1FoxM1 overexpression promotes epithelial-mesenchymal transition and metastasis of hepatocellular carcinoma显示文摘AIM: To investigate the expression of forkhead box protein M1(Fox M1) in the process of epithelial mesenchymal transition in hepatocellular carcinoma(HCC) and its role in metastasis.METHODS: Fox M1 and E-cadherin expression in HCC tissue microarray specimens was evaluated by immunohistochemical staining,and statistical methods were applied to analyze the correlation between FoxM 1 and epithelial-mesenchymal transition(EMT).KaplanMeier analysis of the correlation between the Fox M1 expression level and recurrence or overall survival of HCC patients was performed.The expression of FoxM 1,E-cadherin and snail homologue 1(SNAI1) in HCC cell lines was evaluated by real-time reverse transcriptionpolymerase chain reaction and Western blot.Hepatocyte growth factor(HGF) was used to induce EMT and stimulate cell migration in HCC cells.The expression of Fox M1 and SNAI1 was regulated by transfection with plasmids pc DNA3.1 and si RNAs in vitro.The occurrence of EMT was evaluated by Transwell assay,morphologic analysis and detection of the expression of EMT markers(E-cadherin and vimentin).Luciferase and chromatin immunoprecipitation assays were used to evaluate whether SNAI1 is a direct transcriptional target of FoxM 1.RESULTS: FoxM 1 expression was increased significantly in HCC compared with para-carcinoma(10.7 ± 0.9 vs 8.2 ± 0.7,P < 0.05) and normal hepatic(10.7 ± 0.9 vs 2.7 ± 0.4,P < 0.05) tissues.Overexpression of Fox M1 was correlated with HCC tumor size,tumor number,macrovascular invasion and higher TNM stage,but was negatively correlated with E-cadherin expression in microarray specimens and in cell lines.Fox M1 overexpression was correlated significantly with HCC metastasis and EMT.In vitro,we found that FoxM 1 plays a key role in HGF-induced EMT,and overexpression of Fox M1 could suppress E-cadherin expression and induce EMT changes,which were associated with increased HCC cell invasiveness.Next,we confirmed that FOXM1 directly binds to and activates the SNAI1 promoter,and we identified SNAI1 as a direct transcriptional target of FOXM1.Moreover,inhibiting the expression of SNAI1 significantly inhibited FoxM 1-mediated EMT.CONCLUSION: Fox M1 overexpression promotes EMT and metastasis of HCC,and SNAI1 plays a critical role in FoxM 1-mediated EMT.Fan-Di Meng Ji-Chao Wei Kai Qu Zhi-Xin Wang Qi-Fei Wu Ming-Hui Tai Hao-Chen Liu Rui-Yao Zhang Chang Liu 2015World Journal of Gastroenterology2015,21,1:17
2Quantification of Panax notoginseng saponins metabolites in rat plasma with in vivo gut microbiota-mediated biotransformation by HPLC-MS/MS显示文摘Panax notoginseng saponins(PNS) are the major components of Panax notoginseng, with multiple pharmacological activities but poor oral bioavailability. PNS could be metabolized by gut microbiota in vitro, while the exact role of gut microbiota of PNS metabolism in vivo remains poorly understood. In this study, pseudo germ-free rat models were constructed by using broad-spectrum antibiotics to validate the gut microbiota-mediated transformation of PNS in vivo. Moreover, a high performance liquid chromatography-electrospray ionization tandem mass spectrometry(HPLC-ESI-MS/MS) was developed for quantitative analysis of four metabolites of PNS, including ginsenoside F1(GF1), ginsenoside Rh2(GRh2), ginsenoside compound K(GCK) and protopanaxatriol(PPT). The results showed that the four metabolites could be detected in the control rat plasma, while they could not be determined in pseudo germ-free rat plasma. The results implied that PNS could not be biotransformed effectively when gut microbiota was disrupted. 