus(HAdV)infection can cause a variety of diseases.It is a major pathogen of pediatric acute respiratory tract infections(ARIs)and can be life-threatening in younger children.We described the epidemiology and subtypes shifting of HAdV among children with ARI in Guangzhou,China.Methods We conducted a retrospective study of 161,079 children diagnosed with acute respiratory illness at the Guangzhou Women and Children’s Medical Center between 2010 and 2021.HAdV specimens were detected by real-time PCR and the hexon gene was used for phylogenetic analysis.Results Before the COVID-19 outbreak in Guangzhou,the annual frequency of adenovirus infection detected during this period ranged from 3.92%to 13.58%,with an epidemic peak every four to fve years.HAdV demonstrated a clear seasonal distribution,with the lowest positivity in March and peaking during summer(July or August)every year.A signifcant increase in HAdV cases was recorded for 2018 and 2019,which coincided with a shift in the dominant HAdV subtype from HAdV-3 to HAdV-7.The latter was associated with a more severe disease compared to HAdV-3.The average mortality proportion for children infected with HAdV from 2016 to 2019 was 0.38%but increased to 20%in severe cases.After COVID-19 emerged,HAdV cases dropped to 2.68%,suggesting that non-pharmaceutical interventions probably reduced the transmission of HAdV in the community.Conclusion Our study provides the foundation for the understanding of the epidemiology of HAdV and its associated risks in children in Southern China. | Yi Chen Tao Lin Chang‑Bing Wang Wan‑Li Liang Guang‑Wan Lian Mark Zanin Sook‑San Wong Xin‑Gui Tian Jia‑Yu Zhong Ying‑Ying Zhang Jia‑Hui Xie Ling‑Ling Zheng Fei‑Yan Chen Run Dang Ming‑Qi Zhao Yi‑Yu Yang Rong Zhou Bing Zhu | 2022 | World Journal of Pediatrics2022,18,8: | 5 |
| 19 | Clinical guidelines of non-alcoholic fatty liver disease: A systematic review显示文摘AIM To perform a systematic review to grade guidelines and present recommendations for clinical management of non-alcoholic fatty liver disease(NAFLD).METHODS A database search was conducted on Pub Med for guidelines published before May 2016, supplemented by reviewing relevant websites. The Appraisal of Guidelines for Research and Evaluation(ARGEE) instrument Ⅱ was a tool designed to appraise the methodological rigor and transparency in which a clinical guideline is developed and it is used internationally. it was used to appraise the quality of guidelines in this study. The inclusion criteria include: clinical NAFLD guidelines for adults, published in English, and released by governmental agencies or key organizations.RESULTS Eleven guidelines were included in this study. Since 2007, guidelines have been released in Asia(3 in China, 1 in South Korea, and 1 in Japan), Europe(1 in italy),America(1 in United States and 1 in Chile) and three international agencies [European associations joint, Asia-Pacific Working Party and World Gastroenterology Organization(WGO)]. Using the ARGEE Ⅱ instrument, we found US 2012 and Europe 2016 had the highest scores, especially in the areas of rigor of development and applicability. Additionally, italy 2010 and Korea 2013 also presented comprehensive content, rigorous procedures and good applicability. And WGO 2014 offered various algorithms for clinical practice. | 20 | Accelerated Learning for Restricted Boltzmann Machine with a Novel Momentum Algorithm显示文摘We investigated two commonly used momentum algorithms, Classical momentum(CM) and Nesterov momentum(NM). We found that, when used in Restricted Boltzmann machine(RBM), they have two main problems: The first one is their performances are not obvious and not as good as expected. The second one is they may lose accelerating ability in the later stage of training process. Aiming at these two problems, we proposed the Weight momentum algorithm and evaluated our approach on four datasets.
