1Manganese is critical for antitumor immune responses via cGAS-STING and improves the efficacy of clinical immunotherapy显示文摘CD8^+T cell-mediated cancer clearance is often suppressed by the interaction between inhibitory molecules like PD-1 and PD-L1,an interaction acts like brakes to prevent T cell overreaction under normal conditions but is exploited by tumor cells to escape the immune surveillance.Immune checkpoint inhibitors have revolutionized cancer therapeutics by removing such brakes.Unfortunately,only a minority of cancer patients respond to immunotherapies presumably due to inadequate immunity.Antitumor immunity depends on the activation of the cGAS-STING pathway,as STING-deficient mice fail to stimulate tumor-infiltrating dendritic cells(DCs)to activate CD8^+T cells.STING agonists also enhance natural killer(NK)cells to mediate the clearance of CD8^+T cell-resistant tumors.Therefore STING agonists have been intensively sought after.We previously discovered that manganese(Mn)is indispensable for the host defense against cytosolic dsDNA by activating cGAS-STING.Here we report that Mn is also essential in innate immune sensing of tumors and enhances adaptive immune responses against tumors.Mn-insufficient mice had significantly enhanced tumor growth and metastasis,with greatly reduced tumor-infiltrating CD8^+T cells.Mechanically,Mn^2+promoted DC and macrophage maturation and tumor-specific antigen presentation,augmented CD8^+T cell differentiation,activation and NK cell activation,and increased memory CD8^+T cells.Combining Mn^2+with immune checkpoint inhibition synergistically boosted antitumor efficacies and reduced the anti-PD-1 antibody dosage required in mice.Importantly,a completed phase 1 clinical trial with the combined regimen of Mn^2+and anti-PD-1 antibody showed promising efficacy,exhibiting type I IFN induction,manageable safety and revived responses to immunotherapy in most patients with advanced metastatic solid tumors.We propose that this combination strategy warrants further clinical translation.Mengze Lv Meixia Chen Rui Zhang Wen Zhang Chenguang Wang Yan Zhang Xiaoming Wei Yukun Guan Jiejie Liu Kaichao Feng Miao Jing Xurui Wang Yun-Cai Liu Qian Mei Weidong Han Zhengfan Jiang 2020Cell Research2020,30,11:27
2Constraints on the age of the Tuchengzi Formation by LA-ICP-MS dating in northern Hebei-western Liaoning, China显示文摘Accurately determining the age of the Tuchengzi Formation has direct influence on confirming the boundary between the Jurassic and the Cretaceous systems in northern Hebei-western Liaoning, and on related geological problems in China. However, the Tuchengzi Formation mainly consists of sedi-mentary rocks, with a poor fossil record and especially lack of index fossils. The Tuchengzi Formation is also lack of the type of volcanic rocks that can provide an isotopic age. Therefore, the age of the Tuchengzi Formation has been uncertain. Based on our systematic dating of the tuff interbedded in the Tuchengzi Formation of Chengde and Jinlingsi-Yangshan basins in northern Hebei-western Liaoning, combined with the dating results of previous researchers, here we suggest that the age range of the Tuchengzi Formation in northern Hebei-western Liaoning is from 147 Ma to 136 Ma. 4He-Ar isotope geochemistry of the Yaoling-Meiziwo tungsten deposit,North Guangdong Province:Constraints on Yanshanian crust-mantle interaction and metallogenesis in SE China显示文摘Isotopic abundances and ratios of He and Ar found in inclusion fluids in pyrites formed in the Yaoling-Meiziwo tungsten miner-alization epoch show that the concentration of 4He varies widely,from 1.54×10-7 cm3 STP/g to 2609×10-7 cm3 STP/g.3He is 0.759×10-12 cm3 STP/g-3.463×10-12 cm3 STP/g.3He/4He is 0.0043-4.362 Ra,varying from crustal to mantle values.The concen-tration of 40Ar ranges from 0.624×10-7 cm3 STP/g to 8.89×10-7 cm3 STP/g.The 40Ar/36Ar varies extensively,from 330 to 2952,between atmospheric and crustal or mantle radiogenic values.Mantle-derived He is present in ore-forming fluids and the calcu-lated average proportion of the mantle He is 22%;the maximum is 67%.Our research results show that mantle-derived fluids play a significant role in tungsten mineralization.The fractionation of He and Ar indicate that