1A high-quality chromosome-level genome assembly reveals genetics for important traits in eggplant显示文摘Eggplant(Solanum melongena L.)is an economically important vegetable crop in the Solanaceae family,with extensive diversity among landraces and close relatives.Here,we report a high-quality reference genome for the eggplant inbred line HQ-1315(S.melongena-HQ)using a combination of Illumina,Nanopore and 10X genomics sequencing technologies and Hi-C technology for genome assembly.The assembled genome has a total size of~1.17 Gb and 12 chromosomes,with a contig N50 of 5.26 Mb,consisting of 36,582 protein-coding genes.Repetitive sequences comprise 70.09%(811.14 Mb)of the eggplant genome,most of which are long terminal repeat(LTR)retrotransposons(65.80%),followed by long interspersed nuclear elements(LINEs,1.54%)and DNA transposons(0.85%).The S.melongena-HQ eggplant genome carries a total of 563 accession-specific gene families containing 1009 genes.In total,73 expanded gene families(892 genes)and 34 contraction gene families(114 genes)were functionally annotated.Comparative analysis of different eggplant genomes identified three types of variations,including singlenucleotide polymorphisms(SNPs),insertions/deletions(indels)and structural variants(SVs).Asymmetric SV accumulation was found in potential regulatory regions of protein-coding genes among the different eggplant genomes.Furthermore,we performed QTL-seq for eggplant fruit length using the S.melongena-HQ reference genome and detected a QTL interval of 71.29–78.26 Mb on chromosome E03.The gene Smechr0301963,which belongs to the SUN gene family,is predicted to be a key candidate gene for eggplant fruit length regulation.Moreover,we anchored a total of 210 linkage markers associated with 71 traits to the eggplant chromosomes and finally obtained 26 QTL hotspots.The eggplant HQ-1315 genome assembly can be accessed at http://eggplant-hq.cn.In conclusion,the eggplant genome presented herein provides a global view of genomic divergence at the whole-genome level and powerful tools for the identification of candidate genes for important traits in eggplant.Qingzhen Wei Jinglei Wang Wuhong Wang Tianhua Hu Haijiao Hu Chonglai Bao 2020Horticulture Research2020,7,1:12

9Ginsenoside Rh3 induces pyroptosis and ferroptosis through the Stat3/p53/NRF2 axis in colorectal cancer cells显示文摘Ginsenoside Rh3(GRh3)is a seminatural product obtained by chemical processing after isolation from Chinese herbal medicine that has strong antitumor activity against human tumors.However,its antitumor role remains to be elucidated.The aim of this study is to explore the mechanisms underlying the tumor suppressive activity of GRh3 from the perspective of pyroptosis and ferroptosis.GRh3 eliminates colorectal cancer(CRC)cells by activating gasdermin D(GSDMD)-dependent pyroptosis and suppressing solute carrier family 7 member 11(SLC7A11),resulting in ferroptosis activation through the Stat3/p53/NRF2 axis.GRh3 suppresses nuclear factor erythroid 2-related factor 2(NRF2)entry into the nucleus,leading to the decrease of heme oxygenase 1(HO-1)expression,which in turn promotes NOD-like receptor thermal protein domain associated protein 3(NLRP3)and caspase-1 expression.Finally,caspase-1 activates GSDMD-dependent pyroptosis.Furthermore,GRh3 prevents NRF2 from entering the nucleus,which suppresses SLC7A11,causing the depletion of glutathione(GSH)and accumulation of iron,lipid reactive oxygen species(ROS)and malondialdehyde(MDA),and eventually leading to ferroptosis in CRC cells.In addition,GRh3 effectively inhibits the proliferation of CRC cells in vitro and in nude mouse models.Collectively,GRh3 triggers pyroptotic cell death and ferroptotic cell death in CRC cells via the Stat3/p53/NRF2 axis with minimal harm to normal cells,showing great anticancer potential.Yingchao Wu Dajin Pi Shuyao Zhou Zhongjia Yi Yangyang Dong Wuhong Wang Huan Ye Yiliu Chen Qian Zuo Mingzi Ouyang 2023Acta Biochimica et Biophysica Sinica2023,55,4:1