3Characterization of disinfection byproduct formation potential in 13 source waters in China显示文摘The formation potential of four trihalomethanes (THMFP) and seven haloacetic acids (HAA 7 FP) in 13 source waters taken from four major water basin areas in China was evaluated using the simulated distribution system (SDS) chlorination method.The specific ultraviolet absorbance (SUVA 254:the ratio of UV 254 to dissolved organic carbon (DOC)),which ranged between 0.9 and 5.0 L/(mg·m),showed that the organic compounds in different source waters exhibited different reactivities with chlorine.The HAA 7 FP of source waters ranged from 20 to 448 μg/L and the THMFP ranged from 29 to 259 μg/L.The HAA 7 FP concentrations were higher than the THMFP concentrations in all but one of the samples.Therefore,the risks of haloacetic acids (HAAs) should be of concern in some source waters.TCM (chloroform) and BDCM (bromodichloromethane) were the major THM constituents,while TCAA (trichloroacetic acid) and DCAA (dichloroacetic acid) were the major HAA species.Br-THM (brominated THM species) were much higher than BrHAA (brominated HAA species),and the formation of Br-DBP (Br-THM and Br-HAA) should be of concern when the bromide concentration is over 100 μg/L.Junzhi Zhang Jianwei Yu Wei An Juan Liu Yongjing Wang Youjun Chen Jia Tai Min Yang 2011Journal of Environmental Sciences2011,23,2:13
4Pyogenic liver abscesses associated with nonmetastatic colorectal cancers: An increasing problem in Eastern Asia显示文摘AIM: To elaborate the clinicopathologic features of colorectal cancer-related pyogenic liver abscess (PLA). METHODS: Reported cases of colorectal cancer-related PLAs were collected from the literature published up to October 2011 and evaluated for their clinicopathologic features. Data of collected cases included demographics, clinical presentation, microbial findings and treatment. Categorical variables were compared by 2 analysis and continuous variables were evaluated using Student's t test. RESULTS: A total 96 cases of colorectal cancer-related PLA were collected from the previous literature. Most patients (60%) were male and 40% cases occurred in the age group of 61-70 years. Apart from some special types of PLA, there were significant differences in the microbiological spectrum between Eastern Asia and non-Eastern Asian countries, which implied different risk factors and courses of the disease. Gram negative bacteria especially Klebsiella pneumoniae (K. pneu- moniae ) PLA was predominant in Eastern Asia (80.0%) in contrast to non-Eastern Asian countries (P < 0.01). Meanwhile, most of the Eastern Asian patients exhibited smaller size of liver abscess and atypical presentation. Sigmoid colon and rectum (72.73%) were the main sites of tumor in Eastern Asian patients, whereas tumor sites were uneven among most of the non-Easter Asian PLA patients. 5Panax notoginseng saponins prevent colitis-associated colorectal cancer development: the role of gut microbiota显示文摘Gut microbiota dysbiosis is a risk factor for colorectal cancer(CRC) in inflammatory bowel disease(IBD).In this study, the effects of Panax notoginseng saponins(PNS) on colitis-associated CRC progression were evaluated on an azoxymethane(AOM)/dextran sulfate sodium(DSS) mouse model.In vivo, PNS significantly relieved AOM/DSS-induced colon tumorigenesis and development by reducing the disease activity index(DAI) scores and colon tumor load.The 16S rRNA data of fecal samples showed that the microbiome community was obviously destructed, while PNS could recover the richness and diversity of gut microbiota.Especially, PNS could increase the abundance of Akkermansia spp.which was significantly decreased in model group and negatively correlated with the progression of CRC.Moreover, ginsenoside compound K(GC-K) was evaluated on the effects of human CRC cells,which was the main bio-transformed metabolite of PNS by gut microbiota.Our data showed that PNS played important role in the prevention of the progression of CRC, due to their regulation on the microbiome balance and microbial bio-converted product with antiCRC activity.CHEN Ling CHEN Man-Yun SHAO Li ZHANG Wei RAO Tai ZHOU Hong-Hao HUANG Wei-Hua 2020Chinese Journal of Natural Medicines2020,18,7:12

