2Study of charge structure and radiation characteristic of intracloud discharge in thunderstorms of Qinghai-Tibet Plateau显示文摘The comprehensive observations on lightning discharges were conducted in Naqu area of Qinghai-Tibet Plateau in summer of 2002. The electric structures of thunderstorms and the characteristics of lightning discharges at initial stage were analyzed by using the observation data. The results show that most of intracloud (IC) lightning flashes were polarities inverted in thunderstorms with tripole electric charge structure and occurred between negative charge region located in the middle of the thunderstorm and positive charge region located at the bottom of the thunderstorm. The radiation characteristics of discharge processes in cloud with longer lasting time involved in Cloud-to-Ground (CG) lightning flashes were similar to that of IC discharges.A lot of radiation pulses were produced in these discharge processes. 3Increased Vitamin C Content Accompanied by an Enhanced Recycling Pathway Confers Oxidative Stress Tolerance in Arabidopsis显示文摘Vitamin C(L-ascorbic acid,AsA) has important antioxidant and metabolic functions in both plants and animals.Once used,ascorbic acid can be regenerated from its oxidized form in a reaction catalyzed by dehydroascorbate reductase(DHAR,EC 1.8.5.1).To analyze the physiological role of DHAR catalyzing the reduction of DHA to ascorbate in environmental stress adaptation,we examined whether increasing the level of AsA through enhanced AsA recycling would limit the deleterious effects of oxidative stress.A chimeric construct consisting of the double CaMV35S promoter fused to the Myc-dhar gene was introduced into Arabidopsis thaliana.Transgenic plants were biochemically characterized and tested for responses to oxidative stress.Western blot indicated that the dhar-transgene was successfully expressed.In homozygous T4 transgenic seedlings,DHAR overexpression was increased up to 1.5 to 5.4 fold,which enhanced foliar ascorbic acid levels 2-to 4.25-fold and ratio of AsA/DHA about 3-to 16-fold relative to wild type.In addition,the level of glutathione,the reductant used by DHAR,also increased as did its redox state.When whole plants were treated with high light and high temperature stress or in vitro leaf discs were subjected to 10 μM paraquat,transgenic plants showed a larger AsA pool size,lower membrane damage,and a higher level of chlorophyll compared with controls.These data suggested that increasing the plant vitamin C content through enhanced ascorbate recycling could limit the deleterious effects of environmental oxidative stress.zinan wang Ying Xiao Wansheng Chen Kexuan Tang Lei Zhang 2010Journal of Integrative Plant Biology2010,52,4:16
4Forecasting Different Types of Convective Weather: A Deep Learning Approach显示文摘A deep learning objective forecasting solution for severe convective weather(SCW) including short-duration heavy rain(HR), hail, convective gusts(CG), and thunderstorms based on numerical weather prediction(NWP) data was developed. We first established the training datasets as follows. Five years of severe weather observations were utilized to label the NCEP final(FNL) analysis data. A large number of labeled samples for each type of weather were then selected for model training. The local temperature, pressure, humidity, and winds from 1000 to 200 h Pa, as well as dozens of convective physical parameters, were taken as predictors in our model. A six-layer convolutional neural network(CNN) model was then built and trained to obtain optimal model weights. After that, the trained model was used to predict SCW based on the Global Forecast System(GFS) forecast data as input. The performances of the CNN model and other traditional methods were compared. The results show that the deep learning algorithm had a higher classification accuracy on HR and hail than support vector machine, random forests, and other traditional machine learning algorithms. The objective forecasts by use of the deep learning algorithm also showed better forecasting skills than the subjective forecasts by the forecasters. The threat scores(TSs) of thunderstorm, HR, hail, and CG were increased by 16.1%, 33.2%, 178%, and 55.7%, respectively. 5Broadband interferometer observations of a triggered lightning显示文摘The development of positive leader of an artificially triggered lightning has been analyzed based on the data of electric field change, location of radiation source and frequency spectrum obtained by using the broadband interferometer system. The results indicate that radiation from positive leader could be detected within close distance in spite of the relatively weak radiation, while the radiation from negative breakdown processes was relatively stronger. Positive leader developed with few branches, and the initial progression velocity was of the order of 105 m/s. 7Bidirectional propagation of lightning leader显示文摘The characteristics of initial leader of triggered lightning flashes are studied by use of a highspeed digital camera system with the rate of 1000 frame per second and simultaneous measurements of current and electric field change with 0.1 μs time resolution. The results show that the altitude-triggered lightning was initiated by bidirectional propagation leader because of 'current interruption' by air gap,whereas the grounded-triggered one was not. The onset of downward-moving positive leader was prior to that of the upward-going negative leader for 843 ms in an altitude-triggered lightning under the positive ambient electric field. 