2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3A meta-analysis of lamivudine for interruption of mother-to-child transmission of hepatitis B virus显示文摘AIM:To determine the therapeutic effect of lamivudine in late pregnancy for the interruption of motherto-child transmission(MTCT) of hepatitis B virus(HBV).METHODS:Studies were identified by searching available databases up to January 2011.Inclusive criteria were HBV-carrier mothers who had been involved in randomized controlled clinical trials(RCTs) with lamivudine treatment in late pregnancy,and newborns or infants whose serum hepatitis B surface antigen(HBsAg),hepatitis B e antigen(HBeAg) or HBV DNA had been documented.The relative risks(RRs) for interruption of MTCT as indicated by HBsAg,HBV DNA or HBeAg of newborns or infants were calculated with 95% confidence interval(CI) to estimate the efficacy of lamivudine treatment.RESULTS:Fifteen RCTs including 1693 HBV-carrier mothers were included in this meta-analysis.The overall RR was 0.43(95% CI,0.25-0.76;8 RCTs;Pheterogeneity = 0.04) and 0.33(95% CI,0.23-0.47;6 RCTs;Pheterogeneity = 0.93) indicated by newborn HBsAg or HBV DNA.The RR was 0.33(95% CI,0.21-0.50;6 RCTs;Pheterogeneity = 0.46) and 0.32(95% CI,0.20-0.50;4 RCTs;Pheterogeneity = 0.33) indicated by serum HBsAg or HBV DNA of infants 6-12 mo after birth.The RR(lamivudine vs hepatitis B immunoglobulin) was 0.27(95% CI,0.16-0.46;5 RCTs;Pheterogeneity = 0.94) and 0.24(95% CI,0.07-0.79;3 RCTs;P heterogeneity = 0.60) indicated by newborn HBsAg or HBV DNA,respectively.In the mothers with viral load < 106 copies/mL after lamivudine treatment,the efficacy(RR,95% CI) was 0.33,0.21-0.53(5 RCTs;Pheterogeneity = 0.82) for the interruption of MTCT,however,this value was not significant if maternal viral load was > 106 copies/mL after lamivudine treatment(P = 0.45,2 RCTs),as indicated by newborn serum HBsAg.The RR(lamivudine initiated from 28 wk of gestation vs control) was 0.34(95% CI,0.22-0.52;7 RCTs;Pheterogeneity = 0.92) and 0.33(95% CI,0.22-0.50;5 RCTs;Pheterogeneity = 0.86) indicated by newborn HBsAg or HBV DNA.The incidence of adverse effects of lamivudine was not higher in the mothers than in controls(P = 0.97).Only one study reported side effects of lamivudine in newborns.CONCLUSION:Lamivudine treatment in HBV carriermothers from 28 wk of gestation may interrupt MTCT of HBV efficiently.Lamivudine is safe and more efficient than hepatitis B immunoglobulin in interrupting MTCT.HBV MTCT might be interrupted efficiently if maternal viral load is reduced to < 106 copies/mL by lamivudine treatment.Lei Han Hong-Wei Zhang Jia-Xin Xie Qi Zhang Hong-Yang Wang Guang- Wen Cao 2011World Journal of Gastroenterology2011,17,38:61

5Cancer burden in China:trends,risk factors and prevention显示文摘As the most populous country in the world,China has made strides in health promotion in the past few decades.With the aging population,the burden of cancer in China continues to grow.Changes in risk factors for cancer,especially diet,obesity,diabetes,and air pollution,continue to fuel the shift of cancer transition in China.The burden of upper gastrointestinal cancer in China is decreasing,but still heavy.The rising burden of colorectal,prostate,and breast cancers is also significant.Lung cancer became the top cause of cancer-related deaths,together with smoking as the most important contributor to cancer deaths.The Chinese government has taken several approaches to control cancer and cancer-related risk factors.Many achievements have been made,but some challenges remain.Health China 2030 is ambitious and depicts a bright vision of the future for cancer control in China.The decrease in the cancer burden in China will require cross-sector collaboration and coordinated efforts on primary and secondary preventions by governments,public health organizations,and individuals.In this review,we describe the trends of cancer burden and discuss cancer-related risk factors in China,identifying strategies to reduce the burden of cancer in China.Dianqin Sun He Li Maomao Cao Siyi He Lin Lei Ji Peng Wanqing Chen 2020Cancer Biology & Medicine2020,17,4:62