7Physiological and Molecular Features of Puccinellia tenuiflora Tolerating Salt and Alkaline-Salt Stress显示文摘Saline-alkali soil seriously threatens agriculture productivity; therefore, understanding the mechanism of plant tolerance to alkaline-salt stress has become a major challenge. Halophytic Puccinellia tenuiflora can tolerate salt and alkaline-salt stress, and is thus an ideal plant for studying this tolerance mechanism. In this study, we examined the salt and alkaline-salt stress tolerance of P. tenuiflora, and analyzed gene expression profiles under these stresses. Physiological experiments revealed that P. tenuiflora can grow normally with maximum stress under 600 mmol/L NaCl and 150 mmol/L Na 2 CO 3 (pH 11.0) for 6 d. We identified 4,982 unigenes closely homologous to rice and barley. Furthermore, 1,105 genes showed differentially expressed profiles under salt and alkaline-salt treatments. Differentially expressed genes were overrepresented in functions of photosynthesis, oxidation reduction, signal transduction, and transcription regulation. Almost all genes downregulated under salt and alkaline-salt stress were related to cell structure, photosynthesis, and protein synthesis. Comparing with salt stress, alkaline-salt stress triggered more differentially expressed genes and significantly upregulated genes related to H + transport and citric acid synthesis. 8Improved form of wind wave frequency spectrum显示文摘The lower frequency part of the theoretical wind wave spectrum proposed by the authors (Wen et al. , 1988a, b,c) has been improved and the form of spectrum is appreciably simplified. In addition to the field data collected in the Bohai Sea region and used in the previous papers, those obtained in the Huanghai Sea, the East China Sea and the South China Sea have been employed so that the improved spectra can be verified on a more extensive observational basis. Computed results agree with the observations well. Further comparisons have been made between the proposed spectra and the JONSWAP spectrum. Though the two types of spectrum are close to each other in form, the former shows, as a whole, better agreement with the observation than the latter. By introducing an improved relation between the peak-ness factor and significant wave steepness, the spectrum contains only significant wave height and period as parameters. 9Modified FOLFIRINOX for unresectable locally advanced or metastatic gallbladder cancer,a comparison with GEMOX regimen显示文摘Background:The first-line chemotherapy regimen for advanced gallbladder cancer(GBC)is gemcitabine plus platinum(GP),despite its efficacy is limited.The current investigation is a retrospective study to compare the safety and efficacy between the modified FOLFIRINOX(mFOLFIRINOX)and gemcitabine plus oxaliplatin(GEMOX)as the first-line chemotherapy for unresectable locally advanced or metastatic GBC.Methods:The data of patients with unresectable locally advanced or metastatic GBC,who were treated with mFOLFIRINOX or GEMOX as the first-line therapy between April 2014 and April 2018 at Xinhua Hospital Affiliated to Shanghai Jiaotong University School of Medicine,were retrieved.This retrospective study evaluated the clinical characteristics,survival outcomes and adverse events.Results:A total of 44 patients(n=25 in mFOLFIRINOX,n=19 in GEMOX)were included.There were no significant differences between groups in baseline characteristics.The median progression free survival(mPFS)was 5.0 months in the mFOLFIRINOX group and 2.5 months in the GEMOX group[P=0.021;hazard ratio(HR),0.499;95%CI,0.266 