| 1 | A novel artemisinin derivative, 3-(12-β-artemisininoxy) phenoxyl succinic acid (SM735), mediates immunosuppressive effects in vitro and in vivo显示文摘Aim: To study the immunosuppressive activity of SM735 {[3-(12-β-artemisininoxy)] phenoxyl succinic acid}, a synthetic artemisinin derivative with nonsteroidal antiinflammatory drug structure, with the aim of finding potential immunosuppressive agents. Methods: Concanavalin A (ConA), lipopolysaccharide (LPS), and mixed lymphocyte reaction (MLR), were used to induce the proliferation of splenocytes, and [~3H]-thymidine incorporation was used to evaluate the proliferation of splenocytes. Cytokine production was promoted with ConA, LPS, or PMA plus ionomycin, and was detected with the enzyme-linked immunosorbent assay. Dinitrofluorobenzene (DNFB) and sheep red blood cells (SRBC) were used to induce delayed-type hypersensitivity and quantitative hemolysis of SRBC (QHS) mouse models, as criteria for the evaluation of in vivo immune activity. Results: SM735 strongly inhibited the proliferation of splenocytes induced by ConA, LPS, or MLR, withIC_(50)) values of 0.33 μmol/L, 0.27 μmol/L, and 0.51 μmol/L, respectively. When compared with a CC_(50) value of 53.1 μmol/L, SM735 had a favorable safety range. SM735 dose-dependently inhibited proinflammatory cytokine production [including interleukins (IL)-12, interferon (IFN)-γ and IL-6] induced by LPS or PMA plus ionomycin. Upon ConA stimulation, SM735 suppressed IFN-γ in a dose-dependent manner, but did not affect IL-2 secretion. SM735 also strongly suppressed both T-cell-mediated delayed-type hypersensitivity (DTH) and B-cell-mediated QHS reactions. | 3 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 5 | RESEARCH OF INNER FLOW IN A DOUBLE BLADES PUMP BASED ON OPENFOAM显示文摘The inner flow analysis of centrifugal pumps has gradually become an important issue for the hydraulic design and performance improvement.Nowadays,CFD simulation toolbox of pump inner flow mainly contains commercial tools and open source tools.There are some defects for commercial CFD software for the numerical simulation of 3-D turbulent internal flow in pump,especially in capturing the flow characteristics under the off-design operating conditions.Additionally,it is difficult for researchers to do further investigation because of the undeclared source.Therefore,an open source software like Open Field Operation and Manipulation (OpenFOAM) is increasingly popular with researchers from all over the world.In this paper,a new computational study was implemented based on the original solver and was used to directly simulate the steady-state inner flow in a double blades pump,with the specific speed is 111.In order to disclose the characteristics deeply,three research schemes were conducted.The ratios () of the flow rate are 0.8,1.0 and 1.2,respectively.The simulation results were verified with the Particle Imaging Velocimetry (PIV) experimental results,and the numerical calculation results agree well with the experimental data.Meanwhile,the phenomena of flow separation under the off-design operating conditions are well captured by OpenFOAM.The results indicate that OpenFOAM possesses obvious strong predominance in computing the internal flow field of pump.The analysis results can also be used as the basis for the further research and the improvement of centrifugal pump. | LIU Hou-lin REN Yun WANG Kai WU Deng-hao RU Wei-min TAN Ming-gao | 2012 | Journal of Hydrodynamics2012,24,2: | 11 |
| 6 | Synthesis and Performance of Polyurethane Coated Urea as Slow/controlled Release Fertilizer显示文摘Polyurethane coated urea slow/controlled release fertilizer was prepared based on urea granules, isocyanate, polyols and paraffin. Isocyanate reacted with polyols to synthesize the polyurethane skin layer on urea granules surface. Paraffin serves as a lubricant during syntheses of polyurethane skin layers. The structure and nutrient release characteristics of the polyurethane skin layers were investigated by FTIR, SEM and TG. Urea nitrogen slow-release behavior of the polyurethane coated urea was tested. The experimental results indicated that compact and dense polyurethane skin layers with a thickness of 10-15 μm were formed on urea surface, the urea nitrogen slow-release time can reach 40-50 days. Samples of a light rare earth ore along with mostly ankerite and biotite as dominant gangue minerals as well as bastnaesite, strontianite and goethite as dominated minerals were exposed to further comminution by ball mill and microwave pretreatment fragmentation. Mineralogical characteristics were analyzed by using a mineral liberation analyzer(MLA).The results indicated that tight association mutually penetrates among dominated minerals in the range size of -300 + 212 μm and 212 + 150 μm and gangue minerals in the form of adjacent type. fine vein type. shell type and packing type. Temperature in the ore samples pre-treated by microwave can rapidly rise to 250 ℃ with microwave power of 0-1.5 kW and microwave time of 0-2 min. Applying the microwave pretreatment merely reduces the hardness of the ore causing the