| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 3 | Epidemiological and risk analysis of the H7N9 subtype influenza outbreak in China at its early stage显示文摘Dozens of human cases infected with H7N9 subtype avian influenza virus (AIV) have been confirmed in China since March, 2013. Distribution data of sexes, ages, professions and regions of the cases were analyzed in this report. The results showed that the elderly cases, especially the male elderly, were significantly more than expected, which is different from human cases of H5N1 avian influenza and human cases of the pandemic H1N1 influenza. The outbreak was rated as a Grade Ⅲ (severe) outbreak, and it would evolve into a Grade IV (very severe) outbreak soon, using a method reported previously. The H7N9 AIV will probably circulate in humans, birds and pigs for years. Moreover, with the driving force of natural selection, the virus will probably evolve into highly pathogenic AIV in birds, and into a deadly pandemic influenza virus in humans. Therefore, the H7N9 outbreak has been assumed severe, and it is likely to become very or extremely severe in the future, highlighting the emergent need of forceful scientific measures to eliminate any infected animal flocks. | 4 | Gating system optimization of low pressure casting A356 aluminum alloy intake manifold based on numerical simulation显示文摘To eliminate the shrinkage porosity in low pressure casting of an A356 aluminum alloy intake manifold casting, numerical simulation on fi lling and solidifi cation processes of the casting was carried out using the ProCAST software. The gating system of the casting is optimized according to the simulation results. Results show that when the gating system consists of only one sprue, the fi lling of the molten metal is not stable; and the casting does not follow the sequence solidifi cation, and many shrinkage porosities are observed through the casting. After the gating system is improved by adding one runner and two in-gates, the fi lling time is prolonged from 4.0 s to 4.5 s, the fi lling of molten metal becomes stable, but this casting does not follow the sequence solidifi cation either. Some shrinkage porosity is also observed in the hot spots of the casting. When the gating system was further improved by adding risers and chill to the hot spots of the casting, the shrinkage porosity defects were eliminated completely. | 6 | Enhanced mechanical properties of 6082 aluminum alloy via SiC addition combined with squeeze casting显示文摘In order to improve mechanical properties of 6082 aluminum alloy,the SiC_(p)/Al 6082 composites were prepared by the addition of the micron-sized SiC_(p)articles combined with the squeeze casting.The effects of the SiC_(p)content and squeeze casting on the microstructure and mechanical properties of the 6082 aluminum alloy were investigated by SEM,EDS,TEM,tensile testing and hardness testing analysis methods.Research results exhibited that the SiC_(p)content and squeeze casting had a significant impact on the microstructure and mechanical properties of the 6082 aluminum alloy.The addition of the SiC_(p)refined the grain size of the 6082 aluminum alloy while caused the increase of the porosity with increasing the SiC_(p)content,especially in the permanent mold casting condition.Compared to the permanent mold casting,the squeeze casting obviously reduced pore defects,refined grain size and made the SiC_(p)distribute evenly as well as bond tightly with the Al matrix.The tensile strength,yield strength,elongation,elastic modulus and hardness of the 6082 aluminum alloy obtained with the SiC_(p)and squeeze casting were remarkably improved,and the optimal mechanical properties were obtained with a 2 wt.%SiC_(p),and they increased 10.73%,72.7%,193.9%,23.5%and 25.2%,respectively,compared to those of the6082 aluminum alloy obtained without SiC_(p)and squeeze casting.The fracture surface of the SiC_(p)/Al 6082 composites obtained with the squeeze casting was dense and exhibited a ductile fracture mode. | Wenming Jiang Junwen Zhu Guangyu Li Feng Guan Yang Yu Zitian Fan | 2021 | Journal of Materials Science & Technology2021,,29: | 6 |