to 0.937].The median overall survival(mOS)was 9.5 months in the mFOLFIRINOX group and 7.0 months in the GEMOX group(P=0.019;HR,0.471;95%CI,0.239 to 0.929).Disease control rate(DCR)was 76.0%in the mFOLFIRINOX group and 47.4%in the GEMOX group(P=0.051).The rate of grade 3-4 adverse events was 48%in the mFOLFIRINOX group and 36.8%in the GEMOX group(P=0.459).The incidence of grade 3-4 neutropenia and diarrhea were more common in the mFOLFIRINOX group,while the incidence of grade 3-4 thrombocytopenia and peripheral neuropathy were more common in the GEMOX group.Conclusions:mFOLFIRINOX might improve the poor prognosis of unresectable locally advanced or metastatic GBC,and the results need to be further verified by prospective clinical studies.Xu-Ya Cui Xue-Chuan Li Jiu-Jie Cui Xiang-Song Wu Lu Zou Xiao-Ling Song Tai Ren Yi-Di Zhu Huai-Feng Li Yang Yang Ke Liu Xu-Sheng Han Zi-Yao Jia Wen-Guang Wu Xu-An Wang Wei Gong Li-Wei Wang Mao-Lan Li Ying-Bin Liu 2021Hepatobiliary Surgery and Nutrition2021,10,4:10
10TLR4-HMGB1-, MyD88- and TRIF-dependent signaling in mouse intestinal ischemia/reperfusion injury显示文摘AIM: To characterize high-mobility group protein 1-toll-like receptor 4(HMGB1-TLR4) and downstream signaling pathways in intestinal ischemia/reperfusion(I/R) injury.METHODS: Forty specific-pathogen-free male C57BL/6 mice were randomly divided into five groups(n = 8 per group): sham, control, anti-HMGB1, anti-myeloid differentiation gene 88(My D88), and anti-translocatingchain-associating membrane protein(TRIF) antibody groups. Vehicle with the control Ig G antibody, antiHMGB1, anti-My D88, or anti-TRIF antibodies(all 1 mg/kg, 0.025%) were injected via the caudal vein 30 min prior to ischemia. After anesthetization, the abdominal wall was opened and the superior mesenteric artery was exposed, followed by 60 min mesenteric ischemia and then 60 min reperfusion. For the sham group, the abdominal wall was opened for 120 min without I/R. Levels of serum nuclear factor(NF)-κB p65, interleukin(IL)-6, and tumor necrosis factor(TNF)-α were measured, along with myeloperoxidase activity in the lung and liver. Inaddition,morphologic changes that occurred in the lung and intestinal tissues were evaluated. Levels of m RNA transcripts encoding HMGB1 and NF-κB were measured by real-time quantitative PCR, and levels of HMGB1 and NF-κB protein were measured by Western blot. Results were analyzed using one-way analysis of variance.RESULTS: Blocking HMGB 1, MyD 8 8, and TRIF expression by injecting anti-HMGB1, anti-My D88, or anti-TRIF antibodies prior to ischemia reduced the levels of inflammatory cytokines in serum; NF-κB p65: 104.64 ± 11.89, 228.53 ± 24.85, 145.00 ± 33.63, 191.12 ± 13.22, and 183.73 ± 10.81(P < 0.05); IL-6: 50.02 ± 6.33, 104.91 ± 31.18, 62.28 ± 6.73, 85.90 ± 17.37, and 78.14 ± 7.38(P < 0.05); TNF-α, 43.79 ± 4.18, 70.81 ± 6.97, 52.76 ± 5.71, 63.19 ± 5.47, and 59.70 ± 4.63(P < 0.05) for the sham, control, anti-HMGB1, anti-My D88, and anti-TRIF groups, respectively(all in pg/m L).Antibodies also alleviated tissue injury in the lung and small intestine compared with the control group in the mouse intestinal I/R model. 11BAFF, involved in B cell activation through the NF-κB pathway, is related to disease activity and bone destruction in rheumatoid arthritis显示文摘B cell activating factor of TNF family(BAFF)is a member of TNF ligand superfamily and plays a key role in B cell homeostasis,proliferation,maturation,and survival.In this study,we detected BAFF level,the expressions of BAFF receptors and important molecules in NF-κB pathway in rheumatoid arthritis(RA)patients and analyzed the correlation between BAFF level and clinical variables,laboratory parameters or X-ray scores in order to elucidate the roles of BAFF in RA.A total of 50 RA patients and 50 healthy controls(HCs)were enrolled.We