8Xiuyan crater, China: Impact origin confirmed显示文摘The well-preserved 1.8-km-diameter Xiuyan crater is located in the low mountain-hill region of the northern part of Liaodong Peninsula of northern China. Recently, a 307-m-deep borehole at the centre of crater became available. After penetrating 107 m lacustrine sediments, a breccia lens about 188 m in thickness was encountered. The crater-fill breccia is deposits of rock clasts and fragments more or less shock-metamorphosed. 10Highly stable full-color display device with VLC application potential using semipolarμLEDs and all-inorganic encapsulated perovskite nanocrystal显示文摘A promising approach for the development of effective full-color displays is to combine blue micro LEDs(μLEDs)with color conversion layers.Perovskite nanocrystals(PNCs)are notable for their tolerance to defects and provide excellent photoluminescence quantum yields and high color purity compared to metal chalcogenide quantum dots.The stability of PNCs in ambient conditions and under exposure to blue light can be improved using a SiO;coating.This study proposes a device that could be used for both display and visible light communication(VLC)applications.The semipolar blueμLED array fabricated in this study shows a negligible wavelength shift,indicating a significant reduction in the quantum confined Stark effect.Owing to its shorter carrier lifetime,the semipolar μLED array exhibits an impressive peak 3 dB bandwidth of 655 MHz and a data transmission rate of1.2 Gb/s corresponding to an injection current of 200 m A.The PNC-μLED device assembled from a semipolarμLED array with PNCs demonstrates high color stability and wide color-gamut features,achieving 127.23% and 95.00%of the National Television Standards Committee standard and Rec.2020 on the CIE 1931 color diagram,respectively.These results suggest that the proposed PNC-μLED device is suitable for both display-related and VLC applications.TINGZHU WU YUE LIN YU-MING HUANG MENG LIU KONTHOUJAM JAMES SINGH WANSHENG LIN TINGWEI LU XI ZHENG JIANYANG ZHOU HAO-CHUNG KUO ZHONG CHEN 2021Photonics Research2021,9,11:5

14Genome-wide profiling of RNA editing sites in sheep显示文摘Background: The widely observed RNA-DNA differences(RDDs) have been found to be due to nucleotide alteration by RNA editing. Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an individual.To date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen tissues.Results: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of genes.Conclusions: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. 15The transcription factors TLR1 and TLR2 negatively regulate trichome density and artemisinin levels in Artemisia annua显示文摘The important antimalarial drug artemisinin is biosynthesized and stored in Artemisia annua glandular trichomes and the artemisinin content correlates with trichome density;however,the factors affecting trichome development are largely unknown.Here,we demonstrate that the A.annua R2R3 MYB transcription factor TrichomeLess Regulator 1(TLR1)negatively regulates trichome development.In A.annua,TLR1 overexpression lines had 44.7%–64.0%lower trichome density and 11.5%–49.4%lower artemisinin contents and TLR1-RNAi lines had 33%–93.3%higher trichome density and 32.2%–84.0%higher artemisinin contents compared with non-transgenic controls.TLR1 also negatively regulates the expression of anthocyanin biosynthetic pathway genes in A.annua.When heterologously expressed in Arabidopsis thaliana,TLR1 interacts with GLABROUS3a,positive regulator of trichome development,and represses trichome development.Yeast two-hybrid and pull-down assays indicated that TLR1 interacts with the WUSCHEL homeobox(WOX)protein AaWOX1,which interacts with the LEAFY-like transcription factor TLR2.TLR2 overexpression in Arabidopsis and A.annua showed that TLR2 reduces trichome development by reducing gibberellin levels.Furthermore,artemisinin contents were 19%–43%lower in TLR2-overexpressing A.annua plants compared to controls.These data indicate that TLR1 and TLR2 negatively regulate trichome density by lowering gibberellin levels and may enable approaches to enhance artemisinin yields.Zongyou Lv JinXing Li Shi Qiu Fei Qi Hang Su Qitao Bu Rui Jiang Kexuan Tang Lei Zhang Wansheng Chen 2022Journal of Integrative Plant Biology2022,64,6:2