fracture of rare earth ore, but this does not transform or change the original mineralogy characteristics of the ore samples. On the basis of above study. the liberation value of bastnaesite, strontianite and goethite with microwave pretreatment is greater than with conventional comminution when the liberation class is above 75%. The distribution of particle size of rare earth ore samples is better with microwave pretreatment than with conventional comminution for particle size of 7.4 × 10^(-5) m. | 9 | Protective effects of a novel drug RC28-E blocking both VEGF and FGF2 on early diabetic rat retina显示文摘AIM: To investigate protective effects of a novel recombinant decoy receptor drug RC28-E on retinal damage in early diabetic rats.METHODS: The streptozotocin(STZ)-induced diabetic rats were randomly divided into 6 groups: diabetes mellitus(DM) group(saline, 3 μL/eye); RC28-E at low(0.33 μg/μL, 3 μL), medium(1 μg/μL, 3 μL), and high(3 μg/μL,3 μL) dose groups; vascular endothelial growth factor(VEGF) Trap group(1 μg/μL, 3 μL); fibroblast growth factor(FGF) Trap group(1 μg/μL, 3 μL). Normal control group was included. At week 1 and 4 following diabetic induction,the rats were intravitreally injected with the corresponding solutions. At week 6 following the induction, apoptosis in retinal vessels was detected by TUNEL staining.Glial fibrillary acidic protein(GFAP) expression was examined by immunofluorescence. Blood-retinal barrier(BRB) breakdown was assessed by Evans blue assay.Ultrastructural changes in choroidal and retinal vessels were analyzed by transmission electron microscopy(TEM).Content of VEGF and FGF proteins in retina was measured by enzyme linked immunosorbent assay(ELISA). The retinal expression of intercellular cell adhesion molecule-1(ICAM-1), tumor necrosis factor-α(TNF-α), VEGF and FGF genes was examined by quantitative polymerase chain reaction(qPCR).RESULTS: TUNEL staining showed that the aberrantly increased apoptotic cells death in diabetic retinal vascular network was significantly reduced by treatments of medium and high dose RC28-E, VEGF Trap, and FGF Trap(all P<0.05), the effects of medium and high dose RC28-E or FGF Trap were greater than VEGF Trap(P<0.01). GFAP staining suggested that reactive gliosis was substantially inhibited in all RC28-E and VEGF Trap groups, but the inhibition in FGF Trap group was not as prominent.Evans blue assay demonstrated that only high dose RC28-E could significantly reduce vascular leakage in early diabetic retina(P<0.01). TEM revealed that the ultrastructures in choroidal and retinal vessels were damaged in early diabetic retina, which was ameliorated to differential extents by each drug. The expression of VEGF and FGF2 proteins was significantly upregulated in early diabetic retina, and normalized by RC28-E at all dosages and by the corresponding Traps. | 12 | DEK43 is a P-type pentatricopeptide repeat(PPR)protein responsible for the Cis-splicing of nad4 in maize mitochondria显示文摘Mitochondria, the main energy transducers in plant cells, require the proper assembly of respiratory chain complexes Ⅰ–Ⅴ for their function. The NADH dehydrogenase4(nad4) gene encodes mitochondrial respiratory chain complex Ⅰ subunit Ⅳ, but the mechanism underlying nad4 transcript splicing is unclear. Here, we report that the P-type pentatricopeptide repeat(PPR) protein DEFECTIVE KERNEL 43(DEK43) is responsible for cis-splicing of the nad4 transcript in maize. We demonstrate that DEK43 localizes to both the nucleus and mitochondria. The mutation of Dek43 resulted in embryo-lethal and light-colored defective kernels. Among the 22 mitochondrial group Ⅱ introns, the splicing efficiency of nad4 introns 1 and 3 was reduced by up to 50% compared to the wild type. The levels of complex Ⅰ and supercomplex Ⅰ+Ⅲ2 were also reduced in dek43. Furthermore, in-gel NADH dehydrogenase assays indicated that the activities of these complexes were significantly reduced in dek43. Further, the mitochondrial ultrastructure was altered in the mutant. | 13 | The Association of Socioeconomic Status with the Burden of Cataract-related Blindness and the Effect of Ultraviolet Radiation Exposure: An Ecological Study显示文摘Objective To assess the association of socioeconomic status with the burden of cataract blindness in terms of year lived with disability(YLD) rates and to determine whether ultraviolet radiation(UVR) levels modify the effect of socioeconomic status on this health burden.Methods National and subnational age-standardized YLD rates associated with cataract-related blindness were derived from the Global Burden of Disease(GBD) study 2017. The human development index(HDI) from the Human Development Report was used as a measure of socioeconomic status.Estimated ground-level UVR exposure was obtained from the Ozone Monitoring Instrument(OMI)dataset of the National Aeronautics and Space Administration(NASA).Results Across 185 countries, socioeconomic status was inversely associated with the burden of cataract blindness. Countries with a very high HDI had an 84% lower age-standardized YLD rate [95%confidence interval(CI): 60%–93%, P < 