showed that the serum BAFF level in RA patients was significantly higher than that of HCs,and the percentages of B cell subsets(CD19^(+)B cells,CD19^(+)CD27^(+)B cells,CD19^(+)CD20^(+)CD27^(+)B cells,and CD19^(+)CD20−CD27^(+)B cells)in the serum of RA patients were significantly increased compared with those of HCs.The percentages of CD19^(+)BAFFR^(+)B cells,CD19^(+)BCMA^(+)B cells,and CD19^(+)TACI^(+)B cells in RA patients were significantly increased compared with those in HCs.The expression of important molecules in the NF-κB pathway(MKK3,MKK6,p-P38,p-P65,TRAF2,and p52)was significantly higher in RA patients than in HCs,but p100 level in RA patients was lower than that in HCs.The serum BAFF level was positively correlated with C-reactive protein,rheumatoid factor,disease activity score(in 28 joints),swollen joint counts,tender joint counts,and X-ray scores.When normal B cells were treated with BAFF in vitro,the percentages of the B cell subset and the expression of BAFF receptors were significantly upregulated.BAFF also promoted the expression of MKK3,MKK6,p-P38,p-P65,TRAF2,and p52.In conclusion,this study demonstrates that BAFF level is correlated with the disease activity and bone destruction of RA.BAFF is involved in the differentiation,proliferation,and activation of B cells in RA through NF-κB signaling pathway,suggesting that BAFF might be an ideal therapeutic target for RA.Ling-ling Zhang Hui Xiao Feng Zhang Yu-jing Wu Jin-ling Shu Ying Li Yu Tai Sheng-qian Xu Jian-hua Xu Wei Wei 2021Acta Pharmacologica Sinica2021,42,10:9
12Ultrafine particle emission characteristics of diesel engine by on-board and test bench measurement显示文摘This study investigated the emission characteristics of ultrafine particles based on test bench and on-board measurements. The bench test results showed the ultrafine particle number concentration of the diesel engine to be in the range of (0.56-8.35)×10 8cm-3. The on-board measurement results illustrated that the ultrafine particles were strongly correlated with changes in real-world driving cycles. The particle number concentration was down to 2.0×106cm-3 and 2.7×107cm-3 under decelerating and idling operations and as high as 5.0×108cm-3 under accelerating operation. It was also indicated that the particle number measured by the two methods increased with the growth of engine load at each engine speed in both cases. The particle number presented a 'U' shaped distribution with changing speed at high engine load conditions, which implies that the particle number will reach its lowest level at medium engine speeds. The particle sizes of both measurements showed single mode distributions. The peak of particle size was located at about 50-80nm in the accumulation mode particle range. 13Lignin biosynthesis by suppression of two O-methyl-transferases显示文摘Caffeic acid O-methyltransferase (COMT) and caffeoyl-CoA-3-O-methyltransferase (CCoAOMT) genes encode two methyltransferases at different substrate levels in lignin biosynthesis. We constructed two single antisense expression vectors containing the cDNA of COMT or CCoAOMT genes and one dual antisense expression vector containing cDNAs of both OMTs genes. The antisense constructs were transferred into tobacco mediated by Agrobac-terium tumefacience. PCR-Southern analysis indicated that antisense cDNAs had been integrated into the genome of the transgenic tobacco. The antisense genes were also expressed at transcriptional level displayed by Northern dot analysis. Klason lignin assay of 3-month-old transgenic tobaccos showed that repression in COMT or CCoAOMT alone could result in reduction of lignin content, more reduction was caused by suppression in CCoAOMT than in COMT. 14Genetic aberration in primary hepatocellular carcinoma:correlation between p53 gene mutation and loss-of-heterozygosity on chromosome 