6Long-term efficacy of perioperative chemoradiotherapy on esophageal squamous cell carcinoma显示文摘AIM: To investigate the role of perioperative chemoradiotherapy (CRT) in the treatment of locally advanced thoracic esophageal squamous cell carcinoma (ESCC). METHODS: Using preoperative computed tomography (CT)-based staging criteria, 238 patients with ESCC (stage ⅡⅢ ) were enrolled in this prospective study between January 1997 and June 2004. With informed consent, patients were randomized into 3 groups: preoperative CRT (80 cases), postoperative CRT (78 cases) and surgery alone (S) (80 cases). The 1-, 3-, 5and 10-year survival were followed up. Progressionfree survival (PFS) was chosen as the primary endpoint by treatment arm measured from study entry until documented progression of disease or death from any cause. The secondary endpoint was overall survival (OS) determined as the time (in months) between the date of therapy and the date of death. Other objectives were surgical and adjuvant therapy complications.RESULTS: With median follow-up of 45 mo for all the enrolled patients, significant differences in the 1-, 3-, 5-, 10-year OS (91.3%, 63.5%, 43.5%, 24.5% vs 91%, 62.8%, 42.3%, 24.4% vs 87.5%, 51.3%, 33.8%, 12.5%, P = 0.0176) and PFS (89.3%, 61.3%, 37.5%, 18.1% vs 89.1%, 61.1%, 37.2%, 17.8% vs 84.5%, 49.3%, 25.9%, 6.2%, P = 0.0151) were detected among the 3 arms. There were no significant differences in OS and PFS between the preoperative CRT and postoperative CRT arm (P > 0.05). For the patients who had radical resection, significant differences in median PFS (48 mo vs 61 mo vs 39.5 mo, P = 0.0331) and median OS (56.5 mo vs 72 mo vs 41.5 mo, P = 0.0153) were detected among the 3 arms, but there were no significant differences in OS and PFS between the preoperative CRT and postoperative CRT arm (P > 0.05). The local recurrence rates in the preoperative CRT, postoperative CRT group and S group were 11.3%, 14.1% and 35%, respectively (P < 0.05). No significant differences were detected among the 3 groups when comparing complications but tended to be in favor of the postoperative CRT and S groups (P > 0.05). Toxicities of CRT in the preoperative or postoperative CRT arms were mostly moderate, and could be quickly alleviated by adequate therapy. 8Surgical outcomes of mini-open Wiltse approach and conventional open approach in patients with single-segment thoracolumbar fractures without neurologic injury显示文摘This study aimed to introduce a novel mini-open pedicle screw fixation technique via Wiltse approach,and compared it with the traditional posterior open method.A total of 72 cases of single-segment thoracolumbar fractures without neurologic injury underwent pedicle screw fixation via two different approaches.Among them,37 patients were treated using posterior open surgery,and 35 patients received mini-open operation via Wiltse approach.Crew placement accuracy rate,operative time,blood loss,postoperative drainage,postoperative hospitalization time,radiation exposure time,postoperative improvement in R value,Cobb's angle and visual analog scale(VAS)scores of the two methods were compared.There were no significant differences in the accuracy rate of pedicle screw placement,radiation exposure and postoperative R value and Cobb's angle improvement between the two groups.However,the mini-open method had obvious advantages over the conventional open method in operative time,blood loss,postoperative drainage,postoperative hospitalization time,and postoperative improvement in VAS.The mini-open pedicle screw technique could be applied in treatment of single-segment thoracolumbar fracture without neurologic injury and had advantages of less tissue trauma,short operative and rehabilitative time on the premise of guaranteed accuracy rate and no increased radiation exposure.Haijun Li Lei Yang Hao Xie Lipeng Yu Haifeng Wei Xiaojian Cao 2015The Journal of Biomedical Research2015,29,1:43