there was 4He-enriched air-saturated water(MSAW) in the ore-forming fluid.The ore-forming fluid was a mixture of mantle fluid,crustal magmatic fluid and MSAW.The occurrence of a mantle component in ore-forming fluid indicates the large-scale W and Sn mineralization,including Yaol-ing-Meiziwo,in southeastern China was the result of crust and mantle interaction.The underplating or intrusion of voluminous basaltic magma formed by partial melting of the upper mantle provided the necessary heat to cause partial melting of the crust and the generation of voluminous S-type granitic magmas.Crustal magmatic fluid and mantle fluid with high 3He/4He were released from magma crystallization and fractionation,mixed with the circulating modified air-saturated water,and filled the extensional tectonic fractures,leading to the formation of world-class W and Sn deposits in southeastern China.ZHAI Wei SUN XiaoMing WU YunShan SUN YanYan HUA RenMin YE XianRen 2012Chinese Science Bulletin2012,57,10:16
5MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells.There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting.After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide,secondary neurosphere assay,soft agar colony assay and MTT assay were performed.Compared with U87 cells,the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar,and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony,and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells.Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu 2010The Journal of Biomedical Research2010,24,6:14

8Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 10Design of precision variable-rate spray system for unmanned aerial vehicle using automatic control method显示文摘In order to reduce the use of chemical pesticides in crop plant protection and improve the utilization efficiency of pesticides,it is necessary to study advanced application machinery and application techniques.The use of unmanned aerial vehicle(UAV)for pesticide spraying has the characteristics of less application,strong penetrability,wide applicability and flexible operation scheduling,and has gradually become one of the important development directions in the field of aviation plant protection.However,the operation process of the UAV is often affected by meteorological factors and human manipulation,resulting in poor actual operation with inaccurate spray volume and uneven application.Therefore,to improve the stability and uniformity of the application of the plant protection UAV under variable operating conditions,in this paper a real-time control method was proposed for the application flow rate,and a precision variable-rate spray system was designed based on single-chip microcomputer and micro diaphragm pump that can controls the flow rate of the pump in real time with the changes of the operating state.The response s-peed of the variable-rate spray system was tested.The average control response time of the system was 0.18 s,and the average stability time of the pump flow change was 0.75 s.The test results showed that the system has a quick response to the working state and the adjustment of the target flow of the pump can be quickly completed to realize the variable-rate spray function.The research results can provide a reference for the practical application of plant protection UAV variable-rate spray system.Qi Lian Feng Tan Xiaoming Fu Ping Zhang Xin Liu Wei Zhang 2019International Journal of Agricultural and Biological Engineering2019,12,2:10

12Wind tunnel experimental study on droplet drift reduction by a conical electrostatic nozzle for pesticide spraying显示文摘Droplet drift wastes pesticide,pollutes the environment,and has become one of the focus issues of agricultural crop protection.Electrostatic spray technology reduces drift to a certain degree.In order to investigate the droplet drift pattern of a conical electrostatic nozzle,the droplet drift mass center distance was defined as an experimental index and used to conduct experimental wind tunnel studies on droplet drift.A mathematical model of the droplet drift mass center distance versus electrostatic voltage and wind speed was created via the regression method.The test results showed that the electrostatic voltage had an