16q21-q23 and 9p21-p23显示文摘To elucidate the molecular pathology underlying the development of hepatocellular carcinoma (HCC), we used 41 highly polymorphic microsatellite markers to examine 55 HCC and corresponding non-tumor liver tissues on chromosome 9, 16 and 17. Loss-of-heterozygosity (LOH) is observed with high frequency on chromosomal region 17p13 (36/55, 65%), 9p21-p23 (28/55, 51%), 16q21-q23 (27/55, 49%) in tumors. Meanwhile, microsatellite instability is rarely found in these microsatellite loci. Direct sequencing was performed to detect the tentative mutation of tumor suppressor genes in these regions: p53, MTS1/p16, and CDH1/E-cadherin. Within exon 5-9 of p53 gene, 14 out of 55 HCC specimens (24%) have somatic mutations, and nucleotide deletion of this gene is reported in HCC for the first time. Mutation in MTS1/pl6 is found only in one tumor case. We do not find mutations in CDH1/E-cadherin. Furthermore, a statistically significant correlation is present between p53 gene mutation and loss of chromosome region 16q21q23 and 9p21-p23, which indicates that synergism between p53 inactivation and deletion of 16q21-q23 and 9p21-p23 may play a role in the pathogenesis of HCC. 15Gastric peritoneal carcinomatosis-a retrospective review显示文摘AIM To characterize patients with gastric peritoneal carcinomatosis(PC) and their typical clinical and treatment course with palliative systemic chemotherapy as the current standard of care.METHODS We performed a retrospective electronic chart review of all patients with gastric adenocarcinoma with PC diagnosed at initial metastatic presentation between January 2010 and December 2014 in a single tertiary referral centre.RESULTS We studied a total of 271 patients with a median age of 63.8 years and median follow-up duration of 5.1 mo. The majority(n = 217, 80.1%) had the peritoneum as the only site of metastasis at initial presentation. Palliative systemic chemotherapy was eventually planned for 175(64.6%) of our patients at initial presentation, of which 171 were initiated on it. Choice of first-line regime was in accordance with the National Comprehensive Cancer Network Guidelines for Gastric Cancer Treatment. Thesepatients underwent a median of one line of chemotherapy, completing a median of six cycles in total. Chemotherapy disruption due to unplanned hospitalizations occurred in 114(66.7%), while cessation of chemotherapy occurred in 157(91.8%), with 42 cessations primarily attributable to PC-related complications. Patients who had initiation of systemic chemotherapy had a significantly better median overall survival than those who did not(10.9 mo vs 1.6 mo, P < 0.001). Of patients who had initiation of systemic chemotherapy, those who experienced any disruptions to chemotherapy due to unplanned hospitalizations had a significantly worse median overall survival compared to those who did not(8.7 mo vs 14.6 mo, P < 0.001).CONCLUSION Gastric PC carries a grim prognosis with a clinical course fraught with disease-related complications which may attenuate any survival benefit which palliative systemic chemotherapy may have to offer. 16Paeoniflorin-6′-0-benzene sulfonate alleviates collageninduced arthritis in mice by downregulating BAFF-TRAF2-NF-kB signaling: comparison with biological agents显示文摘Paeoniflorin-6′-0-benzene sulfonate (CP-25) is a new ester derivative of paeoniflorin with improved lipid solubility and oral bioavailability, as well as better anti-inflammatory activity than its parent compound. In this study we explored whether CP-25 exerted therapeutic effects in collagen-induced arthritis (CIA) mice through regulating B-cell activating factor (BAFF)-BAFF receptors-mediated signaling pathways. CIA mice were given CP-25 or injected with biological agents rituximab or etanercept for 40 days. In CIA mice, we found that T cells