| 1 | Manganese is critical for antitumor immune responses via cGAS-STING and improves the efficacy of clinical immunotherapy显示文摘CD8^+T cell-mediated cancer clearance is often suppressed by the interaction between inhibitory molecules like PD-1 and PD-L1,an interaction acts like brakes to prevent T cell overreaction under normal conditions but is exploited by tumor cells to escape the immune surveillance.Immune checkpoint inhibitors have revolutionized cancer therapeutics by removing such brakes.Unfortunately,only a minority of cancer patients respond to immunotherapies presumably due to inadequate immunity.Antitumor immunity depends on the activation of the cGAS-STING pathway,as STING-deficient mice fail to stimulate tumor-infiltrating dendritic cells(DCs)to activate CD8^+T cells.STING agonists also enhance natural killer(NK)cells to mediate the clearance of CD8^+T cell-resistant tumors.Therefore STING agonists have been intensively sought after.We previously discovered that manganese(Mn)is indispensable for the host defense against cytosolic dsDNA by activating cGAS-STING.Here we report that Mn is also essential in innate immune sensing of tumors and enhances adaptive immune responses against tumors.Mn-insufficient mice had significantly enhanced tumor growth and metastasis,with greatly reduced tumor-infiltrating CD8^+T cells.Mechanically,Mn^2+promoted DC and macrophage maturation and tumor-specific antigen presentation,augmented CD8^+T cell differentiation,activation and NK cell activation,and increased memory CD8^+T cells.Combining Mn^2+with immune checkpoint inhibition synergistically boosted antitumor efficacies and reduced the anti-PD-1 antibody dosage required in mice.Importantly,a completed phase 1 clinical trial with the combined regimen of Mn^2+and anti-PD-1 antibody showed promising efficacy,exhibiting type I IFN induction,manageable safety and revived responses to immunotherapy in most patients with advanced metastatic solid tumors.We propose that this combination strategy warrants further clinical translation. | Mengze Lv Meixia Chen Rui Zhang Wen Zhang Chenguang Wang Yan Zhang Xiaoming Wei Yukun Guan Jiejie Liu Kaichao Feng Miao Jing Xurui Wang Yun-Cai Liu Qian Mei Weidong Han Zhengfan Jiang | 2020 | Cell Research2020,30,11: | 27 |
| 2 | The Mechanism of Cortico-Striato-Thalamo-Cortical Neurocircuitry in Response Inhibition and Emotional Responding in Attention Deficit Hyperactivity Disorder with Comorbid Disruptive Behavior Disorder显示文摘The neurocircuitries that constitute the corticostriato-thalamo-cortical(CSTC) circuit provide a framework for bridging gaps between neuroscience and executive function in attention deficit hyperactivity disorder(ADHD), but it has been difficult to identify the mechanisms for regulating emotional problems from the understanding of ADHD comorbidity with disruptive behavior disorders(DBD). Research based on 'cool'' and 'hot''executive functional theory and the dual pathway models,which are thought of as applied response inhibition and delay aversion, respectively, within the neuropsychological view of ADHD, has shed light on emotional responding before and after decontextualized stimuli, while CSTC circuit-related domains have been suggested to explain the different emotional symptoms of ADHD with or without comorbid DBD. This review discusses the role of abnormal connections in each CSTC circuit, especially in the emotion circuit, which may be responsible for targeted executive dysfunction at the neuroscience level. | 3 | Water chemistry controlled aggregation and photo-transformation of silver nanoparticles in environmental waters显示文摘The inevitable release of engineered silver nanoparticles(Ag NPs) into aquatic environments has drawn great concerns about its environmental toxicity and safety. Although aggregation and transformation play crucial roles in the transport and toxicity of Ag NPs, how the water chemistry of environmental waters influences the aggregation and transformation of engineered Ag NPs is still not well understood. In this study, the aggregation of polyvinylpyrrolidone(PVP) coated Ag NPs was investigated in eight typical environmental water samples(with different ionic strengths, hardness, and dissolved organic matter(DOM) concentrations) by using UV–visible spectroscopy and dynamic light scattering. Raman spectroscopy was applied to probe the interaction of DOM with the surface of Ag NPs. Further, the photo-transformation and morphology changes of Ag NPs in environmental waters were studied by UV–visible spectroscopy, inductively coupled plasma mass