2Response surface modeling-based source contribution analysis and VOC emission control policy assessment in a typical ozone-polluted urban Shunde,China显示文摘To develop a sound ozone(O_3) pollution control strategy,it is important to well understand and characterize the source contribution due to the complex chemical and physical formation processes of O_3.Using the 'Shunde' city as a pilot summer case study,we apply an innovative response surface modeling(RSM) methodology based on the Community Multi-Scale Air Quality(CMAQ) modeling simulations to identify the O_3 regime and provide dynamic analysis of the precursor contributions to effectively assess the O_3 impacts of volatile organic compound(VOC) control strategy.Our results show that Shunde is a typical VOC-limited urban O_3 polluted city.The 'Jiangmen' city,as the main upper wind area during July 2014,its VOCs and nitrogen oxides(NO_x) emissions make up the largest contribution(9.06%).On the contrary,the contribution from local(Shunde) emission is lowest(6.35%) among the seven neighbor regions.The local VOCs industrial source emission has the largest contribution comparing to other precursor emission sectors in Shunde.The results of dynamic source contribution analysis further show that the local NO_x control could slightly increase the ground O_3 under low(10.00%) and medium(40.00%)reduction ratios,while it could start to turn positive to decrease ground O_3 under the high NO_x abatement ratio(75.00%).The real-time assessment of O_3 impacts from VOCs control strategies in Pearl River Delta(PRD) shows that the joint regional VOCs emission control policy will effectively reduce the ground O_3 concentration in Shunde.Zhiqiang You Yun Zhu Carey Jang Shuxiao Wang Jian Gao Che-Jen Lin Minhui Li Zhenghua Zhu Hao Wei Wenwei Yang 2017Journal of Environmental Sciences2017,29,1:7

4GOLM1 exacerbates CD8^(+)T cell suppression in hepatocellular carcinoma by promoting exosomal PD-L1 transport into tumor-associated macrophages显示文摘The immunosuppressive microenvironment plays an important role in tumor progression and immunotherapy responses.Golgi membrane protein 1(GOLM1)is correlated to hepatocellular carcinoma(HCC)progression and metastasis.However,little is known about the role of GOLM1 in regulating the immunosuppressive environment and its impact on immunotherapeutic efficacy in HCC.In this study,GOLM1 was positively correlated with infiltrating tumor-associated macrophages(TAMs)expressed high levels of programmed death-ligand 1(PD-L1)and CD8^(+)T cell suppression in HCC tissues.Both gain-and loss-of-function studies determined a close correlation between GOLM1 and immunosuppression.In the mechanism,GOLM1 promoted COP9 signalosome 5-mediated PD-L1 deubiquitination in HCC cells and increased the transport of PD-L1 into exosomes via suppression of Rab27b expression.Furthermore,co-culture with exosomes derived from HCC cells upregulated the expression of PD-L1 on macrophages.Zoledronic acid in combination with anti-PD-L1 therapy reduced PD-L1^(+)TAMs infiltration and alleviated CD8^(+)T cell suppression,resulting in tumor growth inhibition in the mouse HCC model.Together,our study unveils a mechanism by which GOLM1 induces CD8^(+)T cells suppression through promoting PD-L1 stabilization and transporting PD-L1 into TAMs with exosome dependent.Targeting PD-L1^(+)TAM could be a novel strategy to enhance the efficacy of anti-PD-L1 therapy in HCC.Jinhong Chen Zhifei Lin Lu Liu Rui Zhang Yan Geng Minghao Fan Wenwei Zhu Ming Lu Lu Lu Huliang Jia Jubo Zhang Lun-Xiu Qin 2021Signal Transduction and Targeted Therapy2021,6,12:5
5OsPKpa_1 encodes a plastidic pyruvate kinase that affects starch biosynthesis in the rice endosperm显示文摘Pyruvate kinase(PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice(Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpa_1, which encodes a plastidic isoform of PK(PKp). OsPKpa_1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpa_1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, atthe early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein–protein interaction analysis. 