9Pollution characteristics and health risk assessment of benzene homologues in ambient air in the northeastern urban area of Beijing, China显示文摘Ambient benzene homologues were measured at a site in the northeastern urban area of Beijing, China, from August 24 to September 4, 2012 by SUMMA canister sampling followed by laboratory determination using cryogenic cold trap pre-concentration-GC-MS/FID, and their health risks were also assessed. Daily total benzene homologues ranged from 0.99 to 49.71 μg/m3with an average of 11.98 μg/m3. Benzene homologues showed higher concentrations in the morning and evening than that at noontime. Comparison with previous studies revealed a trend of decrease for ambient benzene homologues probably due to the efective emission control in Beijing in recent years. Vehicular exhaust was the main source while volatilization of paints and solvents also made substantial contributions. Health risk assessment showed that BTEX(benzene, toluene, ethylbenzene, o-xylene, m-xylene and p-xylene) and styrene had no appreciable adverse non-cancer health risks for the exposed population, while benzene has potential cancer risk of 1.34E-05. 10Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. 11Achieving over 16% efficiency for single-junction organic solar cells显示文摘To achieve high photovoltaic performance of bulk hetero-junction organic solar cells(OSCs), a range of critical factors including absorption profiles, energy level alignment, charge carrier mobility and miscibility of donor and acceptor materials should be carefully considered. For electron-donating materials, the deep highest occupied molecular orbital(HOMO) energy level that is beneficial for high open-circuit voltage is much appreciated. However, a new issue in charge transfer emerges when matching such a donor with an acceptor that has a shallower HOMO energy level. More to this point, the chemical strategies used to enhance the absorption coefficient of acceptors may lead to increased molecular crystallinity, and thus result in less controllable phase-separation of photoactive layer. Therefore, to realize balanced photovoltaic parameters, the donor-acceptor combinations should simultaneously address the absorption spectra, energy levels, and film morphologies. Here, we selected two non-fullerene acceptors, namely BTPT-4F and BTPTT-4F, to match with a wide-bandgap polymer donor P2F-EHp consisting of an imidefunctionalized benzotriazole moiety, as these materials presented complementary absorption and well-matched energy levels. 12Current cancer burden in China: epidemiology, etiology, and prevention显示文摘Cancer has become the most common cause of death in China.Owing to rapid economic development,improved livelihood,and shifts in risk factors,cancer epidemiology has experienced substantial changes during the past several decades.In this review,we aim to describe the current cancer epidemiology of the main types of cancer in China,report major risk factors associated with cancer development,and summarize the contributions of the Chinese government to controlling the cancer burden.A total of 4,064,000 new cases were diagnosed in China in 2016.The most frequent types are lung cancer(828,100;20.4%),colorectal cancer(408,000;10.0%),and gastric cancer(396,500;9.8%).Lung(657,000;27.2%),liver(336,400,13.9%),and stomach(288,500;12.0%)cancers are the 3 most deadly cancers in the general population.The 5-year survival rate for cancer has dramatically increased in recent decades.However,liver and particularly pancreatic cancers still have the poorest prognosis.The main modifiable risk factors associated with cancer development include infectious agents,smoking,alcohol consumption,obesity,unhealthful dietary habits,and inadequate physical activity.The Chinese government has made unremitting efforts to decrease the cancer burden,including cancer education and investment in cancer screening programs.Maomao Cao He Li Dianqin Sun Siyi He Xinxin Yan Fan Yang Shaoli Zhang Changfa Xia Lin Lei Ji Peng Wanqing Chen 2022Cancer Biology & Medicine2022,19,8:33