spectrometry, and transmission electron microscopy. The results suggested that both electrolytes(especially Ca2+and Mg2+) and DOM in the surface waters are key parameters for Ag NP aggregation, and sunlight could accelerate the morphology change, aggregation, and further sedimentation of Ag NPs. | 4 | Radiofrequency ablation combined with transcatheter arterial chemoembolization therapy versus surgical resection for Barcelona-Clinic Liver Cancer(BCLC) A hepatocellular carcinoma: a meta-analysis显示文摘Purpose: The objective of our study was to compare the effectiveness of the combination of transcatheter arterial chemoembolization(TACE) and radiofrequency ablation(RFA) with that of surgical resection(SR) in Barcelona-Clinic Liver Cancer(BCLC) A hepatocellular carcinoma. Materials and Methods: PubMed, Medline, Embase, and Cochrane Library were searched for comparisons of the two therapies from January 2006 to December 2017. Overall survival rate(OS), recurrence-free survival rate(RFS), complications, and the average length of hospital stay were compared and analyzed. Review Manager v. 5.2 from the Cochrane Collaboration was used for statistical analyses. Results: Seven case-control studies and one randomized controlled trial were identified, of which 717 were treated with a combination of TACE and RFA and 785 were treated with SR. Meta-analysis data revealed that TACE plus RFA had significantly better effectiveness on 1.0-y OS(OR = 0.50, p =.009). The major complications(ORcomplications = 1.88, p =.02) after the combined therapy were significantly lower than those after SR. There were three studies that reported the average length of hospital stay. The hospital stay for the SR group vs the combined therapy group was 19.8 ± 8.4 d vs 7.4 ± 2.2 d, respectively(p <.0001); 18.7 ± 4.9 d vs 11.5 ± 6.9 d, respectively(p <.0001); and 16.6 ± 6.7 d vs 8.5 ± 4.1 d, respectively(p <.0001). There was no significant difference in 3.0-or 5.0-y OS and 1.0-, 3.0-, or 5.0-y RFS. | 6 | Microstructure and strengthening mechanism of hot-extruded ultralight Mg-Li-Al-Sn alloys with high strength显示文摘Duplex-structured Mg-7Li-2Al-1.5Sn alloys with high strength were fabricated and their strengthening mechanism was investigated.The Mg-7Li-2Al-1.5Sn alloys were prepared by casting and extruded at the temperature of 533 K with an extrusion ratio of 25:1.The microstructure and mechanical properties of Mg-7Li-2Al-1.5Sn alloys were systematically investigated by OM,XRD,SEM,TEM,and tensile tests.The results show that Mg-7Li-2Al-1.5Sn alloys are mainly composed ofα-Mg,β-Li,LiMgAl_(2),Mg_(2)Sn and Li_(2)MgSn phases.The yield strength(YS),ultimate tensile strength(UTS)and elongation(EL)of the extruded alloy at room temperature reach 250 MPa,324 MPa and 11.9%,respectively.A lot of Sn-rich precipitates(Mg_(2)Sn and Li_(2)MgSn)are precipitated during extrusion with an average size of∼14 nm,which is beneficial to the grain refinement.Dynamic recrystallization occurs during hot deformation and the nanoprecipitates effectively refine the dynamic recrystallized(DRXed)grains.Besides,the residual dislocations existed in DRXed and un-DRXed grains result in the dislocation strengthening in the extruded alloy.Mg7Li-2Al-1.5Sn alloys possess excellent high-temperature mechanical properties with the YS,UTS and EL of 200 MPa,237 MPa and 26.7%at 423 K,respectively.Sn-rich precipitates with good thermal stability can effectively prevent grain growth,which is good for the improvement of the high-temperature performance of Mg-Li-Al-Sn alloy. | Gang Zhou Yan Yang Hanzhu Zhang Faping Hu Xueping Zhang Chen Wen Weidong Xie Bin Jiang Xiaodong Peng Fusheng Pan | 2022 | Journal of Materials Science & Technology2022,,8: | 7 |

| 9 | Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. | 10 | Super enhancer inhibitors suppress MYC driven transcriptional amplification and tumor progression in osteosarcoma显示文摘Osteosarcoma is the most common primary bone sarcoma that mostly occurs in young adults. The causes of osteosarcoma are heterogeneous and still not fully understood. Identification of novel, important oncogenic factors in osteosarcoma and development of better, effective therapeutic approaches are in urgent need for better treatment of osteosarcoma patients. In this study, we uncovered that the oncogene MYC is significantly upregulated in metastastic osteosarcoma samples. In addition, high MYC expression is associated with poor survival of osteosarcoma patients. Analysis of MYC targets in osteosarcoma revealed that most of the osteosarcoma super enhancer genes are bound by