11A trailer car dynamics model considering brake rigging of a high-speed train and its application显示文摘Brake systems are essential for the speed regulation or braking of a high-speed train.The vehicle dynamic performance under braking condition is complex and directly affects the reliability and running safety.To reveal the vehicle dynamic behaviour in braking process,a comprehensive trailer car dynamics model(TCDM)considering brake systems is established in this paper.The dynamic interactions between the brake system and the other connected components are achieved using the brake disc-pad frictions,brake suspension systems,and wheel-rail interactions.The force and motion transmission from the brake system to the wheel-rail interface is performed by the proposed TCDM excited by track irregularity.In addition,the validity of TCDM is verified by experimental test results.On this basis,the dynamic behaviour of the coupled system is simulated and discussed.The findings indicate that the braking force significantly affects vehicle dynamic behaviour including the wheel-rail forces,suspension forces,wheelset torsional vibration,etc.The dynamic interactions within the brake system are also significantly affected by the vehicle vibration due to track irregularity.Besides,the developed TCDM can be further employed to the dynamic assessment of such a coupled mechanical system under different braking conditions.Zhiwei Wang Linchuan Yang Jiliang Mo Song Zhu Wenwei Jin 2023Railway Engineering Science2023,31,3:0
12New insights into developmental biology of Eimeria tenella revealed by comparative analysis of mRNA N6-methyladenosine modification between unsporulated oocysts and sporulated oocysts显示文摘Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many species.However,no information regarding m^(6)A modification in Eimeria tenella is currently available.In the present study,we surveyed the transcriptome-wide prevalence of m^(6)A in sporulated oocysts and unsporulated oocysts of E.tenella.Methylated RNA immunoprecipitation sequencing(MeRIP-seq)analysis showed that m^(6)A modification was most abundant in the coding sequences,followed by stop codon.There were 3,903 hypermethylated and 3,178 hypomethylated mRNAs in sporulated oocysts compared with unsporulated oocysts.Further joint analysis suggested that m^(6)A modification of the majority of genes was positively correlated with mRNA expression.The mRNA relative expression and m^(6)A level of the selected genes were confirmed by quantitative reverse transcription PCR(RT-qPCR)and MeRIP-qPCR.GO and KEGG analysis indicated that differentially m^(6)A methylated genes(DMMGs)with significant differences in mRNA expression were closely related to processes such as regulation of gene expression,epigenetic,microtubule,autophagy-other and TOR signaling.Moreover,a total of 96 DMMGs without significant differences in mRNA expression showed significant differences at protein level.GO and pathway enrichment analysis of the 96 genes showed that RNA methylation may be involved in cell biosynthesis and metabolism of E.tenella.We firstly present a map of RNA m^(6)A modification in E.tenella,which provides significant insights into developmental biology of E.tenella.Qing Liu Bingjin Mu Yijing Meng Linmei Yu Zirui Wang Tao Jia Wenbin Zheng Wenwei Gao Shichen Xie Xingquan Zhu 2024Journal of Integrative Agriculture2024,23,1:0
13Research on Electromagnetic Loss Characteristics of Submarine Cables显示文摘The electromagnetic losses of submarine cables are mainly caused by the metal shielding layer to prevent the water tree effect and the armor layer that strengthens the strength of the submarine cables.While these losses cause the temperature of submarine cable to rise,and temperature variation will in turn change the conductivity of its metal layer material.In this paper,the electric-magnetic-thermal multi-physical field coupling of the electromagnetic loss variation of the submarine cable is realized by establishing a full coupling system containing Fourier’s law and Maxwell-Ampère’s Law for the photoelectric composite submarine cable.The multi-physical field coupling model is solved and analyzed by using the finite elementmethod.Firstly,the loss of each layer of the optoelectronic composite submarine cable is analyzed,and the lossof eachlayer of the submarine cable and themainfactors leading to the loss of the submarine cable are given.Secondly,the influence of environmental temperature,ampacity and