11Person-independent expression recognition based on person-similarity weighted expression feature显示文摘A new method to extract person-independent expression feature based on higher-order singular value decomposition (HOSVD) is proposed for facial expression recognition. Based on the assumption that similar persons have similar facial expression appearance and shape, the person-similarity weighted expression feature is proposed to estimate the expression feature of test persons. As a result, the estimated expression feature can reduce the influence of individuals caused by insufficient training data, and hence become less person-dependent. The proposed method is tested on Cohn-Kanade facial expression database and Japanese female facial expression (JAFFE) database. 14Thickness-dependent and strain-tunable magnetism in twodimensional van der Waals VSe_(2)显示文摘Two-dimensional(2D)van der Waals(vdW)magnetic materials with reduced dimensionality often exhibit unexpected properties compared to their bulk counterparts.In particular,the mechanical flexibility of 2D structure,enhanced ferromagnetism at reduced layer thickness,as well as robust perpendicular magnetic anisotropy are quite appealing for constructing novel spintronic devices.The vdW vanadium diselenide(VSe_(2))is an attractive material whose bulk is paramagnetic while monolayer is ferromagnetic with a Curie temperature(Tc)above room temperature.To explore its possible device applications,a detailed investigation on the thickness-dependent magnetism and strain modulation behavior of VSe_(2)is highly demanded.In this article,the VSe_(2)nanoflakes were controllably prepared via chemical vapor deposition(CVD)method.The few-layer single VSe_(2)nanoflakes were found to exhibit magnetic domain structures at room temperature.Ambient magnetic force microscopy(MFM)phase images reveal a clear thickness-dependent magnetism and the MFM phase contrast is traceable for the nanoflakes of layer thickness below~6 nm.Moreover,applying strain is found efficient in modulating the magnetic moment and coercive field of 2D VSe_(2)at room temperature.These results are helpful for understanding the ferromagnetism of high temperature 2D magnets and for constructing novel straintronic devices or flexible spintronic devices.Wenjuan Ci Huali Yang Wuhong Xue Ruilong Yang Baohua Lv Peng Wang Run-Wei Li Xiao-Hong Xu 2022Nano Research2022,15,8:0

16Comparing gene expression profiles of Kashin-Beck and Keshan diseases occurring within the same endemic areas of China显示文摘In this study, differentially expressed genes in peripheral blood from patients with Kashin-Beck disease and Keshan disease were compared to further investigate the etiology and pathogenesis of both diseases, which occur in a common endemic area of China. Twenty Kashin-Beck disease patients and 12 healthy controls, and 16 Keshan disease patients and 16 healthy controls, were grouped into four pairs. Patients and controls were selected from common endemic areas for the two diseases. Total RNA was isolated from peripheral blood mononuclear cells from all patients and controls, and gene expression profiles analyzed by oligonucleotide microarrays. Sixteen genes differentially expressed in both Kashin-Beck disease and Keshan disease (versus controls) were identified, and comprised nine genes showing synchronous and seven asynchronous expression. The Comparative Toxicogenomics Database shows that expression and biological function of these genes can be affected by multiple environmental factors, including mycotoxin and selenium content, potential environmental risk factors for the two diseases. 18STUDIES ON BLOOD GLUTATHIONE PEROXIDASE PROTEIN LEVEL OF CHILDREN IN KESHAN DISEASE AND KASHIN-BECK DISEASE AREAS显示文摘Objective To oberve the change in blood glutathione peroxidase (GSH-Px) protein levels of residents in the low-selenium (Se) area by contrasting the blood GSH-Px protein level of the children in the Keshan disease area with those in the Kashin-Beck disease and non-endemic areas. Methods GSH-Px protein levels were measured by enzyme-linked immunosorbent assays (ELISA). The Se content and GSH-Px activity were assayed by the 2,3-diaminonaphthalene spectrofluorimetric method and glutathione reductase-coupled method respectively. Results ①The blood Se content and GSH-Px protein level of children in Keshan disease area (Moding) were significantly lower than those in Xi’an non-endemic area, however, there was no significant difference when compared with the low-Se non-endemic area; ②The blood Se content, GSH-Px activity and GSH-Px protein level of children in the Kashin-Beck disease area (Yulin) were significantly lower than those of children in two non-endemic areas and in the Keshan disease area; ③The blood Se content and GSH-Px activity were positively correlated to the GSH-Px protein level respectively. Conclusion These results indicate that the blood GSH-Px protein level is decreased in the low-Se residents.