MYC. Treatment of osteosarcoma cells with super enhancer inhibitors THZ1 and JQ1 effectively suppresses the proliferation, migration, and invasion of osteosarcoma cells. Mechanistically,THZ1 treatment suppresses a large group of super enhancer containing MYC target genes including CDK6 and TGFB2. | 13 | Dosimetric evaluation of VMAT radiation therapy technique for breast cancer after conservative surgery based on three different types of multileaf collimators显示文摘Objective Radiotherapy combined with conservative surgery plays an important role in the treatment of early-stage breast cancer. Volumetric modulated arc therapy(VMAT) has been introduced into clinical practice. The purpose of this study was to investigate the dosimetric effects of different multileaf collimators(MLC) on VMAT radiotherapy plans for treating breast cancer.Methods Fifteen breast cancer patients who were treated using a conventional technique in our department were selected to participate in this retrospective analysis. VMAT plans based on three types of Elekta MLCs [Beam Modulator(BM) with 4-mm leaf width, Agility with 5-mm leaf width and MLCi2 with 10-mm leaf width] were independently generated for each patient. Plan comparisons were performed based on dose-volume histogram(DVH) analysis including dosimetric parameters such as the homogeneity index(HI), conformity index(CI), Dmax, Dmin, and Dmean for the planning treatment volume(PTV), in addition to dose-volume parameters for the organs at risk(OARs). The delivery efficiency of the three types of MLCs was compared in terms of the beam delivery time and the monitor units(MUs) per fraction for each plan. Results Both target uniformity and conformity were improved in plans for Agility and BM MLC compared with the plan using MLCi2. The mean HI decreased from 1.14 for MLCi2 to 1.13 for BM and 1.10 for Agility, while the mean CI increased from 0.68 for MLCi2 to 0.73 for BM and 0.75 for Agility. Furthermore, at both low and high dose levels, smaller volumes of ipsilateral lung, heart, contralateral lung, and breast were irradiated with Agility MLC than with the other two types of MLCs. | 15 | Cisplatin-induced vestibular hair cell lesion-less damage at high doses显示文摘Cisplatin, a widely used anticancer drug, damages hair cells in cochlear organotypic cultures at low doses, but paradoxically causes little damage at high doses resulting in a U-shaped dose-response function. To determine if the cisplatin dose-response function for vestibular hair cells follows a similar pattern, we treated vestibular organotypic cultures with doses of cisplatin ranging from 10 to 1000 μM. Vestibular hair cell lesions progressively increased as the dose of cisplatin increased with maximum damage occurring around 50-100 μM, but the lesions progressively decreased at higher doses resulting in little hair cell loss at 1000 μM. | 16 | Nonuniform stretch processing for the range profile of a target with micro-motion显示文摘Complex motions such as micro-motions induce complex range-Doppler coupling effect in radar imaging. Hence, it is a challenge to reconstruct a clear range profile of a target with micro-motion. Stretch processing with fine motion compensation cannot perform well while it ties to eliminate the distortion of range profile caused by Doppler effect, because it is difficult to accurately estimate each scattering center velocity during coherent processing interval. Based on nonuniform stretch processing (NSP), a new technology is proposed to decouple range, time and Doppler and then achieve range profile of target with micro-motion. The new technology uses a new synthetic wide-band waveform and has an innovative mathematic interpretation. Statistical properties of the new approach are primarily examined. Typical models of target with micro-motion for high resolution radar are presented. | 17 | Biomechanical analysis of the computer-assisted internal fixation of a femoral neck fracture显示文摘The number and spatial configuration of the screws will affect the stability and prognosis of the fractures.In our study,we assessed the biomechanical effects of the double-head cannulated compression screw(DhCCS)and ordinary cannulated compression screw(OCCS)for the treatment of femoral neck fractures by using computer finite element analysis.The original digital imaging and communications in medicine(DICOM)data of a proximal femur were imported into Materialise’s interactive medical image control system(MIMICS)software for modeling.Both DhCCS and OCCS 3D-models were obtained by using the 3D scan technique.Using the fracture model and