15Late Triassic sedimentary records in the northern Tethyan Himalaya:Tectonic link with Greater India显示文摘The Upper Triassic flysch sediments(Nieru Formation and Langjiexue Group)exposed in the Eastern Tethyan Himalayan Sequence are crucial for unraveling the controversial paleogeography and paleotectonics of the Himalayan orogen.This work reports new detrital zircon U-Pb ages and whole-rock geochemical data for clastic rocks from flysch strata in the Shannan area.The mineral modal composition data suggest that these units were mainly sourced from recycled orogen provenances.The chemical compositions of the sandstones in the strata are similar to the chemical composition of upper continental crust.These rocks have relatively low Chemical Index of Alteration values(with an average of 62)and Index of Compositional Variability values(0.69),indicating that they experienced weak weathering and were mainly derived from a mature source.The geochemical compositions of the Upper Triassic strata are similar to those of graywackes from continental island arcs and are indicative of an acidicintermediate igneous source.Furthermore,hornblende and feldspar experienced decomposition in the provenance,and the sediment became enriched in zircon and monazite during sediment transport.The detrital zircons in the strata feature two main age peaks at 225-275 Ma and 500-600 Ma,nearly continuous Paleoproterozoic to Neoproterozoic ages,and a broad inconspicuous cluster in the Tonian-Stenian(800-1200 Ma).The detrital zircons from the Upper Triassic sandstones in the study area lack peaks at 300-325 Ma(characteristic of the Lhasa block)and 1150-1200 Ma(characteristic of the Lhasa and West Australia blocks).Therefore,neither the Lhasa block nor the West Australia blocks likely acted as the main provenance of the Upper Triassic strata.Newly discovered Permian-Triassic basalt and mafic dikes in the Himalayas could have provided the 225-275 Ma detrital zircons.Therefore,Indian and Himalayan units were the main provenances of the flysch strata.The Tethyan Himalaya was part of the northern passive margin and was not an exotic terrane separated from India during the Permian to Early Cretaceous.This evidence suggests that the Neo-Tethyan ocean opened prior to the Late Triassic and that the Upper Triassic deposits were derived from continental crustal fragments adjacent to the northern passive continental margin of Greater India.Huawen Cao Yong Huang Guangming Li Linkui Zhang Jianyang Wu Lei Dong Zuowen Dai Liu Lu 2018Geoscience Frontiers2018,9,1:27

The first episode of granite emplacement predates the formation of the ophiolite type mafic rock(?500 Ma), and the three subsequent episodes can be temporally correlated to high-pressure(HP) to ultrahigh-pressure(UHP) metamorphism at ca. 500 Ma, retrograde granulite-facies metamorphism at ca. 450 Ma, and amphibolite-facies metamorphism at ca. 420 Ma, respectively. A comprehensive study of these granitic rocks, along with the regional geological background, mafic-ultramafic rocks, and HP-UHP metamorphism, indicates that the four episodes of granitic magmatism are sequentially derived from the partial melting of the earlier subducted oceanic crust at 517 Ma, the thickened continental crust due to continental subduction at ca. 500 Ma, the mid-upper crust in response to slab breakoff at ca. 450 Ma, and the tectonic transition from contraction to extension at ca. 420 Ma. The formation age of 517 Ma for oceanic adakite provides a direct constraint on the time of the oceanic subduction in South Altyn. In addition, there is a ca. 10 Myr interval between the oceanic subduction to the continental deep subduction, suggesting that the Early Paleozoic tectonic evolution might have been a successive process in South Altyn. 