A numerical simulation was performed for the water vapor condensable supersonic flows through Laval nozzles under different flow friction conditions. The comparison between numerical and experimental results shows that the model is accurate enough to investigate the supersonic spontaneous condensation flow of water vapor inside Laval nozzles. | 9 | Fine mapping of the rice bacterial blight resistance gene Xa-4 and its co-segregation marker显示文摘An F2 population developed from the Xa-4 near isogenic lines, IR24 and IRBB4, was used for fine mapping of the rice bacterial blight resistance gene, Xa-4. Some restriction fragment length polymorphism (RFLP) markers on the high-density map constructed by Harushima et al. and the amplified DMA fragments homologous to the conserved domains of plant disease resistance (R) genes were used to construct the genetic linkage map around the gene Xa-4 by scoring susceptible individuals in the population. Xa-4 was mapped between the RFLP marker G181 and the polymerase chain reaction (PCR) marker M55. The R gene homologous fragment marker RS13 was found co-segregating with Xa-4 by analyzing all the plants in the population. | 12 | Research on cracking mechanism of the thin shell mould in expendable pattern shell casting during pattern removal process显示文摘Aiming at the cracking phenomenon of the thin shell mould in the expendable pattern shell casting during the pattern removing process, some systemic researches are presented.The influence of the pattern removing method and temperature on the pattern removing were investigated.The shell mould cracking mechanism was analyzed by using thermo-gravimetric analysis (TGA), and combining the temperature field and the volume change of the expanded polystyrene (EPS) foam pattern being tested.The results indicated that the shell mould was not easily cracked when the pattern removing process was carried out with the furnace being heated little by little because of the shell slowly shrinking with dehydration and shell strength gradually increasing.The shell mould was soon destroyed when it was set directly into the furnace at above 400 oC because of the thin shell mould rapidly shrinking and the foam pattern hindering.However, the shell mould had no cracking when it had been preheated for a long time even if the furnace temperature was above 400 oC and the shell was put into the furnace directly.Moreover, when the shell mould was directly set into the furnace at lower temperatures, 250 to 300 ℃, the shell would shrink slowly and the foam pattern would stay at the maximum expansion stage temperature of 100 to 110 ℃ for a long time; and the shell mould would experience an expansion force from the foam pattern for a long time.The expansion force is related to the pattern removing temperature, holding time, foam pattern thickness and density.Therefore, the foam pattern with higher density could make the shell crack. | Jiang Wenming Fan Zitian Liao Defeng Zhao Zhong | 2010 | China Foundry2010,7,3: | 3 |
| 13 | Increased substitution rate in H5N1 avian influenza viruses during mass vaccination of poultry显示文摘As a means of heated debate,mass vaccination of poultry has been used in some countries to control H5N1 highly pathogenic avian influenza(HPAI),which remains of global economic and public health significance.Theoretically,mass vaccination can act as an evolutionary selective force facilitating the emergence of vaccine-resistant viruses,similar to that widespread use of antibiotics facilitates the emergence of antibiotic-resistant bacteria.To support the hypothesis,the substitution rates in the two subunits,HA1 and HA2,of the viral hemagglutinin gene,were calculated using a Bayesian Markov Chain Monte Carlo(MCMC) approach.It was found that the rate in the HA1 subunit,but not in the HA2 subunit,increased significantly during periods of mass vaccination(2005 2010 in China and 2003 2009 in Indonesia),in contrast to the periods when no mass vaccination programs took place(1996 2004 in China and 2004 2008 in Thailand).Because substitutions in the HA1 subunit rather than in the HA2 subunit can lead to vaccine-resistant viruses,the results support that mass vaccination programs facilitate the emergence of vaccine-resistant viruses,which,in turn,will render mass vaccination programs less effective.Therefore,caution must be taken when adopting mass vaccination as a long-term strategy to control HPAI. | WANG ZhaoGuo JIANG WenMing LIU Shuo HOU GuangYu LI JinPing WANG ZhiYu CHEN JiMing | 2012 | Chinese Science Bulletin2012,57,19: | 3 |