internal fixation assembly model with an inverted triangle,two horizontal and vertical distribution were established in UG software.Next,the displacement and stress distribution were calculated in ANSYS software.The displacement value of the femoral head in the DhCCS group was smaller than that in the OCCS group,and the displacement value in the two horizontal groups was smaller than that in the vertical group.The stress distribution in the DhCCS group was concentrated on the screw rod at the fracture block and thread end,while only at the fracture block in the OCCS group.The stress in the horizontal group was more dispersed on the screws than that in the vertical group.DhCCS has reliable stability for the fixation of femoral neck fractures and applied in the clinical work and 2 horizontal fixation can be used when two screws are selected. | Hui Lu Hongquan Shen Shuqing Zhou Weidong Ni Dianming Jiang | 2020 | Genes & Diseases2020,7,3: | 3 |
| 18 | Construction of molecule-selective mixed matrix membranes with confined mass transfer structure显示文摘Extraordinary mass transfer phenomenon is usually found when the small molecules pass through a confined structure, whose effective size is commensurate with the mean free path of the molecules. Small changes in the confined mass transfer structure(including size, morphology and properties) will lead to significant fluctuations of the mass transfer coefficient. The mass transfer of the penetrant molecules in the dense membranes for pervaporation, gas separation and so on, is located in the scope of confined mass transfer. Incorporating nanofillers into polymer matrix to construct mixed matrix membranes(MMMs) is an effective approach to tune the confined mass transfer structure and enhance the performance of the widely used polymeric membranes. | 19 | Effect of Mn content on the microstructure and mechanical properties of Mg–6Li–4Zn-xMn alloys显示文摘The as-cast Mg–6Li–4Zn-x Mn alloys were prepared and extruded at 280℃with an extrusion ratio of 25:1.The effects of Mn content on the microstructure and mechanical properties of Mg–6Li–4Zn-x Mn alloys were investigated in this study.The XRD results show that Mg–6Li–4Zn–x Mn alloys consisted ofα-Mg(hcp)+β-Li(bcc)duplex structured matrix,Mg Li_(2)Zn and Mn phases.The grains of the extruded Mg–6Li–4Zn–x Mn alloys were refined by dynamic recrystallization during the extrusion process.The EBSD results show that the extruded alloys had basal textures.The grain size of the extruded alloys decreased while the basal texture was strengthened with the increasing Mn addition.The TEM results show that a large amount of nanoscale Mn precipitates existed in the extruded Mg–6Li–4Zn–1.2 Mn alloy,which can effectively inhibit the dynamic recrystallized(DRXed)grains growth during the hot extrusion and is beneficial to the improvement of mechanical properties.Mg–6Li–4Zn–1.2 Mn alloy in this research possesses the best mechanical properties with the ultimate tensile strength and yield strength of 321 MPa,250 MPa,respectively. | Hongju Deng Yan Yang Minmin Li Xiaoming Xiong Guobing Wei Weidong Xie Bin Jiang Xiaodong Peng Fusheng Pan | 2021 | Progress in Natural Science:Materials International2021,31,4: | 3 |
| 20 | Antioxidative stress-induced damage in cochlear explants显示文摘The imbalance of reactive oxygen species and antioxidants is considered to be an important factor in the cellular injury of the inner ear. At present, great attention has been placed on oxidative stress. However,little is known about fighting oxidative stress. In the current study, we evaluated antioxidant-induced cochlear damage by applying several different additional antioxidants. To determine whether excessive antioxidants can cause damage to cochlear cells, we treated cochlear explants with 50 m M M40403, a superoxide dismutase mimetic, 50 m M coenzyme Q-10, a vitamin-like antioxidant, or 50 m M d-methionine, an essential amino acid and the important antioxidant glutathione for 48 h. Control cochlear explants without the antioxidant treatment maintained their normal structures after incubation in the standard serum-free medium for 48 h, indicating the maintenance of the inherent oxidative and antioxidant balance in these cochlear explants. In contrast, M40403 and coenzyme Q-10-treated cochlear explants displayed significant hair cell damage together with slight damage to the auditory nerve fibers.Moreover, d-methiodine-treated explants exhibited severe damage to the surface structure of hair cells and the complete loss of the spiral ganglion neurons and their peripheral fibers.