and B cells exhibited abnormal proliferation;the percentages of CD19^+ total B cells, CD19^+CD27^+-activated B cells, CD19^+BAFFR^+ and CD19^+TACI^+cells were significantly increased in PBMCs and spleen lymphocytes. CP-25 suppressed the indicators of arthritis, alleviated histopathology, accompanied by reduced BAFF and BAFF receptors expressions, inhibited serum immunoglobulin levels, decreased the Bell subsets percentages, and prevented the expressions of key molecules in NF-kB signaling. Furthermore, we showed that treatment with CP-25 reduced CD19+TRAF2^+ cell expressions stimulated by BAFF and decreased TRAF2 overexpression in HEK293 cells in vitro. Thus, CP-25 restored the abnormal T cells proliferation and B-cell percentages to the normal levels, and normalized the elevated levels of IgA, lgG2a and key proteins in NF-kB signaling. In comparison, rituximab and etanercept displayed stronger antiinflammatory activities than CP-25;they suppressed the elevated inflammatory indexes to below the normal levels in CIA mice. In summary, our results provide evidence that CP-25 alleviates CIA and regulates the functions of B cells through BAFF-TRAF2-NF-kB signaling. 17Cloud point extraction and flame atomic absorption spectrometry analysis of palladium, platinum, and gold ions from industrial polluted soil显示文摘Cloud point extraction (CPE) with Tergitol TMN-6 was applied for the extraction of trace amounts of palladium (Pd(Ⅱ)), platinum (Pt(Ⅳ)), and gold (Au(Ⅲ)) in the soil of industrial sewage. Ammonium pyrolysine dithiocarbamate (APDC) was adopted as the chelating agent prior to CPE and then was detected by atomic absorption spectrometry (AAS). Different parameters such as the concentration of surfactants, chelating agent and salt, sample pH, equilibration temperature and time, centrifugation time and rates, and the effect of foreign ions were studied. Under optimum conditions, the low limits of detections are 1.4, 2.8 and 1.2 ng·ml^-1 and the enrichment factors are 21, 12, and 24 for Pd(Ⅱ), Pt(Ⅳ), and Au(Ⅲ, respectively. The relative standard deviations vary from 0.6% to 1.0% (n=11). All correlation coefficients of the calibration curves are >0.9960. 19Celecoxib attenuates hepatocyte apoptosis by inhibiting endoplasmic reticulum stress in thioacetamide-induced cirrhotic rats显示文摘BACKGROUND Endoplasmic reticulum(ER)stress is an important mechanism in the progression of chronic and acute liver diseases,especially in the progression and recovery of liver fibrosis.Excessive and long-term ER stress induces apoptosis.ER stressinduced apoptosis is considered to be an important pathway in the development of liver fibrosis.Cyclooxygenase-2(COX-2)induction is also closely related to ER stress.In our previous studies,we showed that celecoxib,a COX-2 inhibitor,improves liver fibrosis and portal hypertension.However,the role and mechanism of celecoxib in alleviating liver fibrosis remain unclear.AIM To investigate whether celecoxib alleviates liver fibrosis by inhibiting hepatocyte apoptosis via the ER stress response.METHODS Cirrhosis was induced by intraperitoneal injections of thioacetamide(TAA)for 16 wk(injection dose is 200 mg/kg per 3 d for the first 8 wk and 100 mg/kg per 3 d after 8 wk).Thirty-six male Sprague-Dawley rats were randomly divided into three groups,namely,control group,TAA group,and TAA+celecoxib group.In the last 8 wk,TAA-induced cirrhotic rats received celecoxib(20 mg/kg/day)or the vehicle by gastric gavage.After 16 wk,the rats were sacrificed,and serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),and albumin(ALB)were detected.The hepatic fibrosis areas were evaluated by Sirius red staining and the degree of fibrosis was assessed by measuring the level of hydroxyproline.ER stress levels were evaluated by detecting the marker proteins glucose-regulated protein 