17Xinfeng capsule improves pulmonary function in ankylosing spondylitis patients via NF-κB-iNOS-NO signaling pathway显示文摘OBJECTIVE: To study changes in the nuclear factor-κB p65(NF-κB p65)-inducible nitric oxide synthase(i NOS)-nitric oxide(NO) signaling pathway and the effects of Xinfeng capsules(XFC) in patients with ankylosing spondylitis(AS)METHODS: One hundred twenty patients with AS were randomly divided into an XFC group and a Salazopyrin group. Sixty health subjects were included as a normal control group. In the two treatment groups, pulmonary functional parameters,forced vital capacity(FVC), forced expiratory volume in 1 second(FEV1), maximal voluntary ventilation(MVV), peak expiratory flow(PEF), forced expiratory flow at 25% of forced vital capacity(FEF25),forced expiratory flow at 50% of forced vital capacity(FEF50), and forced expiratory flow at 75% of forced vital capacity(FEF75) were determined. Enzyme linked immunosorbent assays were used for detection of the serum oxidative stress indexes,NF-κB p65, i NOS, NO, reactive oxygen species(ROS), reactive nitrogen species(RNS), malondialdehyde(MDA), superoxide dismutase(SOD), catalase(CAT), total antioxidative capacity(TAOC) and interleukin-4(IL-4), IL-10, IL-1β, and tumor necrosis factor-α(TNF-α) contents. Westergren's method was used for determination of erythrocyte sedimentation rate(ESR). High-sensitivity C-reactive protein(Hs-CRP) was detected with a 7060 full-automatic biochemical analyzer(Hitachi, Japan).RESULTS: The clinical therapeutic effect in the XFC group was significantly superior to that in the Salazopyrin group(P<0.01). Compared with the normal control group, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT,TAOC, IL-4, IL-10 were significantly lower, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR,and Hs-CRP significantly higher in patients with AS(P<0.01 or P<0.05). Compared with before treatment, FEV1, MVV, PEF, FEF50, FEF75, SOD, CAT, TAOC,IL-4, and IL-10 were significantly increased, and NF-κB p65, i NOS, NO, ROS, RNS, MDA, IL-1β, TNF-α,ESR, CRP, visual analog scales(VAS), Bath ankylosing spondylitis disease active index, Bath ankylosing spondylitis functional index, and Bath ankylosing spondylitis global index significantly decreased in the two treatment groups after treatment(P<0.01 or P<0.05), with significant differences between the XFC and Salazopyrin groups(P<0.01 or P<0.05). Spearman correlation analysis indicated that FEV1, MVV, PEF, FEF50, and FEF75 were positively correlated with SOD, CAT, TAOC, IL-4, and IL-10, and were negatively correlated with NF-κB p65, i NOS,NO, ROS, RNS, MDA, IL-1β, TNF-α, ESR, and CRP.CONCLUSION: Patients with AS have local pathologic changes in the spinal cord and other joints.They also have decreased pulmonary function,which is negatively correlated with the NF-κB-i NOS-NO signaling pathway, oxidative indexes, and inflammatory factors. XFC improves rigidity and pain in spinal joints and other symptoms, laboratory indexes, and pulmonary function. 18Molecular mechanosensors in osteocytes显示文摘Osteocytes, the most abundant and long-lived cells in bone, are the master regulators of bone remodeling. In addition to their functions in endocrine regulation and calcium and phosphate metabolism, osteocytes are the major responsive cells in force adaptation due to mechanical stimulation. Mechanically induced bone formation and adaptation, disuse-induced bone loss and skeletal fragility are mediated by osteocytes, which sense local mechanical cues and respond to these cues in both direct and indirect ways. The mechanotransduction process in osteocytes is a complex but exquisite regulatory process between cells and their environment, between neighboring cells, and between different functional mechanosensors in individual cells. Over the past two decades, great efforts have focused on finding various mechanosensors in osteocytes that transmit extracellular mechanical signals into osteocytes and regulate responsive gene expression. The osteocyte cytoskeleton, dendritic processes, Integrin-based focal adhesions, connexin-based intercellular junctions, primary cilium, ion channels, and extracellular matrix are the major mechanosensors in osteocytes reported so far with evidence from both in vitro and in vitro studies.