armor layer on the electromagnetic loss of submarine cables is studied,and the main operating factors affecting the electromagnetic loss of submarine cables are summarized.The research shows that the influence of ambient temperature can be ignored,and the loss of shielding layer and armor layer increases with the increase of ampacity,but the impact of shielding layer loss is greater.Finally,this paper studies the electromagnetic loss of each metal layer of the submarine cable and the influence of the laying spacing on the electromagnetic loss.The research results show that the two ways of improving the conductivity of the armor layer and reducing the relative permeability of the armor layer can effectively reduce the loss of each metal layer in the cable structure and increase the current carrying capacity when the tensile strength of the armor layer meets the requirements for single-core and threecore photoelectric composite submarine cables laid horizontally.At the same time,increasing the laying spacing will increase the loss,but it can improve the overall current carrying capacity of the cable.The research in this paper provides a theoretical basis for the design of submarine cable carrying capacity,and also provides a reference for the optimization design of submarine cable structures.Liuhuo Wang Qingcui Liu Wenwei Zhu Yanru Wang Yi Tian Long Zhao 2023Energy Engineering2023,120,11:0
14Timely Data Delivery for Energy-Harvesting IoT Devices显示文摘The devices in the Internet of things(Io T)gain capability of sustainable operation when they harvest energy from ambient sources.Fluctuation in the harvested energy may cause the energy-harvesting IoT devices to suffer from frequent energy shortage,which may bring in intolerable packet delay or packet discarding.It is important to design a low-delay packet delivery scheme that adapts to variation in the harvested energy.In this paper,we present the timely data delivery(TDD)scheme for the IoT devices.Using Markov chain,we develop a probability model for the TDD scheme,which leads to the expected number of packets delivered in an operation cycle,the expected numbers of packets waiting in the data buffer in an operation cycle and an energy-harvesting cycle,and the expected packet delay.Additionally,we formulate the optimization problem that minimizes the packet delay in the TDD scheme,and the solution to the optimization problem yields the optimal parameters for the IoT devices to determine when to harvest energy and when to deliver data under the TDD scheme.The simulation results show that the proposed TDD scheme outperforms the existing schemes in terms of packet delay.LU Wenwei GONG Siliang ZHU Yihua 2022Chinese Journal of Electronics2022,31,2:0

16Rapamycin enhances the anti-tumor activity of cabozantinib in cMet inhibitor-resistant hepatocellular carcinoma显示文摘Cabozantinib,mainly targeting cMet and vascular endothelial growth factor receptor 2,is the second-line treatment for patients with advanced hepatocellular carcinoma(HCC).However,the lower response rate and resistance limit its enduring clinical benefit.In this study,we found that cMet-low HCC cells showed primary resistance to cMet inhibitors,and the combination of cabozantinib and mammalian target of rapamycin(mTOR)inhibitor,rapamycin,exhibited a synergistic inhibitory effect on the in vitro cell proliferation and in vivo tumor growth of these cells.Mechanically,the combination of rapamycin with cabozantinib resulted in the remarkable inhibition of AKT,extracellular signal-regulated protein kinases,mTOR,and common downstream signal molecules of receptor tyrosine kinases;decreased cyclin D1 expression;and induced cell cycle arrest.Meanwhile,rapamycin enhanced the inhibitory effects of cabozantinib on the migration and tubule formation of human umbilical vascular endothelial cells and human growth factor-induced invasion of cMet inhibitor-resistant HCC cells under hypoxia condition.These effects were further validated in xenograft models.In conclusion,our findings uncover a potential combination therapy of cabozantinib and rapamycin to combat cabozantinib-resistant HCC.Chao Gao Shenghao Wang Weiqing Shao Yu Zhang Lu Lu Huliang Jia Kejin Zhu Jinhong Chen Qiongzhu Dong Ming Lu Wenwei Zhu Lunxiu Qin 2022Frontiers of Medicine2022,16,3:0
