| 2 | Machine learning for detecting mesial temporal lobe epilepsy by structural and functional neuroimaging显示文摘Mesial temporal lobe epilepsy(mTLE),the most common type of focal epilepsy,is associated with functional and structural brain alterations.Machine learning(ML)techniques have been successfully used in discriminating mTLE from healthy controls.However,either functional or structural neuroimaging data are mostly used separately as input,and the opportunity to combine both has not been exploited yet.We conducted a multimodal ML study based on functional and structural neuroimaging measures.We enrolled 37 patients with left mTLE,37 patients with right mTLE,and 74 healthy controls and trained a support vector ML model to distinguish them by using each measure and the combinations of the measures.For each single measure,we obtained a mean accuracy of 74%and 69%for discriminating left mTLE and right mTLE from controls,respectively,and 64%when all patients were combined.We achieved an accuracy of 78%by integrating functional data and 79%by integrating structural data for left mTLE,and the highest accuracy of 84%was obtained when all functional and structural measures were combined.These findings suggest that combining multimodal measures within a single model is a promising direction for improving the classification of individual patients with mTLE. | Baiwan Zhou Dongmei An Fenglai Xiao Running Niu Wenbin Li Wei Li Xin Tong Graham J Kemp Dong Zhou Qiyong Gong Du Lei | 2020 | Frontiers of Medicine2020,14,5: | 2 |

| 7 | Application of single-cell RNA sequencing in head and neck squamous cell carcinoma显示文摘Single-cell RNA sequencing has been broadly applied to head and neck squamous cell carcinoma(HNSCC) for characterizing the heterogeneity and genomic mutations of HNSCC benefiting from the advantage of single-cell resolution. We summarized most of the current studies and aimed to explore their research methods and ideas, as well as how to transform them into clinical applications. Through single-cell RNA sequencing, we found the differences in tumor cells’ expression programs and differentiation tracks. The studies of immune microenvironment allowed us to distinguish immune cell subpopulations, the extensive expression of immune checkpoints, and the complex crosstalk network between immune cells and non-immune cells. For cancerassociated fibroblasts(CAFs), single-cell RNA sequencing had made an irreplaceable contribution to the exploration of their differentiation status, specific CAFs markers, and the interaction with tumor cells and immune cells. | 8 | Adjoint Method for an Inverse Problem of CCPF Model显示文摘The problem for determining the exchange rate function of 2D CCPF model by measurements on the partial boundary is considered and solved as one PDE-constraint optimization problem. The optimal variant is the minimum of a cost functional that quantifies the difference between the measurements and the exact solutions. Gradientbased algorithm is used to solve this optimization problem. At each step, the derivative of the cost functional with respect to the exchange rate function is calculated and only one forward solution and one adjoint solution are needed. One method based on the adjoint equation is developed and implemented. | 10 | Pharmacological inhibition of FABP7 by MF 6 counteracts cerebellum dysfunction in an experimental multiple system atrophy mouse model显示文摘Multiple system atrophy(MSA)is a rare,fatal neurodegenerative disease characterized by the accumulation of misfolded asynuclein(asyn)in glial cells,leading to the formation of glial cytoplasmic inclusions(GCl).We previous found that glial fatty acidbinding protein 7(FABP7)played a crucial role in alpha-synuclein(aSyn)aggregation and toxicity in oligodendrocytes,inhibition of FABP7 by a specific inhibitor MF 6 reduced aSyn aggregation and enhanced cell viability in cultured cell lines and mouse oligodendrocyte progenitor cells.In this study we investigated whether MF 6 ameliorated aSyn-associated pathological processes in PLP-haSyn transgenic mice(PLP-aSyn mice),a wildly used MSA mouse model with overexpressing aSyn in oligodendroglia under the proteolipid protein(PLP)promoter.PLP-aSyn mice were orally administered MF6(0.1,1 mg·kg^(-1)·d^(-1))for 32 days starting from the age of 6 months.We showed that oral administration of MF 6 significantly improved motor function assessed in a pole test,and reduced aSyn aggregation levels in both cerebellum and basal ganglia of PLP-aSyn mice.Moreover,MF 6 administration decreased oxidative stress and inflammation levels,and improved myelin levels and Purkinje neuron morphology in the cerebellum.By using mouse brain tissue slices and aSyn aggregates-treated KG-1C cells,we demonstrated that MF 6 reduced aSyn propagation to Purkinje neurons and oligodendrocytes through regulating endocytosis.Overall,these results suggest that MF 6 improves cerebellar functions in MSA by inhibiting asyn aggregation and propagation.We conclude that MF 6 is a promising compound that warrants further development for the treatment of MSA. | An Cheng Wenbin Jia David I.Finkelstein Nadia Stefanova Haoyang Wang Takuya Sasaki Ichiro Kawahata Kohji Fukunaga | 2024 | Acta Pharmacologica Sinica2024,45,1: | 0 |
