| 1 | Paracrine and endocrine actions of bone——the functions of secretory proteins from osteoblasts, osteocytes, and osteoclasts显示文摘The skeleton is a dynamic organ that is constantly remodeled. Proteins secreted from bone cells, namely osteoblasts, osteocytes,and osteoclasts exert regulation on osteoblastogenesis, osteclastogenesis, and angiogenesis in a paracrine manner. Osteoblasts secrete a range of different molecules including RANKL/OPG, M-CSF, SEMA3A, WNT5A, and WNT16 that regulate osteoclastogenesis. Osteoblasts also produce VEGFA that stimulates osteoblastogenesis and angiogenesis. Osteocytes produce sclerostin(SOST) that inhibits osteoblast differentiation and promotes osteoclast differentiation. Osteoclasts secrete factors including BMP6, CTHRC1, EFNB2, S1P, WNT10B, SEMA4D, and CT-1 that act on osteoblasts and osteocytes, and thereby influencea A osteogenesis. Osteoclast precursors produce the angiogenic factor PDGF-BB to promote the formation of Type H vessels, which then stimulate osteoblastogenesis. Besides, the evidences over the past decades show that at least three hormones or 'osteokines'from bone cells have endocrine functions. FGF23 is produced by osteoblasts and osteocytes and can regulate phosphate metabolism. Osteocalcin(OCN) secreted by osteoblasts regulates systemic glucose and energy metabolism, reproduction, and cognition. | 4 | Identification of immunoreactive proteins of Brucella melitensis by immunoproteomics显示文摘Infection with Brucella causes brucellosis, a chronic disease in humans, which induces abortion and sterility in livestock. Among the different Brucella species, Brucella melitensis is considered the most virulent and is the predominant species associated with outbreaks in China. To date, no safe human vaccine is available against Brucella infection. The currently used live vaccines against Brucella in livestock induce antibodies that interfere with the diagnosis of field infection in vaccinated animals, which is harmful to eradication programs. However, there is as yet no complete profile of immunogenic proteins of B. melitensis. Towards the development of a safer, equally efficacious, and field infection-distinguishable vaccine, we used immunoproteomics to identify novel candidate immunogenic proteins from B. melitensis M5. Eighty-eight immunoreactive protein spots from B. melitensis M5 were identified by Western blotting and were assigned to sixty-one proteins by mass spectrometry, including many new immunoreactive proteins such as elongation factor G, F0F1 ATP synthase subunit beta, and OMP1. | 5 | High-resolution crustal structure of the Yinchuan basin revealed by deep seismic reflection profiling:implications for deep processes of basin显示文摘The Yinchuan basin, located on the western margin of the Ordos block, has the characteristics of an active continental rift. A NW-striking deep seismic reflection profile across the center of Yinchuan basin precisely revealed the fine structure of the crust. The images showed that the crust in the Yinchuan basin was characterized by vertical stratifications along a detachment located at a two-way travel time(TWT) of 8.0 s.The most outstanding feature of this seismic profile was the almost flat Mohorovicˇic′ discontinuity(Moho) and a high-reflection zone in the lower crust. This sub-horizontal Moho conflicts with the general assumption of an uplifted Moho under sedimentary basins and continental rifts, and may indicate the action of different processes at depth during the evolution of sedimentary basins or rifts.We present a possible interpretation of these deep processes and the sub-horizontal Moho. The high-reflection zone, which consists of sheets of high-density, mantlederived materials, may have compensated for crustal thinning in the Yinchuan basin, leading to the formation of a sub-horizontal Moho. | 11 | Target-activated and ratiometric photochromic probe for “double-check” detection of toxic thiols in live cells显示文摘Photochromic molecules can achieve reversible isomerization upon alternate light irradiations, which offers a great opportunity to improve the precision of analytes detection and imaging in complicated biological environments. Previous reported photochromic probe exhibited only mono-color switching and an initially fluorescence-ON state that may cause high background signal and impose an adverse impact on the desired sensing precision. To overcome this set-back, we developed a novel photochromic probe with an analyte-activation mode for ratiometric sensing of toxic thiols in both real water samples and live cells. The dynamic dual-fluorescence signal is released only after the fast and selective cleavage of the 2,4-dinitrophenyl sulfonate by the targeted thiophenol derivatives. Consequently, a 'double-check' with synchronized dual-fluorescence blinking for analyte detection is successfully employed upon alternate light triggers with rapid response(k=7.2×10^(-2) s^(-1)), high sensitivity(LOD=6.1 nM) as well as selectivity of thiophenol derivatives over other common thiol species(e.g., GSH, Cys and Hcy). The photochromic probe was successfully introduced to the fast and on-site detection of highly toxic thiophenols in real waste water samples. | 18 | Identification and validation of a major gene for kernel length at the P1 locus in Triticum polonicum显示文摘Kernel size, one of the traits that determine wheat yield, is controlled by multiple quantitative trait loci.Polish wheat(Triticum polonicum) has elongated and plump kernel and is a valuable material for breeding high-yielding wheat cultivars. However, genes or loci determining kernel length(KL) in Polish wheat are unknown. We identified and validated a major KL gene, KL-PW, at the P1 locus in Polish wheat. KL-PW is VRT-A2, which encodes a MIKC-type MADS-box protein(MADS55). An insertion/deletion mutation in intron 1 of VRT-A2;led to an alternatively spliced transcript, VRT-A2;. Quantitative PCR analysis showed that VRT-A2;was more highly expressed in developing seeds than was VRT-A2 Ailanmai.Brassinosteroid(BR) sensitivity experiment and the expression of BR-related genes indicated that VRTA2;functions as a positive regulator of BR responses.