insignificant effect on droplet drift,the wind speed and its interaction with the electrostatic voltage had significant effects on droplet drift.When the wind speed was less than 3 m/s and stable,the crop adsorbability of a droplet had a dominant effect on the droplet drift;the droplet drift decreased with the increase of electrostatic voltage.When the wind speed exceeded 3 m/s and was stable,the reduced droplet particle size had a dominant effect on droplet drift,where droplet drift increased as the electrostatic voltage increased.When the wind speed was 0 m/s and the electrostatic voltage was 12 kV,the minimum droplet drift mass center distance was 35.5 mm,which was 56 mm less than that of conventional nozzle droplet drift.Therefore,a conical electrostatic nozzle is inapplicable for operation in an environment where wind speeds exceed 3 m/s.This study provides a reference for optimizing operational parameters of conical electrostatic nozzles and improving the anti-drift capability of droplets.Zhang Wei Hou Yongrui Liu Xin Lian Qi Fu Xiaoming Zhang Bo Wang Haiyang Wang Yingqian 2017International Journal of Agricultural and Biological Engineering2017,10,3:8
13The geochemical characteristics,geochronology and tectonic significance of the Carboniferous volcanic rocks of the Santanghu area in northeastern Xinjiang,China显示文摘The Santanghu area is located on the northeastern margin of the Junggar Basin,northern Xinjiang,Northwest China.The Carboniferous volcanic rocks in this area are widely distributed in Kaokesaiergaishan,Santanghu,Daheishan and Naomaohu districts,which are located to the north of the Kalameili Fault.These rocks,sourced from a cognate magma,consist of basic,intermediate,and acidic lavas,and pyroclastic rock.The basic volcanic rocks are enriched with large-ion lithophile elements(LILE),but are relatively depleted in high field strength elements(HFSE),and have an obvious negative Nb-Ta-Ti anomaly.They were most probably derived from a depleted mantle source,and during their ascent,these magmas were not contaminated by the crustal material as they underwent magma crystallization differentiation.Based on the Carboniferous volcanic assemblage and geochemical data,it is apparent that the early Carboniferous volcanism occurred in a subduction-related tectonic setting.New LA-ICP-MS zircon U-Pb analyses constrain the age of the andesite within the volcanic rocks as the early Carboniferous(328.9-331.3 Ma).Combined with the regional geological record,comprehensive analysis of the isotope geochronological data indicates that the subduction of the Junggar Ocean predates the early Carboniferous,and that the Santanghu island arc magmatism was induced by the subduction of the Junggar Ocean in the Carboniferous.LI Wei LIU YiQun DONG YunPeng ZHOU XiaoHu LIU XiaoMing LI Hong FAN TingTing ZHOU DingWu XU XueYi CHEN JunLu 2013Science China Earth Sciences2013,56,8:8
14Sedimentary fill history of the Huicheng Basin in the West Qinling Mountains and associated constraints on Mesozoic intracontinental tectonic evolution显示文摘The Qinling Orogenic Belt is divided commonly by the Fengxian-Taibai strike-slip shear zone and the Huicheng Basin into the East and West Qinling mountains,which show significant geological differences after the Indosinian orogeny.The Fengxian-Taibai fault zone and the Meso-Cenozoic Huicheng Basin,situated at the boundary of the East and West Qinling,provide a natural laboratory for tectonic analysis and sedimentological study of intracontinental tectonic evolution of the Qinling Orogenic Belt.In order to explain the dynamic development of the Huicheng Basin and elucidate its post-orogenic tectonic evolution at the junction of the East and West Qinling,we studied the geometry and kinematics of fault zones between the blocks of West Qinling,as well as the sedimentary fill history of the Huicheng Basin.First,we found that after the collisional orogeny in the Late Triassic,post-orogenic extensional collapse occurred in the Early and Middle Jurassic within the Qinling Orogenic Belt,resulting in a series of rift basins.Second,in the Late Jurassic and Early Cretaceous,a NE-SW compressive stress field caused large-scale sinistral strike-slip faults in the Qinling Orogenic Belt,causing intracontinental