0.001] than countries with a low HDI;for high-HDI countries, the proportion was 76%(95% CI: 53%–88%, P < 0.001), and for medium-HDI countries, the proportion was48%(95% CI: 15%–68%, P = 0.010;P for trend < 0.001). The interaction analysis showed that UVR exposure played an interactive role in the association between socioeconomic status and cataract blindness burden(P value for interaction = 0.047).Conclusion Long-term high-UVR exposure amplifies the association of poor socioeconomic status with the burden of cataract-related blindness. | 15 | Efficacy of integrated Traditional Chinese Medicine and anti-retroviral therapy on immunological nonresponse in patients with human immunodeficiency virus/acquired immunodeficiency syndrome: a Meta-analysis of randomized controlled trials显示文摘OBJECTIVE:To explore the efficacy of integrating Traditional Chinese Medicine(TCM)and anti-retroviral therapy(ART),a customized combination of different classes of medications which was also called cock-tail treatment,on the immunological nonresponse(INR)in people living with human immunodeficiency virus(HIV)(PLWH).METHODS:Relevant literature in databases such as China National Knowledge Infrastructure Database(CNKI),Wanfang Digital Journal,Chinese Medical Journal Database(CMJD),Chinese Biomedical Literature Database(CBM),Pub Med,Cochrane,and Embase was reviewed by two independent investigators.Data were extracted from the studies according to the eligible criteria and analyzed using Review Manager 5.3.RESULTS:Nine randomized controlled trials(RCTs)with 1078 patients were analyzed.Our analyses showed that CD4 T cell counts in the treatment group improved compared with that in the control group[mean difference(MD)=13.51,95%confidence interval(CI):7.42-19.60,P<0.0001].There was no significant difference between the treated and control groups after 3 months(MD=25.31,95%CI:-2.78 to 53.41,P=0.08).However,after 6 and 12 months,the response of the treatment group was superior to the control group(MD=27.45,95%CI:7.09-47.81,P=0.008 and MD=27.34,95%CI:6.31–48.37,P=0.01,respectively).The clinical efficacy of the treatment group was also higher than that of the control group(RR=1.75,95%CI:1.16-2.65,P=0.007).However,CD45 RO and CD45 RA T cell counts did not differ significantly between the two groups(MD=12.37,95%CI:-6.71 to 31.45,P=0.20 and MD=5.67,95%CI:-3.00 to14.35,P=0.20,respectively).CONCLUSION:The combined treatment strategy of integrated TCM and Western Medicine promotes long-term reconstitution of the immune system and thus,is beneficial and has potential use for improving INR in PLWH.However,large-scale RCTs are required to provide evidence for optimal intervention strategies. | TAO Zhuang HUANG Xiaojie LIU Ying WANG Ru DONG Jipeng LIANG Biyan ZOU Wen GAO Guojian WANG Zhuo ZHANG Ke WU Hao ZHANG Tong WANG Jian | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 6 |

| 20 | Puerarin Up-regulates Methyl-CpG Binding Protein 2 Phosphorylation in Hippocampus of Vascular Dementia Rats显示文摘Objective: To observe the effect of puerarin on methyl-CpG binding protein 2(MeCP2) phosphorylation(pMeCP2) in the hippocampus of a rat model of vascular dementia(VD). Methods: Thirty-six healthy Sprague-Dawley rats were randomly assigned to the sham-operated group, dementia group and puerarintreated group using a random number table(n=12 per group). The modified permanent bilateral common carotid artery occlusion method was used to establish the VD model. The sham-operated and dementia groups were given 2 m L/d of saline, while the puerarin-treated group was given 100 mg/(kg·d) of puerarin for 17 days. The learning and memory abilities were evaluated by the Morris water maze test. Hematoxylin-eosin staining, immunohistochemical(IHC) staining and Western blot analysis were carried out to observe changes in neuron morphology and in level of pMeCP2 in the hippocampus, respectively. Results: The morphologies of rat hippocampal neurons in the puerarintreated group were markedly improved compared with the dementia group. The escape latency of the dementia group was significantly longer than the sham-operated group(P<0.05), while the puerarin-treated group was obviously shorter than the dementia group(P<0.05). Cross-platform times of the dementia group were significantly decreased compared with the sham-operated group(P<0.05), while the puerarin-treated group was obviously increased compared with the dementia group(P<0.05). IHC staining showed no significant difference in the number of MeCP2 positive cells among 3 groups(P>0.05). The number of pMeCP2 positive cells in the CA1 region of hippocampus in the dementia group was significantly increased compared with the sham-operated group, and the puerarin-treated group was significantly increased compared with the dementia group(both P<0.05). Western blot analysis showed no significant difference of MeCP2 expression among 3 groups(P>0.05). The expression of pMeCP2 in the dementia group was significantly increased compared with the sham-operated group, while it in the puerarin-treated group was significantly increased compared with the dementia group(P<0.05).