78(GRP78),CCAAT/enhancer binding protein homologous protein(CHOP),PKR-like ER protein kinase(PERK),activating transcription factor 6(ATF6),and inositol-requiring enzyme 1 alpha(IRE1α).Apoptosis levels were evaluated by detecting caspase-12 and caspase-3.RESULTS The serum ALT and AST levels in the liver were significantly reduced by celecoxib;however,the serum ALB had no significant changes.Celecoxib significantly reduced the degree of liver fibrosis and the levels of hydroxyproline(-38%and-25.7%,respectively,P<0.01).Celecoxib ameliorated ER stress by reducing the level of GRP78 compared to the TAA group(P<0.05).Consistently,after celecoxib administration,the upregulation of TAA-induced hepatic apoptosis markers(caspase-12 and caspase-3)and CHOP were significantly inhibited.In addition,after celecoxib treatment,the expression of key molecules associated with ER stress(PERK,ATF6,and IRE1)was decreased(P<0.05).CONCLUSION Therapeutic administration of celecoxib effectively reduces hepatic apoptosis in TAA-induced cirrhotic rats.The mechanism of action may be attributed to the suppression of CHOP expression,which subsequently inhibits ER stress.Wei Su Yang Tai Shi-Hang Tang Yan-Ting Ye Chong Zhao Jin-Hang Gao Bi-Guang Tuo Cheng-Wei Tang 2020World Journal of Gastroenterology2020,26,28:4
20Down-regulation of FoxM1 inhibits viability and invasion of gallbladder carcinoma cells, partially dependent on inducement of cellular senescence显示文摘AIM:To investigate the effect of knockdown of forkhead box M1(foxM1)on the proliferation and invasion capacities of human gallbladder carcinoma(Gbc)-sd cells.METHODS:four foxM1 shRNAs were transfected into Gbc-sd cells with lipofectamine 2000 to select the appropriate shRNA for down-regulation of foxM1.A recombinant lentivirus for shfoxM1(lv-shfoxM1),which expresses FoxM1-specific shRNA,and a negative control carrying green fluorescent protein,which expresses a scrambled RNA,were constructed.After transfection with the recombinant adenovirus and screened with puromycin,RT-PcR and Western blot were utilized to evaluate the inhibition efficiency.cell viability was evaluated by MTT assay,and cell migration and invasion were assessed using the Transwell system.cells were suspended in serum-free medium and seeded into Transwell inserts either uncoated(for migration assay)or coated(for invasion assay)with growth factor-reduced Matrigel.To verify the involvement of foxM1 in the senescence of tumor cells,staining of senescenceβ-galactosidase(sAβ-gal),the widely used biomarker of cellular senescence,was also performed.RESULTS:After successful transfection of four foxM1small interfering RNAs(shRNAs)with lipofectamine2000,the shf1822 was selected as the most appropriate shRNA according to its obvious inhibitory effect.The recombinant adenovirus was then constructed with the shf1822 and successfully transfected into the Gbcsd cells,resulting in the significant inhibition of foxM1expression at both the mRNA and protein levels,compared with the negative control(P<0.05).After transfection,down-regulation of foxM1 significantly inhibited cell viability according to the MTT assay(P<0.05).In addition,Transwell migration and invasion assays also suggested the suppression of invasion ability of the transfected cells.sAβ-gal staining showed that downregulation of foxM1 could induce more senescent Gbc cells(P<0.05),suggesting the possible involvement of the senescence process of the foxM1-deficient cells in Gbc.CONCLUSION:foxM1 is functionally involved in viability of Gbc cells,partially dependent on the inducement of cellular senescence,and is a potential target for Gbc therapy.Jie Tao Xin-sen Xu Yan-zhou song Kai Qu Qi-fei Wu Rui-Tao Wang fan-di Meng Ji-chao Wei shun-bin dong Yue-lang zhang Min-hui Tai Ya-feng dong Lin Wang Chang liu 2014World Journal of Gastroenterology2014,20,28:4