escape tectonics at the junction of the East and West Qinling,including eastward finite escape of the East Qinling micro-plate and southwest lateral escape of the Bikou Terrane.Meanwhile,the strike-slip-related Early Cretaceous sedimentary basin was formed with a right-order echelon arrangement in sinistral shear zones along the southern margin of the Huicheng fault.Overall during the Mesozoic,the Huicheng Basin and surrounding areas experienced four tectonic evolutionary stages,including extensional rift basin development in the Early and Middle Jurassic,intense compressive uplift in the Late Jurassic,formation of a strike-slip extensional basin in the Early Cretaceous,and compressive uplift in the Late Cretaceous.LI Wei DONG YunPeng GUO AnLin LIU XiaoMing LIU YiQun ZHA XianFeng ZHANG KuaiLe 2013Science China Earth Sciences2013,56,10:7

18Manganese salts function as potent adjuvants显示文摘Aluminum-containing adjuvants have been used for nearly 100 years to enhance immune responses in billions of doses of vaccines.To date,only a few adjuvants have been approved for use in humans,among which aluminum-containing adjuvants are the only ones widely used.However,the medical need for potent and safe adjuvants is currently continuously increasing,especially those triggering cellular immune responses for cytotoxic T lymphocyte activation,which are urgently needed for the development of efficient virus and cancer vaccines.Manganese is an essential micronutrient required for diverse biological activities,but its functions in immunity remain undefined.We previously reported that Mn^(2+) is important in the host defense against cytosolic dsDNA by facilitating cGAS-STING activation and that Mn^(2+)alone directly activates cGAS independent of dsDNA,leading to an unconventional catalytic synthesis of 2′3′-cGAMP.Herein,we found that Mn^(2+) strongly promoted immune responses by facilitating antigen uptake,presentation,and germinal center formation via both cGAS-STING and NLRP3 activation.Accordingly,a colloidal manganese salt(Mn jelly,MnJ)was formulated to act not only as an immune potentiator but also as a delivery system to stimulate humoral and cellular immune responses,inducing antibody production and CD4^(+)/CD8^(+)T-cell proliferation and activation by either intramuscular or intranasal immunization.When administered intranasally,MnJ also worked as a mucosal adjuvant,inducing high levels of secretory IgA.MnJ showed good adjuvant effects for all tested antigens,including T cell-dependent and T cell-independent antigens,such as bacterial capsular polysaccharides,thus indicating that it is a promising adjuvant candidate.Rui Zhang Chenguang Wang Yukun Guan Xiaoming Wei Mengyin Sha Mengran Yi Miao Jing Mengze Lv Wen Guo Jing Xu Yi Wan Xin-Ming Jia Zhengfan Jiang 2021Cellular & Molecular Immunology2021,18,5:7
19Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 20Theoretical study of the N—H···O red-shifted andblue-shifted hydrogen bonds显示文摘Theoretical calculations are performed to study the nature of the hydrogen bonds in complexes HCHO···HNO, HCOOH···HNO, HCHO···NH3, HCOOH···NH3, HCHO···NH2F and HCOOH···NH2F. The geomet- ric structures and vibrational frequencies of these six complexes at the MP2/6-31+G(d,p), MP2/6-311++G(d,p), B3LYP/6-31+G(d,p) and B3LYP/6-311++G(d,p) levels are calculated by standard and counterpoise-corrected methods, respectively. The results indicate that in complexes HCHO···HNO and HCOOH···HNO the N—H bond is strongly contracted and N—H···O blue-shifted hydrogen bonds are observed. While in complexes HCHO···NH3, HCOOH···NH3, HCHO···NH2F and HCOOH···NH2F, the N—H bond is elongated and N—H···O red-shifted hydrogen bonds are found. From the natural bond orbital analysis it can be seen that the X—H bond length in the X—H···Y hydrogen bond is controlled by a balance of four main factors in the opposite directions: hyperconjugation, electron density redistribu- tion, rehybridization and structural reorganization. Among them hyperconjugation has the effect of elongating the X—H bond, and the other three factors belong to the bond shortening effects. In complexes HCHO···HNO and HCOOH···HNO, the shortening effects dominate which lead to the blue shift of the N—H stretching frequencies.
