| 1 | Effect of L-NAME on nitric oxide and gastrointestinal motility alterations in cirrhotic rats显示文摘AIM: To invsstigare the effect of L-NAME on nitric oxide andgastriubtestubal motility alterations in cirrhotic ratsMETHODS: Rats with cirrhosis induced by carbontetrachloride were randomly divided into two groups, one( n= 13) receiving 0. 5 mg@ kg-1 per clay of NG-nitro-L-argininemethyl ester (L-NAME), a nitric oxide synthase inhibitor,for 10 days, whereas the other group ( n = 13) and control( n = 10) rats were administrated the same volume of 9 g@ L-1saline.Half gastric emptying time and 2 h residual rate weremeasured by SPECT, using 99m Tc-DTPA-labeled bariumsuifate as test meal. Gastrointestinal transition time wasrecorded simultaneously. Serum concentration of nitrcoxide (NO) was determined by the kinetic cadmiunreduction and colorimetric methods. ImmunohistochemicalSABC method was used to observe the expression anddistribution of three types of nitric oxide synthase (NOS)isoforms in the mt gastrointestinal tract. Western blot wasused to detect expression of gastrointestinal NOS isoforms.RESULTS: Half gastric emptying time and trans-gastrointestinal time were significantly prolonged( 124.0 ± 26.4min; 33.7± 8.9min;72.1 ± 15.3 min; P<0.01), (12.4±0.5h; 9.5±0.3 h; 8.2±0.8 h; P<0.01), 2h residual rate wasraised in cirrhotic rots than in controls and cirrhotic ratstreated with L-NAME(54.9± 7.6 % ,13.7 ± 3.2 %, 34.9± 10.3%, P< 0.01). Serum concentration of NO was significantlyincreased in cirrhotic rots than in the other groups (8.20 ± 2.48)μmol@L-1, (5.94± 1.07) μmol@L-1 ,and control (5.66± 1.60) tμmol@L-1, P< 0.01. NOS staining intensities which weremainly located in the gastrointestinal tissues were markedlylower in cirrhotic rats than in the controls and cirrhotic ratsafter treated with L- NAME.CONCLUSION: Gastrointestinal motility was remarkablyinhibited in cirrhotic rats, which could he alleviated by L-NAME. | 3 | A major pathway for carbon and nitrogen losses-Gas emissions during storage of solid pig manure in China显示文摘This study investigated the carbon(C) and nitrogen(N) gas emissions(N_2O,NH_3,CO_2 and CH_4) from solid pig manure management in China.Gas emissions were quantified from static piles over 60 days during summer in China's Yangtze River Basin,using Drager-Tube and static chamber-gas chromatography techniques.High emissions of NH_3 and N_2 O were observed at the early stage of storage,but high emission of CH_4 occured later during storage.Overall,62% of the total C in the original pile was lost; CO_2 and CH_4 emissions accounted for 57 and 0.2% of C lost respectively.Over the same time,41% of the total N in the original pile was lost; NH_3 and N_2 O emissions accounted for 15 and 0.3% of N lost respectively.The volatilization of NH_3 during storage in summer was 4.56 g NH_3 per kg dry weight.The total greenhouse gas(GHG) emissions during storage accounted for 67.93 g CO_2 equivalent per kg dry weight; N_2 O and CH_4 contributed to 46 and 55% of total GHG emissions respectively.Given China's major role in pig production,further attention should given to pig manure management to mitigate its contribution to atmospheric pollution. | SHAN Nan LI Hu LI Jian-zheng Ee Ling Ng MA Yan WANG Li-gang CHEN Qing | 2019 | Journal of Integrative Agriculture2019,18,1: | 3 |
| 4 | Protein and RNA expression of acid-sensing ion channels 2 and 3 in myocardial ischemia rats induced by isoproterenol显示文摘OBJECTIVE:To investigate the impact of electro-acupuncture at the Neiguan(PC 6) acupoint on protein and RNA expression of acid-sensing ion channel 2(ASIC2) and ASIC3 in myocardial ischemia rats.METHODS:Fifty male Sprague-Dawley rats were used,weighing(230 ± 50) g.The rats were randomized into a normal group A,model group B,Neiguan(PC 6) group C,Lieque(LU 7) group D,and A-shi points group E.There were 10 rats in each group.Rats were continuously administered 85 mg/kg intravenous isoproterenol daily to establish the model.Successfully modeled rats in groups C,D,and E were given electro-acupuncture treatment.Each group of rats was sacrificed with chloral hydrate(1 mL/100 g) intraperitoneal injection.The left ventricular myocardium was extracted and placed at- 70 ℃ until use.Western blot analysis and real-time PCR were performed to assay protein and RNA expressions of ASIC2 and ASIC3,respectively.Fold changes in RNA expression were quantified with the 2~^(-△△Ct) method.Blood samples were drawn from the aorta abdominalis and tested for creatine kinase-MB(CK-MB) and lactate dehydrogenase(LDH) levels using enzyme-linked immunosorbent assay.RESULTS:Myocardial ischemia rats given electro-acupuncture at the Neiguan(PC 6) acupoint had significantly lower protein and RNA expression of ASIC2 and ASIC3,and CK-MB and LDH levels,compared with model rats(P < 0.01).CONCLUSION:Electro-acupuncture at the Neiguan(PC 6) acupoint can not only decrease the protein and RNA expression of ASIC2 and ASIC3,but also inhibit the opening of ASICs and reduce the cardiomyocyte damage in myocardial ischemia rats. | Wang Shudong Zhang Lide Dong Baoqiang Zhang Wenshun Ho Chin Ee Guo Na Chen Yiguo | 2015 | Journal of Traditional Chinese Medicine2015,35,2: | 2 |
